An intracellular pH gradient in the anammox bacterium Kuenenia stuttgartiensis as evaluated by 31P NMR.

The cytoplasm of anaerobic ammonium oxidizing (anammox) bacteria consists of three compartments separated by membranes. It has been suggested that a proton motive force may be generated over the membrane of the innermost compartment, the "anammoxosome". 31P nuclear magnetic resonance (NMR) spectroscopy was employed to investigate intracellular pH differences in the anammox bacterium Kuenenia stuttgartiensis. With in vivo NMR, spectra were recorded of active, highly concentrated suspensions of K. stuttgartiensis in a wide-bore NMR tube. At different external pH values, two stable and distinct p

Characterization of the Saccharomyces cerevisiae cytosine transporter using energizable plasma membrane vesicles.

The purine-cytosine permease is a carrier localized in the plasma membrane of the yeast Saccharomyces cerevisiae. The energetics of cytosine transport catalyzed by this permease has been studied in an artificial system obtained by fusion between proteoliposomes containing beef heart cytochrome c oxidase and plasma membrane-enriched fractions of a S. cerevisiae strain overexpressing the permease. Upon addition of an energy donor, a proton-motive force (inside alkaline and negative) is created in this system and promotes cytosine accumulation. By using different phospholipids, it is shown that c

Metabolic potentiation of antibiotic killing by L-arginine in drug-resistant Edwardsiella tarda.

The role of metabolic state reprogramming in modulating antibiotic susceptibility has attracted growing interest as a promising strategy to combat antimicrobial resistance. Our study revealed that L-arginine potentiates chloramphenicol's bactericidal activity by at least two orders of magnitude against multidrug-resistant Edwardsiella tarda via the coordinated modulation of three interconnected metabolic pathways: the tricarboxylic acid cycle disruption, redox homeostasis alteration, and phenylalanine metabolic suppression. Mechanistically, L-arginine-mediated tricarboxylic acid cycle inhibiti

Stimulating Central Carbon Metabolism to Re-sensitize Pseudomonas aeruginosa to Aminoglycosides.

In this issue of Cell Chemical Biology, Meylan et al. (2017) examine how stimulation of central carbon metabolism of Pseudomonas aeruginosa modulates aminoglycoside lethality in tolerant bacteria. They identify fumarate as a tobramycin potentiator that stimulates proton motive force-dependent drug uptake and increases respiration-dependent killing.

Energy dependence of lipopolysaccharide translocation in Salmonella typhimurium.

Energy inhibitors block translocation of pulse-labeled core lipopolysaccharide to outer membrane under conditions which allow maintenance of constant specific radioactivity of intracellular precursor pools throughout the chase period. Under the conditions used, approximately 75% of the total cellular label was membrane-bound at initiation of chase. Translocation of core lipopolysaccharide from inner to outer membrane showed apparent first order kinetics (t1/2 = 1.2 min, 32 degrees C). Translocation was blocked by arsenate (5-10 mM) under conditions where proton motive force was unchanged, whil

Genome-wide screen reveals glycerol-induced aminoglycoside potentiation against Staphylococcus aureus via boosting GlpK-initiated energy metabolism.

Potentiation of existing antibiotics represents a promising strategy for combating the global health crisis of antibiotic resistance. Here we report that 1 min co-treatment with glycerol markedly enhances aminoglycoside efficacy against stationary-phase Staphylococcus aureus cells. This rapid combined treatment also effectively eliminates S. aureus biofilms and persister cells, methicillin-resistant S. aureus clinical isolates in vitro, and S. aureus in a mouse skin infection model. The potentiation is achieved through rapid enhancement of proton motive force-dependent aminoglycoside uptake. A

Membrane-Active Rhamnolipids Overcome Aminoglycoside Resistance.

In this issue of Cell Chemical Biology, Radlinski et al. (2019) identify Pseudomonas-derived rhamnolipids that potentiate aminoglycoside antibiotics in the eradication of antibiotic-tolerant bacterial phenotypes. Microbial physiological and mechanistic studies indicate that rhamnolipids permeabilize S. aureus membrane and drive the uptake of aminoglycosides that is independent of proton-motive force.

Stimulating Aminoglycoside Uptake to Kill Staphylococcus aureus Persisters.

Aminoglycosides are bactericidal drugs which require a proton motive force (PMF) for uptake into the bacterial cell. Low energy cells, such as persisters, maintain a PMF below the threshold for drug uptake and are tolerant to aminoglycosides. In this chapter, we discuss mechanisms to target the bacterial membrane and stimulate aminoglycoside uptake to kill Staphylococcus aureus persisters.

Regulatory plasticity balances photosynthetic electron flow with enhanced pH-dependent cytochrome b6f control in Arabidopsis.

To avoid photodamage of photosystem I under fluctuating light, plants have evolved multiple photoprotective mechanisms. One key mechanism is photosynthetic control, in which acidification of the thylakoid lumen downregulates electron transport through the cytochrome b6f complex, thereby preventing overreduction of photosystem I. The Arabidopsis proton gradient regulation 5 (pgr5) mutant, which is defective in cyclic electron transport around photosystem I, fails to induce photosynthetic control and consequently suffers severe photosystem I photodamage under fluctuating light. Previously, we sh

Activity of Antibacterial/Antifungal Compounds against the Protozoan Parasite, Toxoplasma gondii.

We investigated the antiparasitic activity of several antimicrobial drug leads against Toxoplasma gondii tachyzoites and, in one case, bradyzoites. Carbazole and phenylthiazole aminoguanidine anti-infectives, originally developed as antibacterial and antifungal agents, showed potent activity, with IC50 values as low as 2 μM. This potency was comparable to that observed with the tuberculosis drug candidate SQ109 and a series of its analogs. Notably, SQ109 also significantly reduced the viability of in vivo-derived bradyzoites. All compounds acted, at least in part, as protonophore uncouplers by

Mechanism of Synergy between Piceatannol and Ciprofloxacin against Staphylococcus aureus.

Piceatannol (PIC) is a natural stilbene extracted from grape skins that exhibits biological activities such as antibacterial, antitumor, and antioxidant activities. The present study was carried out to further investigate the effect of PIC on the antibacterial activity of different antibiotics and to reveal the antibacterial mechanism of PIC. We found that PIC had an inhibitory effect against Staphylococcus aureus (S. aureus); its minimum inhibitory concentration (MIC) and minimum bactericidal concentration (MBC) were 128 μg/mL and 256 μg/ mL, respectively. Additionally, we measured the fracti

Overexpression of Stenotrophomonas maltophilia major facilitator superfamily protein MfsA increases resistance to fluoroquinolone antibiotics.

BACKGROUND: Stenotrophomonas maltophilia is an opportunistic human pathogen causing nosocomial infections worldwide. S. maltophilia infection is of particular concern due to its inherent resistance to currently used antibiotics. Proton motive force-driven transporters of the major facilitator superfamily frequently contribute to the efflux of substances, including antibiotics, across cell membranes. METHODS: An mfsA expression plasmid (pMfsA) was constructed and transferred into bacterial strains by electroporation. The antibiotic susceptibility levels of S. maltophilia strains were determine

Peptide Probes Reveal a Hydrophobic Steric Ratchet in the Anthrax Toxin Protective Antigen Translocase.

Anthrax toxin is a tripartite virulence factor produced by Bacillus anthracis during infection. Under acidic endosomal pH conditions, the toxin's protective antigen (PA) component forms a transmembrane channel in host cells. The PA channel then translocates its two enzyme components, lethal factor and edema factor, into the host cytosol under the proton motive force. Protein translocation under a proton motive force is catalyzed by a series of nonspecific polypeptide binding sites, called clamps. A 10-residue guest/host peptide model system, KKKKKXXSXX, was used to functionally probe polypepti

Basic residues R260 and K357 affect the conformational dynamics of the major facilitator superfamily multidrug transporter LmrP.

Secondary-active multidrug transporters can confer resistance on cells to pharmaceuticals by mediating their extrusion away from intracellular targets via substrate/H(+)(Na(+)) antiport. While the interactions of catalytic carboxylates in these transporters with coupling ions and substrates (drugs) have been studied in some detail, the functional importance of basic residues has received much less attention. The only two basic residues R260 and K357 in transmembrane helices in the Major Facilitator Superfamily transporter LmrP from Lactococcus lactis are present on the outer surface of the pro

Tolfenpyrad Modulates Antibiotic Activity on Francisella novicida and Synergizes with Electron Transport Chain Inhibitors.

Francisella tularensis is a highly virulent intracellular pathogen for which treatment options remain limited and vulnerable to resistance development. Tolfenpyrad, a pesticide with selective antibacterial activity against Francisella species, has emerged as a promising compound, yet its mechanism of action remains poorly defined. Here, we investigated how tolfenpyrad modulates the activity of established antibiotics and probed its effects on bacterial energy metabolism in Francisella. In this study, we used combinatorial drug experiments to identify both synergistic and antagonistic drug-drug

[Antibacterial activity and mechanism of the antiparasitic drug resorantel against Staphylococcus epidermidis].

OBJECTIVES: Staphylococcus epidermidis (S. epidermidis) is a common opportunistic pathogen in clinical settings. The widespread misuse of antibiotics has led to a continuous increase in its antimicrobial resistance, greatly complicating treatment. Drug repurposing, characterized by low cost, high safety, and short development cycles, represents an effective strategy for combating drug-resistant bacterial infections. This study aims to investigate the antibacterial activity of the antiparasitic drug resorantel against S. epidermidis and to preliminarily elucidate its antibacterial mechanism. M

Fluoroquinolone resistance of Serratia marcescens: sucrose, salicylate, temperature, and pH induction of phenotypic resistance.

Serratia marcescens is a nosocomial agent with a natural resistance to a broad spectrum of antibiotics, making the treatment of its infections very challenging. This study examines the influence of salicylate, sucrose, temperature, and pH variability on membrane permeability and susceptibility of S. marcescens to norfloxacin (hydrophilic fluoroquinolone) and nalidixic acid (hydrophobic quinolone). Resistance of wild-type S. marcescens UOC-67 (ATCC 13880) to norfloxacin and nalidixic acid was assessed by minimal inhibitory concentration (MIC) assays after growth in the presence of various conce

Substrate dependence of transport coupling and phenotype of a small multidrug resistance transporter in Pseudomonas aeruginosa.

UNLABELLED: Small multidrug resistance (SMR) transporters are key players in the defense of multidrug-resistant pathogens to toxins and other homeostasis-perturbing compounds. However, recent evidence demonstrates that EmrE, an SMR from Escherichia coli and a model for understanding transport, can also induce susceptibility to some compounds by drug-gated proton leak. This runs down the ∆pH component of the proton-motive force (PMF), reducing the viability of the affected bacteria. Proton leak may provide an unexplored drug target distinct from the targets of most known antibiotics. Activating

Adenosine Awakens Metabolism to Enhance Growth-Independent Killing of Tolerant and Persister Bacteria across Multiple Classes of Antibiotics.

Metabolic and growth arrest are primary drivers of antibiotic tolerance and persistence in clinically diverse bacterial pathogens. We recently showed that adenosine (ADO) suppresses bacterial growth under nutrient-limiting conditions. In the current study, we show that despite the growth-suppressive effect of ADO, extracellular ADO enhances antibiotic killing in both Gram-negative and Gram-positive bacteria by up to 5 orders of magnitude. The ADO-potentiated antibiotic activity is dependent on purine salvage and is paralleled with a suppression of guanosine tetraphosphate synthesis and the mas

Functional evidence of Hv1-mediated proton flux in cardiac mitochondria.

Mitochondria are essential for cardiac myocyte function, providing the continuous ATP supply required for contraction and cellular homeostasis. Regulation of the proton motive force, which is critical for ATP synthesis, depends on multiple ion transport mechanisms, including those mediating H+ flux. The voltage-gated proton channel (Hv1), encoded by the Hvcn1 gene, is known to regulate cytosolic pH and membrane potential in several cell types. Here, using isolated mouse cardiac mitochondria, we demonstrate for the first time that Hv1 is functionally expressed in cardiac mitochondria. Pharmacol
