BACKGROUND: We evaluated the toxicity of 5% (w/v) povidone-iodine (PI) applied to the ocular surface of rabbits.
METHODS: Twenty-three white rabbits were divided into four groups; these were a control group and three study groups in which the ocular surface was exposed to PI for different times. In control group, one drop of phosphate-buffered saline (PBS) was applied once for 10 min. In study groups, one drop of 5% (w/v) PI was topically applied once for 1 min, 3 min, and 10 min, and then the animals were observed for 7 days. We compared gentamicin 0.3% one drop every three hours the night before and the morning of surgery ("half-day regimen") with the same drug one drop qid for three days ("three-day regimen") in 15 eyes undergoing intraocular surgery. Povidone-Iodine (Betadine) Paint was used in thirteen patients with a proven fungal infection. Ten patients had pityriasis versicolor, two had trichophyton rubrum and one M. canis. In the pityriasis versicolor group, seven out of the ten patients were either improved or cleared up completely within 7 days. There were no adverse reactions in this group of patients and the material proved quite acceptable to them. Scanning electron micrographs were carried out on three of the patients and demonstrated the decrease in fungal elements after treatment and clinical improvement.

The antimicrobial effect of 20 minutes exposure to 10% povidone-iodine solution and to 5% neomycin-erythromycin solution was evaluated in vitro in 6 suspensions of dog feces.
