Specific RANK Cytoplasmic Motifs Drive Osteoclastogenesis.

Upon receptor activator of NF-κB ligand (RANKL) binding, RANK promotes osteoclast formation through the recruitment of tumor necrosis factor (TNF) receptor-associated factors (TRAFs). In vitro assays identified two RANK intracellular motifs that bind TRAFs: PVQEET560-565 (Motif 2) and PVQEQG604-609 (Motif 3), which potently mediate osteoclast formation in vitro. To validate the in vitro findings, we have generated knock-in (KI) mice harboring inactivating mutations in RANK Motifs 2 and 3. Homozygous KI (RANKKI/KI ) mice are born at the predicted Mendelian frequency and normal in tooth eruption

Diabetes and the license to drive.

People with diabetes are subject to restrictive licensing policies that bar them from driving certain types of motor vehicles. Discriminatory rules regarding the license to drive are based mainly on concerns that treatment with blood glucose-lowering agents, especially insulin--may induce episodes of altered consciousness due to hypoglycemia. The risk of hypoglycemia, however, differs greatly among insulin-requiring diabetics and it is not difficult to identify those few who present a substantial risk. Moreover, the advent of simple, portable devices for blood glucose self-monitoring allows di

Blunted respiratory drive in congenital myopathy.

Two patients with clinically mild congenital myopathies presented with chronic respiratory failure. Muscle weakness alone could not account for the respiratory insufficiency since static respiratory pressures were not markedly impaired, ventilation during exercise was normal, and daytime ventilation was normal if ventilatory assistance was provided at night. The ventilatory responses to inhaled carbon dioxide were very low, suggesting that impairment of the central nervous respiratory chemoreceptor contributed to hypoventilation. These patients and others described in the literature suggest th

[Schizophrenic defect syndromes and drive behavior].

First the terms "impulse" (Antrieb) and "schizophrenic defect" (schizophrener Defekt) are explained and defined. 49 (= 12 per cent) of the 409 schizophrenic in-patients, treated 1970-1972, had the symptoms of a schizophrenic defect. All of them showed disturbances of impulse: more than half the patients a deficiency, nearly a third an unsettled surplus, and about ten percent an alternating impulse. The deficiency as the alternating impulse usually were accompanied with a negativ mood. The surplus of impulse was bound up as equal parts with positive and negative moods. At last any kind of distu

Mesopic and glare driving performance in a driving simulator.

OBJECTIVE: Difficulties in night driving due to deficits in visual function are a common problem among older drivers. Signage, hazards, and road markings can be more difficult to identify for those with presbyopia, especially in nighttime conditions that include glare from headlights. This study evaluated the visual function and driving abilities of participants in various lighting conditions to find efficient and effective testing procedures for predicting how older individuals will perform in night driving conditions. METHODS: A driving simulator study was conducted to investigate the relat

Comparing Powered Wheelchair Driving Characteristics of Real Driving and Two Types of Simulated Driving.

We aimed to gather evidence on the feasibility of using simulator-based driving assessments for prescribing powered mobility devices (PMDs). Therefore, we compared the driving characteristics of real driving and two types of simulated driving. Thirty participants with difficulty walking more than 100 meters independently were enrolled. We developed a full-cabin and desktop simulator and created driving scenarios that closely resembled a real driving route in a park. They participated in three separate driving sessions, each using a powered wheelchair, full-cabin simulator, and desktop simulato

Glaucoma and driving: Are we documenting driving status and advising patients with glaucoma appropriately about their driving?

PURPOSE: Guidelines from the Driver and Vehicle Licensing Agency (DVLA) in the UK state that patients with bilateral glaucomatous visual field defects must stop driving unless confirmed able to meet minimum visual field requirements. Guidelines from the Royal College of Ophthalmologists state that ophthalmologists must consider driving status in every consultation, provide appropriate information if visual problems may affect driving and document any advice that has been given. The authors aimed to investigate whether in glaucoma clinics: (1) driving status is being documented, (2) appropriate

Design of Distributed Drive Control Strategy for Wheel Side Rear Drive Electric Bus Based on Neural Network Algorithm.

The electric bus driven by the wheel-side motor has a natural advantage for the driving stability control of the vehicle because the torque of each driving wheel is independently controllable. Based on the advantages of the artificial neural network control algorithm, this paper designs a direct yaw torque control strategy based on neural network PID control, and combines the steering characteristics of the vehicle to distribute the driving torque. The co-simulation results of Matlab/Simulink and TruckSim show that the designed control strategy can effectively reduce the vehicle's center of ma

[Driving ability and fitness to drive in people with diabetes mellitus].

Chronic sequelae of diabetes that could potentially affect driving include the following: visual retinopathy with associated impaired visual acuity, loss of peripheral vision and poor dark adaptation; neuropathy that may affect lower limb functions needed for safe driving; and acute events, including transient cognitive dysfunction and loss of consciousness related to hypo- or hyperglycemia. Hyperglycemia does not suddenly incapacitate drivers, however its occurrence often leads to tiredness, blurred vision, decreased visual acuity and adjustment of treatment which may precipitate hypoglycaemi

[Driving fitness/driving capacity of patients treated with methadone].

To answer the question whether or not therapeutic methadone doses significantly reduce traffic-related performance of drivers on medically supervised methadone programmes, 34 methadone substitution patients, all of them volunteers, were subjected to a test series: the focus of the study was a psychophysical test battery consisting of 10 individual performance tests to assess essential functions with regard to driving ability, such as concentration, attention, reaction capability, memory, perception and sensorimotor coordination. In evaluating the results of the psychophysical tests, multiple d
