[pH-dependence of kinetic parameters in the superprecipitation reaction and ATPase activity of myometrial actomyosin].

The investigation of pH-dependence of superprecipitation reaction and ATPase activity of myometrium actomyosin in the interval of pH 5.5-8.0 has detected cupola-shaped curves with maximal activity of both processes by pH 6.5. On the basis of calculating the constants of ionization it was supposed that in the case of actomyosin ATPase imidazole groups of two histidins had an essential role in reaction of ATP hydrolysis and in superprecipitation process--imidazol group of histidine and carboxyl group of asparagin acid. The investigation of [ATP]- and [Mg2+]-dependence of superprecipitation react

Changes in morphology of human skin fibroblasts induced by local anaesthetics: role of actomyosin contraction.

Local anaesthetics block action potentials in the membranes of excitable cells but their effects on non-excitable cells are less well known. Some local anaesthetics are applied directly onto the skin, and for this reason the effect of procaine (p-aminobenzoic acid diethylamino-etyl ester hydrochloride) and tetracaine (4-[butylamino]benzoic acid 2-[dimethylamino]ethyl ester) upon the morphology and cytoskeleton organisation of human skin fibroblasts was investigated. The time lapse video recording of fibroblasts cultured in serum-enriched medium revealed that the cells rapidly change shape afte

Coordinated regulation of diverse F-actin organizations orchestrates F-actin pulses in the actomyosin network.

Pulsatile actomyosin networks emerge as a widespread mechanism driving tissue morphogenesis. Compared with myosin-II pulses, F-actin pulses are largely unexplored. By studying Drosophila oogenesis, we report that basal pulsatile actomyosin networks consist of F-actin bundle and meshwork architectures, implicating the existence of different F-actin microstructures and corresponding nucleators. Here, Rac1 and the Scar/WAVE-Arp2/3 complex (branched F-actin nucleator), exhibiting constant levels, are necessary to support F-actin pulsation, whereas pulses of Dia (unbranched F-actin nucleator) trigg

APOL1 C-Terminal Variants May Trigger Kidney Disease through Interference with APOL3 Control of Actomyosin.

The C-terminal variants G1 and G2 of apolipoprotein L1 (APOL1) confer human resistance to the sleeping sickness parasite Trypanosoma rhodesiense, but they also increase the risk of kidney disease. APOL1 and APOL3 are death-promoting proteins that are partially associated with the endoplasmic reticulum and Golgi membranes. We report that in podocytes, either APOL1 C-terminal helix truncation (APOL1Δ) or APOL3 deletion (APOL3KO) induces similar actomyosin reorganization linked to the inhibition of phosphatidylinositol-4-phosphate [PI(4)P] synthesis by the Golgi PI(4)-kinase IIIB (PI4KB). Both AP

α-catenin phosphorylation is actomyosin-sensitive and required for epithelial barrier functions through Afadin.

Zonula adherens junctions (zAJ) are spatially proximal to tight junctions (TJ), in a superstructure known as the apical junctional complex (AJC). A key component of the AJC is a circumferential ring of filamentous (F)-actin, but how actomyosin contractility drives AJC structure and epithelial barrier function is incompletely understood. Here, we show that a central mechanosensitive component of zAJ, α-catenin (α-cat), undergoes force-dependent phosphorylation in an unstructured linker region. This modification in turn primes the α-cat mechanosensitive Middle-region for effector-binding. We cre

Nonlinear Elastic Bottlebrush Polymer Hydrogels Modulate Actomyosin Mediated Protrusion Formation in Mesenchymal Stromal Cells.

The nonlinear elasticity of many tissue-specific extracellular matrices is difficult to recapitulate without the use of fibrous architectures, which couple strain-stiffening with stress relaxation. Herein, bottlebrush polymers are synthesized and crosslinked to form poly(ethylene glycol)-based hydrogels and used to study how strain-stiffening behavior affects human mesenchymal stromal cells (hMSCs). By tailoring the bottlebrush polymer length, the critical stress associated with the onset of network stiffening is systematically varied, and a unique protrusion-rich hMSC morphology emerges only

Cardiac actin changes in the actomyosin interface have different effects on myosin duty ratio.

Hypertrophic cardiomyopathy (HCM) is an inherited cardiovascular disease (CD) that commonly causes an increased size of cardiomyocytes in the left ventricle. The proteins myosin and actin interact in the myocardium to produce contraction through the actomyosin ATPase cycle. The duty ratio (r) of myosin is the proportion of the actomyosin ATPase cycle that myosin is bound to actin and does work. A common hypothesis is that HCM mutations increase contraction in cardiac sarcomeres; however, the available data are not clear on this connection. Based on previous work with human α-cardiac actin (ACT

Actomyosin forces and the energetics of red blood cell invasion by the malaria parasite Plasmodium falciparum.

All symptoms of malaria disease are associated with the asexual blood stages of development, involving cycles of red blood cell (RBC) invasion and egress by the Plasmodium spp. merozoite. Merozoite invasion is rapid and is actively powered by a parasite actomyosin motor. The current accepted model for actomyosin force generation envisages arrays of parasite myosins, pushing against short actin filaments connected to the external milieu that drive the merozoite forwards into the RBC. In Plasmodium falciparum, the most virulent human malaria species, Myosin A (PfMyoA) is critical for parasite re

[Combined effect of Pb2+ and divalent mental complexes on superprecipation and ATPase activity of myometrial actomyosin].

The inhibiting effect of Pb2+ (0.1-1.0 mM) on superprecipitation and ATPase activity of myometrium actomyosin has been shown. It is found out that of 11 studied chelators of bivalent cations only monosodium salt of 4-guanidino-1-hydroxybutyliden-1.1-bisphosphonic acid can efficiently renews the value of superprecipitation and ATPase activity of actomyosin in presence of lead ions. It is supposed that this renewal is underlied by binding of guanidin group of Pb ions.

Sudden death case of single coronary artery with special reference to the effect of ischemia on actomyosin.

A normal single left coronary artery was found in a high school boy who died suddenly after Rugby football practice. He had an infarct lesion in the posterior wall where there was no perfusion because of the absence of the right coronary artery. There was no significant lesion in the anterior wall except for contraction band necrosis. Myoglobin diffusion paralleled the extent of necrosis. The case was useful in evaluating the effects of ischemia on a human subject because variations due to personality and postmortem changes were not involved. Actomyosin ATPase activity was not different among

[Effect of heavy metal ions on superprecipitation and ATPase activity of uterine smooth muscle actomyosin activity].

The inhibiting effect of Pb2+, Zn2+ and Cd2+ on Mg(2+)-dependent superprecipitation and ATPase activity of myometrium actomyosin. The inhibiting effect of heavy metals cations on the both processes satisfies the succession: Pb2+ > Zn2+ > Cd2+. Cadmium and zinc ions in concentration of 1 mM stimulate the initial velocity (v0) of Mg(2+)-dependent superprecipitation by 25% and 80%, respectively, while the lead ions under the same concentration inhibit the initial velocity by 40%. It is possible that these results evidence for the direct effect of ions of heavy metals on active-myosin interaction

[Effect of temperature on superprecipitation of actomyosin of myometrial smooth muscle and reactions of ATP hydrolysis catalyzed by this protein].

The experiments carried out on the preparation of myometrium actomyosin were carried out. The reaction of superprecipitation of this protein and ATP hydrolysis were studied as affected by temperature. For the both processes the value of temperature optimum is 45 degrees C. The value of activation energy Ea is equal to 20 +/- 3 and 23 +/- 3 kJ/mol for the reaction of superprecipitation and ATP hydrolysis, respectively. Preincubation of actomyosin (45-60 degrees C, 1-5 min) in the medium not including ions of Ca, Mg and ATP with following introduction of the components to this medium and ten-min

Protocol to identify mechanosensitive nuclear proteins using tunable actomyosin contractility and proximity biotinylation in mammalian cells.

Mechanical forces influence a range of cellular behaviors; however, how these forces are sensed and converted into biochemical changes remains incompletely understood. A key aspect of mechanotransduction is the regulation of subcellular protein localization. Here, we present a protocol describing the engineering of cell lines with tunable actomyosin contractility combined with a proximity biotinylation strategy confined to the nucleus followed by liquid chromatography-tandem mass spectrometry (LC-MS/MS) analysis. This approach allows the identification of proteins whose nuclear localization is

TGF-β signaling modulates adhesion and actomyosin contractility to pattern epithelial permeability in Drosophila ovaries.

Epithelial morphogenesis and homeostasis depend on dynamic remodeling of cell-cell junctions. Tricellular junctions (TCJs) control epithelial permeability and plasticity, yet how TCJs are remodeled remains unclear. In the Drosophila ovarian follicular epithelium, TCJs open transiently in a process called patency to allow passage of yolk proteins for uptake by the oocyte. Here, we investigated how a TGF-β signaling gradient represses patency along the follicular epithelium. We show that TGF-β signaling blocks patency cell-autonomously by strengthening E-Cadherin (E-Cad, also known as Shg)-based

InsP3R signaling and actomyosin-dependent mitochondrial dynamics play essential roles in mitochondrial stress-induced longevity.

Certain forms of mitochondrial impairment confer longevity, while disease-associated mitochondrial dysfunction triggers pathogenesis. The adaptive pathways that distinguish benefit from pathology remain unclear. Here we reveal that longevity induced by mitochondrial Complex I/nuo-6 mutation in C. elegans is dependent on the endoplasmic reticulum (ER) Ca2+ channel, InsP3R. To explain this connection, we test multiple candidate links between Ca2+ and mitochondrial homeostasis previously established in vitro, including mitochondrial calcium uniporter (MCU)-dependent stimulation of respiration and

Nuclear compression overrides matrix softness to regulate stem cell fate through actomyosin-driven YAP mechanotransduction.

Mechanical cues play a pivotal role in regulating stem cell fate, yet how cells integrate compressive forces in compliant microenvironments remains poorly understood. Here, we show that nuclear compression overrides soft matrix cues to induce osteogenic differentiation in mesenchymal stem cells through an integrin/vinculin-independent, cytoskeleton-mediated mechanism. Using a microfabricated polydimethylsiloxane (PDMS) confiner, we applied defined vertical confinement (3 and 10 μm) to mesenchymal stem cells (MSCs) cultured on substrates of varying stiffness. On soft substrates, compression tri

Quality improvement of ready-to-eat chicken breast after two-stage heating: The role of actomyosin dissociation.

Preventing quality deterioration in ready-to-eat (RTE) sauce-braised chicken breast during high-temperature sterilization (121 °C for 10 min) remains challenging. Herein, we propose a new strategy to mitigate quality deterioration induced by two-stage heating (cooking and high-temperature sterilization) by regulating actomyosin dissociation through adjustment of the cooking temperature (50-98 °C). The results indicated that samples treated at 50 °C exhibited the highest level of actin released from actomyosin compared to those treated at 98 °C. This was accompanied by a reduction in actomyosin

Reconstitution of actomyosin networks in cell-sized liposomes dissects distinct mechanisms of membrane blebbing and symmetry breaking.

The actin cortex, a thin layer of actomyosin network beneath the plasma membrane, regulates various cell functions by generating active forces and inducing membrane deformations, including blebs. Although upstream signaling is involved in regulating cell shape, the extent to which downstream actomyosin molecules can control the shape remains elusive. Here, using a minimal reconstituted system combined with an agent-based computational model, we show that actin-membrane coupling strength determines the magnitude of membrane deformation, while its balance with actin network connectivity governs
