Comparative sequence, structure and redox analyses of Klebsiella pneumoniae DsbA show that anti-virulence target DsbA enzymes fall into distinct classes.

Bacterial DsbA enzymes catalyze oxidative folding of virulence factors, and have been identified as targets for antivirulence drugs. However, DsbA enzymes characterized to date exhibit a wide spectrum of redox properties and divergent structural features compared to the prototypical DsbA enzyme of Escherichia coli DsbA (EcDsbA). Nonetheless, sequence analysis shows that DsbAs are more highly conserved than their known substrate virulence factors, highlighting the potential to inhibit virulence across a range of organisms by targeting DsbA. For example, Salmonella enterica typhimurium (SeDsbA,

PDIA6 exacerbates hepatocellular carcinoma via enhancing proteasome-dependent degradation of AKAP12.

The continuously rising incidence and persistently high mortality of hepatocellular carcinoma (HCC) have created an urgent need for a deeper understanding of the molecular mechanisms underlying this disease. In the present study, leveraging multiple HCC transcriptome databases, we employed expression analysis, correlation analysis, Gene Set Variation Analysis (GSVA), and cox regression analysis to identify that aberrant upregulation of PDIA6 represents a promising prognostic biomarker associated with adverse clinical outcomes in HCC. Using a series of in vitro oncology research approaches, as

Immune-evasion-associated prognostic gene expression score stratifies survival and immune microenvironment features in breast cancer.

Breast cancer is characterized by marked prognostic and immune heterogeneity, and tumor immune evasion may contribute to variable responses to immunotherapy. This study aimed to characterize immune-evasion-associated transcriptomic patterns in breast cancer and to develop an exploratory prognostic score linked to immune microenvironment features. A curated set of 182 immune-evasion-associated genes (IEGs) was analyzed in The Cancer Genome Atlas Breast Invasive Carcinoma (TCGA-BRCA) cohort. Fifteen differentially expressed IEGs distinguished two molecular clusters with different overall surviva

AGR2, an androgen-inducible secretory protein overexpressed in prostate cancer.

AGR2, the human homologue of Xenopus anterior gradient 2 (XAG2), was identified by a suppression subtractive hybridization-based technique as an androgen-inducible gene. There are two AGR2 transcripts, which encode the same secretory protein of 175 amino acids. The androgen induction was time- and dose-dependent, with more than a 10-fold increase in the level of AGR2 mRNA after 48 hr of treatment with 10(-9) M R1881. Expression of AGR2 mRNA was specifically detected in limited human tissue rich in epithelial cells, including the prostate gland. Analysis of 46 microdissected primary prostate ad

Reductions of the components of the calreticulin/calnexin quality-control system by proteasome inhibitors and their relevance in a rodent model of Parkinson's disease.

Evidence indicates that the ubiquitin-proteasome system and the endoplasmic retculum (ER) quality-control system work in concert to ensure that proteins are correctly folded in the ER and that misfolded proteins are retrotransported to the cytosol for degradation by proteasomes. Dysfunction of either system results in developmental abnormalities and even death in animals. This study investigates whether and how proteasome inhibition impacts the components of the calreticulin (CRT)/calnexin (CNX) glycoprotein folding machinery, a typical ER protein quality-control system, in the context of earl

TXNDC5 in POAG: Promoting Extracellular Matrix Protein Accumulation and Raising Intraocular Pressure.

PURPOSE: Excessive accumulation of extracellular matrix (ECM) proteins in the trabecular meshwork (TM) leads to increased resistance to outflow of aqueous humor (AH) and the consequent increase in intraocular pressure (IOP) in primary open-angle glaucoma (POAG). Currently, there is no effective treatment to address this pathogenic mechanism of POAG. Therefore it is essential to screen for key proteins that are involved in TM ECM accumulation and further develop drugs targeting these mechanisms. METHODS: Proteins were screened for significant up-regulation in TGFβ2-induced TM by label-free qua

Downregulation of ERp57 expression is associated with poor prognosis in early-stage cervical cancer.

OBJECTIVE: We investigated the clinical significance of ERp57 in the progression of cervical cancer. METHODS: mRNA and protein expression of ERp57 in cervical neoplasias were examined. RESULTS: ERp57 mRNA expression was significantly decreased in cervical cancers. Immunohistochemistry revealed that ERp57 expression in 123 cervical cancers was down-regulated compared to cervical intraepithelial neoplasias or normal tissues (p < 0.001). Low ERp57 expression was significantly associated with worse overall survival (HR = 12.19, p = 0.018). CONCLUSIONS: Low ERp57 expression independently predict

Induction of endoplasmic reticulum stress in thymic lymphocytes by the envelope precursor polyprotein of a murine leukemia virus during the preleukemic period.

Infection of thymic lymphocytes by a mink cell focus-forming murine leukemia virus induces apoptosis during the preleukemic period of lymphomagenesis. In this study, we observed that during this period, the viral envelope precursor polyprotein accumulated to high levels in thymic lymphocytes from mice inoculated with virus. Envelope accumulation occurred with the same kinetics as the induction of endoplasmic reticulum (ER) stress, which resulted in the upregulation of the 78-kDa glucose-regulated protein (GRP78). In thymic lymphomas, GRP78 levels were higher than those in virus-infected preleu

The P4HB/LGALS3BP/NF-kB axis promotes the progression of MIA to IAC in lung adenocarcinoma.

Prolyl 4-hydroxylase β-subunit (P4HB) is a class of molecular chaperones that inhibits the misfolding of endoplasmic reticulum proteins, and its role in microinvasive adenocarcinoma (MIA) progresses to invasive adenocarcinoma (IAC) is currently unclear. We mined the single-cell dataset GSE189357 to analyze the role of P4HB in LUAD progression. Subsequently, P4HB was overexpressed in H1299 and A549 cell lines, and Galactose lectin-3 binding protein (LGALS3BP) expression was increased and NF-kB pathway was inhibited by PCR and WB. Meanwhile, we also knock down of P4HB in H1299 and A549 cell line

Hepatic PDIA5 improves hyperglycaemia and insulin resistance through stabilisation of ATF6.

AIMS/HYPOTHESIS: Dysregulation of hepatic glucose production, primarily driven by aberrantly elevated gluconeogenesis, is a core pathological feature of type 2 diabetes. However, the molecular mechanisms governing this process remain incompletely understood. Protein disulfide isomerase A5 (PDIA5) is ubiquitously expressed, but its specific function in hepatic metabolism is unknown. We hypothesised that PDIA5 plays a critical role in regulating hepatic gluconeogenesis, and that its dysregulation contributes to hyperglycaemia. METHODS: We assessed hepatic PDIA5 expression in liver biopsies from

The Cys-N-degron pathway modulates pexophagy through the N-terminal oxidation and arginylation of ACAD10.

In the N-degron pathway, N-recognins recognize cognate substrates for degradation via the ubiquitin (Ub)-proteasome system (UPS) or the autophagy-lysosome system (hereafter autophagy). We have recently shown that the autophagy receptor SQSTM1/p62 (sequestosome 1) is an N-recognin that binds the N-terminal arginine (Nt-Arg) as an N-degron to modulate autophagic proteolysis. Here, we show that the N-degron pathway mediates pexophagy, in which damaged peroxisomal fragments are degraded by autophagy under normal and oxidative stress conditions. This degradative process initiates when the Nt-Cys of

Peptide Loading Complex in Cancer: From Peptide Translocation, Editing, and Loading into MHC-I to a Potential Therapeutic Target.

The peptide-loading complex (PLC) is a pivotal endoplasmic reticulum (ER) protein machinery that facilitates peptide translocation, editing, and loading onto major histocompatibility complex class I (MHC-I) to allow recognition of infected or transformed cells by CD8⁺ cytotoxic T lymphocytes (CTLs). PLC comprises the peptide transporter complex formed of transporters associated with antigen processing (TAP1/2), the adapter protein tapasin, chaperones such as calreticulin (CRT) and ERp57, and β2-microglobulin (β2M), ensuring the proper assembly of MHC-I/peptide complexes for subsequent antigen

Full-length recombinant antibodies from Escherichia coli: production, characterization, effector function (Fc) engineering, and clinical evaluation.

Although several antibody fragments and antibody fragment-fusion proteins produced in Escherichia coli (E. coli) are approved as therapeutics for various human diseases, a full-length monoclonal or a bispecific antibody produced in E. coli has not yet been approved. The past decade witnessed substantial progress in expression of full-length antibodies in the E. coli cytoplasm and periplasm, as well as in cell-free expression systems. The equivalency of E. coli-produced aglycosylated antibodies and their mammalian cell-produced counterparts, with respect to biochemical and biophysical propertie

HLA-B27 Subtypes Predisposing to Ankylosing Spondylitis Accumulate in an Endoplasmic Reticulum-Derived Compartment Apart From the Peptide-Loading Complex.

OBJECTIVE: It was previously shown that HLA-B27 subtypes predisposing to spondyloarthritis (SpA), i.e., B*27:02, B*27:05, and B*27:07, displayed an increased propensity to form intracellular oligomers and to accumulate at a high density in cytoplasmic vesicles, as compared to the non-SpA-associated HLA-B*07:02 and HLA-B*27:06. This study was undertaken to characterize the nature and content of HLA-B-containing vesicles and to further examine their relevance to SpA predisposition. METHODS: Vesicles containing HLA-B proteins were detected in transfected HeLa cells and in cells from SpA patients

Antigen-processing machinery in human dendritic cells: up-regulation by maturation and down-regulation by tumor cells.

It has been known for some time that functional properties of dendritic cells (DC), and in particular their ability to process and present Ags to T cells, can be modulated by cytokine-induced maturation and by interactions with tumor cells. However, the molecular basis for these functional changes is unknown. We have investigated whether changes in expression of Ag-processing machinery (APM) components in DC are associated with alterations in their ability to present tumor-derived Ags to T cells. Using a panel of mAbs specific for individual APM components and a quantitative flow cytometry-bas
