Expression of sphingosine 1-phosphate 1-3 on penile cavernous tissue in hypertensive and normotensive rats.

OBJECTIVE: To investigate the expression of sphingosine 1-phosphate 1-3 (S1P1-3) on penile cavernous tissues in hypertensive and normotensive rats and its association with erectile function. METHODS: Ten 14-week-old male spontaneously hypertensive rats (SHRs) and 10 male age-matched normotensive Wistar-Kyoto rats were randomly designated as hypertensive rat group and normotensive control group, respectively. Mean blood pressure, serum testosterone, and maximum intracavernosal pressure/mean arterial pressure (ICPmax-to-MAP ratio) were measured. The expression of S1P1-3, endothelial nitric oxid

Islands of cartilage and bone at the margin of the carotid bodies of rats.

Islands of mature cartilage or bone were observed in 6 respectively 7 out of 1395 carotid bodies of rats studied. They were found only in spontaneously hypertensive rats of the Okamoto-Aoki stain and situated regularly at the margin of the carotid bodies. The bone islands were bigger than those of cartilage. The presence of such tissue components in the carotid bodies of rats is discussed in connection with the embryology of this peripheral arterial chemoreceptor, especially its mesodermal origin.

Short-term effect of ligature-induced periodontitis on cardiovascular variability and inflammatory response in spontaneously hypertensive rats.

BACKGROUND: We previously reported that periodontal disease (PD) induces high arterial pressure variability (APV) consistent with sympathetic overactivity and elicits myocardial inflammation in Balb/c mice. However, it is unknown whether PD can change APV and heart rate variability (HRV) in spontaneously hypertensive (SHR) and normotensive Wistar-Kyoto (WKY) rats. This study aimed to evaluate the hemodynamic level, HRV, and APV associating with myocardial inflammation and plasma concentrations of oxide nitric (NO) in SHR and WKY rats with PD. METHODS: Three weeks after bilateral ligation of t

Genetic Models in Applied Physiology. HXB/BXH rat recombinant inbred strain platform: a newly enhanced tool for cardiovascular, behavioral, and developmental genetics and genomics.

This review deals with the largest set of rat recombinant inbred (RI) strains and summarizes past and recent accomplishments with this platform for genetic mapping and analyses of divergent and complex traits. This strain, derived by crossing the spontaneously hypertensive rat, SHR/Ola, with a Brown Norway congenic, BN-Lx, carrying polydactyly-luxate syndrome, is referred to as HXB/BXH. The RI strain set has been used for linkage and association studies to identify quantitative trait loci for numerous cardiovascular phenotypes, including arterial pressure, stress-elicited heart rate, and press

The paraganglia within the carotid bifurcation regions of young and old spontaneously hypertensive rats (SHR) after exposure to chronic hypobaric hypoxia. II. The carotid body-like organs.

The following paraganglia in the carotid bifurcations regions of spontaneously hypertensive rats (SHR) were studied: Endoneural paraganglia within the external carotid nerve, the carotid sinus nerve, the glossopharyngeal nerve, and the pharyngeal branch of the vagus nerve, the so-called periadventitial type I cells, and so-called miniglomera. Number and distribution of these paraganglia vary among different individuals. After chronically hypobaric hypoxia the volume of these paraganglia was increased but their number remained unchanged. The increase of volume was dependent on the duration of h

Meteorin-like (METRNL) attenuates hypertensive induced cardiac hypertrophy by inhibiting autophagy via activating BRCA2.

Hypertension, a prevalent cardiovascular ailment globally, can precipitate numerous complications, notably hypertensive cardiomyopathy. Meteorin-like (METRNL) is demonstrated to possess potential protective properties on cardiovascular diseases. However, its specific role and underlying mechanism in hypertensive myocardial hypertrophy remain elusive. Spontaneously hypertensive rats (SHRs) served as hypertensive models to explore the effects of METRNL on hypertension and its induced myocardial hypertrophy. The research results indicate that, in contrast to Wistar-Kyoto (WKY) rats, SHRs exhibit

Unilateral chemical ablation of the adrenal gland lowers blood pressure and alleviates target organ damage in spontaneously hypertensive rats.

UNLABELLED: Adrenal gland hormones play a critical role in the development and maintenance of hypertension. Adrenal ablation has been used to treat primary aldosteronism but not essential hypertension. The present study aimed to investigate the effectiveness and safety of unilateral adrenal gland ablation in treating spontaneously hypertensive rats (SHR). SHR and Wistar-Kyoto rats (WKY) were subjected to unilateral adrenal ablation with injection of anhydrous ethanol or a sham procedure. Blood pressure was monitored by both tail-cuff plethysmography and telemetry until 6 months after the proce

An Evaluation of Cardiac Health in the Spontaneously Hypertensive Rat Colony: Implications of Evolutionary Driven Increases in Concentric Hypertrophy.

BACKGROUND: The Spontaneously Hypertensive Rat (SHR) Colony was established in 1963 and is the most commonly used rodent model for studying heart failure (HF). Ideally, animal models should recapitulate the clinical disease as closely as possible. Any drift in a genetic model may create a new model that no longer adequately represents the human pathology. Further, instability overtime may lead to conflicting data between laboratories and/or irreproducible results. While systolic blood pressure (SBP) is closely monitored during inbreeding, the sequelae of HF (e.g., cardiac hypertrophy) are not.

Amplified cardiorespiratory activity by hypoxia in conscious spontaneously hypertensive rats.

Activation of peripheral respiratory chemoreceptors provokes respiratory and cardiovascular reflexes, providing a novel understanding of pathogenic mechanism of hypertension. Here we hypothesize that activation of peripheral respiratory chemoreceptors by hypoxia causes enhanced cardiorespiratory activity in conscious spontaneously hypertensive rats (SHRs). Using whole body plethysmography in combination with radio telemetry, pulmonary ventilation, arterial blood pressure and heart rate were examined in SHRs and Wistar-Kyoto (WKY) rats. We found that exposure to hypoxia induced greater increase

Induction of apoptosis by hypertension via endoplasmic reticulum stress.

BACKGROUND/AIMS: Endoplasmic reticulum (ER) stress is one of the intrinsic apoptosis pathways, and cardiac apoptosis can occur in cardiovascular diseases, such as hypertension. However, the mechanisms by which ER stress leads to apoptosis remain enigmatic, particularly in the progression from cardiac hypertrophy to diastolic heart failure due to hypertension. METHODS: We used spontaneously hypertensive rats (SHRs) to investigate possible signalling pathways for ER stress. RESULTS: We found that cardiac protein and mRNA levels of glucoseregulated protein 78 were up-regulated. In addition, the

The increased concentration of 2,3-diphosphoglycerate in red blood cells of spontaneously hypertensive rats.

It has been recognised that high haemoglobin oxygen capacity is essential for the development of high blood pressure in spontaneously hypertensive rats. In the present study we have found increased concentration of 2,3 diphosphoglycerate (2,3-DPG) in red blood cells of spontaneously hypertensive rats (SHR) of Okamoto-Aoki strain. As 2,3-DPG is the major factor decreasing haemoglobin affinity to oxygen, our finding suggests that at given value of pO2 oxygen delivery to the tissue of SHR would be increased. Therefore increased concentration of 2,3-DPG in red blood cells of SHR would be of the pa

Dehydration natriuresis in Dahl S rats: no evidence for renal excretory deficit.

There is conflicting evidence for the existence of a renal sodium excretory deficit in the salt-sensitive hypertensive Dahl S (DS) rat strain. While presentation of acute sodium loads, in vivo or in vitro, suggests that DS kidneys cannot excrete sodium as efficiently as kidneys from the salt insensitive genetic control Dahl R (DR) rat strain, metabolic studies of Dahl rats on a high-sodium diet are unable to differentiate between DS and DR rats. The natriuretic response to acute sodium loads is dependent on the integrity of structures in or near the anteroventral 3rd ventricle (AV3V) region. T

Neurochemical alterations of intrinsic cardiac ganglionated nerve plexus caused by arterial hypertension developed during ageing in spontaneously hypertensive and Wistar Kyoto rats.

The acknowledged hypothesis of the cause of arterial hypertension is the emerging disbalance in sympathetic and parasympathetic regulations of the cardiovascular system. This disbalance manifests in a disorder of sustainability of endogenous autonomic and sensory neural substances including calcitonin gene-related peptide (CGRP). This study aimed to examine neurochemical alterations of intrinsic cardiac ganglionated nerve plexus (GP) triggered by arterial hypertension during ageing in spontaneously hypertensive rats of juvenile (prehypertensive, 8-9 weeks), adult (early hypertensive, 12-18 wee

The cytokine alterations/abnormalities and oxidative damage in the pancreas during hypertension development.

The aim of the present study was to compare the content of cytokines, chemokines, and oxidative stress markers in the pancreas of spontaneously hypertensive rats (SHRs) and Wistar Kyoto Rats (WKYs) serving as controls. Enzyme-like immunosorbent assay (ELISA) and biochemical methods were used to measure pancreatic levels of interleukin-1ß, interleukin-6, tumor necrosis factor α, transforming growth factor β, RANES, monocyte chemoattractant protein 1, interferon gamma-induced protein 10, malondialdehyde, and sulfhydryl groups. The results showed that the pancreatic concentrations of all studied

Increased plasma level of endothelin-1 in the Okamoto spontaneously hypertensive rat.

The objectives of this study were to determine plasma levels of endothelin (ET) in a genetic model of hypertension and in control rats during control conditions and in response to short-term volume expansion with saline. Okamoto spontaneously hypertensive rats (SHR) and control Wistar-Kyoto (WKY) rats were used in this study. One group of each strain served as control animals, and another group of each strain underwent volume expansion with saline (5% of body weight infused during a period of 30 minutes). The levels of ET-1 and ET-3 were measured in plasma by using a double-antibody radioimmun

Paradoxical effect of naloxone on the hemorrhagic hypotension in normotensive and spontaneously hypertensive Wistar-Kyoto rats.

The influence of naloxone on recovery from hemorrhagic hypotension was investigated in conscious normotensive and spontaneously hypertensive (SHR) Wistar-Kyoto rats. In both groups arterial blood pressure (MBP) was reduced to 50% of the resting level by arterial bleeding. Administration of naloxone (0.02 mg/kg iv) significantly impaired restitution of MBP in WKY and SHR in comparison to control experiments on WKY and SHR receiving no naloxone. The magnitude of this effect was similar in both strains, however in SHR, it appeared with a significant delay. The results indicate that small doses of

Electromyography data on spontaneously hypertensive rats.

1. To investigate the state of nerve-muscle excitability in 20 spontaneously hypertensive rats and 22 Wistar rats local electromyography was carried out in periods of rest and under the maximum strain of 10 min ischaemia of m.flexor femoris (Trusso's test). 2. The electromyography phenomenon found in the SHR group corresponds to typical tetany electromyography correlatives. 3. Thus, changes occur to the nerve-muscle excitability state according to the neurogenic tetany type.

Adhesive nonfibrotic bioelectronic interfaces on diverse peripheral nerves for long-term functional neuromodulation.

Bioelectronic devices implanted on peripheral nerves offer potential for the treatment and rehabilitation of clinical diseases. However, the foreign body reaction and the subsequent fibrous capsule formation at the device-peripheral nerve interface severely limit their efficacy and longevity in vivo. Here, we describe a robust bioadhesive strategy that can establish nonfibrotic bioelectronic interfaces on diverse peripheral nerves-occipital, vagus, deep peroneal, sciatic, tibial, and common peroneal nerves-for up to 12 weeks. Our approach inhibits the infiltration of immune cells into the inte
