Modulation of cyclic AMP in purified rat mast cells. II. Studies on the relationship between intracellular cyclic AMP concentrations and histamine release.

Changes in intracellular and extracellular rat mast cell adenosine 3':5' monophosphate (cAMP) concentrations during stimulation of histamine release by 48/80 were studied. There was a rapid and progressive fall in intracellular cAMP beginning within 10 sec after the addition of 48/80. The lowest cAMP values were obtained at 10 min, with return to control levels by 30 min. The fall in cAMP was dose-related with progressive decreases in 10-min cAMP measurements as the 48/80 concentration was increased from 0.25 to 1.00 mug/ml. There was a graded increase in histamine release over the same concen

Human leukocyte cyclic AMP and cyclic GMP levels during chemotaxis in delayed type hypersensitivity.

Ten nickel-allergic patients and six healthy control subjects participated in a study of the morphology, kinetics and evolution of the cAMP and cGMP concentrations of migrated leukocytes, using an improved skin chamber technique. Also studied was the effect of nickel exposure in the chamber medium during development of an eczematous reaction in the nickel-allergic patients. Nickel exposure had a specific effect on the morphology, from the 24th hour to the end of the 48 h observation period, with a significant increase in the percentages of basophils, eosinophils and lymphocytes and a decrease

Cyclic di-AMP signaling and regulation in Staphylococcus aureus.

Cyclic di-AMP (c-di-AMP) is an essential bacterial second messenger that coordinates multiple cellular processes and is closely linked to central physiology in many bacteria. In Staphylococcus aureus, both depletion and excessive accumulation of c-di-AMP impair bacterial fitness, indicating that this signaling pathway must be tightly regulated. Recent studies have identified c-di-AMP receptor proteins in S. aureus, revealing how c-di-AMP maintains osmotic balance through ion transport and influences cell envelope function and stress adaptation. These regulatory effects also influence bacterial

Protective effect of cyclic AMP against cisplatin-induced nephrotoxicity.

We reported earlier that reactive oxygen species are implicated in necrotic injury induced by a transient exposure of cultured renal tubular cells to a high concentration of cisplatin but not in apoptosis occurring after continuous exposure to a low concentration of cisplatin. We report here the protective effect of cyclic AMP against cisplatin-induced necrosis in cultured renal tubular cells as well as cisplatin-induced acute renal failure in rats. Several pharmacological agents that stimulate cyclic AMP signaling, including the nonhydrolyzable cyclic AMP analogue dibutyryl cyclic AMP, forsko

Hypoxanthine effects on cyclic AMP levels in human lymphocytes.

We have measured hypoxanthine effect on cAMP levels in PBL in basal conditions (no agonist), and with the addition of 2-(p-[2-carboxyethyl] phenylethylamino)-5'-N-ethylcarboxamidoadenosine (CGS-21680, a specific A2 receptor agonist). We have found that hypoxanthine, at 25 microM and 50 microM concentrations, increases cAMP levels in PBL in basal and A2 agonist stimulated conditions.

Cyclic AMP and anionic currents in porcine ciliary epithelium.

INTRODUCTION: To investigate whether in the ciliary epithelium of isolated porcine ciliary body cyclic 3',5' adenosine monophosphate (cAMP) activates transmembrane anionic currents. METHODS: Changes in membrane potential induced either by the adenylcyclase activator forskolin (10 microM; n = 4) or the stable membrane permeable cAMP analog 8-bromo-adenosine 3',5'-cyclic monophosphothioate (8-br-cAMP; 30 microM; n = 4) were measured with intracellular microelectrodes. The effect of the drugs were assessed in the absence or in the presence of the non-selective anionic channel/transporter inhibit

Cyclic AMP-regulating agents inhibit endotoxin-mediated cartilage degradation.

The influence of cyclic AMP on cartilage degradation was investigated by using phosphodiesterase inhibitors [theophylline and 3-isobutyl-1-methylxanthine (IBMX)], forskolin (which activates the catalytic subunit of adenylate cyclase) and cyclic AMP analogues (dibutyryl and 8-bromo). Breakdown was assessed by quantification of proteoglycans released into the media of 8-day bovine nasal-septum cartilage cultures. Theophylline (1-20 mM), IBMX (0.01-2 mM) and dibutyryl cyclic AMP (0.1-2 mM) had little or no influence on the rate of proteoglycan release from unstimulated (no-endotoxin) cartilages.

Cyclic AMP enhances progesterone action in human myometrial cells.

Cyclic AMP (cAMP) has been shown to promote progesterone and glucocorticoid action in a variety of cellular settings. In this study, we have used human myometrial cells to investigate whether cAMP potentiates the ability of progesterone to repress IL-1β-driven COX-2 expression. We found that forskolin enhanced progesterone-repression of IL-1β-driven COX-2 expression in association with delayed IL-1β-induced nuclear phospho-p65 entry and reduced NF-κB binding to the COX-2 promoter. Further, forskolin enhanced the progesterone-induced expression of FKBP5 and 11βHSD1, progesterone-driven activity

Panic disorder patients have reduced cyclic AMP in platelets.

Little is known about the intracellular mechanisms involved in the pathophysiology of panic disorder (PD). Abnormalities in the cyclic AMP system have been described in several psychiatric disorders but there are no studies in panic patients. We evaluated not only the levels of platelet cyclic AMP, but also cyclic GMP and nitric oxide synthase (NOS) activity in patients with PD at baseline and after treatment with clomipramine and in healthy volunteers. Platelet cyclic AMP was determined by enzymeimmunoassay, cyclic GMP by radioimmunoassay and NOS activity by the conversion of (3)H-arginine to

Cyclic AMP-dependent signaling aberrations in macronodular adrenal disease.

The adrenal glands are a major source of steroid hormone biosynthesis. In normal physiology, the pituitary hormone corticotropin (ACTH) regulates the secretion of glucocorticoids via its G protein-coupled receptor (ACTHR), the product of the MC2R gene. Aldosterone is another major product of the adrenal gland; its regulation is controlled mainly by the renin-angiotensin system, although ACTH plays a role, too, especially under certain pathological conditions. The adrenal gland also secretes lesser amounts of androgens and intermediate metabolites of all these steroids. Unregulated secretion of

Mechanism of cyclic AMP-induced hyperpolarization in canine colon.

The mechanism of forskolin (FSK)-induced hyperpolarization was investigated in strips of canine colonic circular muscle. FSK responses were compared to those of the K+ channel opener lemakalim (LEM). Both FSK (10 microM) and LEM (10 microM) hyperpolarized cells near the myenteric border by 10 to 20 mV. Responses to both agents were abolished by 35 mM external K+, indicating a probable mediation by K+ channels. FSK increased the open probability of Ca(++)-activated K+ channels in isolated colonic myocytes. However, in muscle strips charybdotoxin (100 nM) and tetraethylammonium (10 mM) failed to

Ca(2+)-cyclic AMP interactions in sustained cellular responses.

As early as 1970 it was apparent that the cyclic AMP (cAMP) and Ca2+ messenger systems often interact to regulate cellular responses. Work over the past 20 years has greatly expanded our knowledge of these interactions, and has shown that these signalling systems interact in complex ways to regulate sustained cellular responses such as aldosterone secretion, smooth muscle contraction and insulin secretion. The latter system is considered in detail because it illustrates several types of interactions, both positive and negative, which help to determine the normal response of beta-cells to physi

Increases in plasma cyclic AMP dependent on endogenous catecholamines.

Administration of tyramine (with or without phentolamine) as well as induction of ether anesthesia or insulin hypoglycemia caused a sharp increase in plasma cyclic AMP in rats. Based on the findings that the treatment of rats with reserpine, 6-hydroxydopamine, cocaine or propranolol totally abolished tyramine-induced increases in plasma cyclic AMP, it was concluded that catecholamines released from sympathetic neuronal terminals by tyramine could activate adenylate cyclase via the stimulation of postsynaptic beta-adrenoceptors. In contrast, catecholamines secreted from adrenal medulla were lar

Cerebrospinal fluid concentration of cyclic AMP in cerebrovascular diseases.

Concentration of cyclic adenosine 3',5'-monophosphate (cAMP) and activities of some enzymes were measured in cerebrospinal fluid (CSF) from 38 patients with various cerebrovascular diseases. Cerebral infarction of the carotid area (less than 14 days after the attack) revealed a significant increase in CSF cAMP level in comparison to a transient ischemic attack (TIA) and cephalagia without any pathological findings (control group). A trend towards elevated values was observed also in cerebral hemorrhage, whereas the CSF cAMP concentrations in subarachnoid hemorrhage, TIA, syncope, and cerebral

[Variations in cyclic AMP level and specific activities of adenylate cyclase and cyclic AMP phosphodiesterase during the cell cycle of an Actinomycete (author's transl)].

The variations in the concentrations of intra- and extracellular cyclic AMP and in he specific activities of adenylate cyclase (EC 4.6.1.1) and cyclic AMP phosphodiesterase (EC 3.1.4.17) have been monitored in synchronized cultures of Nocardia restricta, a prokaryote belonging to the group of Actinomycetes. At the beginning of the cell cycle, during a first period of RNA and protein synthesis, there is an increasing synthesis of adenylate cyclase which can be suppressed in the presence of chloramphenicol or rifampicin. Simultaneously, the specific activity of cyclic AMP phosphodiesterase decre

Inhibitory effect of methacholine on drug-induced relaxation, cyclic AMP accumulation, and cyclic AMP-dependent protein kinase activation in canine tracheal smooth muscle.

Functional antagonism between bronchoconstricting and bronchodilating pathways was examined in canine tracheal smooth muscle. Trachealis strips were contracted with either 0.3 microM (EC55) or 3.0 microM (EC80) methacholine before being relaxed by the cumulative addition of isoproterenol, prostaglandin E2, or forskolin. The EC50 for all three relaxants was increased 10-fold in tissues contracted with 3.0 microM methacholine vs. those contracted with 0.3 microM methacholine. Moreover, contracting tissues with the higher concentration of methacholine reduced the maximum relaxation induced by pro

Vasopressin Expression in Cultured Neurons is Stimulated by Cyclic AMP.

Abstract Vasopressin and oxytocin genes are expressed in mutually exclusive sets of magnocellular neurons in the hypothalamus. Cell specificity and regulation are probably controlled by extra- and intracellular signals acting on one or the other gene. In order to identify factors that regulate peptide expression, we have used primary dissociated cultures derived from 14-day old foetal rats. Vasopressin expression was monitored by combined immunocytochemistry and in situ hybridization. Treatment of cultures with forskolin and/or the phosphodiesterase inhibitor 3-isobutyl-1-methylxanthine (IBMX)

Effect of cyclic AMP on collagen production by corneal fibroblasts.

Subcultured bovine corneal fibroblasts accumulated cyclic AMP intracellularly and in the culture media in response to incubation with 3-isobutyl-1-methylxanthine, isoproterenol, cholera toxin or forskolin. The duration of the intracellular cyclic AMP effect was rather short using isoproterenol (less than three hours) but cholera toxin and forskolin maintained cyclic AMP stimulations for 24 hours at greater than 3-fold and 27-fold over control, respectively. Agents which stimulated adenylate cyclase by different mechanisms or mimicked the action of cyclic AMP decreased collagen secretion by the
