Prolonged myalgia in Sindbis virus infection: case description and in vitro infection of myotubes and myoblasts.

BACKGROUND: Sindbis virus (SINV) is a mosquito-borne alphavirus found in Eurasia, Africa, and Oceania. Clinical SINV infection is characterized by febrile rash and arthritis and sometimes prolonged arthralgia and myalgia. The pathophysiological mechanisms of musculoskeletal and rheumatic disease caused by SINV are inadequately understood. METHODS: We studied the muscle pathology of SINV infection ex vivo by examining a unique muscle biopsy obtained from a patient with chronic myalgia and arthralgia 6 months after acute SINV infection and assessed potential genetic predisposing factors by dete

Vector Competence of Northern European Culex pipiens Biotype pipiens and Culex torrentium to West Nile Virus and Sindbis Virus.

The West Nile Virus (WNV) and Sindbis virus (SINV) are avian-hosted mosquito-borne zoonotic viruses that co-circulate in some geographical areas and share vector species such as Culex pipiens and Culex torrentium. These are widespread in Europe, including northern parts and Finland, where SINV is endemic, but WNV is currently not. As WNV is spreading northwards in Europe, we wanted to assess the experimental vector competence of Finnish Culex pipiens and Culex torrentium mosquitoes to WNV and SINV in different temperature profiles. Both mosquito species were found susceptible to both viruses a

Binding of hnRNP I-vRNA Regulates Sindbis Virus Structural Protein Expression to Promote Particle Infectivity.

Alphaviruses cause significant outbreaks of febrile illness and debilitating multi-joint arthritis for prolonged periods after initial infection. We have previously reported that several host hnRNP proteins bind to the Sindbis virus (SINV) RNAs, and disrupting the sites of these RNA-protein interactions results in decreased viral titers in tissue culture models of infection. Intriguingly, the primary molecular defect associated with the disruption of the hnRNP interactions is enhanced viral structural protein expression; however, the precise underlying mechanisms spurring the enhanced gene exp

Identification of Interactions between Sindbis Virus Capsid Protein and Cytoplasmic vRNA as Novel Virulence Determinants.

Alphaviruses are arthropod-borne viruses that represent a significant threat to public health at a global level. While the formation of alphaviral nucleocapsid cores, consisting of cargo nucleic acid and the viral capsid protein, is an essential molecular process of infection, the precise interactions between the two partners are ill-defined. A CLIP-seq approach was used to screen for candidate sites of interaction between the viral Capsid protein and genomic RNA of Sindbis virus (SINV), a model alphavirus. The data presented in this report indicates that the SINV capsid protein binds to speci

Diagnostics of Pogosta disease: antigenic properties and evaluation of Sindbis virus IgM and IgG enzyme immunoassays.

Sindbis virus (SINV) is a mosquito-borne causative agent of a fever-rash arthritis, Pogosta disease, as verified recently by virus isolation from acutely ill patients. Pogosta disease occurs annually, but it emerges as unique epidemics every 7 years in Finland; over 10,000 patient samples have been analyzed for SINV antibodies, with over 2000 diagnosed acute SINV infections. However, the performance of these serological tests with a large number of samples has not been described before. The aim of the present study was to characterize and evaluate methods developed for the serodiagnostics of S

Epidemiology of Sindbis virus infections in Finland 1981-96: possible factors explaining a peculiar disease pattern.

Pogosta disease (PD), an epidemic rash-arthritis occurring in late summer is caused by Sindbis virus (SINV) and is transmitted to humans by mosquitoes. Altogether 2183 PD cases were serologically confirmed 1981-96 in Finland, with an annual incidence of 2.7/100000 (18 in the most endemic area of Northern Karelia). The annual average was 136 (varying from 1 to 1282) with epidemics occurring in August-September with a 7-year interval. Studies on 6320 patients with suspected rubella (1973-89) revealed 107 PD cases. The depth of snow cover and the temperature in May-July seemed to predict the numb

A High-Throughput Antiviral Assay Based on a Sindbis Virus-GFP for the Discovery of Inhibitors of Alphavirus Replication.

The re-emergence of alphaviruses (family Togaviridae), such as chikungunya virus, poses significant public health risks, with direct impact on quality of life and work productivity. There are no approved antiviral drugs for the treatment of infections with alphaviruses. Several alphaviruses are classified as risk group 3 agents which require handling in high-containment laboratories. To facilitate antiviral screening efforts against alphaviruses, we established a high-throughput antiviral screening assay using reporter Sindbis virus [SINV-GFP; expresses the green fluorescent protein] as a surr

Molecular evidence of neuroinvasive Sindbis virus infection in humans: detection in cerebrospinal fluid by next generation sequencing.

Sindbis virus (SINV) is a mosquito-borne alphavirus causing seasonal outbreaks in northern Europe, Africa, and Russia. Neurological involvement in humans is poorly documented, and detection in cerebrospinal fluid (CSF) has not previously been reported. This study provides the first unequivocal evidence of human CNS involvement by detecting SINV RNA directly in the cerebrospinal fluid of four autochthonous patients presenting with acute neurological symptoms in south eastern Tuscany, Italy, July-August 2025. Utilizing metagenomic Next Generation Sequencing (mNGS), we reconstructed complete vira

Infection of Aedes aegypti Mosquitoes with Midgut-Attenuated Sindbis Virus Reduces, but Does Not Eliminate, Disseminated Infection.

Arboviruses are transmitted by specific vectors, and the reasons for this specificity are not fully understood. One contributing factor is the existence of tissue barriers within the vector such as the midgut escape barrier. We used microRNA (miRNA) targeting of Sindbis virus (SINV) to study how replication in midgut cells contributes to overcoming this barrier in the mosquito Aedes aegypti. SINV constructs were designed to be attenuated specifically in midgut cells by inserting binding sites for midgut-specific miRNAs into either the 3' untranslated region (MRE3'miRT) or the structural open r

A metagenomics survey of viral diversity in mosquito vectors allows the first detection of Sindbis virus in Burkina Faso.

Arboviruses (i.e., Arthropod-borne viruses) pose a threat to human health worldwide. This taxonomically-diverse group includes numerous viruses that recurrently spread into new regions. Therefore, periodic surveys of the arboviral diversity in a given region can help optimize the diagnosis of arboviral infections. However, such surveys are infrequent, especially in low-income countries. Consequently, case investigation is often limited to a fraction of the arboviral diversity. This situation is likely to result in undiagnosed cases. Here, we investigated the diversity of mosquito-borne arbovir

Characterization of the local and systemic B cell response of normal and athymic nude mice with Sindbis virus encephalitis.

During Sindbis virus (SV) encephalitis in mice B cells are an important component of the mononuclear inflammatory response and recovery depends primarily on the development of antiviral antibody. To begin to characterize various parameters of the local B cell response during SV encephalitis we have defined B cell isotype expression in brain sections, splenocytes and peripheral blood mononuclear cells (PBMC) in normal and athymic nude mice using an immunoperoxidase technique. Early (days 3-5) in SV encephalitis brain perivascular B cells are IgM or IgM/IgD-bearing lymphocytes, later (days 10-14

Double-subgenomic Sindbis virus recombinants expressing immunogenic proteins of Japanese encephalitis virus induce significant protection in mice against lethal JEV infection.

A series of double-subgenomic Sindbis virus (dsSIN) recombinants that express cassettes encoding the immunogenic proteins of Japanese encephalitis virus (JEV) [prM-E, prM-E-NS1, NS1-NS2A, 80%E (encodes the amino-terminal 80% part of E), and NS1] were constructed and analyzed for their ability to confer protective immunity in mice against lethal challenge with neurovirulent JEV. The cassettes were introduced into both 5' [second subgenomic promoter of the vector precedes the SIN structural open reading frame (SP-ORF)] and 3' (the promoter follows the SP-ORF) dsSIN vectors. The longest cassette

Sindbis virus self-amplifying replicon is compatible with modified nucleotides mediating expression and vaccine responses in vivo.

The advent of effective formulations based on RNA encapsulated within lipid nanoparticles has led to the evaluation of this technology for its ability to treat and prevent a wide range of diseases. Expansion of the RNA-based tool kit available to vaccine and drug developers will accelerate this development and hopefully produce more robust and cost-effective therapies. With its more sustained expression profile, self-amplifying RNA offers many advantages to standard messenger RNA, allowing for the administration of lower RNA doses in vivo and potentially reducing dosing frequency for indicatio

Development of a multi-epitope vaccine candidate against Sindbis virus through integrated immunoinformatics approaches and molecular dynamics simulations.

Sindbis virus (SINV), belonging to the genus Alphavirus, is the causative agent of Pogosta disease in humans. The clinical infection is characterized by fever, malaise, rash, myalgia, and arthralgia, which is generally self-limiting. Chronic infection with SINV triggers autoimmune conditions that lead to persistent arthritis. Despite its clinical relevance, no licensed vaccine is currently available for the prevention of SINV infection. To the best of our knowledge, this study presents the first in silico design and evaluation of a multi-epitope vaccine candidate against SINV. Using an integra

A sindbis virus replicon-based DNA vaccine encoding the rabies virus glycoprotein elicits immune responses and complete protection in mice from lethal challenge.

A sindbis virus replicon-based DNA vaccine encoding rabies virus glycoprotein (G) was developed by subcloning rabies G gene into a sindbis virus replicon-based vaccine vector (pAlpha). The self-amplification of RNA transcripts and translation efficiency of rabies G was analyzed in pAlpha-Rab-G-transfected mammalian cells using RT-PCR, SDS-PAGE and Western blot analysis. The transfected cells also showed induction of apoptosis which is an important event in the enhancement of immune responses. Further, immune responses induced with replicon-based rabies DNA vaccine (pAlpha-Rab-G) was compared w

Enhanced surveillance to assess the presence of Sindbis and Batai virus in mosquito populations at an urban zoo in the United Kingdom.

BACKGROUND: Sindbis virus (SINV) and Batai virus (BATV) are emerging zoonotic arboviruses with a growing number of detections in Europe. Recent SINV outbreaks in northern Europe and high BATV seroprevalence in sheep, goats, and cattle in Germany emphasise the threat they pose to both animal and human health. Despite their presence in countries of similar latitude and climate, neither of these viruses have been detected in the UK. METHODS: Zoos are strategic sentinel sites for disease surveillance because they are well monitored and possess a high diversity of animal species. Located in southe

Pathogenesis of temperature-sensitive mutants of Sindbis virus in the embryonated egg. III. Autologous, homologous, and heterologous interference.

Two temperature-sensitive (ts) mutants of Sindbis virus able to synthetize RNA, but not two ts mutants defective in viral RNA synthesis, reduced to lethality of the superinfecting heat-resistant parent strain of Sindbis virus in embryonated chicken eggs as compared with results in controls. Autologous interference (of mutant with parent) was observed under a variety of conditions. Time-course studies in one selected system showed that lower titers of superinfecting parent virus were detected in doubly infected eggs than in controls eggs infected with only parent virus. Expression of genes encoding vesicular stomatitis and Sindbis virus glycoproteins in yeast leads to formation of disulfide-linked oligomers.

Saccharomyces cerevisiae strains transformed with plasmids containing cDNAs coding for the glycoproteins of vesicular stomatitis or Sindbis viruses can be induced to produce large amounts of glycosylated virus glycoproteins. Studies reported here show that these proteins from high molecular weight disulfide-linked oligomers in the yeast endoplasmic reticulum. Oligomers were also found for two genetically altered forms of VSV G; one of these was lacking the membrane anchor domain and the other had the cysteine in the cytoplasmic tail replaced with serine.
