The hydride isonitrile complex [FeH(CNCEt3 )(1 a)]BF4 (2) containing a chiral P2 (NH)2 macrocycle (1 a), in the presence of 2-propanol as hydrogen donor, catalyzes the base-free asymmetric transfer hydrogenation (ATH) of prostereogenic ketones to alcohols and the hemihydrogenation of benzils to benzoins, which contain a base-labile stereocenter. Benzoins are formed in up to 83 % isolated yield with enantioselectivity reaching 95 % ee. Ketones give the same enantioselectivity observed with the parent catalytic system [Fe(CNCEt3 )2 (1 a)] (3 a) that operates with added NaOt Bu.

In human astrocytoma cells (1321N1), trifluoperazine potently inhibited histamine-induced phosphoinositide hydrolysis, while it slightly inhibited carbachol-induced phosphoinositide hydrolysis. Trifluoperazine as well as diphenhydramine inhibited [3H]mepyramine binding in astrocytoma cell membranes with Ki values of 0.052 microM and 0.005 microM, respectively. However, trifluoperazine only slightly inhibited [3H]quinuclidinyl benzilate binding with a Ki value of 3 microM. These results indicate that trifluoperazine specifically inhibits histamine-induced phosphoinositide hydrolysis as a histam

The muscarinic acetylcholine receptors were identified in membrane preparations from human tissues by the specific binding of 1-[benzilic-4,4'-3H] quinuclidinyl benzilate. Saturation binding isotherms of this radioligand yielded a total amount of receptors of 435 +/- 208, 159 +/- 65 and 913 +/- 89 fmol/mg protein, respectively, in the hippocampus, pons and submandibular gland. Non linear least squares analysis of competition binding studies with the antagonists pirenzepine and AF-DX 116 indicates that the majority of receptors are of the M1-type in the hippocampus (83%, high affinity for piren

Binding properties for the muscarinic antagonist quinuclidinyl benzilate (QNB), the nicotinic antagonist tubocurarine and nicotine to crude synaptosomal fractions from mouse and rat brain have been studied. Marked regional differences in number of binding sites for labelled QNB (3H-QNB) were found with a high concentration in the striatum, cortex and a low concentration in the cerebellum. The specific binding sites for labelled tubocurarine (3H-tubocurarine) showed less regional variation. An increased number of muscarinic binding sites (with no change in receptor affinity) was found in rats f

Systematic studies of the structure-activity relationships of atropine-like anticholinergic drugs have provided valuable information about the nature of the muscarinic receptor. In this study, the pharmacological activities of the (Z) and (E)-isomers of 2-phenylcyclohexyl diethylaminoethyl ether (1 and 2, respectively) in the isolated rat left atrium were investigated and compared with their activities in the isolated rat ileum preparation. Compound 1 was found to be one of the most ileal selective muscarinic antagonists reported to date. Other data concerning possible differences in the recep

Urea-treatment of the microsome fraction of the heart of guinea-pigs caused selective reduction in the apparent affinity of an agonist (carbachol), but not an antagonist (atropine), to muscarinic acetylcholine receptors (mAChR), measured as inhibition of binding of 3H-quinuclidinyl benzilate (3H-QNB). This effect was similar to that of Gpp (NH)p. The effects of urea-treatment and Gpp (NH)p were not additive. On the other hand, treatment of the microsome fraction with 5,5'-dithiobis (2-nitrobenzoic acid) (DTNB) increased the apparent affinity of agonist, but not antagonist. The effect of DTNB p

1. Kinetic, saturation and inhibition radioligand binding experiments with [3H]-N-methylscopolamine and [3H]quinuclidinyl benzilate were used to characterize the muscarinic cholinergic receptor in opossum (Didelphis virginiana, Kerr) submandibular salivary gland membranes. 2. The receptor density in opossum submandibular gland was found to be more than 3-fold higher than in rat, and 22-fold higher than in human, submandibular glands. 3. Inhibitor equilibrium dissociation constants for the antagonists pirenzepine, dicyclomine, atropine, N-methylscopolamine and AF-DX 116 revealed that the muscar

Muscarinic acetylcholine receptors were studied in slices of unaffected skin and in lymphocyte membranes from 3 patients with cholinergic urticaria and 5 healthy subjects using receptor autoradiography and the radioligand [3H]quinuclidinyl benzilate binding technique. Autoradiography revealed that muscarinic acetylcholine receptors were located in regions with sweat glands. The distribution of receptors did not differ qualitatively between patients and controls. However, the maximum number of muscarinic acetylcholine receptors was reduced in skin from patients with cholinergic urticaria, by 44

1 The synthesis of tritium labelled propylbenzilylcholine mustard ([(3)H]-PrBCM; N-2'-chloroethyl-N-[2'', 3''-(3)H(2)] propyl-2-aminoethyl benzilate) is described.2 The uptake by muscle strips was measured and shown to be considerably increased by previous immersion of the muscle in distilled water.3 A considerable part of the uptake is inhibited selectively by atropine, but not by nicotinic antagonists. A number of muscarinic agonists also inhibit uptake and their apparent affinity constants have been determined.4 The uptake by atropine-sensitive sites is temperature-insensitive, whereas the

The ability of chlorpromazine (CPZ) to inhibit muscarinic-cholinergic secretory events was studied in vitro in rat parotid acinar cells. CPZ inhibited carbachol-induced amylase release in a dose-dependent fashion but had no effect on that elicited by isoproterenol. The inhibition of parotid protein synthesis induced by carbachol, but not that induced by A23187, was blocked by CPZ. CPZ exhibited a dose-dependent inhibition of [3H] quinuclidinyl benzilate (QNB) binding to muscarinic receptors, and altered the KD of the receptor for the ligand. These results are consistent with an ability of CPZ

The contracted segment of terminal large intestine in both Hirschsprung's disease in humans and in the piebald-lethal strain of mice with congenital megacolon may result from muscarinic cholinergic receptor denervation supersensitivity. This hypothesis was tested by direct binding studies using the muscarinic receptor ligand, tritiated quinuclidinyl benzilate. There were no differences in either the receptor density, dissociation constant, or agonist/antagonist Hill coefficients in the proximal, middle, or distal colon of mice with congenital megacolon as compared with normal littermates. The effects of various substrates on the binding of agonists to muscarinic receptors were studied in the rat hypothalamus and adenohypophysis by competition experiments using the highly specific tritiated muscarinic antagonist N-methyl-4-piperidyl benzilate. It was found that agonist binding properties were affected only by the steroid sex hormones (beta-estradiol and progesterone), both of which resulted in a decrease in the proportion of high-affinity binding sites and a decrease in the dissociation constant. This suggests a link between the muscarinic system and the mechanism by which these

1. The effects of thyroid status on the specific binding of the muscarinic ligand (-)-[3H]quinuclidinyl benzilate (QNB) and of the beta-adrenoreceptor ligand (-)-[3H]dihydroalprenolol (DHA) in the adult rat lung were investigated. 2. The specific binding of (-)-[3H]quinuclidinyl benzilate (QNB) to lung membranes was saturable and the equilibrium dissociation constant (KD) determined from Scatchard analysis was 54 pM. Kinetic analysis of the binding of [3H]QNB yielded a KD of 42 pM. [3H]QNB binding was inhibited by muscarinic agonists and antagonists, the order of their potency was l-hyoscyamin

The biochemical and morphological effects of postnatal acetylcholinesterase (AChE) inhibition were examined in rat pups dosed with paration, at time points critical to hippocampal neurogenesis and synaptogenesis (i.e., day 5-20). In treated pups, sacrificed on day 21, hippocampal histopathology, as assessed by light and electron microscopy, consisted of cellular disruption and necrosis in the dentate gyrus (DG), and CA4 regions. Synaptic disruption in the DG molecular layer was suggested by histochemical preparation using both the Timm's and AChE stains. In parathion-treated pups, sampled at d

Binding sites with high affinity and specificity for [(3)H]quinuclidinyl benzilate (QNB) are present in homogenates of rat brain. The characteristics of the binding sites resemble those of muscarinic cholinergic receptors. Specific binding is saturable with respect to [(3)H]QNB and tissue concentration and is time-, temperature-, and pH-dependent. The bimolecular rate of association (2.0 x 10(8) M(-1) min(-1)) and dissociation (1.2 x 10(-2) min(-1)) at 35 degrees indicate a dissociation constant of 60 pM and a density of 65 pmol/g of brain. A novel series of macrocyclic complexes of the type [M(C(48)H(32)N(4))X]X(2); where M=Cr(III), Fe(III) and Mn(III); X=Cl(-), NO(3)(-), CH(3)COO(-) have been synthesized by template condensation of 1,8-diaminonaphthalene and benzil in the presence of trivalent metal salts in methanolic medium. The complexes have been characterized with the help of elemental analyses, conductance measurements, magnetic measurements, electronic, NMR, infrared, far infrared and mass spectral studies. On the basis of these studies, a five coordinate square pyramidal geometry for all of these complexes has been prop

In July 1995 about 15,000 people attempted to walk from the enclave of Srebrenica to free territory in Bosnia. Two-thirds were captured or killed. Many of the remainder experienced hallucinations on the march, leading them to believe they were the victims of chemical weapons. This paper reports extended structured interviews with 35 survivors, including three doctors, carried out a year later on behalf of Human Right Watch. The literature on the likeliest CW agent, 3-quinuclidinyl benzilate (BZ), and on stress as a cause of hallucinations, is reviewed. While CW exposure cannot be ruled out, it
