Raman microscopy as a tool to study changes in chemical composition upon structural differentiation of Ambystoma embryo.

Ambystoma mexicanum is often used in regeneration studies because it reaches sexual maturity as aquatic larvae within 360 hours, and it can regenerate its appendages. In developing a spectroscopic approach for regeneration studies, in this contribution, we show that Raman confocal microscopy can be used as a diagnostic tool to describe the chemical composition of axolotl embryonal tissues at different stages of development. Experimental observations, supported by quantum chemistry studies on molecules reported or considered to be present in the tissues of the living organism, show that in the

Ultrasound description of the coelomic cavity of the axolotl (Ambystoma mexicanum) in a clinically healthy population: a pilot study.

Axolotls (Ambystoma mexicanum) are extensively studied for their relevance in human medical research. Despite being critically endangered in the wild, they have gained popularity as household pets. Although they have been kept in captivity for over a century, detailed descriptions of their coelomic organ anatomy remain limited. Also, this species exhibits significant variations compared to other amphibians. Ultrasound is a non-invasive and painless medical imaging technique, ideally suited for investigating internal organs or structures. This study focused on describing the ultrasound appearan

Neurotrophic regulation of epidermal dedifferentiation during wound healing and limb regeneration in the axolotl (Ambystoma mexicanum).

Adult urodeles (salamanders) are unique in their ability to regenerate complex organs perfectly. The recently developed Accessory Limb Model (ALM) in the axolotl provides an opportunity to identify and characterize the essential signaling events that control the early steps in limb regeneration. The ALM demonstrates that limb regeneration progresses in a stepwise fashion that is dependent on signals from the wound epidermis, nerves and dermal fibroblasts from opposite sides of the limb. When all the signals are present, a limb is formed de novo. The ALM thus provides an opportunity to identify

Immunocytochemical localization and immunochemical characterization of an insulin-related peptide in the pancreas of the urodele amphibian, Ambystoma mexicanum.

The pancreas of the axolotl, Ambystoma mexicanum, was investigated by immunocytochemical methods for the presence of immunoreactivity to a number of antisera raised against mammalian insulins. All anti-insulin antisera tested revealed substantial amounts of reaction products confined solely to the aldehyde-fuchsinophilic B cells of the endocrine pancreas. The reactive cell population was detected by use of one polyclonal antiserum against bovine insulin and eight different monoclonal antibodies against insulins from various mammalian species. Six of these antibody clones have known specificity

Lectin-binding pattern on the surface epidermis of Ambystoma tigrinum larvae. A light- and electron-microscopic study.

The surface epidermis of Ambystoma tigrinum larvae was examined at the light- and electron-microscope levels using five different lectin conjugates as probes for the detection of sugar residues on the cell membranes. Concanavalin A (Con-A), wheat-germ agglutinin (WGA), Ricinus communis agglutinin I (RCA-I), Dolichos biflorus agglutinin and soybean agglutinin (SBA) conjugates clearly labelled the surface cells, especially their apical surfaces. At electron microscopy, the labelling on plasma membranes was found to exhibit regional differences. Among the lectins tested WGA displayed a particular

Short-term storage of tiger salamander (Ambystoma tigrinum) spermatozoa: The effect of collection type, temperature and time.

The aims of this project were to characterize tiger salamander (Ambystoma tigrinum) spermatozoa motility over time, when excreted as either milt or spermic urine prior to packaging into a spermatophore, and to determine the effect of temperature on sperm motility. A split-plot design was utilized to assess the motility of the two pre-spermatophore sample types at two temperatures, 0°C and 20°C (n = 10 for each treatment). Spermiation was induced through exogenous hormone treatment of luteinizing hormone releasing hormone analog in order to collect both milt and spermic urine, which were evalua

Leucine-enkephalin-like immunoreactivity is localized in luteinizing hormone-producing cells in the axolotl (Ambystoma mexicanum) pituitary.

In this study, we used immunohistochemical techniques to determine the cell type of leucine-enkephalin (Leu-ENK)-immunoreactive cells in the axolotl (Ambystoma mexicanum) pituitary. Immunoreactive cells were scattered throughout the pars distalis except for the dorso-caudal portion. These cells were immuno-positive for luteinizing hormone (LH), but they were immuno-negative for adrenocorticotrophic, growth, and thyroid-stimulating hormones, as well as prolactin. Immunoelectron microscopy demonstrated that Leu-ENK-like substance and LH co-localized within the same secretory granules. Leu-ENK se

Tooth development in Ambystoma mexicanum: phosphatase activities, calcium accumulation and cell proliferation in the tooth-forming tissues.

Prerequisites of tooth formation, cell proliferation in the tooth-forming tissues, calcium accumulation and the enzymatic activities of alkaline (ALP) and acid phosphatases (ACP) were investigated by immunohistochemical and histochemical methods in various developmental stages of the Mexican Axolotl, Ambystoma mexicanum. During the growth of replacement teeth, the tooth-forming tissues continually recruit cells from the surrounding regions. The basal layer of the oral epithelium, the dental lamina and sometimes even the outer enamel epithelium provide cells for the differentiated inner enamel

Preliminary Experience Using Axolotl (Ambystoma mexicanum) Dermis Patches as a Biologic Agent for Wound Management After Neurosurgical Procedures.

OBJECTIVE: In the United States, chronic wounds affect more than 6.5 million people annually-a mean cost of $23,755 among neurosurgery patients. Current wound management solutions have disadvantages, including rejection, disease transmission from mammalian sources, and cultural issues prohibiting some products. Here, we describe preliminary use of xenograft tissue derived from axolotl (Ambystoma mexicanum) dermis for use in wound management after neurosurgical procedures. METHODS: Our prospective database was retrospectively searched for consecutive patients who underwent wound closures using

Establishment of a Practical Sperm Cryopreservation Pathway for the Axolotl (Ambystoma mexicanum): A Community-Level Approach to Germplasm Repository Development.

The axolotl (Ambystoma mexicanum) draws great attention around the world for its importance as a biomedical research model, but housing and maintaining live animals is increasingly expensive and risky as new transgenic lines are developed. The goal of this work was to develop an initial practical pathway for sperm cryopreservation to support germplasm repository development. The present study assembled a pathway through the investigation of axolotl sperm collection by stripping, refrigerated storage in various osmotic pressures, cryopreservation in various cryoprotectants, and in vitro fertili

Epizootic of Clinostomum marginatum (Trematoda: Clinostomidae) in Ambystoma tigrinum from Colorado, USA: Investigation through Genomics, Histopathology, and Noninvasive Imagery.

Trematodes in the genus Clinostomum develop into large metacercariae that can sometimes achieve high intensity in their second intermediate hosts, potentially causing pathology. In 2022, there was 100% (15/15) infection prevalence of Clinostomum marginatum, with a mean of 3,125 metacercariae per salamander (range: 279-4,075) and a median of 2,949. Dissection and histopathology results indicated that C. marginatum was found in nearly all body tissues and organs of the salamanders. Parasitic infection was closely associated with chronic inflammation and fibrosis around the cysts, with heavily in

Abnormal limb regeneration in the short-toes mutant of the axolotl, Ambystoma mexicanum: studies of age, level of amputation, and extracellular matrix.

Limb regeneration in the short-toes axolotl is impaired. Our goal was to characterize the regeneration process in this mutant by histological and immunocytochemical methods. Previous research indicates that age and a defective basement membrane may be instrumental factors in short-toes axolotl regeneration (Del Rio-Tsonis et al. [1992] Proc. Natl. Acad. Sci. U.S.A., 89:5502-5506). The present results show that limb regeneration can occur even in older (1-2-year-old) short-toes axolotls. The process was always significantly delayed, but the time required for complete regeneration varied. Even s

Analysis of the three-dimensional distributions of alpha-actinin, ankyrin, and filamin in developing hearts of normal and cardiac mutant axolotls (Ambystoma mexicanum).

alpha-Actinin is an actin binding protein that assists in the stabilization of the plasma membrane and helps to fix organelles in position in a variety of cell types. In muscle, it is a major component of the Z-lines of organized myofibrils. Ankyrin binds to various elements of the cytoskeletal system including microtubules, microfilaments, and intermediate filaments and may help to anchor these structures to the cell membrane. Filamin is a well-characterized actin-associated protein first isolated from chicken smooth muscle. In addition, filamin is a gel-forming protein which aids in the form

Changes to Skin Microbiome Community Dynamics Support Resistance to Enzootic and Epizootic Strains of Batrachochytrium dendrobatidis (Bd) in the Spotted Salamander (Ambystoma maculatum).

Global spread of animal pathogens has contributed to species declines and extinctions. In regions where a particular disease is enzootic, pathogen inhibition may arise through protection provided by host-associated microbiomes. Amphibians skin microbiomes can inhibit growth of the fungal pathogen Batrachochytrium dendrobatidis (Bd), preventing emergence of disease through a range of microbe-mediated antifungal mechanisms, allowing hosts to resist Bd infection. However, it remains unclear how skin microbiomes may shift in community composition or structure following infection by different Bd st

A scanning electron microscopy and histological study on the effects of the mutant eyeless (e/e) gene upon the hypothalamus in the Mexican axolotl Ambystoma mexicanum Shaw.

A scanning electron microscopy, histological, and immunochemical investigation examined the effects of the mutant gene (e) upon hypothalamic development in the Mexican axolotl. The adult eyeless mutant is sterile. Previous studies indicated that this reproductive defect was due to the mutation's effect upon the hypothalamus. The present study demonstrated the pleiotropic effects of the eyeless gene upon development of the hypothalamus. Scanning electron microscopy studies looked at the early ontogeny of the hypothalamohypophyseal system. The major morphological difference observed in the hypot

Localization of amylin-like immunoreactivity in melanocyte-stimulating hormone-containing cells of the pars intermedia but not those of the pars distalis in the axolotl (Ambystoma mexicanum) pituitary.

Immunohistochemical techniques were employed to investigate the distribution of amylin-like immunoreactivity in the axolotl (Ambystoma mexicanum) pituitary. Amylin-immunoreactive cells were observed in the pars intermedia, and these cells were found to be immunoreactive for α-melanocyte-stimulating hormone (αMSH) as well. In contrast, αMSH-immunoreactive cells in the pars distalis were immuno-negaitive for amylin. These light microscopic findings were confirmed by immunoelectron microscopy. Amylin-immunoreactive signals were located on the haloes of presumable secretory granules in association

THE ANTAGONISM BETWEEN THYROID AND PARATHYROID GLANDS.

From the facts stated in this paper it is evident that the thymus gland of mammals contains a substance which is capable of producing tetany when fed to the larvae of certain species of salamanders (Ambystoma opacum and Ambystoma maculatum). As long as the larvae have not developed their own thymus glands, they are able, by means of some mechanism, to counterbalance the tetanic action of the thymus substance introduced in their food. When, however, the secretion from their own thymus glands is added to the thymus material introduced with the food, this mechanism of preventing tetany becomes in
