Fenofibrate Reverses Dysfunction of EPCs Caused by Chronic Heart Failure.

The enhanced activity of endothelial progenitor cells (EPCs) by AMP-activated protein kinase (AMPK) agonists might explain the reversal of chronic heart failure (CHF)-mediated endothelial dysfunction. We studied baseline circulating EPC numbers in patients with heart failure and clarified the effect of fenofibrate on both circulating angiogenic cell (CAC) and late EPC activity. The numbers of circulating EPCs in CHF patients were quantified by flow cytometry. Blood-derived mononuclear cells were cultured, and CAC and late EPC functions, including fibronectin adhesion, tube formation, and migra

Fenofibrate prevents skeletal muscle loss in mice with lung cancer.

The cancer anorexia cachexia syndrome is a systemic metabolic disorder characterized by the catabolism of stored nutrients in skeletal muscle and adipose tissue that is particularly prevalent in nonsmall cell lung cancer (NSCLC). Loss of skeletal muscle results in functional impairments and increased mortality. The aim of the present study was to characterize the changes in systemic metabolism in a genetically engineered mouse model of NSCLC. We show that a portion of these animals develop loss of skeletal muscle, loss of adipose tissue, and increased inflammatory markers mirroring the human c

Morphometric study of human hepatic cell modifications induced by fenofibrate.

We have studied liver biopsies obtained in 12 hyperlipoproteinemic (HLP) patients (type II, 6; type IV, 6) treated with diet and fenofibrate, and in 15 patients (type II, 11; type IV, 4) receiving diet only. Electron microscopy of liver biopsies and the morphometric analysis according to the method of Weibel and Rohr showed mitrochondrial changes in patients treated with fenofibrate, these changes depending on the type of hyperlipoproteinemia. In type II HLP, we found a decreased volume of normal mitochondria (fenofibrate, 125.72 +/- 17.04 X 10(-3) cm3/cm3; diet only, 185.84 +/- 8.96 10(-3), P

Preparation and Characterization of Fenofibric Acid-Loaded Electrospun Fibrous Mats.

Fenofibric acid-loaded microfibrous mats were produced by electrospinning using polyvinylpyrrolidone (PVP) as a carrier polymer. Fenofibric acid, obtained through alkaline hydrolysis of fenofibrate, was incorporated into PVP solutions and processed under optimized electrospinning conditions to obtain uniform, defect-free fibers. The morphology and average fiber diameter were examined by scanning electron microscopy, revealing homogeneous, continuous fibers. Thermal analysis revealed the disappearance of the characteristic melting endotherm of fenofibric acid following electrospinning, indicati

Cardiometabolic benefits of fenofibrate in heart failure related to obesity and diabetes.

BACKGROUND: Heart failure (HF) is a serious and common condition affecting millions of people worldwide, with obesity being a major cause of metabolic disorders such as diabetes and cardiovascular disease. This study aimed to investigate the effects of fenofibrate, a peroxisome proliferator-activated receptor alpha (PPARα) agonist, on the obese- and diabetes-related cardiomyopathy. METHODS AND RESULTS: We used db/db mice and high fat diet-streptozotocin induced diabetic mice to investigate the underlying mechanisms of fenofibrate's beneficial effects on heart function. Fenofibrate reduced fib

PPARα-dependent increase of mouse urine output by gemfibrozil and fenofibrate.

While gemfibrozil and fenofibrate are prescribed for anti-dyslipidemia treatment, a rational basis for the use of these drugs for treatment of dyslipidemia with concurrent metabolic syndrome has not been established. In this study, wild-type and Pparα-null mice were fed gemfibrozil- or fenofibrate-containing diets for 14 days. Urine output (24 h) was monitored, and urine, serum, and liver and kidney tissues were subjected to toxicity assessment. A 2-month challenge followed by a 2-week wash-out was performed for gemfibrozil to determine urine output and the potential toxicity. A therapeuticall

Rhabdomyolysis associated with fenofibrate monotherapy in a patient with chronic myelogenous leukemia.

Rhabdomyolysis associated with fenofibrate monotherapy is extremely rare. Here, we report a rare case of rhabdomyolysis of the psoas muscle in an 82-year-old man with chronic myelogenous leukemia (CML). He was prescribed fenofibrate because of a hypertriglyceridemia. The patient reported generalized muscle pain and right abdominal pain while receiving fenofibrate monotherapy. An abdominal computed tomography scan and an abdominal ultrasound showed a large and low attenuation and high echogenicity, respectively, in the right middle abdominal area. Laboratory values included a serum creatine con

Effects of fenofibrate on C-reactive protein levels in hypertriglyceridemic patients.

We investigated the effects of fenofibrate on C-reactive protein (CRP) levels in patients with hypertriglyceridemia. Patients with a triglyceride level >or=200 mg/dL were randomly assigned to receive either 200 mg of fenofibrate (n = 54) or general measures (n = 54). A third group of patients with hypercholesterolemia received a statin (n = 54). Patients with a CRP level >or=10 mg/L were excluded. CRP levels were measured before and after 2 months of therapy. Fenofibrate did not reduce CRP levels (1.74 +/- 1.74 vs. 1.54 +/- 1.66 mg/L, P = 0.27) nor did general measures (P = 0.85). Statin reduc

Potential use of fenofibrate and other fibric acid derivatives in the clinic.

The fibric acid derivatives continue to have a place in the treatment of hyperlipidemia. The third generation of these drugs, including fenofibrate, appears to offer some advantages over those currently available in the United States. These drugs should be prescribed only after dietary and lifestyle changes have been offered as the preferable treatment. In severe hypertriglyceridemia, clofibrate, gemfibrozil, or fenofibrate may reduce the very low-density lipoprotein and chylomicron levels adequately. Dysbetalipoproteinemia may also be completely controlled by a combination of diet and any one

A chitosan-based hydrogel loaded with fenofibrate for diabetic wound healing.

Diabetic wounds represent a common chronic condition, posing significant challenges in the treatment process due to bacterial infections, increased generation of reactive oxygen species (ROS) and exacerbated inflammation. Fenofibrate (FEN) is a clinical medication used for lipid regulation. In this study, it was utilized for the first time as an effective component of wound dressings for treating diabetic ulcers, exploring its novel applications further. Therefore, we prepared a polyvinyl alcohol/chitosan/FEN (PCF) hydrogel using a freeze-thaw method and conducted physicochemical characterizat

Harnessing the pleiotropic effects of atorvastatin-fenofibrate combination for cardiovascular stents.

Atorvastatin and fenofibrate have been conventionally employed as lipid-lowering agents. They also exhibit beneficial effects in the treatment of endothelial dysfunction, oxidative stress and vascular inflammation due to their pleiotropic effects that include vasodilatory and anti-inflammatory effects. These pleiotropic effects may serve to overcome the drawbacks of late stent thrombosis and delayed endothelialization that plague conventional drug eluting stents. However, the combination has not been explored yet as therapeutic coatings in drug eluting stents. The present study aims to investi
