Stimulatory effect of bombesin on phosphoinositide metabolism in the rat pineal gland.

The pineal gland is under complex peptidergic nervous control originating from hypothalamic nuclei. The daily rhythm of bombesin-like peptide in the hypothalamus suggests that this neuropeptide, similarly as other neuropeptides, might be involved in modulation of the physiological activity of the pineal gland. In our experiments we studied the mechanism of signal transduction of bombesin in the isolated pineal glands of rats. The phosphoinositide signalling system was examined by measuring 32P-labelling of phosphatidylinositol (PI), phosphatidylinositol phosphate (PIP) and phosphatidylinositol

Effect of motilin, somatostatin and bombesin on gastroduodenal myoelectric activity in sheep.

The effects of motilin, erythromycin, somatostatin and bombesin on antroduodenal myoelectric activity were investigated in conscious sheep. Myoelectric recordings were obtained from electrodes chronically implanted on the antrum and duodenal bulb. Peptides or erythromycin were infused intravenously (i.v.) during 5 min. Antagonists were injected i.v. as a bolus. Neither motilin (2.5-80 ng/kg/min) nor erythromycin (2-16 micrograms/kg/min) modified the antroduodenal myoelectric activity, although a single bolus of these compounds (250 ng/kg and 50 micrograms/kg respectively) increased the antral

Effect of bombesin on core temperature in rats: studies involving radiotelemetry.

The effect of centrally administered bombesin on core temperature was investigated in rats which had been chronically fitted with a cannula in the third cerebral ventricle and with an intraperitoneal temperature-monitoring radiotransmitter. Intracerebroventricularly administered bombesin (0.01, 0.1 or 1 microgram) resulted in hypothermia, the duration and depth of the effect being a function of the dose. The use of telemetry minimized experimental interventions in this study; the results demonstrate that the hypothermic response to bombesin is not dependent upon exposure to a low ambient tempe

Effects of intracerebroventricular bombesin on gastric ulcers and gastric glycoproteins in rats.

Effects of intracerebroventricular injection of bombesin on gastric mucosal glycoproteins and acid secretion were studied to clarify its action mechanism for experimental ulcers in rats. Bombesin (10 micrograms) prevented stress ulcers and indomethacin ulcers. Aspirin (300 mg/kg p.o.) reduced the high molecular weight glycoproteins extracted from the gastric mucosa, and increased the degraded glycoproteins extracted from gastric juice in pylorus-ligated rats. Bombesin inhibited the aspirin-induced changes of the glycoproteins. Bombesin also inhibited basal gastric acid secretion. These results

Role of dopaminergic system(s) in mediation of the behavioural effects of bombesin.

To test the effects of dopamine receptor blockade on bombesin (BN)-induced behavioural changes, adult male Sprague-Dawley rats were administered fluphenazine (0.01, 0.025, 0.1, 0.25 mg/kg, IP) followed 30 min later by BN (1 micrograms in 5 microliter) or saline (5 microliter) intracerebroventricularly (ICV). Subsequent behavioural effects were monitored in chambers equipped with strategically located infrared beam grids, controlled by a microprocessor. The following behaviours were monitored: locomotor activity (distance traversed), floor activity (horizontal or lateral displacement) and reari

Bombesin receptor subtypes in human cancers: detection with the universal radioligand (125)I-[D-TYR(6), beta-ALA(11), PHE(13), NLE(14)] bombesin(6-14).

PURPOSE: Bombesin and bombesin receptors have been shown to play a role in cancer. Whereas the gastrin-releasing peptide (GRP) receptor is a bombesin receptor subtype frequently expressed by tumors, the other three subtypes, the neuromedin B (NMB), BB3, and BB4 receptors, have been poorly investigated in human tissues. EXPERIMENTAL DESIGN: We investigated 161 human tumors for their bombesin receptor subtype expression using in vitro receptor autoradiography with the universal bombesin radioligand (125)I-[D-Tyr(6), beta-Ala(11), Phe(13), Nle(14)]bombesin(6-14) in displacement experiments with

An Ugi-like Biosynthetic Pathway Encodes Bombesin Receptor Subtype-3 Agonists.

Isocyanide functional groups can be found in a variety of natural products. Rhabduscin is one such isocyanide-functionalized immunosuppressant produced in Xenorhabdus and Photorhabdus gammaproteobacterial pathogens, and deletion of its biosynthetic gene cluster inhibits virulence in an invertebrate animal infection model. Here, we characterized the first "opine-glycopeptide" class of natural products termed rhabdoplanins, and strikingly, these molecules are spontaneously produced from rhabduscin via an unprecedented multicomponent "Ugi-like" reaction sequence in nature. The rhabdoplanins also

Gastrin-releasing peptide, a bombesin-like neuropeptide, promotes cutaneous wound healing.

BACKGROUND: Little is known about the effects of neuropeptides on wound healing. OBJECTIVE: To investigate the effect of gastrin-releasing peptide (GRP), one of the bombesin-like neuropeptides, on wound healing. METHODS: The effects of GRP on cultured keratinocyte proliferation and migration were measured by BrdU uptake and in vitro scratch assay, respectively. Various concentrations of GRP ointments (0, 10(-9), 10(-8), 10(-7), 10(-6) M) were topically applied to 1.0 mm wounds on porcine flanks. RESULTS: GRP stimulated keratinocyte growth and locomotion in a dose-dependent manner. Topical a

Identification of functional GRP-preferring bombesin receptors on human melanoma cells.

Bombesin was originally isolated from amphibian skin, whereas its mammalian counterpart, gastrin-releasing peptide (GRP), was first identified in the nervous system of the gastrointestinal tract. Whether GRP is present in the human skin is not known. Bombesin-like peptides are also known to modulate growth. We therefore investigated whether human melanoma cell lines express functional GRP-preferring bombesin receptors and whether they alter growth or other specific cellular functions of these tumour cells. GRP receptor mRNA was found in HBL, D-10, Me-28 and A375-6 cell lines, but only A375-6 c

Endocrine effects of new bombesin/gastrin-releasing peptide antagonists in rats.

Four new and specific pseudononapeptide bombesin/gastrin-releasing peptide (GRP) receptor antagonists, containing the D-forms of Trp or Trp analogue (Tpi) at position 6, were studied for their effects on the endocrine pancreas and GRP-(14-27)-induced gastrin release in pentobarbital-anesthetized rats. One of the analogues, D-Tpi6,Leu13-psi (CH2NH)Leu14-bombesin-(6-14) (RC-3095), was injected into the lateral brain ventricle just preceding intracerebroventricular administration of GRP-(14-27) to evaluate its antagonistic effect on GRP-induced serum growth hormone (GH) suppression. Cloning of novel bombesin precursor cDNAs from skin of Bombina maxima.

Amphibian skin is a rich resource of bioactive peptides like proline-rich bombesin from frog Bombina maxima. A novel cDNA clone encoding a precursor protein that comprises proline-rich bombesin and a novel peptide, designated as bombestatin, was isolated from a skin cDNA library of B. maxima. The predicted primary structure of the novel peptide is WEVLLNVALIRLELLSCRSSKDQDQKESCGMHSW, in which two cysteines form a disulfide bond. A BLAST search of databases did not detect sequences with significant similarity. Bombestatin possesses dose-dependent contractile activity on rat stomach strips. The d

Synthesis and Evaluation of Novel 68Ga-Labeled GRPR-Targeted PET Tracers Derived from [d-Phe6,Pro14]Bombesin(6-14) and [d-Phe6,des-Met14]Bombesin(6-14) Sequences.

Gastrin-releasing peptide receptor (GRPR) is a G-protein-coupled receptor that is overexpressed in several malignancies, rendering it an ideal target for cancer imaging and therapy. In this report, we conducted structure-activity relationship studies by optimizing the substitution of Pro14 based on an agonist sequence, [d-Phe6,Pro14]Bombesin(6-14), and optimizing the substitution of the C-terminal Leu13-NH2 based on an antagonist sequence, [d-Phe6,des-Met14]Bombesin(6-14). PEP-1-PEP-5 derived from the agonist sequence and PEP-6-PEP-14 derived from the antagonist sequence were synthesized by a

Characterization of scratching responses in rats following centrally administered morphine or bombesin.

The aim of this study was to characterize scratching behavior elicited by central administration of morphine or bombesin in rats, and to determine the role of opioid receptors in scratching induced by both pruritogenic agents. Central administration included intracisternal (i.c.), intrathecal (i.t.), and intracerebroventricular (i.c.v.) routes. Scratching events made with hind paws were counted by observers blinded to treatment conditions. Intracisternal morphine (0.01-0.1 microg) produced dose-dependent increases in scratching; the maximum response to i.c. morphine 0.1 microg was approximatel

The effect of prolactin and bombesin on the growth of meningioma-derived cells in monolayer culture.

Meningioma growth is thought to be stimulated by the sex hormones progesterone and possibly estrogen. We report here stimulation of growth of meningioma-derived cells in culture by prolactin. Fourteen human tumors taken from surgery were initially grown in Ham's medium F10 with 15% fetal calf serum. The tumors were then trypsinized and resuspended in medium in a multi-well plate with either prolactin or bombesin; the cells were incubated for 1 week, washed, and resuspended for cell counting. The growth-stimulating effect of prolactin at 10 and 200 micrograms/ml was compared with bombesin at 5

Receptor heterogeneity for bombesin-like peptides in the rat central nervous system.

As an initial characterization of bombesin binding sites in the rat central nervous system, we examined the pharmacological specificity of binding of bombesin (BBS) and several BBS analogs to rat cortex and compared these results to those for rat pancreas. In addition, we used in vitro receptor autoradiography to evaluate binding of 125I-[Tyr4]bombesin and 125I-Bolton-Hunter neuromedin B (NMB) to several regions of the rat brain. The results of the pharmacological study indicated that the pancreas and cortex had different binding affinities for BBS-like peptides. While cortical binding sites h

Chronic administration of bombesin stimulates antral gastrin cell proliferation in the rat.

Bombesin tetradecapeptide, administered in gelatin twice daily for 1 wk, significantly increased the antral gastrin content in rats by stimulating antral gastrin cell proliferation. The labeling index of gastrin cells after five daily injections of [3H]thymidine given during bombesin treatment was significantly enhanced as compared with controls, while no modification in the pattern of antral somatostatin cell labeling was revealed. Total antral gastrin cell population at the end of bombesin treatment also increased significantly. Morphometric measurements at the electron microscope level show

1. The changes in blood pressure in response to parenteral administration of bombesin, the active tetradecapeptide of the skin of the European discoglossid frogs Bombina bombina and Bombina variegata variegata have been investigated in some experimental animals.2. In most species, the polypeptide elicited hypertension which was usually gradual in onset and slow to disappear. Blood pressure increases rarely exceeded 40-50 mmHg. At the beginning of an experiment some dose-response relationship could often be observed, but later tachyphylaxis developed. During an intravenous infusion of bombesin

Healing-promoting effect of bombesin treatment on chronic gastric ulcer in rats.

To evaluate whether bombesin treatment has a facilitatory effect on the healing of chronic gastric ulcer, following the induction of ulcer by serosal application of acetic acid, rats were given bombesin (30 microg/kg/day; subcutaneously) or vehicle three times a day for 7, 14 or 21 days until they were decapitated. Neither food intake nor gastric emptying rate in either vehicle-treated or bombesin-treated groups was not statistically different from control rats. Similarly, ulcer indices and gastric myeloperoxidase (MPO) activities at the first and second weeks of injury were not different amon

Central pretreatment with CRF receptor antagonists, alpha-helical CRF-(9-41) (alpha-CRF-(9-41)) or [D-Phe12, C(alpha)MeLeu37]CRF-(12-41) (CalphaMeCRF), blocked the effects of centrally administered bombesin on food intake and related behaviors and partially attenuated the satiety effects of systemically administered bombesin.
