In vitro multi-organ invasion model for exploring cardiac resistance to cancer metastasis.

Despite the systemic spread of cancer, the ventricular myocardium is one of the least common sites of metastasis-a phenomenon that remains poorly understood. To examine this, we develop the Multi-organ Invasion Device (MInD), an organ-on-a-chip platform that enables multi-organ culture under flow. Organ compartments are connected in MInD using PermeoTubes-3D-printed porous conduits that support cancer cell intravasation, migration, and extravasation. In dual-organ devices, where highly aggressive breast cancer cells are co-cultured with either hepatic or cardiac tissue, invasion into cardiac t

Cisplatin-Resistant Lung Adenocarcinoma Cells Exhibit Increased Proangiogenic Capacity in a Microphysiological Model of Tumor Neovascularization.

Carcinomas commonly recur and progress rapidly after a period of remission following platinum-based therapy. This clinical scenario suggests that surviving drug-resistant tumor cells are dormant or slow-cycling before re-entering a rapid growth phase. Remodeling of the recurrent tumor microenvironment (TME) contributes to high rates of metastasis, but little is known about differences in TME remodeling before therapy and after recurrence. This study explores selection for cisplatin-resistant subpopulations of A549 lung adenocarcinoma cells in culture to derive populations for modeling features

Protective effect of osmanthus water extract on liver dysfunction caused by DBP based on organoids and organ chips technologies.

This study aimed to investigate the protective mechanism of Osmanthus fragrans water extract (OSF) against liver injury induced by dibutyl phthalate (DBP). We utilized liver organoids and liver organ chip technology to replicate the liver microenvironment in vivo. Metabolomic analysis revealed that DBP induced oxidative stress and lipid metabolism disorders; however, following intervention with OSF, the associated abnormal metabolites were significantly reduced. Molecular docking studies demonstrated the binding of its active ingredients to key targets. The primary findings indicate that OSF c

Impact of microgravity on a three-dimensional microphysiologic culture of the human kidney proximal tubule epithelium: cell response to serum and vitamin D.

The microgravity environment aboard the International Space Station (ISS) provides a unique stressor that can help understand underlying cellular and molecular drivers of pathological changes observed in astronauts with the ultimate goals of developing strategies to enable long-term spaceflight and better treatment of diseases on Earth. We used this unique environment to evaluate the effects of microgravity on kidney proximal tubule epithelial cell (PTEC) response to serum exposure and vitamin D biotransformation capacity. To test if microgravity alters the pathologic response of the proximal

Miniaturized engineered heart tissues from hiPSC-derived triple cell type co-cultures to study human cardiac function.

Human heart tissues grown as three-dimensional spheroids and consisting of different cardiac cell types derived from pluripotent stem cells (hiPSCs) recapitulate aspects of human physiology better than standard two-dimensional models in vitro. They typically consist of less than 5000 cells and are used to measure contraction kinetics although not contraction force. By contrast, engineered heart tissues (EHTs) formed around two flexible pillars, can measure contraction force but conventional EHTs often require between 0.5 and 2 million cells. This makes large-scale screening of many EHTs costly

Extracellular matrix properties, interstitial flow, and VEGF gradients shape trophoblast behavior in a pumpless Trophoblast Invasion-on-Chip (TIoC).

Placental trophoblast invasion into the maternal endometrium is a critical step in establishing the maternal-fetal interface during implantation. In vitro, this complex process can be partially modeled using a Trophoblast Invasion-on-Chip (TIoC) platform. In this study, we used an in-house-developed, pumpless, recirculating "loop-in-loop" microfluidic device that generates interstitial flow (IF) within a tissue compartment to optimize indirect co-culture of cytotrophoblast BeWo cells with human umbilical vein endothelial cells (HUVECs). Eight extracellular matrix (ECM) conditions were tested,

Acoustic cell patterning reveals geometry- and substrate-dependent vasculogenesis and human embryo model development.

Precise spatial organization of cells is critical for engineering functional tissues and constructing human developmental models. Here, we report the development of acoustic cell patterning (ACP), an ultrasound-based method capable of rapidly generating spatial patterns of multiple cell types on both rigid and compliant substrates without requiring chemical or physical modification of the substrates. Spatially controlled co-patterning of human endothelial and stromal fibroblast cells through ACP reveals geometry- and substrate-dependent vascular morphogenesis. Applied to human pluripotent stem

Engineering Epithelial-Mesenchymal Microtissues to Study Cell-Cell Interactions in Development.

Intercellular signaling drives human development, but there is a paucity of in vitro models that recapitulate important tissue architecture while remaining operationally simple and scalable. As an example, formation of the upper lip and palate requires the orchestrated proliferation and fusion of embryonic facial growth centers and is dependent on paracrine epithelial-mesenchymal signaling through multiple pathways including the Sonic Hedgehog (SHH), transforming growth factor-beta (Tgf-β), bone morphogenic protein (BMP), and epidermal growth factor (EGF) pathways. We have developed a robust,

Melt electrowritten microfiber-hydrogel composite scaffolds with local cross-bridging reinforcements for aligned muscle tissue engineering.

Effective regeneration of skeletal muscle with highly aligned fiber architecture remains a significant challenge in tissue engineering. Structural alignment of muscle constructs along with mechanical integrity are crucial for effective engineering of grafts and microphysiological systems. This study introduced a composite microfiber-hydrogel platform using melt electrowriting (MEW) with a reinforcing architecture for aligned fiber scaffolds that maintains directional consistency while providing mechanical cohesion. Three MEW scaffold designs (Isotropic, Aligned T with perpendicular fiber reinf

Secondary bile acid lithocholic acid ameliorates colitis-like inflammation in a human intestine-on-chip system.

Inflammatory bowel disease is a multifactorial disease of the gastrointestinal tract without curative treatment. Previous studies highlighted that altered fecal bile acid levels correlate with intestinal microbiota composition changes and inflammation in inflammatory bowel disease. Lithocholic acid is a secondary bile acid drastically reduced during active inflammatory bowel disease but mediates beneficial effects at the mucosal intestinal barrier during intestinal homeostasis. In a dextran sodium sulfate-induced colitis-on-chip model, it was investigated whether the administration of lithocho

Endothelial-immune cells-on-chip for multiplex evaluation of drug responses through patient plasma screening.

Most organ-on-chip models focus on parenchymal cell function, overlooking the critical role of endothelial-immune interactions in metabolic disease pathophysiology. Drugs for metabolic conditions, such as SGLT2 inhibitors, provide not only metabolic improvements but also vascular-protective effects, underscoring the need for human platforms that capture immunovascular dynamics. Here, we developed a human endothelial-immune cells-on-chip system integrating patient-derived blood outgrowth endothelial cells, T cells, and hepatocytes to model the liver sinusoidal microenvironment. Perfluorocarbon ameliorates high-altitude pulmonary edema and blood-gas barrier dysfunction in a canine model and a lung-on-chip model by suppressing the HIF-1α/HO-1 pathway.

High-altitude pulmonary edema (HAPE) is an increasing problem with high morbidity and mortality due to its rapid onset and progression. Perfluorocarbon (PFC) is reported to have promising effects on acute lung injury (ALI) by offering oxygenation and anti-inflammatory effects. However, PFC's effect on HAPE is still unknown. In our study, we investigated the effect of PFC, first in a HAPE canine model and then in a lung-on-chip model. We exposed beagle dogs to a simulated altitude of 6000 m for 48 h to establish the HAPE model and treated them with vaporized PFC. Next, we created a lung-on-chip

Microfluidic liver-chip to predict clinical drug-drug interaction net effects for precipitant drugs with dual CYP3A4 inhibition and induction potential.

Accurate predictions of complex clinical drug-drug interactions (DDIs), arising from dual induction and time-dependent inhibition (TDI) of CYP3A4, has remained challenging with conventional in vitro and static/dynamic modeling approaches. In this work, we aimed to anticipate the hepatic DDI effects of 6 CYP3A4 precipitant drugs using a liver-chip coupled to microfluidic perfusion, which enabled simulating clinically relevant pharmacokinetic (PK) time-concentration profiles. Midazolam clearance was measured in the liver-chip to determine the CYP3A4-mediated DDI net effect following exposure to

A Microphysiological Model of Progressive Human Hepatic Insulin Resistance.

BACKGROUND & AIMS: Hepatic insulin resistance is a fundamental phenomenon observed in both Type 2 diabetes (T2D) and metabolic (dysfunction) associated fatty liver disease (MAFLD). The relative contributions of nutrients, hyperinsulinemia, hormones, inflammation, and other cues are difficult to parse in vivo as they are convoluted by interplay between the local and systemic events. Here, we used a well-established human liver microphysiological system (MPS) to establish a physiologically-relevant insulin-responsive metabolic baseline and probe how primary human hepatocytes respond to controlle

In vitro safety "clinical trial" of the cardiac liability of drug polytherapy.

Only a handful of US Food and Drug Administration (FDA) Emergency Use Authorizations exist for drug and biologic therapeutics that treat severe acute respiratory syndrome-coronavirus 2 (SARS-CoV-2) infection. Potential therapeutics include repurposed drugs, some with cardiac liabilities. We report on a chronic preclinical drug screening platform, a cardiac microphysiological system (MPS), to assess cardiotoxicity associated with repurposed hydroxychloroquine (HCQ) and azithromycin (AZM) polytherapy in a mock phase I safety clinical trial. The MPS contained human heart muscle derived from induc

A 3D Microfluidic Blood-Brain Barrier Chip for Real-Time Assessment of Micro/Nanoplastics Permeability and Neuroinflammatory Injury.

The micro/nanoplastics (MNPs) have been evidenced to exert detrimental effects on the blood-brain barrier (BBB) and the central nervous system (CNS). However, there is still a lack of effective research models on the mechanism of nerve injury caused by microplastics particles. This study focuses on analyzing the particle size characteristics of MNPs precipitated from plastic water bottles under different conditions of storage and uses 3D BBB microfluidic chips to assess the permeability and dynamic neurotoxicity of MNPs. The results showed that there was a significant increase in the average d

Self-assembly of tumor-related vascularized colorectal tumoroid-on-a-chip for precision medicine drug testing.

The complexity of tumors cannot be accurately simulated by current tumoroid models. One of these limitations is poor vascularization in current tumor models, particularly the lack of tumor-specific blood vessels. Therefore, it is challenging to have a thorough discussion of many in vitro studies regarding the tumor vascular environment. To overcome this limitation, this study combines tumoroid and microfluidic technologies to develop a vascularized tumoroid-on-a-chip. This physiologically relevant colorectal tumoroid-on-a-chip enables self-assembly of tumor-associated vasculature and provides

A human 3D BBB chip model of acute stroke simulating a reversible penumbra.

Ischemic stroke is a leading cause of mortality and disability worldwide. However, existing models often fail to replicate key aspects of human pathophysiology, particularly blood-brain barrier (BBB) dysfunction and the salvageable ischemic penumbra. We developed a three-dimensional BBB chip model of acute ischemic stroke that reproduces penumbra-like, partially reversible BBB injury. This platform integrates a microfluidic BBB chip (Emulate) with parallel Transwell inserts to facilitate complementary structural, molecular, and functional analyses. Ischemia-like injury was induced using 2.5 μM

Biomimetic Urethra-on-a-Chip Platform for Modelling Fibrosis: 3D-Printing and Near-Field Electrospinning in BAM-Functionalized Microenvironment.

Urethral stricture, a prevalent urological disorder characterized by fibrosis of periurethral tissues, severely compromises urinary function and patient quality of life. Despite various clinical interventions, recurrence remains frequent, largely due to the lack of physiologically relevant in vitro models for mechanistic investigation and drug screening. Here, we present a biomimetic urethra-on-a-chip platform that integrates microfluidics, three-dimensional (3D) printing, and near-field electrospinning to recapitulate the structural and biochemical complexity of the native urethra. Mucopenetrative Lipid-Polymer nanoparticles show Potent Anti-Inflammatory activity in a human Lung-on-Chip model.

Airway mucus presents a significant barrier to inhaled drug delivery, particularly for nanoparticle-based interventions, with this barrier exacerbated in chronic respiratory diseases (CRDs) due to hyperviscous secretions and persistent inflammation. In this study, a dual-functional lipid-polymer hybrid nanoparticle was developed to combine rapid mucolysis with sustained anti-inflammatory activity, and its performance was evaluated using both conventional in vitro assays and a physiologically relevant lung-on-a-chip model.
