Stress-related white matter microstructure alterations and chronic pain.

Posttraumatic stress symptoms (PTSS) are commonly experienced in people with chronic pain. Reduced white matter microstructural integrity in the uncinate fasciculus and the cingulum has separately been reported in chronic pain and posttraumatic stress disorder (PTSD) studies. However, the relationship between chronic pain, PTSS and white matter integrity remains unclear. This study aims to disentangle the relationship between PTSS severity and white matter microstructural integrity common across different chronic pain conditions. Thirty-six subjects with chronic pain and 20 without chronic pai

White matter disconnection in acquired criminality.

Structural brain imaging is increasingly introduced as evidence in criminal trials. A key imaging abnormality identified in criminal populations is alteration to the right uncinate fasciculus but it remains unclear whether these changes play a causal role in criminal behavior. Lesion studies of acquired criminality offer the opportunity to assess the causal role of focal disruption of specific white matter connections in criminal behavior. We studied lesion locations of focal brain damage associated with new onset criminal behavior compared to lesions associated with 21 diverse neuropsychiatri

Ventral temporal-frontal pathways in the human brain based on diffusion MRI tractography.

In the primate brain, two ventral fiber pathways connect the temporal cortex with the frontal cortex: the Uncinate Fasciculus (UF) and the Temporal-Frontal extreme capsule Fasciculus (TFexcF). The definition and reconstruction of these pathways in the human brain have been inconsistent between research groups and have led to conflicting findings about their functional roles. Here, we define the UF and TFexcF in the human brain based on a review of the temporal-frontal connections demonstrated in the macaque monkey via invasive tracer studies and a novel protocol for reconstructing them in the

Applications of magnetic resonance imaging in the diagnosis of posttraumatic stress disorder.

The application of magnetic resonance imaging (MRI) techniques in diagnosing posttraumatic stress disorder (PTSD) represents a significant research focus, providing deeper insights into trauma-related psychological disorders. PTSD often arises from chronic stress, a state in which the body struggles to adapt to overwhelming stimuli. Patients frequently experience persistent flashbacks (intrusive memories or sensory recollections of the traumatic event) and nightmares, which evoke intense emotional responses and disrupt daily life. These recurring episodes of stress and anxiety severely impact

White matter integrity and cognition in mild traumatic brain injury following motor vehicle accident.

The aim of this study is to explore the white matter structure integrity in patients with mild traumatic brain injury (mTBI) using diffusion tensor imaging (DTI), and to analyze the relationship between the white matter structure integrity and cognitive impairment of patients with mTBI. Twenty-five patients with mTBI and 25 healthy control subjects were studied with conventional MR imaging and diffusion tensor imaging. Fractional anisotropy (FA) and mean diffusivity (MD) maps of patients with mTBI were calculated and compared, with these control maps using tract-based spatial statistics (TBSS)

Frontotemporal networks and behavioral symptoms in primary progressive aphasia.

OBJECTIVE: To determine if behavioral symptoms in patients with primary progressive aphasia (PPA) were associated with degeneration of a ventral frontotemporal network. METHODS: We used diffusion tensor imaging tractography to quantify abnormalities of the uncinate fasciculus that connects the anterior temporal lobe and the ventrolateral frontal cortex. Two additional ventral tracts were studied: the inferior fronto-occipital fasciculus and the inferior longitudinal fasciculus. We also measured cortical thickness of anterior temporal and orbitofrontal regions interconnected by these tracts. T

White matter correlates of psychopathic traits in a female community sample.

Psychopathy comprises interpersonal, affective, lifestyle and antisocial facets that vary dimensionally in the population and are associated with criminal offending and adverse psychosocial outcomes. Evidence associating these facets with white matter microstructure of the uncinate fasciculus and the cingulum tracts is inconsistent and derives principally from studies of male offenders. In a sample of 99 young women presenting a range of scores on the Psychopathy Checklist: Screening Version, we used Diffusion Tensor Imaging, tractography and Tract-Based Spatial Statistics to investigate micro

Multimodal MRI characterization of brain injury in heatstroke: integrating gray matter morphometry and advanced diffusion tractometry.

OBJECTIVE: To investigate gray-white matter injury in heatstroke using multimodal MRI, and precisely localize damaged white matter segments via automated fiber quantification (AFQ) with clinical correlation. RESULTS: Compared with the healthy control group, VBM revealed reduced volume in bilateral cerebellar anterior lobes and left fusiform gyrus. TBSS showed widespread white matter abnormalities. AFQ identified: (a) Decreased fractional anisotropy in left corticospinal tract and right inferior fronto-occipital fasciculus; (b) increased mean diffusivity in left thalamic radiation, bilateral c

White Matter-Based Structural Brain Network of Anxiety.

Anxiety disorders are characterized by excessive fear and anxiety and related behavioral disturbances. Because diffusion tensor imaging is sensitive to detect subtle pathology of the brain, it has been used to characterize differences in white matter microstructure for a broad spectrum of psychiatric disorders. The neurobiological underpinnings of a trait anxiety seem to be associated with the uncinate fasciculus, a major pathway between the amygdala and orbitofrontal cortex. Apparent WM micro-alterations in patients with panic disorder are present in diverse and widespread regions, although a

White Matter Features Associated With Autistic Traits in Obsessive-Compulsive Disorder.

Obsessive-compulsive disorder (OCD) is among the most debilitating psychiatric disorders. Comorbid autism spectrum disorder (ASD) or autistic traits may impair treatment response in OCD. To identify possible neurostructural deficits underlying autistic traits, we performed white matter tractography on diffusion tensor images (DTI) and assessed autistic trait severity using the Autism-Spectrum Quotient (AQ) in 33 OCD patients. Correlations between AQ and the DTI parameters, fractional anisotropy (FA), mean diffusivity (MD), axial diffusivity (AD), and radial diffusivity (RD) were examined in ma

Abnormal language pathway in children with Angelman syndrome.

Angelman syndrome is a genetic disorder characterized by pervasive developmental disability with failure to develop speech. We examined the basis for severe language delay in patients with Angelman syndrome by diffusion tensor imaging. Magnetic resonance imaging/diffusion tensor imaging was performed in 7 children with genetically confirmed Angelman syndrome (age 70 ± 26 months, 5 boys) and 4 age-matched control children to investigate the microstructural integrity of arcuate fasciculus and other major association tracts. Six of 7 children with Angelman syndrome had unidentifiable left arcuate

Dissociations in white matter tracts and neuropsychological findings in a 17 years old patient with Subacute sclerosing panencephalitis.

The present multimodal diffusion tensor imaging and neuropsychological study investigated the integrity of the white matter fascicles in a 17 years-old patient diagnosed with subacute sclerosing panencephalitis (SSPE). A brief neuropsychological testing showed that word and pseudoword repetition, naming, semantic and phonological fluency, long-term memory, working memory were impaired. A review of the literature on Diffusion weighted imaging (DWI) and Diffusion Tensor Imaging (DTI) evidenced that, studies investigating the integrity of white matter in this condition being a rare disease, are v

Early life stress, cortisol, frontolimbic connectivity, and depressive symptoms during puberty.

Early life stress (ELS) is a risk factor for the development of depression in adolescence; the mediating neurobiological mechanisms, however, are unknown. In this study, we examined in early pubertal youth the associations among ELS, cortisol stress responsivity, and white matter microstructure of the uncinate fasciculus and the fornix, two key frontolimbic tracts; we also tested whether and how these variables predicted depressive symptoms in later puberty. A total of 208 participants (117 females; M age = 11.37 years; M Tanner stage = 2.03) provided data across two or more assessment modalit

Longitudinal Structural and Diffusion-Weighted Neuroimaging of Young Children Born Preterm.

BACKGROUND: Children born preterm are at risk for diffuse injury to subcortical gray and white matter. METHODS: We used a longitudinal cohort study to examine the development of subcortical gray matter and white matter volumes, and diffusivity measures of white matter tracts following preterm birth. Our participants were 47 children born preterm (24 to 32 weeks gestational age) and 28 children born at term. None of the children born preterm had significant neonatal brain injury. Children received structural and diffusion weighted magnetic resonance imaging scans at ages five, six, and seven y

White-matter tract abnormalities and antisocial behavior: A systematic review of diffusion tensor imaging studies across development.

Antisocial behavior (AB), including aggression, violence, and theft, is thought be underpinned by abnormal functioning in networks of the brain critical to emotion processing, behavioral control, and reward-related learning. To better understand the abnormal functioning of these networks, research has begun to investigate the structural connections between brain regions implicated in AB using diffusion tensor imaging (DTI), which assesses white-matter tract microstructure. This systematic review integrates findings from 22 studies that examined the relationship between white-matter microstruct

Depression and anxiety mediate the relationship between frontotemporal white matter integrity and quality of life in distressed young adults.

Depression and anxiety have been linked to poor quality of life (QoL) - one's subjective perception of relationships, physical health, daily functioning, general sense of well-being and life satisfaction. Elucidating abnormal white matter microstructure associated with mood and other symptoms and QoL is important to facilitate treatment. Ninety-six young adults (18-25 years old) seeking help for psychological distress, irrespective of presence or absence of psychiatric diagnosis completed diffusion weighted and anatomical scans, clinical and behavioral measures, and QoL assessment. We examined

White matter integrity and key structures affected in Alzheimer's disease characterized by diffusion tensor imaging.

White matter (WM) degeneration is suggested to predict the early signs of Alzheimer's disease (AD). The exact structural regions of brain circuitry involved are not known. This study aims to examine the associations between WM tract integrity, represented by the diffusion tensor imaging (DTI) measures, and AD diagnosis and to denote the key substrates in predicting AD. It included DTI measures of mean diffusivity (MD), fractional anisotropy, radial diffusivity and axial diffusivity of 18 main WM tracts in 84 non-Hispanic white participants from the Alzheimer's Disease Neuroimaging Initiative d

Impairment of speech production predicted by lesion load of the left arcuate fasciculus.

BACKGROUND AND PURPOSE: Previous studies have suggested that patients' potential for poststroke language recovery is related to lesion size; however, lesion location may also be of importance, particularly when fiber tracts that are critical to the sensorimotor mapping of sounds for articulation (eg, the arcuate fasciculus) have been damaged. In this study, we tested the hypothesis that lesion loads of the arcuate fasciculus (ie, volume of arcuate fasciculus that is affected by a patient's lesion) and of 2 other tracts involved in language processing (the extreme capsule and the uncinate fasci
