Deletion of a branch-point consensus sequence in the LMX1B gene causes exon skipping in a family with nail patella syndrome.

Nail patella syndrome (NPS) has been shown to result from loss of function mutations within the transcription factor LMX1B. In a large NPS family a 17 bp intronic deletion encompassing a consensus branchpoint sequence was observed to segregate with the NPS phenotype. RNA analysis demonstrated that deletion of the branchpoint sequence resulted in skipping of the downstream exon. A mechanism to explain this phenomenon is presented.

Fingerprinting of mixed bacterial strains and BIOLOG gram-negative (GN) substrate communities by enterobacterial repetitive intergenic consensus sequence-PCR (ERIC-PCR).

PCR-based genomic fingerprinting by use of enterobacterial repetitive intergenic consensus primers (ERIC-PCR) was evaluated for its use in fingerprinting DNA of mixed Gram-negative bacterial strains and BIOLOG Gram-negative (GN) microplate substrate communities. ERIC-PCR fingerprints of six different pure bacterial strains and a combined mixture of the strains were compared with fingerprints obtained by two more established methods: amplified ribosomal DNA restriction analysis (ARDRA) and random amplified polymorphic DNA analysis (RAPD-PCR). The ERIC-PCR fingerprint of the mixed strains was hi

Design and immunogenicity evaluation of pan-recombinant vaccines based on the consensus sequence of severe acute respiratory syndrome coronavirus-2 spike protein.

Although multivalent vaccines are being developed to address the rapid mutations of severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2), universal vaccines are needed to protect against evolving variants and seasonal epidemics. We designed and evaluated the efficacy of three universal SARS-CoV-2 recombinant vaccine candidates based on a consensus sequence of spike protein from multiple variants. The candidates were designed with specific substitutions, including S-GSAS/6P and S-R/x2, to enhance immunogenicity by stabilizing the prefusion spike-protein conformation. Using murine models

High-risk genomic consensus validation for patients with newly diagnosed multiple myeloma using next-generation sequencing.

The prognostic heterogeneity of multiple myeloma is mainly driven by the genomic features of myeloma cells. The International Myeloma Society (IMS)/International Myeloma Working Group (IMWG) recently proposed a high-risk (HR) genomic model to have a consensus definition of genomic risk. We performed next-generation sequencing in the form of a panel on samples from 6528 patients with newly diagnosed multiple myeloma (NDMM) and 1583 patients at first relapse between 2019 and 2024. We observed that 22.4% of patients at diagnosis and 36.7% of patients at first relapse were classified as high risk

2025 update of the National French consensus on gene lists for the diagnosis of muscle diseases using high-throughput sequencing.

Diagnosing rare muscle diseases can be challenging due to their genetic heterogeneity. The French National Network for Rare Neuromuscular Diseases (FILNEMUS) has previously established a pioneering nationwide strategy based on gene lists organized in 13 phenotype-specific gene panels. We now revise these lists and add recently described genes. Using data collected from all FILNEMUS diagnostic laboratories, we also establish a "Major Muscle Genes" panel that includes genes responsible for the most frequent genetic muscle diseases. The updated diagnostic strategy of the FILNEMUS network will hel

[Rabies Virus Glycoprotein with a Consensus Amino Acid Sequence and a Lysosome Targeting Signal Causes Effective Production of Antibodies in DNA-Immunized Mice].

Safe and effective anti-rabies vaccines are intensely sought worldwide. DNA vaccines have already shown their efficacy and safety and have occupied a special place in the field. Two prototype anti-rabies DNA vaccines were compared for the potential to induce virus-specific antibody production. One vector contained a codon-optimized gene with a territory-adapted consensus sequence of the rabies virus glycoprotein. The other one expressed the same glycoprotein in fusion with a c-CD63 lysosome targeting motif at the C terminus. ELISA of serum samples from immunized mice showed that the c-CD63 var

A Possible Mechanism of Zika Virus Associated Microcephaly: Imperative Role of Retinoic Acid Response Element (RARE) Consensus Sequence Repeats in the Viral Genome.

Owing to the reports of microcephaly as a consistent outcome in the fetuses of pregnant women infected with ZIKV in Brazil, Zika virus (ZIKV)-microcephaly etiomechanistic relationship has recently been implicated. Researchers, however, are still struggling to establish an embryological basis for this interesting causal handcuff. The present study reveals robust evidence in favor of a plausible ZIKV-microcephaly cause-effect liaison. The rationale is based on: (1) sequence homology between ZIKV genome and the response element of an early neural tube developmental marker "retinoic acid" in human

A conserved nuclear receptor consensus sequence (DR-4) mediates transcriptional activation of the chicken CYP2H1 gene by phenobarbital in a hepatoma cell line.

Phenobarbital-responsive DNA elements were identified in the 5'-flanking region of the chicken CYP2H1 gene by in reporter gene assays in a chicken hepatoma cell line (leghorn male hepatoma (LMH)). A 264-base pair (bp) enhancer sequence (phenobarbital-responsive unit (PBRU)) responded to phenobarbital and a variety of phenobarbital-type inducers. Analysis of putative transcription factor binding sites within the 264-bp element revealed a nuclear receptor half-site repeat (DR-4) neighboring a putative nuclear factor-1 site. This motif resembles phenobarbital response elements in the flanking reg

[Expert consensus on the clinical application of Single-Molecule Real-Time Sequencing in the precise prevention and control of Thalassemia (2025 Edition)].

Thalassemia is a highly prevalent genetic hemoglobinopathy, with approximately 350 million people worldwide carrying variants of the globin genes. The carrier rate in southern China is as high as 10% ~ 25%. The "multi-level sequential screening and diagnosis" approach, which uses hematological screening, hotspot mutation testing, and genetic testing based on other technologies to diagnose rare types of thalassemia, has proven to be highly effective for the prevention and control of thalassemia. However, its cumbersome process, high cost for testing and medical labor, and high demand for geneti

Meta-analysis and multidisciplinary consensus statement: exome sequencing is a first-tier clinical diagnostic test for individuals with neurodevelopmental disorders.

PURPOSE: For neurodevelopmental disorders (NDDs), etiological evaluation can be a diagnostic odyssey involving numerous genetic tests, underscoring the need to develop a streamlined algorithm maximizing molecular diagnostic yield for this clinical indication. Our objective was to compare the yield of exome sequencing (ES) with that of chromosomal microarray (CMA), the current first-tier test for NDDs. METHODS: We performed a PubMed scoping review and meta-analysis investigating the diagnostic yield of ES for NDDs as the basis of a consensus development conference. We defined NDD as global dev

Advanced endometrial cancer in the gulf cooperation council: a modified-Delphi consensus on unmet needs, molecular testing, and treatment sequencing.

BACKGROUND: Advanced endometrial cancer (EC), including primary unresectable stage III-IV, metastatic, and recurrent disease, is an increasing clinical challenge in the Gulf Cooperation Council (GCC). Rising cardiometabolic risk, heterogeneous referral pathways, variable access to molecular diagnostics, and inconsistent uptake of biomarker-driven therapy contribute to variation in care. This consensus aimed to identify GCC-specific unmet needs and develop regionally adapted recommendations for the diagnosis and management of advanced or recurrent EC. METHODS: A structured, modified Delphi pro

Efficient reconstruction of full-length RNA isoforms using ISAtools and large-scale PacBio circular consensus sequencing data.

Accurate reconstruction and quantification of full-length RNA isoforms remain challenging in long-read RNA sequencing due to sequencing artifacts, complex splicing, and incomplete annotations. Although Pacific Biosciences circular consensus sequencing (PacBio CCS) provides high-fidelity long reads, scalable and annotation-flexible analysis frameworks remain limited. Here, we present ISAtools, an efficient framework specifically designed for PacBio CCS data. ISAtools introduces a splice site chain representation that unifies read alignments and transcript annotations into a compact format for s

Comparative Genomics of Herpes Simplex Virus 2 Isolated from a Maternal-Neonatal Dyad Reveals High Consensus Sequence Homology as Well as Minor Variant Diversity.

Neonatal herpes simplex virus disease most often occurs following mother-to-neonate transmission during childbirth. However, genomic differences between the maternal and neonatal viral populations are poorly understood. Here, we performed deep whole-genome sequencing of clinical HSV-2 samples obtained from a maternal-neonatal dyad to investigate viral population dynamics during transmission.

Molecular basis of group A xeroderma pigmentosum: a missense mutation and two deletions located in a zinc finger consensus sequence of the XPAC gene.

The molecular basis of group A xeroderma pigmentosum (XP) was investigated, and 3 mutations located in a zinc finger consensus sequence (nucleotide 313-387) of the XP group A complementing (XPAC) gene were identified in 2 Caucasian patients GM2990 and GM2009 who had typical symptoms of group A XP. The first mutation was a C deletion at nucleotide 374. Patient GM2990 was a homozygote for this mutation. The second mutation was a 5-bp deletion (CTTAT) at nucleotides 349-353. The third mutation was a G to T transversion at nucleotide 323 that alters the Cys-108 codon (TGT) to a Phe codon (TTT). Pa

Surveillance of monochorionic twins for detection of twin-twin transfusion syndrome and twin anemia polycythemia sequence: a North American Fetal Therapy Network consensus statement.

The Society for Maternal-Fetal Medicine recently released updated clinical guidelines on the ultrasound surveillance and clinical management of monochorionic twin pregnancies. The North American Fetal Therapy Network has supported but has not endorsed these guidelines. In this document, we suggest an enhanced ultrasound surveillance strategy for timely detection of twin-twin transfusion syndrome and twin anemia polycythemia sequence in monochorionic pregnancies. We provide specific recommendations that depart from the Society for Maternal-Fetal Medicine guidelines. The rationale for these reco

An Arg760Cys mutation in the consensus sequence of the von Willebrand factor propeptide cleavage site is responsible for a new von Willebrand disease variant.

We describe a von Willebrand disease (VWD) variant characterized by the persistence of von Willebrand factor (VWF) propeptide as a result of a C>T transition at nucleotide 2527 in exon 17 of the VWF gene. This mutation, which was present in the proband and his father, predicts the substitution of Cys for Arg at position 760 of pre-pro-VWF, 4 residues before the propeptide cleavage site belonging to a consensus sequence for substrate recognition by the processing enzyme paired dibasic amino acid-cleaving enzyme (PACE)/furin. The promoter of a novel human papillomavirus (HPV77) associated with skin cancer displays UV responsiveness, which is mediated through a consensus p53 binding sequence.

An aetiological role has been proposed for human papillomavirus (HPV) in skin carcinogenesis within the immunosuppressed patient population. To examine this possibility, we have focused on an HPV type that, to date, has been identified only in the cutaneous lesions of renal transplant recipients despite a high degree of sequence homology with other HPVs commonly found in warts in the general population. We report that the non-coding region of this virus, HPV type 77, contains a consensus binding site for the tumour suppressor protein p53, and we show by gel-retardation analysis that this seque
