A time course study of cadmium effect on membrane permeability of single human bladder cancer cells using scanning electrochemical microscopy.

Cd(2+) is carcinogenic to both humans and experimental animals. We present quantitative time-course imaging of Cd(2+)-induced variation in the membrane permeability of single live human bladder cancer cells (T24) to ferrocenemethanol using scanning electrochemical microscopy (SECM). High temporal resolution combined with non-invasive nature renders a time-lapse SECM depth scan, a promising method to quantitatively investigate the effectiveness, kinetics, and mechanism of metal ions based on the responses of single live cells in real time. Under unstimulated conditions, T24 cells have constant

[Effect of dielectric permeability of culture media on the kinetics of the ATP hydrolysis reaction, catalyzed by Ca2+-transporting ATPase, solubilized from smooth muscle cell membrane].

Transport Ca2+,Mg(2+)-ATPase was solubilized from myometrial plasma membrane and purified by calmodulin-sepharose 4B affinity chromatography; the effect of dielectric permeability of incubation medium on kinetics of ATP hydrolysis by this enzyme was studied. Various concentrations of dimethylsulfoxide, ethanol, acetone, and dioxane (up to 10%) caused various dose-dependent inhibition of enzymatic ATP hydrolysis. The correlation between specific activity (A) of Ca2+,Mg(2+)-ATPase and dielectric permeability of the incubation medium (D) did not depend on the nature of organic solvents and was sa

Porphyromonas gingivalis outer membrane vesicles increase vascular permeability by inducing stress fiber formation and degrading vascular endothelial-cadherin in endothelial cells.

Porphyromonas gingivalis (Pg) is a keystone bacterium associated with systemic diseases, such as diabetes mellitus and Alzheimer's disease. Outer membrane vesicles (OMVs) released from Pg have been implicated in systemic diseases by delivering Pg virulence factors to host cells in distant organs and inducing cellular dysfunction. Pg OMVs also have the potential to enter distant organs via the bloodstream. However, the effects of Pg OMVs on the vascular function are poorly understood. Here, we showed that Pg OMVs increase vascular permeability by promoting stress fiber formation and lysosome/en

Heat shock transcription factor 2 reduces mitochondrial pathway apoptosis in intestinal epithelial cells by inhibiting the increase in mitochondrial membrane permeability in ulcerative colitis.

The destruction of intestinal mucosal mechanical barrier homeostasis caused by excessive apoptosis of intestinal epithelial cells (IECs) is an important reason for the occurrence and development of ulcerative colitis (UC). The increase in mitochondrial membrane permeability caused by the opening of the mitochondrial membrane permeability transition pore (mPTP) is a key link in the initiation of mitochondrial pathway apoptosis. Our previous studies revealed that heat shock transcription factor 2 (HSF2), which is highly expressed in the intestinal mucosa of UC patients, can inhibit the expressio

Influenza A virus infection of vascular endothelial cells induces GSK-3β-mediated β-catenin degradation in adherens junctions, with a resultant increase in membrane permeability.

Multiorgan failure with vascular hyperpermeability is the final outcome in the progression of seasonal influenza virus pneumonia and influenza-associated encephalopathy, and it is also common in infection with highly pathogenic avian influenza virus. However, the precise molecular mechanism by which influenza virus infection causes vascular endothelial cell hyperpermeability remains poorly defined. We investigated the mechanisms of hyperpermeability of human umbilical vein endothelial cells infected with influenza A virus (IAV)/Puerto Rico/8/34 (PR8) (H1N1). The levels of β-catenin, a key regu

Increased susceptibility of Pseudomonas aeruginosa to macrolides and ketolides in eukaryotic cell culture media and biological fluids due to decreased expression of oprM and increased outer-membrane permeability.

BACKGROUND: Macrolides show high minimum inhibitory concentrations (MICs) against Pseudomonas aeruginosa when tested in recommended media (cation-adjusted Muller-Hinton broth [CA-MHB]). Nevertheless, azithromycin is successfully used in cystic fibrosis patients, supposedly because of "nonantibiotic effects." METHODS: CA-MHB and Roswell Park Memorial Institute (RPMI) 1640 medium (used for growing eukaryotic cells) were compared for measuring azithromycin MICs (with or without Phe-Arg-β-naphthylamide [PAβN], an efflux inhibitor), [(14)C]-clarithromycin accumulation, azithromycin-induced protein

Inhibitory effects of zinc chloride (ZnCl2), n-acetyl-L-cysteine (NAC), and calcium/calmodulin dependent protein kinase II inhibitor (KN93) on Cd2+-induced abnormal cell morphology and membrane permeability.

Cadmium (Cd) could reduce abnormal cell morphology and membrane permeability, however, there are few studies on the detoxification of Cd-reduced cell membrane toxicity. In the present study, we firstly studied the effects of zinc chloride (ZnCl2), n-acetyl-L-cysteine (NAC), and calcium/calmodulin dependent protein kinase II inhibitor (KN93) on cell membrane permeability, respectively; then, we studied the inhibitory effects of ZnCl2, NAC, and KN93 on Cd2+-induced abnormal cell membrane permeability by scanning electrochemical microscopy (SECM) scanning imaging, transverse scanning curve and DP

Cell-permeable ceramides preferentially inhibit coated vesicle formation and exocytosis in Chinese hamster ovary compared with Madin-Darby canine kidney cells by preventing the membrane association of ADP-ribosylation factor.

Differential effects of acetyl(C2-) ceramide (N-acetylsphingosine) were studied on coated vesicle formation from Golgi-enriched membranes of Chinese hamster ovary (CHO) and Madin-Darby canine kidney (MDCK) cells. C2-ceramide blocked the translocation of ADP-ribosylation factor-1 (ARF-1) and protein kinase C-alpha (PKC-alpha) to the membranes from CHO cells, but not those of MDCK cells. Consequently, C2-ceramide blocked the stimulation of phospholipase D1 (PLD1) by the cytosol and guanosine 5'-[gamma-thio]triphosphate (GTP[S]) in membranes from CHO cells. Basal specific activity of PLD1 and the

The membrane permeable calcium chelator BAPTA-AM directly blocks human ether a-go-go-related gene potassium channels stably expressed in HEK 293 cells.

BAPTA-AM is a well-known membrane permeable Ca(2+) chelator. The present study found that BAPTA-AM rapidly and reversibly suppressed human ether a-go-go-related gene (hERG or Kv11.1) K(+) current, human Kv1.3 and human Kv1.5 channel currents stably expressed in HEK 293 cells, and the effects were not related to Ca(2+) chelation. The externally applied BAPTA-AM inhibited hERG channels in a concentration-dependent manner (IC(50): 1.3 microM). Blockade of hERG channels was dependent on channel opening, and tonic block was minimal. Steady-state activation V(0.5) of hERG channels was negatively shi

Exposure of tumor necrosis factor-alpha to luminal membrane of bovine brain capillary endothelial cells cocultured with astrocytes induces a delayed increase of permeability and cytoplasmic stress fiber formation of actin.

Tumor necrosis factor-alpha (TNF-alpha), a proinflammatory cytokine, has long been known to be involved in the pathogenesis of central nervous system infections and of certain neurodegenerative diseases. However, the possible role of the blood-brain barrier (BBB), the active interface between the blood circulation and brain tissue, remained unknown during these pathological conditions. In our in vitro reconstructed BBB model, 1-hr exposure of recombinant human TNF-alpha (in concentrations of 50, 250, and 500 U/ml, respectively) to the luminal membrane of bovine brain capillary endothelial cell

The effect of abnormal hemoglobins on the membrane regulation of cell hydration.

Several hemoglobinopathies are associated with abnormalities in the permeability of the red cell membrane, in some cases leading to permanent alterations of the intracellular milieu. Homozygous sickle cell disease is the most thoroughly studied example. Deoxygenation of sickle cells causes a transient increase in the permeability to monovalent cations and Ca; prolonged deoxygenation can lead to a permanent accumulation of Ca and loss of total cations and water. Although the mechanisms for the permeability changes are not yet defined, mechanical stress on the membrane, with subsequent damages b

Transport parameters in the human red cell membrane: solute-membrane interactions of hydrophilic alcohols and their effect on permeation.

A systematic study has been made of the three coefficients that describe the human red cell membrane transport of a series of short straight-chain hydrophilic alcohols: the permeability coefficient, omega i, the reflection coefficient, sigma i, and the hydraulic conductivity, Lp. Ethylene glycol transport is saturable with Km = 220 +/- 50 mM; there is a second, low-affinity, ethylene glycol site which inhibits water transport (K = 570 +/- 140 mM, max. inhib. = 90 +/- 10%). sigma eth gly = 0.71 +/- 0.04 which is significantly less than 1 (n = 6, P less than 0.001), as are sigma i for six other

Real-Time NanoBRET Target Engagement Reveals Permeability-Activity Relationships in BET-Targeting Degraders.

Membrane permeability is critical to the function of proteolysis targeting chimeras (PROTACs), yet common methods such as the parallel artificial membrane permeability assay (PAMPA) and Caco-2 transwell assays are limited by their experimental configuration and fail to directly measure a compound's ability to access the cytosolic space of a target cell. Here, we present a method to measure the membrane permeability of unmodified PROTACs and other E3 ligase ligands directly into the cytosol of living cells. By utilizing NanoBRET live-cell target engagement in real time, we quantify permeability

[Erythrocyte membrane permeability of sodium in different types of chronic experimental hypertension].

The permeability of the red blood cell membrane for sodium in rats with spontaneous genetic hypertension, in rats with renovascular and DOCA-salt-induced hypertension as well as the effect of high salt diet on the permeability were studied using the flame photometry method. The increase permeability of the red blood cells in the rats with spontaneous genetic hypertension was observed, while no changes occurred in the cases of DOCA-induced and renovascular hypertension. Salt load as well as deoxycorticosterone administration had no effect on the permeability of the red blood cell membrane.

Differential involvement of mitochondrial permeability transition in cytotoxicity of 1-methyl-4-phenylpyridinium and 6-hydroxydopamine.

Defects in mitochondrial function have been shown to participate in the induction of neuronal cell injury. The aim of the present study was to assess the influence of the mitochondrial membrane permeability transition inhibition against the toxicity of 1-methyl-4-phenylpyridinium (MPP(+)) and 6-hydroxydopamine (6-OHDA) in relation to the mitochondria-mediated cell death process and role of oxidative stress. Both MPP(+) and 6-OHDA induced the nuclear damage, the changes in the mitochondrial membrane permeability, leading to the cytochrome c release and caspase-3 activation, the formation of rea

MicroRNA-29a regulates intestinal membrane permeability in patients with irritable bowel syndrome.

BACKGROUND: The molecular mechanisms underlying the pathophysiology of irritable bowel syndrome (IBS) are poorly understood. One mechanism may involve increased intestinal permeability that is reversed with glutamine supplementation. Our goal was to evaluate the expression of glutamine synthetase and its complementary miRNA in blood microvesicles and gut tissues of IBS patients with increased intestinal membrane permeability. METHODS: We evaluated 19 diarrhoea-predominant IBS patients and 10 controls for intestinal membrane permeability using the lactulose/mannitol method. miRNA expression wa

Bacteriostatic mechanisms of CO2 against high-oxygen packaged meat-borne Pseudomonas fragi during chilled storage: Membrane physiological and microstructural changes.

The CO2 inside high-oxygen modified atmosphere packaging (HiOx-MAP) greatly retards the predominant Pseudomonas fragi (P. fragi) growth in chilled meat, whereas the CO2-regulated mechanism on P. fragi cell membrane properties is still unclear. This study investigated the influences of CO2 on both type and wild-type P. fragi growth and cell membrane characteristics during the chilled storage of HiOx-MAP (TMAP = 50% O2/40% CO2/10% N2; CMAP = 50% O2/50% N2). Compared to CMAP, TMAP significantly reduced P. fragi counts by limiting its motility and disturbing cell membrane structure, composition an

Cosurfactant-Induced Disorder in Polymersome Membrane Enhances Diffusion of Cargo Molecules.

Giant polymersomes are widely used as artificial cells due to their resemblance to natural cells; however, their intrinsically low membrane permeability remains a major limitation for applications such as drug delivery and bioreactors. Here, we report a microfluidic approach to fabricate permeable polymersomes using PB-b-PEO as a macromolecular surfactant and oleyl alcohol as a cosurfactant. Incorporation of the cosurfactant into the membrane induces membrane structural disorder in polymer packing, leading to enhanced membrane permeability. Using advanced nuclear magnetic resonance (NMR) spect

Recombinant Dengue virus protein NS2B alters membrane permeability in different membrane models.

BACKGROUND: One of the main phenomena occurring in cellular membranes during virus infection is a change in membrane permeability. It has been observed that numerous viral proteins can oligomerize and form structures known as viroporins that alter the permeability of membranes. Previous findings have identified such proteins in cells infected with Japanese encephalitis virus (JEV), a member of the same family that Dengue virus (DENV) belongs to (Flaviviridae). In the present work, we investigated whether the small hydrophobic DENV protein NS2B serves a viroporin function. METHODS: We cloned t
