Crohn's disease: a defensin deficiency syndrome?

This comprehensive review promotes the novel concept that a defensin deficiency, i.e. lack of mucosal peptide antibiotics, may play a pivotal role in the aetiopathogenesis of Crohn's disease. Such an impaired function of this chemical barrier is consistent with the epidemiological relationship of good domestic hygiene with the incidence of inflammatory bowel diseases. The disregulated adaptive immune system, formerly believed to be the major cause in the development of Crohn's disease, may reflect only the primary break of the mucosal defence since the immune response is mostly directed agains

Immunohistochemical study on the distribution of β-defensin 1 and β-defensin 2 throughout the respiratory tract of healthy rats.

The distributions of β-defensin 1 and 2 in secretory host defense system throughout respiratory tract of healthy rats were immunohistochemically investigated. In the nasal epithelium, a large number of non-ciliated and non-microvillous cells (NCs) were immunopositive for both β-defensin 1 and 2, whereas a small number of goblet cells (GCs) were immunopositive only for β-defensin 1. Beta-defensin 2-immunopositive GCs were few. In the nasal glands, a small number of acinar cells and a large number of ductal epithelial cells were immunopositive for both β-defensins. In the laryngeal and tracheal

α-defensin 1-3 and α-defensin 4 as predictive markers of imatinib resistance and relapse in CML patients.

OBJECTIVE: Imatinib mesylate is a tyrosine kinase inhibitor used as first line treatment in chronic myeloid leukaemia. Despite a remarkable effectiveness, treatment failure cases have been reported in 20 percent of CML patients. The identification of biomarkers which can predict the response to imatinib is our point of interest. METHODS: Gene expression profiling microarray was carried out on secondary imatinib resistant patients. Longitudinal studies were performed on imatinib treated responder/resistant patients. Then, Q-RT/PCR studies were realized on patients prior imatinib initiation. R

Expression of the peptide antibiotics human beta defensin-1 and human beta defensin-2 in normal human skin.

Normal human skin is remarkably resistant to infection from the large numbers of microorganisms that routinely colonize its surface. In addition to the role of skin as a mechanical barrier, it has long been recognized that skin and other epithelia can produce a range of anti-microbial chemicals that play an important part in eliminating potential cutaneous pathogens. Anti-microbial peptides are an important evolutionarily conserved innate host defense mechanism in many organisms. Human beta defensin-1 and -2 are cysteine-rich, cationic, low molecular weight anti-microbial peptides that have re

Purification and characterization of a scorpion defensin, a 4kDa antibacterial peptide presenting structural similarities with insect defensins and scorpion toxins.

Insect defensins are a group of inducible small-sized antibacterial peptides with three intramolecular disulfide bridges. NMR studies have recently shown that they share striking structural similarities with scorpion toxins. We have investigated in a scorpion species, Leiurus quinquestriatus, the potential presence of antibacterial molecules and report the isolation and structural characterization of a novel insect defensin homologue, which we refer to as scorpion defensin. This peptide shows a remarkably high degree of sequence homology with a defensin recently characterized in a species belo

Engineering defensins: Heterologous expression strategies from genetic engineering to synthetic biology.

Defensins are a structurally diverse class of host defense peptides with broad-spectrum antimicrobial and immunomodulatory activities. Their broader applications in biomedicine and biotechnology, however, remain constrained by limited natural availability and difficulties associated with heterologous production, including low yields, host toxicity, inefficient folding, and loss of biological activity. In this review, we examine defensin production from an integrated engineering perspective, emphasizing the interplay between expression regulation, host physiology, and molecular design. We estab

Antiplasmodial activity of tick defensins in a mouse model of malaria.

Malaria is a mosquito-borne disease affecting millions of people mainly in Sub-Saharan Africa, Asia and some South American countries. Drug resistance to first-line antimalarial drugs (e.g. chloroquine, sulfadoxine-pyrimethamine and artemisinin) is a major constrain in malaria control. Antimicrobial peptides (AMPs) have shown promising results in controlling Plasmodium spp. parasitemia in in vitro and in vivo models of infection. Defensins are AMPs that act primarily by disrupting the integrity of cell membranes of invasive microbes. We previously showed that defensins from the tick Ixodes ric

Follicular fluid expression of alpha-defensins and their role in ovulation.

INTRODUCTION: Defensins are cytotoxic peptides and have a well-defined role in host defense. Human alpha defensins 1-3 (HNP1-3) are primarily produced by peripheral neutrophils and constitute about 50% of the azurphil granule protein. Studies have suggested that peripheral neutrophils and the resident neutrophils in the ovary enhance the release of IL-8 and TNF-alpha that play a role in ovulation and influence fertilisation rate and IVF outcome. The production of HNP1-3 by follicular fluid and its role in ovulation has never studied. The aim of this study was to demonstrate the presence of HNP

Interaction of human defensins with Escherichia coli. Mechanism of bactericidal activity.

Defensins are small, cysteine-rich antimicrobial peptides that are abundant in human, rabbit, and guinea pig neutrophils (PMN). Three defensins (human neutrophil peptide defensin [HNP]-1, HNP-2, and HNP-3) constitute between 30 and 50% of the total protein in azurophil granules of human PMN. We examined the mechanism of HNP-mediated bactericidal activity against Escherichia coli ML-35 (i-, y-, z+) and its pBR322-transformed derivative, E. coli ML-35p. Under conditions that supported bactericidal activity, HNP-1 sequentially permeabilized the outer membrane (OM) and inner membrane (IM) of E. co

Differential Expression of Putative Ornithodoros turicata Defensins Mediated by Tick Feeding.

Additional research on soft ticks in the family Argasidae is needed to bridge the knowledge gap relative to hard ticks of the family Ixodidae; especially, the molecular mechanisms of Ornithodoros biology. Ornithodoros species are vectors of human and animal pathogens that include tick-borne relapsing fever spirochetes and African swine fever virus. Soft tick vector-pathogen interactions involving components of the tick immune response are not understood. Ticks utilize a basic innate immune system consisting of recognition factors and cellular and humoral responses to produce antimicrobial pept

A Centipede Toxin Family Defines an Ancient Class of CSαβ Defensins.

Disulfide-rich peptides (DRPs) play diverse physiological roles and have emerged as attractive sources of pharmacological tools and drug leads. Here we describe the 3D structure of a centipede venom peptide, U-SLPTX15-Sm2a, whose family defines a unique class of one of the most widespread DRP folds known, the cystine-stabilized α/β fold (CSαβ). This class, which we have named the two-disulfide CSαβ fold (2ds-CSαβ), contains only two internal disulfide bonds as opposed to at least three in all other confirmed CSαβ peptides, and constitutes one of the major neurotoxic peptide families in centipe

Defensins enable macrophages to inhibit the intracellular proliferation of Listeria monocytogenes.

Listeria monocytogenes is a facultative intracellular pathogen that infects a large diversity of host cells, including macrophages. To avoid the phagosome microbicidal environment, L. monocytogenes secretes a pore-forming toxin (listeriolysin O, LLO) that releases the bacterium into the cytoplasm. We hypothesized that the α-defensins (HNPs) and/or humanized θ-defensin (RC-1) peptides produced by human and non-human primate neutrophils, respectively, cooperate with macrophages to control L. monocytogenes infection. Our results establish that HNP-1 and RC-1 enable macrophages to control L. monoc

Human beta-defensins--at the front line of the peritonsillar abscess.

Antimicrobial proteins like human beta-defensins 1-4 (hBD1-4) protect the surface of organs against different bacteria. Little is still known about these proteins within an abscess formation. The purpose of this study was to analyse and describe the distribution of the antimicrobial proteins hBD 1-4 within the peritonsillar abscess. A total of 17 peritonsillar abscesses were analysed. Immunohistochemical stainings were performed, characteristic pictures were taken, and the mean colour intensity was measured using a specific imaging software. A statistical analysis compared the areas of interes

Discovery and characterization of new epididymis-specific beta-defensins in mice.

The male urogenital tract epithelium is exposed to several pathogens, but only a few are potent enough to cause infection in a healthy individual. The exact mechanisms that protect the male reproductive tract from ascending pathogenic micro-organisms are still poorly characterized. We recently reported a method to identify novel epididymis-specific genes by analyzing the expressed sequence tags (ESTs) present in the mouse epididymal cDNA library of the UniGene collection at National Center for Biotechnology Information (NCBI). In the present study, we discovered in silico two novel epididymal

Molecular cloning and characterization of three beta-defensins from canine testes.

Mammalian beta-defensins are small cationic peptides possessing broad antimicrobial and physiological activities. Because dogs are particularly resilient to sexually transmitted diseases, it has been proposed that their antimicrobial peptide repertoire might provide insight into novel antimicrobial therapeutics and treatment regimens. To investigate this proposal, we cloned the full-length cDNA of three canine beta-defensin isoforms (cBD-1, -2, and -3) from canine testicular tissues. Their predicted peptides share identical N-terminal 65-amino-acid residues, including the beta-defensin consens

Expression of alpha- and beta-defensins in human ocular surface tissue.

PURPOSE: Defensins are antimicrobial peptides and are classified into three groups, alpha-, beta- and theta-defensins. We investigated the localization of human neutrophil peptides (HNP) 1-3, which are members of the alpha-defensin subfamily, and the expression of human beta-defensin 1 (hBD-1) and hBD-2 in the ocular surface tissues. METHODS: Corneal or conjunctival tissues were obtained from 17 patients undergoing surgery. The reasons for surgery were thermal burns (two patients), acid burn (one patient), corneal perforation (one patient), corneal abscess (one patient), and pterygium (two pa
