The bell pepper is a vegetable with high antioxidant content, and its consumption is important because it can reduce the risk of certain diseases in humans. Plants can be affected by different types of stress, whether biotic or abiotic. Among the abiotic factors, there is saline stress that affects the metabolism and physiology of plants, which causes damage, decreasing productivity and quality of fruits. The objective of this work was to evaluate the application of selenium, silicon and copper nanoparticles and saline stress on the bioactive compounds of bell pepper fruits. The bell pepper pl

Stripe rust is a fungal disease that has devastated the barley production for a long time. The present study focused on the role of β-glucan, PR proteins, diamine oxidase (DAO), polyamine oxidase (PAO), key enzymes and metabolites of phenol and proline metabolism in the stripe rust resistance of barley. RD2901 with resistant behavior against stripe rust showed increased levels of PR proteins, phenylalanine ammonia lyase (PAL), tyrosine ammonia lyase (TAL) along with the accumulation of β-glucan and lignin which strengthen the plant cell wall during plant-pathogen interaction. It also depicted

The organ-specific enzyme histidase was measured as an indicator of skin involvement in the blood of 52 patients with primary and secondary lymphedema of the upper and lower limbs. A stable type of the disease (degree I) was characterized by washing histidase off the derma into the blood. Mild and moderate histidase washing associated with skin dystrophy occurred with progression of the disease (stage II and III, respectively). In advanced lymphedema (degree 4) histidase does not enter blood from the skin. This fact is explained by total impairment of skin cell elements and their replacement f

Hepatic histidase activity in adult female rats is twice that in adult male rats. Hypophysectomy and thyroidectomy result in a significant increase in hepatic histidase activities in males, but not in females. This effect on histidase is reversed by the exogenous administration of tri-iodothyronine, but not by ectopic pituitary glands or purified pituitary hormones.

The synthesis of non-canonical amino acids (ncAAs) has recently made tremendous progress in the field of biocatalysis, with enzymes such as transaminases, dehydrogenases, and ammonia lyases being used. Frameworks of β-branched aromatic D-amino acids are crucial for the efficacy of pharmaceuticals, influencing spatial conformation, binding modes with receptors and pharmacokinetic properties. However, there is a scarcity of studies on the efficient synthesis of β-branched D-amino acids, which has significantly hindered the advancement of structure-activity relationship (SAR) studies. Here, we pr

This research was conducted to determine the relationship between plant defense responses and the extent of treatment applied to either the aerial parts or roots of the plant. The experimental treatments included different methods of application (spraying versus soil drenching), varying treatment areas (one-sixth, one-third, half, or all of the plant's aerial parts and roots) with SA, and infecting the plants with root-knot nematodes. Evaluation of plant growth and nematode pathogenicity indices in the greenhouse section, H2O2 accumulation rate, and phenylalanine ammonia lyase enzyme activity

Heavy metal stress can lead to many adverse effects that inhibit cellular processes at various levels of metabolism, causing a decrease in plant productivity. In response to environmental stressors, phenolic compounds fulfill significant molecular and biochemical functions in plants. Increasing the biosynthesis of phenolic compounds in plants subjected to heavy metal stress helps protect plants from oxidative stress. A pot experiment was carried out to determine the effect of the accumulation of copper (Cu) and lead (Pb) salts at concentrations of 200, 500, and 1000 ppm on seed germination, th

trans-Cinnamic acid (tCA) is a precursor in the synthesis of many high-value compounds with bio-active qualities useful in applications like medicine, polymers, and cosmetics. Currently tCA is produced by industrial chemical synthesis from fossil fuels or cost-prohibitive isolation from terrestrial plants. Cyanobacteria, a type of photosynthetic bacteria, can be readily engineered to convert sunlight and carbon dioxide into metabolites of interest at relatively high amounts compared to terrestrial plants. The purpose of this study is to advance the industrial and commercial value of cyanobacte

Tea (Camellia sinensis (L.) O. Kuntze) leaves are a major source of flavonoids that mainly belong to the flavan-3-ols or catechins and are implicated in a wide range of health benefits. Although the catechins in tea leaves were identified long ago, the regulatory mechanisms governing catechin biosynthesis remain unclear. In the present work, the dynamic changes of catechin levels and the expression profiles of catechin-related genes in albino tea plants were intensively examined. The amounts of most catechins decreased to their lowest levels in the albino phase, when epigallocatechingallate wa

The urea cycle is a metabolic pathway for the disposal of excess nitrogen, which arises primarily as ammonia. Nitrogen is essential for growth and life-maintenance, but excessive ammonia leads to life-threatening conditions. The urea cycle disorders (UCDs) comprise diseases presenting with hyperammonemia that arise in either the neonatal period (about 50% of cases) or later. Congenital defects of the enzymes or transporters of the urea cycle cause the disease. This cycle utilizes five enzymes, two of which, carbamoylphosphate synthetase 1 and ornithine transcarbamylase are present in the mitoc

The detoxification of ammonia occurs mainly through conversion of ammonia to urea in the liver via the urea cycle and glutamine synthesis. Congenital portosystemic shunts (CPSS) in dogs cause hyperammonemia eventually leading to hepatic encephalopathy. In this study, the gene expression of urea cycle enzymes (carbamoylphosphate synthetase (CPS1), ornithine carbamoyltransferase (OTC), argininosuccinate synthetase (ASS1), argininosuccinate lyase (ASL), and arginase (ARG1)), N-acetylglutamate synthase (NAGS), Glutamate dehydrogenase (GLUD1), and glutamate-ammonia ligase (GLUL) was evaluated in do

Low phenylalanine diet has been the key treatment of phenylketonuria for more than 50years, allowing efficient management of thousands of PKU patients to date. However, non-compliance exists, mainly after adolescence. A medication for PKU received approval in Europe in 2009 (sapropterine dihydrochloride or Kuvan(®)) and can benefit to patients responsive to this drug. Other treatment options are available in some countries (glycomacropeptides, large neutral amino acids) or are currently under investigation (phenylalanine ammonia lyase, chaperones molecules, gene therapy).

Several halogenated alkenes are metabolized in part to cysteine S-conjugates, which are mitochondrial toxicants of kidney and, to a lesser extent, other organs. Toxicity is due to cysteine S-conjugate beta-lyases, which convert the cysteine S-conjugate into pyruvate, ammonia and a reactive sulphur-containing fragment. A section of the human population is exposed to halogenated alkenes. To understand the health effects of such exposure, it is important to identify cysteine S-conjugate beta-lyases that contribute to mitochondrial damage. Mitochondrial aspartate aminotransferase [Cooper, Bruschi,

Urea cycle defects are a group of metabolic disorders caused by enzymatic disruption of the urea cycle pathway, transforming nitrogen to urea for excretion from the body. Severe cases present in early infancy with life-threatening metabolic decompensation, and these episodes of hyperammonemia can be fatal or result in permanent neurologic damage. Despite the progress in pharmacologic treatment, long-term survival is poor especially for severe cases. Liver transplant is an alternative treatment option, providing sufficient enzymatic activity and decreasing the risk of metabolic decompensation.

The concentrations of ammonia in the plasma of the mudskipper Boleophthalmus boddaerti exposed to cyanide for 1-6 days were significantly greater than the respective values of the controls. This was due to an increase in the production of NH3 in the muscle and an increase in the retention of NH3 and/or NH4+ in the blood of the cyanide-exposed fish when compared to controls. Cyanide exposure significantly increased the specific activity of muscle AMP deaminase. Since adenylosuccinate synthetase and lyase were also present in the muscle, exposure to cyanide might increase the production of NH3 f

Argininosuccinate lyase (ASL) deficiency (McKusick 207900) is a rare autosomal recessive disorder affecting the urea cycle. The cardinal symptom in the neonatal form is progressive hyperammonemia, which is often life-threatening. However, clinical symptoms in the late onset form are quite heterogeneous. As well as measurement of ASL activity, analysis of the ASL gene is necessary to clarify the genetic basis of various phenotypes. We report a patient with late onset argininosuccinate lyase deficiency (ASLD) who had hepatomegaly and mildly increased level of ammonia. By mutation analysis of the

A simple and reliable method is described which is suitable for estimation of a whole blood phenylalanine concentration for the patient with PKU in various settings including the physician's office and the home. Excellent correlations were obtained between this method and weighed phenylalanine standards, as well as with measurement of phenylalanine in serum, plasma, and whole blood, using the McCaman-Robins fluorometric assay. Increasing the frequency and rapidity of feedback to the patient should improve metabolic control, just as home glucose monitoring has for the patient with diabetes mell

We report the development of a cofactor-free CO2 fixation platform based on a three-enzyme cascade comprising Aspergillus niger ferulic acid decarboxylase (AnFDC), Anabaena variabilis phenylalanine ammonia-lyase (AvPAL), and Proteus mirabilis l-amino acid deaminase (PmLAAD). Unlike canonical ATP- or NADPH-dependent CO2 assimilation pathways, this system uses a prFMN-dependent carboxylation mechanism, enabling efficient CO2 incorporation under ambient conditions without energy-intensive cofactors. Systematic screening identified AnFDC as the optimal decarboxylase for styrene carboxylation, whil
