Extracellular Vesicle Isolation from Botrytis cinerea Culture and B. cinerea-Infected Arabidopsis Plants.

Extracellular vesicles (EVs) are universally conserved and essential players in the communication between cells across nearly all organisms of life. Botrytis cinerea produces and secretes EVs that are critical in its virulence on plants. Numerous studies in various fungal species reveal that EVs are important across different pathosystems to facilitate fungal infection. For characterization of the cargo and role of EVs in fungal growth and communication, it is necessary to carefully and precisely purify fungal EVs from fungal culture or the plant interaction interface. Here, we present a metho

Antifungal Mechanism of Biphenyl Phytoalexin Derivative Sorbauphylin A against Botrytis cinerea in Grapes.

Botrytis cinerea causes significant losses in crops, especially in grapes. The biphenyl derivative sorbauphylin A (4'-n-pentyl-[1,1'-biphenyl]-3,5-diol), derived from plant phytoalexins, exerts potent antifungal effects with low toxicity. This study demonstrated that sorbauphylin A significantly inhibited the growth of B. cinerea (EC50 = 15 μg mL-1), suppressed spore germination, and markedly reduced infection in grapes and leaves. The results of fluorescence staining and enzyme activity detection indicated that sorbauphylin A damaged cell membranes and walls and triggered a reactive oxygen sp

SlARG2 contributes to MeJA-induced defense responses to Botrytis cinerea in tomato fruit.

BACKGROUND: Arginase plays key roles in methyl jasmonate (MeJA)-mediated quality maintenance in vegetables and fruits. MeJA treatment induced the Arginase 2 (SlARG2) expression, which is one of the most important encoding genes of arginase. In addition, the treatment with MeJA induced resistance to pathogenic infection in many plants. However, the functions of SlARG2 in MeJA-induced defense to Botrytis cinerea are unclear. In our work, control and SlARG2-silenced tomato fruits (Solanum lycopersicum) were treated with 0.05 mmoL L-1 MeJA before storage to assay the roles of SlARG2 in MeJA-induce

Characterization of Postharvest Fungicide-Resistant Botrytis cinerea Isolates From Commercially Stored Apple Fruit.

Botrytis cinerea causes gray mold and is an economically important postharvest pathogen of fruit, vegetables, and ornamentals. Fludioxonil-sensitive B. cinerea isolates were collected in 2011 and 2013 from commercial storage in Pennsylvania. Eight isolates had values for effective concentrations for inhibiting 50% of mycelial growth of 0.0004 to 0.0038 μg/ml for fludioxonil and were dual resistant to pyrimethanil and thiabendazole. Resistance was generated in vitro, following exposure to a sublethal dose of fludioxonil, in seven of eight dual-resistant B. cinerea isolates. Three vigorously gro

Induced systemic resistance against Botrytis cinerea by Micromonospora strains isolated from root nodules.

Micromonospora is a Gram positive bacterium that can be isolated from nitrogen fixing nodules from healthy leguminous plants, where they could be beneficial to the plant. Their plant growth promoting activity in legume and non-legume plants has been previously demonstrated. The present study explores the ability of Micromonospora strains to control fungal pathogens and to stimulate plant immunity. Micromonospora strains isolated from surface sterilized nodules of alfalfa showed in vitro antifungal activity against several pathogenic fungi. Moreover, root inoculation of tomato plants with these

beta-Aminobutyric acid-induced protection of Arabidopsis against the necrotrophic fungus Botrytis cinerea.

The non-protein amino acid beta-aminobutyric acid (BABA) protects numerous plants against various pathogens. Protection of Arabidopsis plants against virulent pathogens involves the potentiation of pathogen-specific defense responses. To extend the analysis of the mode of action of BABA to necrotrophs we evaluated the effect of this chemical on Arabidopsis plants infected with the gray mold fungus Botrytis cinerea. BABA-treated Arabidopsis were found to be less sensitive to two different strains of this pathogen. BABA protected mutants defective in the jasmonate and ethylene pathways, but was

Differential Responses of Botrytis Isolates to Pristine, Luna Tranquility, Switch, and Their Active Ingredients.

Botrytis cinerea is an adaptable, high-risk pathogen capable of developing resistance to several site-specific fungicides. To manage gray mold, blueberry growers in Washington State and Oregon follow spray regimes that include rotation of site-specific fungicides, tank-mixing site-specific fungicides with a multisite fungicide such as captan, and using premix fungicides to slow down selection of B. cinerea populations with resistance to site-specific fungicides. In this study, we conducted conidial germ tube growth assays on media to assess responses of Botrytis isolates to discriminatory dose

Photodynamic inactivation of Botrytis cinerea by an anionic porphyrin: an alternative pest management of grapevine.

Botrytis cinerea is a necrotic plant fungus that causes gray mold disease in over 200 crops, including grapevine. Due to its genetic plasticity, this fungus presents strong resistance to many fungicides. Thus, new strategies against B. cinerea are urgently needed. In this context, antimicrobial photodynamic treatment (APDT) was considered. APDT involves the use of a photosensitizer that generates reactive oxygen species upon illumination with white light. Tetra-4-sulfonatophenyl porphyrin tetra-ammonium (TPPS) was tested on B. cinerea using light. 1.5 µM TPPS completely inhibited mycelial grow

Resistance to Increasing Chemical Classes of Fungicides by Virtue of "Selection by Association" in Botrytis cinerea.

Previous research has shown that Botrytis cinerea isolates with resistance to multiple chemical classes of fungicides exist in eastern strawberry fields. In this study, the fungicide resistance profiles of 2,130 isolates from flowers of commercial strawberry fields located in multiple states was determined over four consecutive strawberry production seasons. Producers were asked to alternate single-site fungicides that were considered low risk in their specific location based on resistance monitoring results in their fields. This recommendation led to an increase of chemical class diversity us

The glutaredoxin ATGRXS13 is required to facilitate Botrytis cinerea infection of Arabidopsis thaliana plants.

Botrytis cinerea is a major pre- and post-harvest necrotrophic pathogen with a broad host range that causes substantial crop losses. The plant hormone jasmonic acid (JA) is involved in the basal resistance against this fungus. Despite basal resistance, virulent strains of B. cinerea can cause disease on Arabidopsis thaliana and virulent pathogens can interfere with the metabolism of the host in a way to facilitate infection of the plant. However, plant genes that are required by the pathogen for infection remain poorly described. To find such genes, we have compared the changes in gene express

Identification of allergens homologous to Alt a 1 from Stemphylium botryosum and Ulocladium botrytis.

Several studies have demonstrated that proteins homologous to the Alt a 1 major allergen of Alternaria alternata are expressed in other members of the Pleosporaceae family. However, since no direct biochemical data have been reported concerning the presence of Alt a 1 allergen homologues capable of binding IgE in the excretion-secretion products of Stemphylium and Ulocladium, our objective was to explore their presence in Stemphylium botryosum and Ulocladium botrytis. S. botryosum and U. botrytis culture filtrate extracts were analyzed by two-dimensional (2D)-electrophoresis and 2D-immunoblott

PanEuCOPT-01: Photodynamic Inactivation of fungal mycelial spheres of plant-pathogenic Botrytis cinerea.

This protocol describes an assay for Photodynamic Inactivation of the plant-pathogenic filamentous fungal species Botrytis cinerea. Grey mold (B. cinerea) occurs worldwide and affects over 200 different crops. Several resistant strains of the species have been collected on fields in past years. Here, mycelial spheres are used as a three-dimensional model to mimic the mold's hyphal structures. The protocol is intended for comparative evaluation of antifungal efficacy of different variables, such as different photosensitizers, concentration dependence, and illumination parameters.

Endophytic microorganisms from 'Bordô' grapes as biological control agents against Colletotrichum and Botrytis.

Colletotrichum spp. and Botrytis cinerea are the main causal agents of grape bunch rot in Brazil. Although chemical control is widely used, it has limitations, such as the selection of resistant pathogen populations and environmental concerns. Biological control emerges as a promising alternative; however, commercial products for viticulture remain limited. We hypothesized that by prospecting endophytic microorganisms from berries of Vitis hosts more resistant to rots, it would be possible to identify isolates with greater biocontrol potential. In this study, 52 endophytic isolates from berrie

Design, synthesis and antifungal activity of novel 1,3,4-Oxadiazole derivatives against Botrytis cinerea.

BACKGROUND: Botrytis cinerea, a highly destructive fungal pathogen with a broad host range, inflicts massive pre- and post-harvest losses on crops like tomatoes and blueberries, endangering global agricultural sustainability and food security. Escalating pesticide resistance reduces the efficacy of conventional fungicides, creating an urgent need for novel, efficient, low-toxicity antifungal agents. 1,3,4-oxadiazole derivatives possess diverse bioactivities, making them ideal candidates for development. RESULTS: Thirty novel 1,3,4-oxadiazole derivatives were rationally designed via scaffold h

The transcription factor ORA59 represses hypoxia responses during Botrytis cinerea infection and reoxygenation.

Transcription factors belonging to the large ethylene response factor (ERF) family are involved in plant responses to biotic and abiotic stresses. Among the ERFs, OCTADECANOID-RESPONSIVE ARABIDOPSIS 59 (ORA59) integrates ethylene and jasmonic acid signaling to regulate resistance to necrotrophic pathogens. The ERF group ERFVII encodes oxygen-labile proteins that are required for oxygen sensing and are stabilized by hypoxia established at the site of Botrytis (Botrytis cinerea) infection. Here, we show that ORA59 represses ERFVII protein activity to induce the expression of hypoxia-responsive g

Synthesis, Fungicidal Activity, and Structure Activity Relationship of β-Acylaminocycloalkylsulfonamides against Botrytis cinerea.

In order to discover new antifungal agrochemicals that could have highly active and novel motifs, thirty-six new 2-acylaminocycloalkylsulfonamides (IV) were synthesized. Their structures were characterized and confirmed by 1H NMR, 13C NMR, IR, MS, elemental analysis and X-ray single crystal diffraction. In vitro and in vivo activities against various Botrytis cinerea strains were evaluated. Bioassay results revealed that most of the title compounds exhibited excellent in vitro fungicidal activity, in which compound IV-26 showed the highest activity against sensitive, low-resistant, moderate-re

Characterisation of QoI-resistant field isolates of Botrytis cinerea from citrus and strawberry.

BACKGROUND: In 2004, field isolates of Botrytis cinerea Pers. ex Fr., resistant to strobilurin fungicides (QoIs), were first found in commercial citrus orchards in Wakayama Prefecture, Japan. Subsequently, QoI-resistant isolates of this fungus were also detected in plastic strawberry greenhouses in Saga, Ibaraki and Chiba prefectures, Japan. Biological and molecular characterisation of resistant isolates was conducted in this study. RESULTS: QoI-resistant isolates of B. cinerea grew well on PDA plates containing kresoxim-methyl or azoxystrobin at 1 mg L(-1), supplemented with 1 mM of n-propyl

Fungicide-Loaded Liposomes for the Treatment of Fungal Diseases in Agriculture: An Assessment of Botrytis cinerea.

In this work, liposomes loaded with the fungicide, Fludioxonil (FLUD), for the containment of fungal diseases in agriculture were developed. Three types of vesicles with different compositions were compared: (I) plain vesicles, composed of soy phosphatidylcholine and cholesterol; (II) PEG-coated vesicles, with an additional polyethylene glycol coating; and (III) cationic vesicles, containing didodecyldimethylammonium bromide. Nanometric-sized vesicles were obtained both by the micelle-to-vesicle transition method and by the extrusion technique, and encapsulation efficiency, drug loading conten

Structural and biosynthetic studies on eremophilenols related to the phytoalexin capsidiol, produced by Botrytis cinerea.

A thorough study of the fermentation broth of three strains of Botrytis cinerea which were grown on a modified Czapek-Dox medium supplemented with 5 ppm copper sulphate, yielded five undescribed metabolites. These metabolites possessed a sesquiterpenoid (+)-4-epi-eremophil-9-ene carbon skeleton which was enantiomeric to that of the phytoalexin, capsidiol. The isolation of these metabolites when the fungus was stressed, suggests that they may be potential effectors used by B. cinerea to circumvent plant chemical defences against phytopathogenic fungi. The biosynthesis of these compounds has bee

Characteristics of inositol phosphorylceramide synthase and effects of aureobasidin A on growth and pathogenicity of Botrytis cinerea.

Inositol phosphorylceramide (IPC) synthase is the key enzyme with highly conserved sequences, which is involved in fungal sphingolipid biosynthesis. The antibiotic aureobasidin A (AbA) induces the death of fungi through inhibiting IPC synthase activity. The mutations of AUR1 gene coding IPC synthase in fungi and protozoa causes a resistance to AbA. However, the mechanism of AbA resistance is still elusive. In this paper, we generated two mutants of Botrytis cinerea with AbA-resistance, BcAUR1a and BcAUR1b, through UV irradiation. BcAUR1a lost an intron and BcAUR1b had three amino acid mutation
