Comparison and development of scanning electron microscope techniques for delicate plant tissues.

Cell deformation often occurs during sample preparation and imaging with scanning electron microscope (SEM), especially with delicate samples, which influences the accuracy of the results. Here we investigate the influence of several preparation methods on cell deformation, using water content and tissue hardness as indicators to classify "delicate" samples of plant species. The degree of deformation in samples resulting from five preparation methods was measured at the tissue and single-cell levels, revealing that a cryo- and methanol-fixation produced lower degrees of tissue dimension deform

Preclinical development of plant-based oral immune modulatory therapy for haemophilia B.

Anti-drug antibody (ADA) formation is a major complication in treatment of the X-linked bleeding disorder haemophilia B (deficiency in coagulation factor IX, FIX). Current clinical immune tolerance protocols are often not effective due to complications such as anaphylactic reactions against FIX. Plant-based oral tolerance induction may address this problem, as illustrated by the recent first regulatory approval of orally delivered plant cells to treat peanut allergy. Our previous studies showed that oral delivery of plant cells expressing FIX fused to the transmucosal carrier CTB (cholera toxi

How glyphosate affects plant disease development: it is more than enhanced susceptibility.

Glyphosate has been shown to affect the development of plant disease in several ways. Plants utilize phenolic and other shikimic acid pathway-derived compounds as part of their defense against pathogens, and glyphosate inhibits the biosynthesis of these compounds via its mode of action. Several studies have shown a correlation between enhanced disease and suppression of phenolic compound production after glyphosate. Glyphosate-resistant crop plants have also been studied for changes in resistance as a result of carrying the glyphosate resistance trait. The evidence indicates that neither the r

Stability and pre-formulation development of a plant-produced anthrax vaccine candidate.

Second generation anthrax vaccines focus on the use of recombinant protective antigen (rPA) to elicit a strong, toxin neutralizing antibody responses in immunized subjects. The main difference between the rPA vaccines compared to the current licensed vaccine, anthrax vaccine absorbed (AVA), is the rPA vaccines are highly purified preparations of only rPA. These second generation rPA vaccines strive to elicit strong immune responses with substantially fewer doses than AVA while provoking less side effects. Many of the rPA candidates have shown to be effective in pre-clinical studies, but most o

DOF Transcription Factors: Regulatory Hubs Coordinating Plant Growth, Development and Stress Tolerance.

Under global climate warming, plants evolve unique growth and developmental strategies to cope with frequent abiotic stresses. The plant-specific DNA-BINDING WITH ONE FINGER (DOF) transcription factors (TFs) originated in green algae and are phylogenetically divided into four subfamilies (A, B, C, D), governing plant growth development and stress adaptation. Subfamily B mainly regulates vegetative growth, subfamily C is predominantly involved in seed development and vegetative growth, and subfamily D1 (CYCLING DOF FACTOR, CDF) participates extensively in vegetative growth, flowering, and stres

Role of cell wall-associated kinases in plant development and defense responses.

Plant life is coordinated by several developmental and defense associated mechanisms and cell wall is actively important in these mechanisms. The plant cell wall also serves as an important component system that monitors plant growth and defense apart from only protecting the cell. However, resident sensors of the plant cell membrane and pattern recognition receptors (PRRs) also constitute major response mechanisms in plants while they are both as well developed for maintaining cell wall integrity (CWI). These PRRs are molecules such as ligands, peptides or glycans that are released for sensin

Transcription factor PbNAC71 regulates xylem and vessel development to control plant height.

Dwarfism is an important agronomic trait in fruit breeding programs. However, the germplasm resources required to generate dwarf pear (Pyrus spp.) varieties are limited. Moreover, the mechanisms underlying dwarfism remain unclear. In this study, "Yunnan" quince (Cydonia oblonga Mill.) had a dwarfing effect on "Zaosu" pear. Additionally, the dwarfism-related NAC transcription factor gene PbNAC71 was isolated from pear trees comprising "Zaosu" (scion) grafted onto "Yunnan" quince (rootstock). Transgenic Nicotiana benthamiana and pear OHF-333 (Pyrus communis) plants overexpressing PbNAC71 exhibit

Roles of AGD2a in Plant Development and Microbial Interactions of Lotus japonicus.

Arabidopsis AGD2 (Aberrant Growth and Death2) and its close homolog ALD1 (AGD2-like defense response protein 1) have divergent roles in plant defense. We previously reported that modulation of salicylic acid (SA) contents by ALD1 affects numbers of nodules produced by Lotus japonicus, but AGD2's role in leguminous plants remains unclear. A combination of enzymatic analysis and biological characterization of genetic materials was used to study the function of AGD2 (LjAGD2a and LjAGD2b) in L. japonicus. Both LjAGD2a and LjAGD2b could complement dapD and dapE mutants of Escherichia coli and had a

Proteolytic Processing of SERK3/BAK1 Regulates Plant Immunity, Development, and Cell Death.

Plants have evolved many receptor-like kinases (RLKs) to sense extrinsic and intrinsic cues. The signaling pathways mediated by multiple Leucine-rich repeat (LRR) RLK (LRR-RLK) receptors require ligand-induced receptor-coreceptor heterodimerization and transphosphorylation with BRI1-ASSOCIATED RECEPTOR KINASE1 (BAK1)/SOMATIC EMBRYOGENESIS RECEPTOR KINASES family LRR-RLKs. Here we reveal an additional layer of regulation of BAK1 via a Ca2+-dependent proteolytic cleavage process that is conserved in Arabidopsis (Arabidopsis thaliana), Nicotiana benthamiana, and Saccharomyces cerevisiae The prote

Development of real-time radioisotope imaging systems for plant nutrient uptake studies.

Ionic nutrition is essential for plant development. Many techniques have been developed to image and (or) measure ionic movement in plants. Nevertheless, most of them are destructive and limit the analysis. Here, we present the development of radioisotope imaging techniques that overcome such restrictions and allow for real-time imaging of ionic movement. The first system, called macroimaging, was developed to visualize and measure ion uptake and translocation between organs at a whole-plant scale. Such a device is fully compatible with illumination of the sample. We also modified fluorescent

Development of automated headspace gas chromatography determination of dithiocarbamates in plant matrixes.

The determination of dithiocarbamates in plant matrixes is generally carried out by spectrophotometric (European Norm EN 12396-1, 1996) or gas chromatography headspace (European Norm EN 12396-2, 1999) methods. However, the former method presents a risk of carbon disulfide loss during hydrolysis and distillation and its sensitivity is low, whereas the latter method is time-consuming. In comparison to these European methods and in compliance with norm V03-110, we have developed an automated gas chromatography headspace method. This method offers a good level of accuracy and precision and is spec

Revisiting plant-microbe interaction mechanisms for developing efficient biostimulants.

The diverse and often complex challenges arising from climate change, severe soil degradation and agricultural intensification due to increased use of synthetic agrochemicals, have exacerbated the need for sustainable agricultural systems that can sustain crop productivity with less reliance on agrochemicals. Plant microbe interactions really help in this transition as they influence plant growth, nutrient access, adaptation to stress, and defense against pathogens. Plants interact with many belowground microbes in the rhizosphere through an incredibly diverse set of root exudates that act as

This study provides a comprehensive overview of the current knowledge regarding phototoxic terrestrial plants and their phototoxic and photosensitizing metabolites. Within the 435,000 land plant species, only around 250 vascular plants have been documented as phototoxic or implicated in phototoxic occurrences in humans and animals. This work compiles a comprehensive catalog of these phototoxic plant species, organized alphabetically based on their taxonomic family. The dataset encompasses meticulous details including taxonomy, geographical distribution, vernacular names, and information on the

Diversity of copper-containing nanoparticles and their influence on plant growth and development.

Copper (Cu) is an important microelement for plants, but in high concentrations it can be toxic. Cu-containing nanoparticles (Cu NPs) are less toxic, their use for plants is safer, more effective and economical than the use of Cu salts. This review presents detailed information on the chemical diversity of Cu NPs and various methods of their synthesis. The mechanisms of the effect of Cu NPs on plants are described in detail, and examples of research in this area are given. The main effects of Cu NPs on plants are reviewed including on their growth and development (organogenesis, mitosis, accum

Comparing and Contrasting the Multiple Roles of Butenolide Plant Growth Regulators: Strigolactones and Karrikins in Plant Development and Adaptation to Abiotic Stresses.

Strigolactones (SLs) and karrikins (KARs) are both butenolide molecules that play essential roles in plant growth and development. SLs are phytohormones, with SLs having known functions within the plant they are produced in, while KARs are found in smoke emitted from burning plant matter and affect seeds and seedlings in areas of wildfire. It has been suggested that SL and KAR signaling may share similar mechanisms. The α/β hydrolases DWARF14 (D14) and KARRIKIN INSENSITIVE 2 (KAI2), which act as receptors of SL and KAR, respectively, both interact with the F-box protein MORE AXILLARY GROWTH 2

Constitutive elevation of cAMP in Arabidopsis thaliana enhances plant growth, development, and productivity.

Cyclic adenosine 3',5'-monophosphate (cyclic AMP; cAMP) has emerged as a bona fide signalling molecule in plants. Although its regulatory roles in diverse cellular processes are increasingly recognised, the transient nature of cAMP signalling has limited our understanding of its systemic functions at the whole-plant level. In this study, we engineered Arabidopsis thaliana plants to constitutively express adenylate cyclase (AC) activity, thereby enabling sustained elevation of endogenous cAMP levels. The AC transgenic lines exhibited a 1.3- to 1.8-fold increase in endogenous cAMP content relati

Fuelling the top - Metabolic control of shoot apical meristem function and plant development.

Plant development relies on a complex network of transcription factors and hormone signalling pathways that guide pattern formation and developmental transitions in the meristems. Yet, how this core network is regulated to enable developmental plasticity remains poorly understood. A key question is how information about nutrient availability influences growth decisions, with nutrients acting not only as substrates for energy production and anabolism but also as regulatory signals. Here, we review recent advances in plant nutrient signalling during development, focussing on how carbon and nitro

A single UV-C pulse modulates Gibberellin homeostasis and Plant Development in Arabidopsis.

Plants are rarely exposed to ultraviolet C (UV-C) radiation because it is absorbed by the ozone layer. Nevertheless, low doses of UV-C can induce physiological and developmental responses, although the underlying hormonal mechanisms are not well understood. We show that a single one-minute UV-C pulse (20 W/m2) is associated with temporally distinct differences in gibberellin (GA) homeostasis in Arabidopsis. At early sampling times, levels of GA12, a precursor of bioactive GAs, are lower in UV-C-treated plants compared with controls. This is accompanied by lower transcript levels of KS and KAO2
