Safety of the tau-directed monoclonal antibody BIIB092 in progressive supranuclear palsy: a randomised, placebo-controlled, multiple ascending dose phase 1b trial.

BACKGROUND: Progressive supranuclear palsy is a rare neurodegenerative disease associated with dysfunctional tau protein. BIIB092 is a humanised monoclonal antibody that binds to N-terminal tau and is thus being assessed as a potential novel treatment for progressive supranuclear palsy. We aimed to investigate the safety and tolerability of BIIB092 in individuals with progressive supranuclear palsy. METHODS: This 12-week, double-blind, randomised, placebo-controlled, multiple ascending dose, phase 1b trial was done at 13 outpatient sites in the USA. Participants aged 41-86 years with probable

[Possibilities of traffic-injury prevention using seat belts and their disadvantages].

As a result of the increasing number of serious injuries, and/or deaths caused by automobile accidents, seatbelts, along with other safety items are installed in cars. The idea of having automobile seatbelts came from the aircraft seatbelt. In our country there is no law requiring installation of seatbelts in all automobiles. Most imported cars, however do have built-in seat builts which are more or less used by the drivers and passengers. Wearing seatbelts is a necessity because, according to statistics, of 100,000 registered vehicles, 340.8 drivers and/or passengers lose their lives. In Swed

Tolerability of the Oscar 2 ambulatory blood pressure monitor among research participants: a cross-sectional repeated measures study.

BACKGROUND: Ambulatory blood pressure monitoring (ABPM) is increasingly used to measure blood pressure (BP) in research studies. We examined ease of use, comfort, degree of disturbance, reported adverse effects, factors associated with poor tolerability, and association of poor tolerability with data acquisition of 24-hour ABPM using the Oscar 2 monitor in the research setting. METHODS: Sixty adults participating in a research study of people with a history of borderline clinic BP reported on their experience with ABPM on two occasions one week apart. Poor tolerability was operationalized as

Bruising characteristics discriminating physical child abuse from accidental trauma.

OBJECTIVE: Our objective was to conduct a pilot study to identify discriminating bruising characteristics and to model those findings into a decision tool for screening children at high risk for abuse. METHODS: A case-control study of children 0 to 48 months of age who were admitted to a PICU because of trauma was performed. Case subjects (N = 42) were victims of physical abuse, and control subjects (N = 53) were children admitted because of accidental trauma during the same time period. Bruising characteristics (total number and body region) and patient age were compared for children with ab

[Diagnostic assessment and treatment concepts for thoracic trauma].

Most patients with chest trauma can be successfully treated with tube thoracostomy and appropriate pain medication. Initial care of these patients is usually straightforward and performed by an emergency doctor or an emergency room surgeon, e.g. a general surgeon. If more extensive therapy of these polytraumatized patients appears to be required, tertiary care should be done in specialized centers or clinics with network structures. An appropriate structured network of surgical centers guarantees sufficient and efficient care of patients with severe chest trauma. In a best-case scenario the sp

Pseudo-entrapment of extraocular muscles in patients with orbital fractures.

Diplopia is a prominent finding in patients who have suffered orbital fractures. If the patient's double vision or ocular motility restriction was caused by soft tissue entrapment into the fracture site, surgery is frequently performed in order to release this entrapment and restore normal eye movement. However, the presence of diplopia should not necessarily be an indication for surgery. Brief case reports are hereby presented to illustrate that the symptoms of diplopia and motility restriction are not always attributable to the presence of orbital fractures that require surgical repair. NanoScript-Enabled Nonviral Transient Repression of Phosphatase and Tensin Homolog for Axonal Regeneration and Central Nervous System Injury Repair.

Spinal cord injury (SCI) remains a debilitating neurological disorder with limited therapeutic options, as existing treatments primarily address symptoms rather than address the complex interplay of cellular and molecular barriers to regeneration. These barriers collectively hinder functional recovery, including inhibitory glial scarring, chronic neuroinflammation, intrinsic neuronal regenerative deficits, and disruption of the blood-spinal cord barrier (BSCB). To address these limitations, we developed NanoScript-PTEN (NS-PTEN), a nonviral nanoparticle platform that delivers synthetic transcr

Endovascular repair for complicated Azzizadeh Grade III blunt traumatic aortic injury: a single-center cohort study of thoracic injuries with a separate abdominal subgroup.

BACKGROUND: The optimal management of Azzizadeh Grade III blunt traumatic aortic injury (BTAI) is debated, with a clinical dilemma persisting particularly for hemodynamically stable patients. This study evaluates a risk-stratified protocol guiding intervention versus surveillance in a cohort that included both stable and unstable patients. METHODS: We retrospectively analyzed consecutive patients with Grade III BTAI (2014-2023) managed per a multidisciplinary protocol: thoracic endovascular aortic repair (TEVAR) for complicated thoracic injuries, and endovascular aortic repair (EVAR) for comp

Establishment and External Validation of a Nomogram for Predicting In-Hospital Mortality in Patients With Maxillary Fractures Combined With Basilar Skull Fractures.

BACKGROUND: Maxillary fractures rarely cause death alone; when combined with basilar skull fractures, they indicate high-energy craniofacial trauma and are often accompanied by severe intracranial, airway, and systemic injuries. PURPOSE: The purpose of this study was to develop and validate a nomogram for predicting in-hospital mortality in patients with maxillary fractures combined with basilar skull fractures. STUDY DESIGN, SETTING, AND SAMPLE: A retrospective cohort study utilized a training set from the Medical Information Mart for Intensive Care IV (MIMIC-IV) database (US, 2008-2022) an

Distal Radial Fracture as an Early Marker of Bone Fragility: A Population-Based Study in Japan.

BACKGROUND: Distal radial fractures (DRFs) are common fragility fractures in older adults and often indicate skeletal deterioration. However, whether DRFs reflect underlying systemic frailty or occur predominantly in relatively active individuals with preserved physical function remains unclear. METHODS: This retrospective cohort study analyzed data from the Shizuoka Kokuho Database, which includes the health and long-term care (LTC) claims of 2.4 million residents. Individuals aged 65 to 89 years who were newly diagnosed with a DRF between 2012 and 2022 were compared with patients with an up

A Deployable Viscoelastic Coagulation Monitor Enables Point-of-Care Assessment of Coagulopathy in Swine With Polytrauma.

INTRODUCTION: Absence of pre-hospital coagulation tests challenges prompt management of hemostasis after trauma. The Viscoelastic Coagulation Monitor (VCM, Entegrion, Durham, NC) is a hand-held coagulation test for point-of-care. We evaluated VCM in a translational swine polytrauma model, hypothesizing that VCM correlates with a laboratory reference method, the TEG 5000 (Haemonetics, Boston, MA), and can identify coagulopathic phenotypes relevant to trauma. Our secondary hypothesis was that pre-warming of VCM disposable test cartridges using a heating plate versus pre-warming of cartridges by

Short- and long-term changes in neurological, behavioural, and blood biomarkers following repeated mild traumatic brain injury in rats-potential biological sex-dependent effects.

INTRODUCTION: Chronic traumatic encephalopathy (CTE) is a progressive neurodegenerative disease resulting from repeated mild traumatic brain injuries (rmTBI). The necessity for diagnosis of CTE, which can so far only be confirmed after post-mortem, is a pressing need. New approaches to early diagnose this disease are crucial to facilitate the translation of novel treatment strategies to the clinic. Several studies have found suitable candidate biomarkers, but the results are not straightforward. As biological sex is suggested to be a major confounding factor, we explored how sex influences beh
