Bloody diarrhoea cases caused by Shigella and amoeba in Jordan.

This study was done to evaluate the occurrence of bloody diarrhoea in Jordan in the period 1988-2000, with particular interest in Shigilla and amoeba cases in relation to the surveillance control system and hygiene policy. The cases were collected from various medical diagnostic laboratories. In addition to the clinical presentation, stool specimens or rectal swabs were collected and sub-cultured on the appropriate media. Shigella spp. and amoeba were identified using standard methods. Anti-microbial susceptibility tests were performed. One-way ANOVA and Least Significant Difference (LSD) were

Brain-Eating Amoebae in the United Arab Emirates?

Given the warm climate, excessive water-related recreational activities, and large Muslim population practicing ablution, it is surprising that primary amoebic meningoencephalitis is not reported in the United Arab Emirates. In a medical district, up to 43% of meningitis cases did not determine the causative organism, suggesting that cases likely go undiagnosed.

Endosymbiotic bacteria associated with nematodes, ticks and amoebae.

Endosymbiosis is a mutualistic, parasitic or commensal symbiosis in which one symbiont is living within the body of another organism. Such symbiotic relationship with free-living amoebae and arthropods has been reported with a large biodiversity of microorganisms, encompassing various bacterial clades and to a lesser extent some fungi and viruses. By contrast, current knowledge on symbionts of nematodes is still mainly restricted to Wolbachia and its interaction with filarial worms that lead to increased pathogenicity of the infected nematode. In this review article, we aim to highlight the ma

Role of the Legionella pneumophila rtxA gene in amoebae.

Legionella pneumophila infects humans, causing Legionnaires' disease, from aerosols generated by domestic and environmental water sources. In aquatic environments L. pneumophila is thought to replicate primarily in protozoa. A 'repeats in structural toxin' (RTX) gene, rtxA, from L. pneumophila was identified recently that plays a role in entry and replication in human macrophages and also has the ability to infect mice. However, the role of this gene in the interaction of L. pneumophila with environmental protozoa and its distribution in different Legionella species has not been examined. Sout

[Human pathology caused by free-living amoebae].

Naegleria fowleri, Acanthamoeba spp. and Balamuthia mandrillaris are free-living amoebae that occasionally may induce pathology in human beings. CNS disease due to N. fowleri, called "primary" amoebic meningoencephalitis, is acquired after exposure to polluted waters in swimming pools, rivers, and lakes. The clinical course is acute, often fulminant and characterized pathologically by necrotizing hemorrhagic meningoencephalitis, involving mainly the base of the brain, brainstem and cerebellum. In contrast, some Acanthamoeba spp. and B. mandrillaris cause opportunistic, chronic "granulomatous"

[Review article: occurrence, parasitism and pathogenetic potency of free-living amoeba].

In areals of human life free living amoebae (FLA) are widely distributed and various possibilities to infect men as well as animals do exist under specific circumstances. While Naegleria species, respectively, Naegleria fowleri give rise to fulminant fatal processes of CNS Acanthamoeba infections mostly have a chronical course and immunodeficiency could be a favouring factor for acanthamoebiasis. Loss of Cln3 impacts protein secretion in the social amoeba Dictyostelium.

Neuronal ceroid lipofuscinosis (NCL), also referred to as Batten disease, is the most common form of childhood neurodegeneration. Mutations in CLN3 cause the most prevalent subtype of the disease, which manifests during early childhood and is currently untreatable. The precise function of the CLN3 protein is still not known, which has inhibited the development of targeted therapies. In the social amoeba Dictyostelium discoideum, loss of the CLN3 homolog, Cln3, reduces adhesion during early development, which delays streaming and aggregation. The results of the present study indicate that this

The Social Amoeba Dictyostelium discoideum Is Highly Resistant to Polyglutamine Aggregation.

The expression, misfolding, and aggregation of long repetitive amino acid tracts are a major contributing factor in a number of neurodegenerative diseases, including C9ORF72 amyotrophic lateral sclerosis/frontotemporal dementia, fragile X tremor ataxia syndrome, myotonic dystrophy type 1, spinocerebellar ataxia type 8, and the nine polyglutamine diseases. Protein aggregation is a hallmark of each of these diseases. In model organisms, including yeast, worms, flies, mice, rats, and human cells, expression of proteins with the long repetitive amino acid tracts associated with these diseases reca

Potentially pathogenic amoeba-associated microorganisms in cooling towers and their control.

Cooling towers provide a favorable environment for the proliferation of microorganisms. Cooling towers generate a biofilm and often aerosolize contaminated water, thereby increasing the risk of microorganism dissemination by human inhalation. This pathogen dissemination was first revealed by the epidemics of Legionnaires' disease that were directly related to the presence of cooling towers, and since then, the ecology of Legionella pneumophila has been well studied. Each country has specific standards regarding the acceptable amount of microorganisms in cooling tower systems. However, those st

Disseminated Balamuthia mandrillaris amoeba infection in an AIDS patient from Brazil.

This report describes a 32-year-old male AIDS patient. He presented with a clinical picture characterized by severe headache, blurred vision, and fever that had lasted for 10 days. At admission, no remarkable neurologic abnormalities were observed. Cranial tomography showed a ring-enhanced lesion with edema and a mass effect in the right occipital lobe. The initial diagnosis was toxoplasmosis, and treatment of this was administered. However, 5 days later, the patient's clinical status worsened and he died. The necropsy showed necrotizing and hemorrhagic encephalitis, with trophozoites similar

Inhibition of induced pinocytosis in Amoeba proteus by membrane stabilizing drugs.

The effect of membrane stabilizing drugs on cation induced pinocytosis was studied in Amoeba proteus. Initially the presence of local anesthetic drugs during a pinocytosis cycle had a stimulating effect on channel formation, however, the capacity to develop pinocytotic channels was reversibly inhibited after a period of treatment with these drugs. Imipramine, vinblastine and the phenothiazines had effects similar to local anaesthetics. The local anesthetics inhibited pinocytosis in the following order: dibucaine greater than tetracaine greater than bupivacaine greater than lidocaine greater th

Zinc Oxide Nanoconjugates against Brain-Eating Amoebae.

Naegleria fowleri and Balamuthia mandrillaris are opportunistic protists, responsible for fatal central nervous system infections such as primary amoebic meningoencephalitis (PAM) and granulomatous amoebic encephalitis (GAE) with mortality rates higher than 90%. Threatening a rise in cases is the increase in temperature due to global warming. No effective treatment is currently available. Herein, nanotechnology was used to conjugate Zinc oxide with Ampicillin, Ceftrixon, Naringin, Amphotericin B, and Quericitin, and the amoebicidal activity and host cell cytotoxicity of these resulting compoun

Various brain-eating amoebae: the protozoa, the pathogenesis, and the disease.

Among various genera of free-living amoebae prevalent in nature, some members are identified as causative agents of human encephalitis, in which Naegleria fowleri followed by Acanthamoeba spp. and Balamuthia mandrillaris have been successively discovered. As the three dominant genera responsible for infections, Acanthamoeba and Balamuthia work as opportunistic pathogens of granulomatous amoebic encephalitis in immunocompetent and immunocompromised individuals, whereas Naegleria induces primary amoebic meningoencephalitis mostly in healthy children and young adults as a more violent and deadly

Antibiotic susceptibility of Neochlamydia hartmanellae and Parachlamydia acanthamoebae in amoebae.

Parachlamydia acanthamoebae and Neochlamydia hartmanellae are Chlamydia-related bacteria naturally infecting free-living amoebae. These strict intracellular bacteria might represent emerging pathogens. Recent studies report an association with lower respiratory tract infections, especially with pneumonia where they have been identified as a potential causative agent in 1-2% of cases. In this study, we defined the antibiotic susceptibility of N. hartmanellae, two strains of P. acanthamoebae and two yet unclassified Parachlamydiaceae strains using a quantitative approach. We confirmed the result

Germination and amplification of anthrax spores by soil-dwelling amoebas.

While anthrax is typically associated with bioterrorism, in many parts of the world the anthrax bacillus (Bacillus anthracis) is endemic in soils, where it causes sporadic disease in livestock. These soils are typically rich in organic matter and calcium that promote survival of resilient B. anthracis spores. Outbreaks of anthrax tend to occur in warm weather following rains that are believed to concentrate spores in low-lying areas where runoff collects. It has been concluded that elevated spore concentrations are not the result of vegetative growth as B. anthracis competes poorly against ind
