Telomere-dependent chromosomal instability.

Telomeres are specialized DNA-protein structures at the ends of the linear chromosomes. In mammalian cells, they are composed of multifold hexameric TTAGGG repeats and a number of associated proteins. The double-stranded telomeric DNA ends in a 3' single stranded overhang of 150 to 300 base pair (bp) which is believed to be required for a higher order structure (reviewed in (Blackburn, 2001)). One important model is that the telomeres form loop structures, the T-loops, and by invasion of the 3' overhang into the duplex region of the double stranded part protect the DNA against degradation and

Fission yeast RPA-TERT-Tpz1TPP1 complex promotes telomere extension and suppresses telomere recombination.

Telomerase maintains chromosome ends by extending telomeric DNA, yet how recruited telomerase becomes productively engaged remains poorly understood. Recent studies found that Replication Protein A (RPA) contributes to telomerase stimulation through interaction with TERT in humans and with the TPP1 ortholog Est3 in budding yeast, suggesting a direct role in telomerase activation. Here, we provide genetic and structural modeling evidence for a RPA-Trt1TERT-Tpz1TPP1 ternary complex that promotes telomere extension while suppressing recombination in fission yeast. Guided by results from genetic s

Telomere-to-telomere assembly detects genomic diversity in Canadian strains of Borrelia burgdorferi.

Borrelia burgdorferi, the bacteria causing Lyme disease, has a complex genome comprising a linear chromosome and multiple linear and circular plasmids. The atypical hairpin telomeres and the highly paralogous plasmids complicate genome assembly. We develop a genome assembly pipeline using both long and short reads to overcome these challenges. Using long reads, we assemble the hairpin telomeres of the linear replicons, an lp28-1a plasmid subtype, and circular plasmids of nine B. burgdorferi strains from five regions across Northwest Ontario and Manitoba, Canada. Although similar across the cor

Effect of exercise on telomere length and telomere proteins expression in mdx mice.

In Duchenne muscular dystrophy (DMD), telomere shortening has been postulated to contribute to the failure of regenerative activity promoting the premature senescence of satellite cells. The aim of the present study was to investigate the telomere length and the expression of telomeric repeat-binding factor-1 (TRF1), poly (ADP-ribose) polymerase-1 (PARP1) and mouse telomerase reverse transcriptase (MTERT) in gastrocnemius, tibialis anterior and diaphragm muscles of the murine model of DMD, the mdx mouse and whether a chronic protocol of forced exercise impacts on them. Our results confirmed a

A Complete Telomere-To-Telomere Assembly of Plectropomus leopardus and Phylogenomic Insights Into Perciformes.

The leopard coral grouper, Plectropomus leopardus Lacepède, 1802, is an ecologically and economically important species in the family Epinephelidae. However, the absence of a complete reference genome has impeded advanced genetic research on this species, and the phylogenetic position of Epinephelidae within Perciformes remains unresolved. Here, we generated a complete telomere-to-telomere (T2T) genome assembly (887.20 Mb; contig N50: 39.03 Mb; QV: 67) for P. leopardus with no need for gap-filling. Leveraging this genome combined with other available genome resources, we conducted a large-scal

Giardia telomeres and telomerase.

Giardia duodenalis, the protozoan responsible for giardiasis, is a significant contributor to millions of diarrheal diseases worldwide. Despite the availability of treatments for this parasitic infection, therapeutic failures are alarmingly frequent. Thus, there is a clear need to identify new therapeutic targets. Giardia telomeres were previously identified, but our understanding of these structures and the critical role played by Giardia telomerase in maintaining genomic stability and its influence on cellular processes remains limited. In this regard, it is known that all Giardia chromosome

Assessing telomeric phenotypes.

The concept of telomeres as being the end-part of eukaryotic chromosomes was first described by H. J. Muller and B. McClintock. Their pioneering work opened the path for multiple new researches and assays on a thrilling subject, with implications for various domains such as aging, replication, immortality, and cancer. Yeast has been a model of choice to study telomere length, senescence, telomerase activity, telomere cloning, and sequencing with important new techniques being discovered in this species and adapted afterward for other organisms. The main functions of telomeres include the prote

Telomeres in the haemopoietic system.

The limited life span of most blood cells requires the continuous production of cells, which in adults exceeds 10(12) cells/day. This impressive production of cells (approximately 4 x 10(16) cells over a lifetime) is achieved by the proliferation and differentiation of committed progenitor cells, which themselves are derived from a population of pluripotent stem cells with self-renewal potential. Paradoxically, the large majority of stem cells in adult bone marrow are quiescent cells. One possibility is that stem cells, like other somatic cells, have only a limited replicative potential (< 100

Telomere-to-telomere genome of Stylosanthes guianensis uncovers symbiotic adaptation to phosphorus-deficient soils.

BACKGROUND: Stylosanthes guianensis, a representative tropical legume, exhibits remarkable adaptation to low-phosphorus acidic soils. As a symbiotic species, it forms root nodule associations with rhizobia to fix atmospheric nitrogen, potentially enhancing phosphate use efficiency. This study aims to decipher the mechanisms linking root nodule symbiosis to low-phosphate adaptation in Stylosanthes guianensis. RESULTS: We present the first gap-free, telomere-to-telomere genome of Stylosanthes guianensis (1.20 Gb), containing 82.28% repetitive sequences and 34,728 genes, with 99.30% BUSCO comple

Two telomere-to-telomere Nelumbo genome assemblies reveal domestication history and empower precision breeding.

Lotus (Nelumbo) is an ancient aquatic plant of major ecological, economic, and cultural importance, yet its genomic architecture and domestication history remain incompletely resolved. Here, we generate telomere-to-telomere reference genomes for the two extant species, Asian lotus (N. nucifera) and American lotus (N. lutea). Comparative genomics analysis reveals divergence in centromeric regions and chromosomal structural variation between the two species. Population analysis of 832 globally distributed lotus accessions supports tropical Asia as the primary dispersal cradle of Asian lotus. We

Two telomere-to-telomere gapless genomes reveal insights into Capsicum evolution and capsaicinoid biosynthesis.

Chili pepper (Capsicum) is known for its unique fruit pungency due to the presence of capsaicinoids. The evolutionary history of capsaicinoid biosynthesis and the mechanism of their tissue specificity remain obscure due to the lack of high-quality Capsicum genomes. Here, we report two telomere-to-telomere (T2T) gap-free genomes of C. annuum and its wild nonpungent relative C. rhomboideum to investigate the evolution of fruit pungency in chili peppers. We precisely delineate Capsicum centromeres, which lack high-copy tandem repeats but are extensively invaded by CRM retrotransposons. Through ph

Telomere Disorders and the Liver.

Telomeres are repetitive sequences at the ends of chromosomes that contribute to genome stability and prevent replicative senescence. Telomere biology disorders (TBDs) encompass a spectrum of genetic conditions characterized by accelerated telomere shortening, which affects different organs, including the liver. TBD-related hepatic manifestations are often unrecognized and can range from asymptomatic liver test abnormalities to steatohepatitis, hepatopulmonary syndrome, or cirrhotic and noncirrhotic portal hypertension requiring liver transplantation. This review summarizes the current literat

Telomeres and Human Disease.

Telomeres, the long nucleotide repeats, and protein complex at chromosome ends, are central to genomic integrity. Telomere length (TL) varies widely between populations due to germline genetics, environmental exposures, and other factors. Very short telomeres caused by pathogenic germline variants in telomere maintenance genes cause the telomere biology disorders, a spectrum of life-threatening conditions including bone marrow failure, liver and lung disease, cancer, and other complications. Cancer predisposition with long telomeres is caused by rare pathogenic germline variants in components

Telomeres, aging and reproduction.

PURPOSE OF REVIEW: Women's fertility decay starts at the mid 30 s. However, the current delay of childbearing leads to ovarian aging and the need of assisted reproduction technologies (ART). Telomere biology is one of the main pathways involved in organismal aging. Thus, this review will focus on the knowledge acquired during the last 2 years about the telomere pathway and its influence on female fertility and the consequences for the newborn. RECENT FINDINGS: New research on telomere biology reaffirms the relationship of telomere attrition and female infertility. Shorter maternal telomeres,

Telomere--the twilight to immortality.

Besides forming a very important component of the chromosome, the telomeres have extremely significant modes of action and functions, right from maintaining a basic infrastructure and integrity of the chromosome vis a vis the other chromosomes, telomeres are responsible for the cell divisions and replicative senescence of the cell. The number of mitotic divisions which a cell will go through in its life span while passing through the cell cycle is governed inturn by these telomeres, the crux of the entire functioning of these chromosomal components suggests that they are the ticking clocks of

Telomeres and marrow failure.

Telomeres, repeat sequences at the ends of chromosomes, are protective chromosomal structures highly conserved from primitive organisms to humans. Telomeres inevitably shorten with every cell cycle, and telomere attrition has been hypothesized to be fundamental to normal senescence of cells, tissues, and organisms. Molecular mechanisms have evolved to maintain their length and protective function; telomerase (TERT) is a reverse transcriptase enzyme that uses an RNA molecule (TERC) as the template to elongate the 3' ends of telomeres. Shelterin is a collection of DNA-binding proteins that cover

Telomeres and chromosome instability.

Genomic instability has been proposed to play an important role in cancer by accelerating the accumulation of genetic changes responsible for cancer cell evolution. One mechanism for chromosome instability is through the loss of telomeres, which are DNA-protein complexes that protect the ends of chromosomes and prevent chromosome fusion. Telomere loss can occur as a result of exogenous DNA damage, or spontaneously in cancer cells that commonly have a high rate of telomere loss. Mouse embryonic stem cells and human tumor cell lines that contain a selectable marker gene located immediately adjac

A telomere-to-telomere gapless genome assembly of the Tibetan wild ass (Equus kiang).

As a First-Class Protected species in China, the Tibetan wild ass (Equus kiang) is endemic to the Qinghai-Tibet Plateau and exhibits remarkable adaptation to extreme high-altitude environments characterized by low oxygen, cold temperatures, and high UV radiation. To explore its genetic basis and evolutionary adaptations, we generated the first telomere-to-telomere (T2T) gapless genome using an integrated sequencing strategy that combined PacBio HiFi, ultra-long Oxford Nanopore (ONT), Hi-C, and BGI short-read technologies.
