A reverse vaccinology approach to swine dysentery vaccine development.

Swine dysentery (SD) is a mucohaemorrhagic colitis of pigs resulting from infection of the large intestine with the anaerobic intestinal spirochaete Brachyspira hyodysenteriae. Whole-cell bacterin vaccines are available to help control SD, but their performance has been inconsistent. This study aimed to use a reverse vaccinology approach to identify B. hyodysenteriae proteins for use as recombinant vaccine components. Nineteen open reading frames (ORFs) predicted to encode potential vaccine candidate molecules were identified from in silico analysis of partial genomic sequence data. The distri

Reverse vaccinology and vaccines for serogroup B Neisseria meningitidis.

Whole genome sequence data are increasingly available for a wide range of human pathogens. The use of bioinformatic tools allows the comprehensive in silico screening of genome data for surface-expressed proteins, in order to identify candidate vaccine antigens. In vitro confirmation of surface location and the use of animal models to test immunogenicity further refine the list of proteins likely to be of use as vaccine antigens. This process, first applied to serogroup B Neisseria meningitidis, has been termed as reverse vaccinology. Reverse vaccinology offers the ability to undertake a rapid

Changing Priorities in Vaccinology: Antibiotic Resistance Moving to the Top.

Antimicrobial resistance (AMR) is currently the most alarming issue for human health. AMR already causes 700,000 deaths/year. It is estimated that 10 million deaths due to AMR will occur every year after 2050. This equals the number of people dying of cancer every year in present times. International institutions such as G20, World Bank, World Health Organization (WHO), UN General Assembly, European Union, and the UK and USA governments are calling for new antibiotics. To underline this emergency, a list of antibiotic-resistant "priority pathogens" has been published by WHO. It contains 12 fam

Reverse vaccinology--in search of a genome-derived meningococcal vaccine.

Although significant advances have been made toward the control of bacterial meningitis in children with the development of capsular polysaccharide protein conjugate vaccines, this approach has proven problematic for the serogroup B meningococcus. Non-capsular vaccines based upon outer membrane vesicles of Neisseria meningitidis have been useful in control of clonal serogroup B outbreaks, although due to variability of PorA, these vaccines may be less useful in control of endemic disease. Genome-based vaccine discovery was evaluated in an attempt to produce a candidate capable of conferring a

Reverse Vaccinology: An Approach for Identifying Leptospiral Vaccine Candidates.

Leptospirosis is a major public health problem with an incidence of over one million human cases each year. It is a globally distributed, zoonotic disease and is associated with significant economic losses in farm animals. Leptospirosis is caused by pathogenic Leptospira spp. that can infect a wide range of domestic and wild animals. Given the inability to control the cycle of transmission among animals and humans, there is an urgent demand for a new vaccine. Inactivated whole-cell vaccines (bacterins) are routinely used in livestock and domestic animals, however, protection is serovar-restric

The Input of Structural Vaccinology in the Search for Vaccines against Bunyaviruses.

A significant increase in the number of viruses causing unexpected illnesses and epidemics among humans, wildlife and livestock has been observed in recent years. These new or re-emerging viruses have often caught the scientific community off-guard, without sufficient knowledge to combat them, as shown by the current coronavirus pandemic. The bunyaviruses, together with the flaviviruses and filoviruses, are the major etiological agents of viral hemorrhagic fever, and several of them have been listed as priority pathogens by the World Health Organization for which insufficient countermeasures e

The Africa COVID-19 legacy-Improved vaccinology for pandemic preparedness.

The COVID-19 pandemic caused global disruption that reverberated across Africa and revealed deep systemic vulnerabilities. Nonetheless, the scientific response to SARS-CoV-2 catalyzed unprecedented global scientific collaboration, established new collaborative networks, and accelerated local capacity building across Africa. This legacy extends beyond COVID-19, informing responses to subsequent mpox and Ebola outbreaks, and establishes a framework to respond to future infectious disease threats in Africa.

African Swine Fever Vaccinology: The Biological Challenges from Immunological Perspectives.

African swine fever virus (ASFV), a nucleocytoplasmic large DNA virus (NCLDV), causes African swine fever (ASF), an acute hemorrhagic disease with mortality rates up to 100% in domestic pigs. ASF is currently epidemic or endemic in many countries and threatening the global swine industry. Extensive ASF vaccine research has been conducted since the 1920s. Like inactivated viruses of other NCLDVs, such as vaccinia virus, inactivated ASFV vaccine candidates did not induce protective immunity. However, inactivated lumpy skin disease virus (poxvirus) vaccines are protective in cattle. Unlike some e

Systems vaccinology: its promise and challenge for HIV vaccine development.

PURPOSE OF REVIEW: The use of systems biology approaches to understand and predict vaccine-induced immunity promises to revolutionize vaccinology. For centuries vaccines were developed empirically, with very little understanding of the mechanisms by which they mediate protective immunity. The so-called systems vaccinology approach employs high-throughput technologies (e.g. microarrays, RNA-seq and mass spectrometry-based proteomics and metabolomics) and computational modeling to describe the complex interactions between all the parts of immune system, with a view to elucidating new biological

The Development of a Vaccine Against Meningococcus B Using Reverse Vaccinology.

The discovery of vaccine antigens through whole genome sequencing (WGS) contrasts with the classical hypothesis-driven laboratory-based analysis of microbes to identify components to elicit protective immunity. This radical change in scientific direction and action in vaccine research is captured in the term reverse vaccinology. The complete genome sequence of an isolate of Neisseria meningitidis serogroup B (MenB) was systematically analyzed to identify proteins predicted to be secreted or exported to the outer membrane. This identified hundreds of genes coding for potential surface-exposed a

Systems vaccinology: learning to compute the behavior of vaccine induced immunity.

The goal of systems biology is to access and integrate information about the parts (e.g., genes, proteins, cells) of a biological system with a view to computing and predicting the behavior of the system. The past decade has witnessed technological revolutions in the capacity to make high throughput measurements about the behavior of genes, proteins, and cells. Such technologies are widely used in biological research and in medicine, such as toward prognosis and therapy response prediction in cancer patients. More recently, systems biology is being applied to vaccinology, with the goal of: (1)

[Vaccinology in armed conflicts: a punctual intervention in poliomyelitis eradication in Afghanistan].

The situations of armed conflict suppose a handicap for the eradication of the poliomyelitis due to the difficulties to access the susceptible population. In Afghanistan, after the military operation which put an end to the Taliban regime, Spanish military sanitarians made an intervention in order to immunize the children of the zone where they were conducting their mission, against poliomyelitis. After having solved planning and logistic problems, two rounds of vaccination with IPV were made. A local translator helped to fill out a questionnaire about the state of immunization in that zone, p

Multiviral Quartet Nanocages Elicit Broad Anti-Coronavirus Responses for Proactive Vaccinology.

Defending against future pandemics may require vaccine platforms that protect across a range of related pathogens. The presentation of multiple receptor-binding domains (RBDs) from evolutionarily-related viruses on a nanoparticle scaffold elicits a strong antibody response to conserved regions. Here we produce quartets of tandemly-linked RBDs from SARS-like betacoronaviruses coupled to the mi3 nanocage through a SpyTag/SpyCatcher spontaneous reaction. These Quartet Nanocages induce a high level of neutralizing antibodies against several different coronaviruses, including against viruses not re

Discovery of Novel Leptospirosis Vaccine Candidates Using Reverse and Structural Vaccinology.

Leptospira spp. are diderm (two membranes) bacteria that infect mammals causing leptospirosis, a public health problem with global implications. Thousands of people die every year due to leptospirosis, especially in developing countries with tropical climates. Prophylaxis is difficult due to multiple factors, including the large number of asymptomatic hosts that transmit the bacteria, poor sanitation, increasing numbers of slum dwellers, and the lack of an effective vaccine. Several leptospiral recombinant antigens were evaluated as a replacement for the inactivated (bacterin) vaccine; however

Identification of novel vaccine candidates against Acinetobacter baumannii using reverse vaccinology.

Acinetobacter baumannii (Ab) is a global emerging bacterium causing nosocomial infections such as pneumonia, meningitis, bacteremia and soft tissue infections especially in intensive care units. Since Ab is resistant to almost all conventional antibiotics, it is now one of the 6 top-priorities of the dangerous microorganisms listed by the Infectious Disease Society of America. The development of vaccine is one of the most promising and cost-effective strategies to prevent infections. In this study, we identified potential protective vaccine candidates using reverse vaccinology. We have analyze

Novel Vaccine Candidates against Brucella melitensis Identified through Reverse Vaccinology Approach.

Global health therapeutics is a rapidly emerging facet of postgenomics medicine. In this connection, Brucella melitensis is an intracellular bacterium that causes the zoonotic infectious disease, brucellosis. Presently, no licensed vaccines are available for human brucellosis. Here, we report the identification of potential vaccine candidates against B. melitensis using a reverse vaccinology approach. Based on a systematic screening of exoproteome and secretome of B. melitensis 16 M, we identified eight proteins as potential vaccine candidates, including LPS-assembly protein LptD, a polysaccha
