Landscape genetics reveals focal transmission of a human macroparasite.

Macroparasite infections (e.g., helminths) remain a major human health concern. However, assessing transmission dynamics is problematic because the direct observation of macroparasite dispersal among hosts is not possible. We used a novel landscape genetics approach to examine transmission of the human roundworm Ascaris lumbricoides in a small human population in Jiri, Nepal. Unexpectedly, we found significant genetic structuring of parasites, indicating the presence of multiple transmission foci within a small sampling area ( approximately 14 km(2)). We analyzed several epidemiological variab

Epstein-Barr virus in multiple sclerosis pathogenesis: The path towards mechanistically faithful models.

Multiple Sclerosis (MS) is a chronic, disabling autoimmune disease of the central nervous system (CNS). While its aetiology is multifactorial, compelling evidence now implicates Epstein-Barr virus (EBV) as a primary aetiological agent. This review summarises the extensive epidemiological and mechanistic data supporting a causal link between EBV infection and MS. Epidemiological studies demonstrate that EBV infection confers >30-fold increased risk for MS, with seroconversion preceding the onset of neuroaxonal damage. The leading proposed mechanism is molecular mimicry, where antibodies and T c

Sleep Apnoea Adverse Effects on Cancer: True, False, or Too Many Confounders?

Obstructive sleep apnoea (OSA) is a prevalent disorder associated with increased cardiovascular, metabolic and neurocognitive morbidity. Recently, an increasing number of basic, clinical and epidemiological reports have suggested that OSA may also increase the risk of cancer, and adversely impact cancer progression and outcomes. This hypothesis is convincingly supported by biological evidence linking certain solid tumours and hypoxia, as well as by experimental studies involving cell and animal models testing the effects of intermittent hypoxia and sleep fragmentation that characterize OSA. Ho

Inflammation in prostate carcinogenesis.

About 20% of all human cancers are caused by chronic infection or chronic inflammatory states. Recently, a new hypothesis has been proposed for prostate carcinogenesis. It proposes that exposure to environmental factors such as infectious agents and dietary carcinogens, and hormonal imbalances lead to injury of the prostate and to the development of chronic inflammation and regenerative 'risk factor' lesions, referred to as proliferative inflammatory atrophy (PIA). By developing new experimental animal models coupled with classical epidemiological studies, genetic epidemiological studies and m

Multi-event capture-recapture models estimate the diagnostic performance of serological tests for myxoma and rabbit haemorrhagic disease viruses in the absence of reference samples.

Estimation of the diagnostic performance of serological tests often relies on another test assumed as a reference or on samples of known infection status, yet both are seldom available for emerging pathogens in wildlife. Longitudinal disease serological data can be analysed through multi-event capture-mark-recapture (MECMR) models accounting for the uncertainty in state assignment, allowing us to estimate epidemiological parameters such as incidence and mortality. We hypothesized that by estimating the uncertainty in state assignment, MECMR models estimate the diagnostic performance of serolog

Role of pregnancy and female sex steroids on aneurysm formation, growth, and rupture: a systematic review of the literature.

OBJECTIVE: Women have been shown to have a higher risk of cerebral aneurysm formation, growth, and rupture than men. The authors present a review of the recently published neurosurgical literature that studies the role of pregnancy and female sex steroids, to provide a conceptual framework with which to understand the various risk factors associated with cerebral aneurysms in women at different stages in their lives. METHODS: The PubMed database was searched for "("intracranial" OR "cerebral") AND "aneurysm" AND ("pregnancy" OR "estrogen" OR "progesterone")" between January 1980 and February

Psychological stress and type 1 diabetes mellitus: what is the link?

Type 1 diabetes mellitus (T1DM) is a chronic disease characterized by the destruction of insulin-producing β-cells of the pancreas. The current paradigm in this disease's etiopathogenesis points toward the interplay of genetic and environmental factors. Among the environmental variables, dietary factors, intestinal microbiota, toxins, and psychological stress have been implicated in disease onset. Areas covered: This review aims to investigate the relationship between psychological stress and T1DM by presenting evidence from epidemiological studies, animal models, and to provide the mechanism

Actions of estrogen and estrogen receptors in nonclassical target tissues.

Hormonal effects on classical endocrine target organs such as the female reproductive tract, mammary gland, ovary, and neuroendocrine system have been thoroughly studied, with significant advancements in our understanding of estrogen actions and disease conditions from both cell culture as well as new experimental animal models. Knowledge of the highly appreciated effects of estrogen in nonclassical endocrine organ systems, arising from epidemiological and clinical findings in the cardiovascular, immune, GI tract, and liver, is only now becoming clarified from the development and use of knock-

Is Panton-Valentine leukocidin the major virulence determinant in community-associated methicillin-resistant Staphylococcus aureus disease?

Methicillin-resistant Staphylococcus aureus (MRSA) remains a major problem in hospitals, and it is now spreading in the community. A single toxin, Panton-Valentine leukocidin (PVL), has been linked by epidemiological studies to community-associated MRSA (CA-MRSA) disease. However, the role that PVL plays in the pathogenesis of CA-MRSA has not been tested directly. To that end, we used mouse infection models to compare the virulence of PVL-positive with that of PVL-negative CA-MRSA representing the leading disease-causing strains. Unexpectedly, strains lacking PVL were as virulent in mouse seps

Inferring the disruption of rabies circulation in vampire bat populations using a betaherpesvirus-vectored transmissible vaccine.

Transmissible vaccines are an emerging biotechnology that hold prospects to eliminate pathogens from wildlife populations. Such vaccines would genetically modify naturally occurring, nonpathogenic viruses ("viral vectors") to express pathogen antigens while retaining their capacity to transmit. The epidemiology of candidate viral vectors within the target wildlife population has been notoriously challenging to resolve but underpins the selection of effective vectors prior to major investments in vaccine development. Here, we used spatiotemporally replicated deep sequencing to parameterize comp

The psychological factor 'self-blame' predicts overuse injury among top-level Swedish track and field athletes: a 12-month cohort study.

BACKGROUND: Athletes' psychological characteristics are important for understanding sports injury mechanisms. We examined the relevance of psychological factors in an integrated model of overuse injury risk in athletics/track and field. METHODS: Swedish track and field athletes (n=278) entering a 12-month injury surveillance in March 2009 were also invited to complete a psychological survey. Simple Cox proportional hazards models were compiled for single explanatory variables. We also tested multiple models for 3 explanatory variable groupings: an epidemiological model without psychological v

Detecting changepoints in dynamical systems: Modeling time-varying transmission of seasonal influenza.

Seasonal influenza epidemics exhibit complex transmission dynamics influenced by time-varying extrinsic factors such as social behavior and seasonal effects. Estimating changes in transmission rates is critical to enable accurate forecasting of the epidemic curve. This study presents a framework for detecting changepoints in the transmission parameter ([Formula: see text]), applied as a piecewise constant function within a deterministic compartmental model. Using hospitalized case data from four recent influenza A seasons in Ireland (2019/2020 and 2022-2025), we applied iterated filtering and

A robust compartmental modeling framework for infectious disease monitoring and analysis via fractional differential equations.

This study presents a comprehensive framework for infectious disease monitoring using fractional differential equations, specifically developing the SEIQRDP (Susceptible, Exposed, Infected, Quarantined, Recovered, Deceased, Protected) model. Traditional compartmental models are extended by incorporating fractional calculus, with orders α∈(0,2], which provides enhanced flexibility in capturing memory effects and non-local behaviors inherent in disease transmission dynamics. The framework demonstrates improved accuracy when fitted to Canadian COVID-19 data compared to classical integer-order mod

Global region-specific influenza early warning by establishing an epidemic intensity threshold system: a modeling study.

BACKGROUND: Influenza exhibits marked geographical heterogeneity in epidemic patterns, yet there is no globally standardized approach for determining epidemic thresholds. This study aimed to establish a region-specific epidemic intensity threshold system and evaluate its performance in influenza early warning and peak timing simulation. METHODS: Weekly influenza surveillance data from 167 countries (2009-2020) were obtained from the WHO Global Influenza Surveillance and Response System (GISRS). Three indicators were calculated for each of the 18 influenza transmission zones: ILI consultation

Stochastic models indicate rapid smallpox spread and mass mortality of Indigenous Australians after colonial exposure.

The impact of smallpox (variola) on Aboriginal communities in Australia beginning in 1789 was catastrophic and continues to cause intergenerational trauma. Historically biased perspectives and contemporary misinformation of the disease's introduction and spread impede modern-day truth-telling and efforts towards reconciliation and national healing. Understanding whether the disease entered and spread from pre-colonial Makassan (Indonesian) trade along the northern coast, or from the British First Fleet's arrival in southeastern Australia in 1788, is necessary to estimate the demographic impact

Estimating the potential impact of surveillance test-and-treat posts to reduce malaria in border regions in sub-Saharan Africa: a modelling study.

BACKGROUND: The last malaria cases in near-elimination settings are often found in international border regions due to the presence of hard-to-reach populations, conflict, uneven intervention coverage, and human migration. Test-and-treat border posts are an under-researched form of active case detection used to interrupt transmission chains between countries. METHODS: An individual-based, mathematical metapopulation model of Plasmodium falciparum was used to estimate the effectiveness of border screening posts on total cases in malaria-endemic sub-Saharan Africa. RESULTS: The implementation

Formulation of a new sir Model with Non-local Mobility.

In this manuscript, we develop a mobility-based Susceptible-Infectious-Recovered (SIR) model to elucidate the dynamics of pandemic propagation. While traditional SIR models within the field of epidemiology aptly characterize transitions among susceptible, infected, and recovered states, they typically neglect the inherent spatial mobility of particles. To address this limitation, we introduce a novel dynamical SIR model that incorporates nonlocal spatial motion for three distinct particle types, thereby bridging the gap between epidemiological theory and real-world mobility patterns. Risk of evolution driven population-wide emergence of mpox: The paradoxic effect of moderate interventions.

BACKGROUND: The global mpox outbreak in 2022 was declared a public health emergency of international concern. While in non-endemic countries disease spread remained limited mostly to a high risk group, a main public health concern is that through evolution, mpox gains the ability to widely spread in the entire population. METHODS: We construct a stochastic epidemiological model of SEIR type, to investigate the spread of mpox primarily propagating within a core population - consisting of MSM individuals having multiple sexual partners - before affecting the general population. We examine how e

Modeling Pathogen Dispersal in Marine Fish and Shellfish.

In marine ecosystems, oceanographic processes often govern host contacts with infectious agents. Consequently, many approaches developed to quantify pathogen dispersal in terrestrial ecosystems have limited use in the marine context. Recent applications in marine disease modeling demonstrate that physical oceanographic models coupled with biological models of infectious agents can characterize dispersal networks of pathogens in marine ecosystems. Biophysical modeling has been used over the past two decades to model larval dispersion but has only recently been utilized in marine epidemiology. I
