Land use regression (LUR) and dispersion/chemical transport models (D/CTMs) are frequently applied to predict exposure to air pollution concentrations at a fine scale for use in epidemiological studies. Moreover, the use of satellite aerosol optical depth data has been a key predictor especially for particulate matter pollution and when studying large populations. Within the STEAM project we present a hybrid spatio-temporal modeling framework by (a) incorporating predictions from dispersion modeling of nitrogen dioxide (NO2), ozone (O3) and particulate matter with an aerodynamic diameter equal

The emergence of mosquito-transmitted viruses poses a global threat to human health. Combining mechanistic epidemiological models based on temperature-trait relationships with climatological data is a powerful technique for environmental risk assessment. However, a limitation of this approach is that the local microclimates experienced by mosquitoes can differ substantially from macroclimate measurements, particularly in heterogeneous urban environments. To address this scaling mismatch, we modeled spatial variation in microclimate temperatures and the thermal potential for dengue transmission

While obesity is generally associated with unfavorable health outcomes, there are notable exceptions. In cancer immunotherapy, epidemiological studies consistently link obesity to improved checkpoint inhibitor responses. Desharnais et al. show in mouse models that this paradox is driven by diet-dependent interactions with the gut microbiome, not obesity itself.

The correlation between cholera epidemics and climatic drivers, in particular seasonal tropical rainfall, has been studied in a variety of contexts owing to its documented relevance. Several mechanistic models of cholera transmission have included rainfall as a driver by focusing on two possible transmission pathways: either by increasing exposure to contaminated water (e.g. due to worsening sanitary conditions during water excess), or water contamination by freshly excreted bacteria (e.g. due to washout of open-air defecation sites or overflows). Our study assesses the explanatory power of th

Age-related neurodegenerative diseases such as Alzheimer's disease (AD) and Parkinson's disease (PD) are associated with substantial mortality and morbidity. The etiology and pathogenesis of these diseases are not fully understood, which impedes the development of new therapies. Emerging epidemiological evidence has revealed that environmental toxicants substantially contribute to the pathogenesis of diseases through mechanisms involving compromised endogenous detoxification systems. This review synthesizes epidemiological and clinical evidence from human studies elucidating the crucial role o

Understanding the spatio-temporal spread of infectious diseases is important to improve control, yet most studies lack granular epidemiological data and are complicated by heterogeneities in pre-existing immunity. The COVID-19 pandemic provides a unique opportunity to study the invasion of a novel pathogen in a naive population. Here we apply correlation statistics and mathematical modelling to daily city-level COVID-19 case data between 2020 and 2022 to dissect the spatial diffusion of different virus variants across the United States. We find that between-city correlation in case incidences

Mpox, originating primarily in African rodents, has led to human outbreaks over recent years. This study presents a mathematical model for Mpox, distinguishing between individuals with and without HIV who are susceptible. We explore scenarios involving both rodent-to-human transmission and those without it. In the absence of this transmission route, the model undergoes a backward bifurcation, suggesting that reducing the basic reproduction number below one would not eliminate the disease unless further control strategies are used. With the account of rodent-to-human transmission, if Mpox is en

Seasonal disease outbreaks are common in many infectious diseases such as seasonal influenza, Zika, dengue fever, Lyme disease, malaria, and cholera. Seasonal outbreaks are often due to weather patterns affecting pathogens or disease-carrying vectors or by social behavior. We investigate disease emergence and extinction in seasonal stochastic epidemic models. Specifically, we study disease emergence through seasonally varying parameters for transmission, recovery, and vector births and deaths in time-nonhomogeneous Markov chains for SIR, SEIR, and vector-host systems. A branching process appro

The current global initiative to eliminate lymphatic filariasis is a major renewed commitment to reduce or eliminate the burden of one of the major helminth infections from resource-poor communities of the world. Mathematical models of filariasis transmission can serve as an effective tool for guiding the scientific development and management of successful community-level intervention programmes by acting as analytical frameworks for integrating knowledge regarding parasite transmission dynamics with programmatic factors. However, the power of these tools for supporting control interventions w

Otitis media (OM) represents a complex set of clinically defined disease phenotypes. Epidemiological and pathological data suggest that inflammation of the middle ear occurs on a continuum of disease. Here I review epidemiological data and apply mathematical modelling to better define the interrelation of OM phenotypes, including the relationship of upper respiratory tract infection to acute OM (AOM), of AOM to OM with effusion, and of recurrent OM to chronicity of effusion. I use these mathematical models to represent OM graphically as a 'landscape of disease'. This novel nosology may enable

Dengue is an arboviral disease of high public health relevance, characterized by pronounced temporal variability, nonlinearity, and recurrent outbreaks, which pose challenges to epidemiological surveillance and decision-making. This study evaluated the performance of machine learning methods for short-term forecasting of the weekly dengue morbidity rate in the 27 Brazilian capital cities, comprising the 26 state capitals and Brasília, Federal District, with horizons up to 4 weeks. Epidemiological, climatic, and socioeconomic data were compiled for these capital cities and used to compare a Gat

Modelling is a useful tool for the understanding and hence, potentially, control of disease. In an emergency situation, obtaining data to validate detailed models may be difficult. However, modelling can be useful in aiding the management of disease, if the objectives are not too ambitious. For example, import risk analysis can use models to minimise the risk of outbreaks. Modelling can also be used to maximise the power of surveillance data to ensure that any outbreak is detected as rapidly as possible. Methods of modelling spread of disease, and measures to prevent this, including SIR-style

Mathematical models of viral transmission and control are important tools for assessing the threat posed by deliberate release of the smallpox virus and the best means of containing an outbreak. Models must balance biological realism against limitations of knowledge, and uncertainties need to be accurately communicated to policy-makers. Smallpox poses the particular challenge that key biological, social and spatial factors affecting disease spread in contemporary populations must be elucidated largely from historical studies undertaken before disease eradication in 1979. We review the use of m

BACKGROUND & OBJECTIVES: New diagnostic tests for tuberculosis, especially those based on nucleic acid amplification, offer the possibility of early and accurate diagnosis of active TB. In this study we use mathematical modelling to explore the potential epidemiological impact of these new tests, with particular reference to India.
METHODS: A behavioural model of patient-doctor interactions embedded in an epidemiological model of Mycobacterium tuberculosis transmission, linked to field data, was used to investigate the effects of early diagnosis in preventing future TB cases.
RESULTS: New di

Epidemiological studies suggest that about 3.5% of the adult United States population meet criteria for panic disorder, and that an additional 2-5% suffer from agoraphobia with or without panic. Both biological and behavioral models have been proposed to elucidate the nature of this disorder. The authors review research findings regarding these models, as well as pharmacological and cognitive-behavioral treatment approaches. The complexity of factors contributing to the disorder suggests the need for an integrated treatment approach involving considerations of combination drug therapy and psyc

BACKGROUND: Hemorrhagic fever with renal syndrome (HFRS) is a severe natural focal infectious disease that remains a major public health concern in Shandong Province, China. Understanding the epidemiological characteristics of HFRS and forecasting its incidence are crucial to provide guidance for policymaking and implement timely interventions against this disease.
OBJECTIVE: This study examined the long-term epidemiological characteristics of HFRS based on 15 years of surveillance data; the analysis used 4 time series models to predict its incidence trends. The aim was to provide a scientifi

Routine use of genetic data in healthcare is much-discussed, yet little is known about its performance in epidemiological models including traditional risk factors. Using severe COVID-19 as an exemplar, we explore the integration of polygenic risk scores (PRS) into disease models alongside sociodemographic and clinical variables. PRS were optimized for 23 clinical variables and related traits previously-associated with severe COVID-19 in up to 450,449 UK Biobank participants, and tested in 9,560 individuals diagnosed in the pre-vaccination era. Associations were further adjusted for (i) sociod

BACKGROUND: Epidemiological studies of disease exposure risk are frequently based on observational, cross-sectional data, and use statistical approaches as crucial tools for formalising causal processes and making predictions of exposure risks. However, an acknowledged limitation of traditional models is that the inferred relationships are correlational, cannot easily distinguish direct from indirect determinants of disease risk, and are often considerable simplifications of complex interrelationships. This may be particularly important when attempting to infer causality in patterns of co-infe

OBJECTIVES: The taxonic versus dimensional status of mood symptoms has been the subject of debate among mental health professionals. Conventional diagnostic models suggest that mood disorders are categorical; however, the inability of categorical models to adequately account for subthreshold unipolar and bipolar presentations and the heterotypic continuity of symptoms in unipolar and bipolar cases has resulted in growing support for dimensional views. The current study sought to evaluate the relative viabilities of categorical and dimensional models of mood symptoms within a taxometric framewo

A coupled epidemic-behavior model is proposed to assess how vaccination, testing, and treatment jointly influence early outbreak control, accounting for individual choices based on perceived risk, intervention costs, and policy incentives. In a uniformly mixing population, individuals make strategic decisions regarding vaccination and testing participation based on the trade-offs between personal costs and communal benefits, while the intensity of public health testing adjusts according to infection prevalence; the transmission model includes vaccine efficacy, the probability of false negative
