Effect of Immediate Dentin Sealing on the Bond Strength of Resin Cement to Diabetic Dentin.

This in vitro study evaluated the shear bond strength (SBS) of resin cement to diabetic and non-diabetic dentin following immediate dentin sealing (IDS) or conventional bonding without IDS, under two cementation protocols: immediate cementation (IC) and one-week delayed cementation (DC) after provisionalization. From a biomimetic perspective, IDS represents a tissue-preserving adhesive approach aimed at maintaining a stable resin-dentin interface; however, systemic conditions such as diabetes mellitus may alter the dentin substrate and affect adhesive performance. A total of 120 extracted huma

Effect of bioactive glasses used as dentin desensitizers on the dentin-pulp complex in rats.

Bioactive glasses have been recommended for the occlusion of dentinal tubules in treating cervical dentin hypersensitivity. This study evaluates an in vivo model of dentin exposure, and tests the efficacy of bioglass treatments. Thirty male Wistar rats received gingival recession surgery on the upper left first molar. The treatments were applied over the surface of the exposed dentin every 4 days for 28 days. The groups were as follows: Naive; Gingival recession; Cavity varnish; Biosilicate®; Strontium bioglass; and Potassium bioglass. Changes in the dentin-pulp complex, and the presence of su

Protein S100-A7 Derived from Digested Dentin Is a Critical Molecule for Dentin Pulp Regeneration.

Dentin consists of inorganic hard tissue and organic dentin matrix components (DMCs). Various kinds of bioactive molecules are included in DMCs and some of them can be released after digestion by endogenous matrix metalloproteinases (MMPs) in the caries region. Digested DMCs induced by MMP20 have been reported to promote pulpal wound healing processes, but the released critical molecules responsible for this phenomenon are unclear. Here, we identified protein S100-A7 as a critical molecule for pulpal healing in digested DMCs by comprehensive proteomic approaches and following pulp capping expe

Transgenic expression of dentin phosphoprotein (DPP) partially rescued the dentin defects of DSPP-null mice.

Mutations in the dentin sialophosphoprotein (DSPP) gene cause dentinogenesis imperfecta. After synthesis, DSPP is proteolytically processed into NH2- and COOH-terminal fragments. The NH2-terminal fragment of DSPP is highly glycosylated but not phosphorylated, whereas the COOH-terminal fragment (named "dentin phosphoprotein" or "DPP") is highly phosphorylated but not glycosylated. These two fragments are believed to perform distinct roles in dentin formation. To analyze the functions of DPP in dentinogenesis, we created "Dspp-/-;DPP Tg mice", which expressed transgenic DPP driven by a Type I co

Calcium Sodium Phosphosilicate in Management of Dentin Hypersensitivity and Strategies for Reconstitution of the Dentin Matrix.

Dentin hypersensitivity (DH) prevalence ranges from 72.5%-98% and is common from the third decade of life. Despite the variety of products available, dentine hypersensitivity remains a persistent issue. From the vast array of existing products, ceramics specifically Calcium sodium phosphosilicate (CSPS) based glasses seem to be the most versatile material to manage dentin hypersensitivity. Literature clearly indicates that Calcium sodium phosphosilicate (CSPS) permits a multitude of possibilities as it allows integration with soft and hard tissues. The ability to generate a flexible structure

Aspects of dentinal and pulpal pain. Pain of dentinal and pulpal origin--a review for the clinician.

Recent advances in understanding the mechanisms of pain arising from the dental pulp serve to benefit patients by improving the clinician's ability to diagnose and treat pain. There are two types of pain arising from the pulp which are mediated by entirely different nerve fibres, each with their own individual characteristics. One is a short, sharp fast pain which is induced by stimuli which cause a rapid fluid flow within the dentinal tubules. Such stimuli include cold, heat, air, drilling, and osmotic stimuli. Once the affected teeth are identified, they can often be treated by sealing the o

A silk fibroin-based hydrogel desensitizer achieving 660 μm dentin tubule occlusion for dentin hypersensitivity treatment.

Dentin hypersensitivity (DH) causes sharp pain due to external stimuli transmitted through dentinal tubules (DTs) in exposed dentin. Current desensitizers struggle to provide sufficient DT occlusion depth and density, limiting long-term effectiveness under daily erosion and friction. Inspired by fish schooling behavior, we introduce an adaptive hydrogel desensitizer (STU) composed of "sardines" (tannic acid and silk fibroin) and "predators" (urea). The "sardines" form a tightly packed hydrogel "shoal" through hydrogen bonds, while the "predators" disrupt these bonds, transforming STU into a lo

Mechanism of dentin hardness measurements using a dentin hardness measuring device with a light-emitting diode.

SIGNIFICANCE: Managing caries is imperative in a rapidly aging society. Current diagnoses use qualitative indices. However, a quantitative evaluation of hardness in a clinical setting may lead to more accurate diagnoses. Previously, hardness meter using indenter with light for tooth monitoring (HAMILTOM) was developed to quantitatively measure tooth hardness. Herein, the physical interpretation of dentin hardness measured using HAMILTOM and the dentin hardness measurement mechanism are discussed. AIM: This study evaluates the mechanism of dentin hardness measurements using HAMILTOM physically

Biomimetic oligopeptide formed enamel-like tissue and dentin tubule occlusion via mineralization for dentin hypersensitivity treatment.

OBJECTIVE: Dentin hypersensitivity (DH) is a common oral disease with approximately 41.9% prevalence. Reconstruction of dental hard tissues is the preferred treatment for relieving DH. Here, we applied biomineralization method using oligopeptide simulating cementum protein 1 (CEMP1) to regenerate hard tissues on demineralized dentin. METHODS: The self-assembly and biomineralization property of the oligopeptide were detected by scanning electron microscopy (SEM), circular dichroism spectroscopy, and transmission electron microscopy. Oligopeptide's binding capacity to demineralized dentin was e

Dentine hypersensitivity: effect of interactions between metal salts, fluoride and chlorhexidine on the uptake by dentine.

A large number of compounds may be applied to the teeth for preventive or therapeutic purposes, most notably in dentine hypersensitivity. The uptake of any one compound by tooth substances may clearly be affected by another to produce synergistic, additive or antagonistic effects. This study determined whether uptake interactions occurred between fluoride, chlorhexidine, strontium, tin and zinc. All compounds, in particular tin, showed considerable affinity for dentine. Fluoride reduced the uptake of zinc but not tin, strontium or chlorhexidine. Chlorhexidine increased the uptake of zinc, but

Cysteine cathepsins in human carious dentin.

Matrix metalloproteinases (MMPs) are important in dentinal caries, and analysis of recent data demonstrates the presence of other collagen-degrading enzymes, cysteine cathepsins, in human dentin. This study aimed to examine the presence, source, and activity of cysteine cathepsins in human caries. Cathepsin B was detected with immunostaining. Saliva and dentin cysteine cathepsin and MMP activities on caries lesions were analyzed spectrofluorometrically. Immunostaining demonstrated stronger cathepsins B in carious than in healthy dentin. In carious dentin, cysteine cathepsin activity increased

Radiological features of hereditary opalescent dentin.

A case of hereditary opalescent dentin is described, which showed the same clinical and radiological features as those reported previously. However, there was no evidence of osteogenesis imperfecta. While a diagnosis of type II dentinogenesis imperfecta was therefore made, this case also had features characteristic of type III dentinogenesis imperfecta. Treatment by means of an overdenture is described.

Reparative dentin: factors affecting its deposition.

Results of this study showed no correlation between the thickness or amount of reparative dentin deposited and the type of dental restorative material placed at either 5 or 8 weeks in controlled Class V cavity preparations in monkey teeth. Factors such as preparation trauma from the bur, operator hand instrumentation, and microleakage of bacterial toxins played a greater role in the stimulation of reparative dentin than did material irritation or toxicity. Some differences in the thickness of the reparative dentin deposited were noted when teeth were grouped according to the amount of remainin
