Isolation and cultivation of a novel sulfate-reducing magnetotactic bacterium belonging to the genus Desulfovibrio.

Magnetotactic bacteria (MTB) synthesize magnetosomes composed of membrane-enveloped magnetite (Fe3O4) and/or greigite (Fe3S4) nanoparticles in the cells. It is known that the magnetotactic Deltaproteobacteria are ubiquitous and inhabit worldwide in the sediments of freshwater and marine environments. Mostly known MTB belonging to the Deltaproteobacteria are dissimilatory sulfate-reducing bacteria that biomineralize bullet-shaped magnetite nanoparticles, but only a few axenic cultures have been obtained so far. Here, we report the isolation, cultivation and characterization of a dissimilatory s

Hydrated silica exterior produced by biomimetic silicification confers viral vaccine heat-resistance.

Heat-lability is a key roadblock that strangles the widespread applications of many biological products. In nature, archaeal and extremophilic organisms utilize amorphous silica as a protective biomineral and exhibit considerable thermal tolerance. Here we present a bioinspired approach to generate thermostable virus by introducing an artificial hydrated silica exterior on individual virion. Similar to thermophiles, silicified viruses can survive longer at high temperature than their wild-type relatives. Virus inactivation assays showed that silica hydration exterior of the modified virus effe

Amelogenin in Enamel Tissue Engineering.

In this chapter the basic premises, the recent findings and the future challenges in the use of amelogenin for enamel tissue engineering are being discoursed on. Results emerging from the experiments performed to assess the fundamental physicochemical mechanisms of the interaction of amelogenin, the main protein of the enamel matrix, and the growing crystals of apatite, are mentioned, alongside a moderately comprehensive literature review of the subject at hand. The clinical importance of understanding this protein/mineral interaction at the nanoscale are highlighted as well as the potential f

Fungal transformations of uranium oxides.

The biogeochemical activities of free-living and symbiotic fungi must be acknowledged in attempts to understand uranium cycling and dispersal in the environment. Although the near-surface geochemistry of uranium is very complex and a wide variety of mineral phases is known, uranium trioxide (UO3) and triuranium octaoxide (U(3)O(8)) can be used as well characterized models in the study of biotransformations. We have used a complex methodological approach involving advanced solid state speciation and scanning electron microscopy to study the ability of saprotrophic, ericoid and ectomycorrhizal f

Calcite microcrystals in the pineal gland of the human brain: first physical and chemical studies.

A new form of biomineralization has been studied in the pineal gland of the human brain. It consists of small crystals that are less than 20 microm in length and that are completely distinct from the often observed mulberry-type hydroxyapatite concretions. A special procedure was developed for isolation of the crystals from the organic matter in the pineal gland. Cubic, hexagonal, and cylindrical morphologies have been identified using scanning electron microscopy. The crystal edges were sharp whereas their surfaces were very rough. Energy dispersive spectroscopy showed that the crystals conta

A Young ahsg/fetuin-a Inactive Retrocopy Reflects Recent Retrotransposon Activity in the Xenopus laevis Lineage.

The vertebrate ahsg (alpha 2-HS glycoprotein, also coined fetuin-a) homologs are highly expressed in the liver, and their secreted protein products exert complex systemic effects, including the regulation of biomineralization of soft and skeletal tissues. Here, we report a previously uncharacterized ahsg retrocopy in the allotetraploid frog species Xenopus laevis. We show that this young retrocopy was born from the ahsg.L homeologue less than 10 Mya, and landed in the S subgenome in a locus located between asic2.S and smarcd2.S. The ahsg.L-retrocopy ends with a poly(A) tail, is intronless, and

Hydrothermal Transformation of Eggshell Calcium Carbonate into Apatite Micro-Nanoparticles: Cytocompatibility and Osteoinductive Properties.

The eggshell is a biomineral consisting of CaCO3 in the form of calcite phase and a pervading organic matrix (1-3.5 wt.%). Transforming eggshell calcite particles into calcium phosphate (apatite) micro-nanoparticles opens the door to repurposing the eggshell waste as materials with potential biomedical applications, fulfilling the principles of the circular economy. Previous methods to obtain these particles consisted mainly of two steps, the first one involving the calcination of the eggshell. In this research, direct transformation by a one-pot hydrothermal method ranging from 100-200 °C was

Fibroblast-like synoviocytes induce calcium mineral formation and deposition.

Calcium crystals are present in the synovial fluid of 65%-100% patients with osteoarthritis (OA) and 20%-39% patients with rheumatoid arthritis (RA). This study sought to investigate the role of fibroblast-like synoviocytes (FLSs) in calcium mineral formation. We found that numerous genes classified in the biomineral formation process, including bone gamma-carboxyglutamate (gla) protein/osteocalcin, runt-related transcription factor 2, ankylosis progressive homolog, and parathyroid hormone-like hormone, were differentially expressed in the OA and RA FLSs. Calcium deposits were detected in FLSs

PROTEIN TEMPLATES IN HARD TISSUE ENGINEERING.

Biomineralization processes such as formation of bones and teeth require controlled mineral deposition and self-assembly into hierarchical biocomposites with unique mechanical properties. Ideal biomaterials for regeneration and repair of hard tissues must be biocompatible, possess micro and macroporosity for vascular invasion, provide surface chemistry and texture that facilitate cell attachment, proliferation, differentiation of lineage specific progenitor cells, and induce deposition of calcium phosphate mineral. To expect in-vivo like cellular response several investigators have used extrac

Silver ion incorporation and nanoparticle formation inside the cavity of Pyrococcus furiosus ferritin: structural and size-distribution analyses.

Highly symmetrical protein cage architectures from three different iron storage proteins, heavy and light human ferritin chains (HuHFt and HuLFt) and ferritin from the hyperthemophilic bacterium Pyrococcus furiosus (PfFt), have been used as models for understanding the molecular basis of silver ion deposition and metal core formation inside the protein cavity. Biomineralization using protein cavities is an important issue for the fabrication of biometamaterials under mild synthetic conditions. Silver nanoparticles (AgNPs) were produced with high yields within PfFt but not within HuHFt and HuLF

Expression of the small leucine-rich proteoglycan osteoadherin/osteomodulin in human dental pulp and developing rat teeth.

Because the extracellular matrices of dentin and bone are composed mainly of type I collagen, their characteristics are determined by the nature of noncollagenous proteins (NCPs). Among these NCPs, some proteoglycans (PGs) belong to the small leucine-rich proteoglycans (SLRPs). Recently, osteoadherin (OSAD) has been described as a new member of this family, that is expressed by mature bovine osteoblasts. Here, we report the expression of OSAD messenger RNA (mRNA) in human dental tissues and during the development of rat molars, using in situ hybridization. For this purpose, we constructed a pr

Tumor necrosis factor-α promotes Staphylococcus aureus-induced osteomyelitis through downregulating endothelial nitric oxide synthase.

BACKGROUND: Infections of Staphylococcus aureus (S. aureus) often result in osteomyelitis, which is the acute or chronic infections of the bone marrow or bones. TNF-α is long recognized as a key factor contributing to the pathogenesis of osteomyelitis, but little is known about the underlying molecular mechanism. METHODS: Expression levels of TNF-α, and several candidate genes, including endothelial nitric oxide synthase (eNOS), known to be downregulated by TNF-α were analysed in MC3T3-E1 cells with S. aureus infection and osteomyelitis patient blood. MicroRNA(miR)-129-5p was predicted and ex

Microstructural Anisotropy in Bone-Mimetic Silk Constructs Promotes Self-Mineralization.

Understanding how organic matrix architecture regulates biomineralization remains a central challenge in mineralized tissue regeneration. Emerging evidence highlights the importance of coupled phenomena, including nanoconfinement, ionic and pH regulation, and hierarchical structural remodeling, in directing mineral formation. Silk fibroin (SF) is a versatile structural protein widely used in biomaterials due to its tunable mechanics, scalable processing into micro- and nanoscale constructs, controllable biodegradation, and ability to support cell infiltration and tissue integration, including

Zinc-selenium synergistic nanoplatform for augmented cancer immunotherapy via trace-elements-mediated immunomodulation.

Essential trace elements (ETEs) are crucial nutrients in maintaining the immune function of the body. Designing nanomedicines based on ETEs has become an emerging strategy for enhancing immunotherapy by utilizing the metabolism of constituent ETEs and their immunomodulatory functions. However, their medical applications are challenged by the dosage-dependent balance between therapeutic necessity and toxicity. The narrow safety zones of ETEs pose great challenges for high efficacy without exceeding strict safety thresholds. Nanomedicinal strategies based on multiple ETEs hold promising potentia

Black phosphorus-loaded mineralized UV-responsive chitosan hydrogel for enhanced osteogenesis and antibacterial activity.

Favorable osteogenesis and effective antibacterial activity are two essential requirements for ideal scaffolds in bone tissue engineering, particularly for infected bone defects. Herein, we developed a novel hydrogel system based on mineralized ultraviolet-responsive methacrylated chitosan hydrogel (CSMA) integrated with black phosphorus (BP) nanosheets exhibiting a photothermal effect. BP was incorporated into CSMA and subsequently subjected to in situ biomineralization to form a BP-loaded mineralized hydrogel (BP@CSMA/CaCO3). The physicochemical and biological properties of BP@CSMA/CaCO3 wer

Reproductive aging drives deterministic microbiota assembly to mitigate uterine oxidative phosphorylation impairment via spermidine production in laying hens.

BACKGROUND: Reproductive aging represents a critical physiological bottleneck characterized by a progressive decline in tissue homeostasis and physiological function. While the gut microbiota is known to shift during host aging, the ecological forces governing the assembly of the reproductive microbiota and its functional feedback on uterine homeostasis remain poorly understood. RESULTS: We demonstrated that uterine aging drives a transition from stochastic to deterministic microbial community assembly, selecting for a microbiota enriched in Rhodococcus in aged laying hens. A Multicolor 3D-STORM High-Resolution Visualization Protocol for Collagen Mineralization in Self-Assembled Recombinant Type I Collagen Fibrils.

This protocol describes a multicolor three-dimensional stochastic optical reconstruction microscopy (3D-STORM) method for nanoscale visualization of collagen mineralization in a recombinant type I collagen self-assembled fibril model. The method enables simultaneous imaging of collagen, non-collagenous proteins (e.g., chondroitin sulfate), and calcium phosphate mineral phases. Sample preparation involves amino‑silanization and collagen self‑assembly, followed by mineralization using a calcium phosphate medium that forms amorphous calcium phosphate (ACP) at an early stage (30 min) and matures i

Synthesis and functionalization of scalable and versatile 2D protein films via amyloid-like aggregation.

Two-dimensional (2D) protein films can be used to modify the properties of surfaces, and find applications predominantly in the fields of biomaterials, lithography, optics and electronics. However, it is difficult to produce scalable homogeneous and robust protein films with an easy, low-cost, green and efficient method. Further challenges include encapsulating and releasing functional building blocks in the film without inactivating them, and maintaining or improving the bioactivities of proteins used for the formation of the films. Here we detail the process to prepare large 2D protein films
