Unveiling drastic influence of cross-interactions in hydrothermal carbonization of spirulina with cellulose, lignin or poplar on nature of hydrochar and activated carbon.

Spirulina platensis contains abundant nitrogen-containing organics, which might react with derivatives of cellulose/lignin during hydrothermal carbonization (HTC), probably affecting yield, property of hydrochar, and pore development in activation of hydrochar. This was investigated herein by conducting co-HTC of spirulina platensis with cellulose, lignin, and sawdust at 260 °C and subsequent activation of the resulting hydrochars with K2C2O4 at 800 °C. The results showed that cross-condensation of spirulina platensis-derived proteins with cellulose/lignin-derived ketones and phenolics did tak

GM2 gangliosidosis associated with a HEXA missense mutation in Japanese Chin dogs: a potential model for Tay Sachs disease.

GM2 gangliosidosis is a fatal lysosomal storage disease caused by a deficiency of β-hexosaminidase (EC 3.2.1.52). There are two major isoforms of the enzyme: hexosaminidase A composed of an α and a β subunit (encoded by HEXA and HEXB genes, respectively); and, hexosaminidase B composed of two β subunits. Hexosaminidase A requires an activator protein encoded by GM2A to catabolize GM2 ganglioside, but even in the absence of the activator protein, it can hydrolyze the synthetic substrates commonly used to assess enzyme activity. GM2 gangliosidosis has been reported in Japanese Chin dogs, and we

Expression of the GM2-activator protein in the methylotrophic yeast Pichia pastoris, purification, isotopic labeling, and biophysical characterization.

The GM2-activator protein (GM2AP) belongs to a group of five small, nonenzymatic proteins that are essential cofactors for the degradation of glycosphingolipids in the lysosome. It mediates the interaction between the water-soluble enzyme beta-hexosaminidase A and its membrane-embedded substrate, ganglioside GM2, at the lipid-water interphase. Inherited defects in the gene encoding this glycoprotein cause a fatal neurological storage disorder, the AB variant of GM2 gangliosidosis. With the aim to establish a convenient eukaryotic system that allows the efficient production of functionally fold

Loss of epithelial Gq and G11 signaling inhibits TGFβ production but promotes IL-33-mediated macrophage polarization and emphysema.

Heterotrimeric guanine nucleotide-binding protein (G protein) signaling links hundreds of G protein-coupled receptors with four G protein signaling pathways. Two of these, one mediated by Gq and G11 (Gq/11) and the other by G12 and G13 (G12/13), are implicated in the force-dependent activation of transforming growth factor-β (TGFβ) in lung epithelial cells. Reduced TGFβ activation in alveolar cells leads to emphysema, whereas enhanced TGFβ activation promotes acute lung injury and idiopathic pulmonary fibrosis. Therefore, precise control of alveolar TGFβ activation is essential for alveolar ho

G Protein Pathway Suppressor 1 Promotes Influenza Virus Polymerase Activity by Activating the NF-κB Signaling Pathway.

Influenza virus relies heavily on cellular machinery to replicate in host cells. Therefore, to better understand the influenza virus life cycle, it is important to identify which host proteins are involved and how they function in virus replication. Previously, we identified G protein pathway suppressor 1 (GPS1) to be a matrix protein 2 (M2)-interacting host protein. GPS1 is a component of the COP9 signalosome, which regulates the NF-κB signaling pathway. Here, we found that the downregulation of GPS1 expression reduced influenza virus replication by more than 2 log units. Although GPS1 was no

Comparative impact of pH-shifting and heat treatment on the structural and emulsifying properties of Clanis bilineata tsingtauica larvae protein.

This study investigated the individual and synergistic effects of pH-shifting (pH 2.0, 7.0, and 12.0) and heat treatment (25-90 °C) on the structural, physicochemical, and emulsifying properties of Clanis bilineata tsingtauica larvae protein (CBTLP). Results demonstrated that alkaline-shifting (pH 12.0) at ambient temperature (25 °C) is a highly efficient modification strategy, achieving an emulsifying activity index (EAI) of approximately 90 m2/g and solubility exceeding 80%, which is comparable to the functional levels of acid-shifting (pH 2.0) combined with 90 °C heating (EAI: 85 m2/g; solu

Cyclodextrin-based metal-organic frameworks (MOFs) as delivery systems for quercetin: Effects of carrier size and protein corona formation.

Understanding how carrier size influences protein corona formation and delivery behavior is important for the rational design of γ-cyclodextrin-based metal-organic frameworks (γCDMOFs). In this study, micron-scale (microMOF) and nanoscale γCDMOFs (nanoMOF) were compared as quercetin carriers using pepsin and human serum albumin as model proteins. NanoMOF (∼350 nm) exhibited a higher specific surface area (1016.01 m2/g vs. 769.79 m2/g) and encapsulation efficiency (85.37% vs. 65.24%) than microMOF (∼2.16 μm). Protein adsorption showed size-dependent differences, with nanoMOF exhibiting greater

Juvenile-onset motor neuron disease caused by novel mutations in β-hexosaminidase.

A 12 year-old female presented with a seven-year history of progressive muscle weakness, atrophy, tremor and fasciculations. Cognition was normal. Rectal biopsy revealed intracellular storage material and biochemical testing indicated low hexosaminidase activity consistent with juvenile-onset G(M2)-gangliosidosis. Genetic evaluation revealed compound heterozygosity with two novel mutations in the hexosaminidase β-subunit (c.512-3 C>A and c.1613+15_1613+18dup). Protein analysis was consistent with biochemical findings and indicated only a small portion of β-subunits were properly processed. The

Crystal structure of human GM2-activator protein with a novel beta-cup topology.

GM2 activator protein (GM2-AP) belongs to a small group of non- enzymatic lysosomal proteins that act as cofactors in the sequential degradation of gangliosides. It has been postulated that GM2-AP extracts single GM2 molecules from membranes and presents them in soluble form to beta-hexosaminidase A for cleavage of N-acetyl-d-galactosamine and conversion to GM3. The high affinity of GM2-AP for GM2 is based on specfic recognition of the oligosaccharide moiety as well as the ceramide lipid tail. Genetic defects in GM2-AP result in an atypical form of Tay-Sachs disease known as variant AB GM2 gan

The specificity of human N-acetyl-beta-D-hexosaminidases towards glycosphincolipids is determined by an activator protein.

It has been very difficult to correlate, on the basis of in vitro measurements of substrate specificities, the glycosphingolipid storage patterns observed in different variants of infantile GM2 gangliosidosis with the hexosaminidase (hex) isoenzyme deficiencies underlying these diseases. However, the in vitro enzyme assays included detergents, which greatly enhanced the enzymic degradation of lipids by breaking down the large lipid micelles that cannot otherwise be attacked by the hydrolases. In vivo, the role of detergent is taken over by water-soluble, low molecular weight proteins, so-calle

Novel Sodium Carbonate Activation for Manufacturing Sludge-Based Biochar and Assessment of Its Organic Adsorption Property in Treating Wool Scouring Wastewater.

Under the concept of green and low-carbon development, efficient and environmentally friendly biochar preparation methods have attracted much attention. This study assessed a novel sodium carbonate activator combined with acid modification for sludge-based biochar (SB) production and its adsorption of organics in wool scouring wastewater. Under 600 °C, the optimal carbonization temperature, the residual weight percentage of biochar carbonized material increases from 27% to 73% after Na2CO3 activation compared to ZnCl2 activation. Compared to HCl-modified ZnCl2-activated biochar (Zn-Cl-SB), HCl

Muscarinic m2 receptor-mediated actin polymerization via PI3 kinase γ and integrin-linked kinase in gastric smooth muscle.

BACKGROUND: Actin polymerization plays an important role in smooth muscle contraction. Integrin-linked kinase (ILK) was shown to mediate actin polymerization in airway smooth muscle. The role of ILK in actin polymerization in response to m2 receptor activation was not in gastric smooth muscle. METHODS: Phosphorylation of paxillin, neuronal Wiskott-Aldrich syndrome protein (N-WASp), and association of paxillin with GEF proteins (Cool2/αPix [Cool2/PAK-interacting exchange factor alpha], Cool1/βPix [Cool1/PAK-interacting exchange factor beta], and DOCK 180 [Dedicator of cytokinesis]) and N-WASp

Juvenile GM2 gangliosidosis (AMB variant): inability to activate hexosaminidase A by activator protein.

Two sibling from a consanguineous Puerto Rican marriage were found to have a juvenile-onset type of lipidosis first noted at age 2 1/2 by expressing difficulties with motor function and developmental delay. They continued to deteriorate, showing muscle atrophy, spasticity, and loss of speech, and death occurred at ages 7 and 8. Examination of the brains from these patients revealed that the concentration of GM2 ganglioside was about 56% of the total gangliosides. Hexosaminidase and percent hexosaminidase A (HEX A) and other lysosomal enzymes were normal in cultured skin fibroblasts, liver, and

[A molecular mechanism for supersensitization of adenylyl cyclase system in cloned opioid receptor-transfected cells following sustained opioid treatment].

Chronic opioid treatment has been shown to develop supersensitization of adenylyl cyclase (AC) system or cAMP overshoot. In this study, we investigated the molecular mechanism of supersensitization of AC system using CHO cells expressing one of the cloned mu-, delta- and kappa-opioid receptors. In naive cells, acute treatment with an opioid agonist, but not antagonist, suppressed forskolin-stimulated cAMP accumulation. In the cells sustainedly (4 hr) treated with the agonist, the challenge by antagonist induced the cAMP overshoot over the naive level (supersensitization of AC system), but had

Cold plasma pretreatment assisted whey protein isolate-gum arabic Maillard conjugates produce water-soluble microcapsules for clove essential oil release.

Clove essential oil (C) is poorly water-soluble, which limits its application in aqueous food systems. This study developed a novel strategy to fabricate microcapsules with improved water solubility by pretreating whey protein isolate (WPI) with cold plasma followed by Maillard glycation with gum arabic (GA) prior to complex coacervation and spray drying. Cold plasma pretreatment for 20 s significantly improved the degree of grafting (DG, from 22.14% to 31.92%) and emulsifying activity (from 32.88 m2/g to 52.40 m2/g) of WPI-GA Maillard conjugates (WPI-GAM). The resulting microcapsules exhibite

Autoantibodies against cardiac G-protein-coupled receptors define different populations with cardiomyopathies but not with hypertension.

It was previously shown that the second extracellular loop of cardiovascular G-protein-coupled receptors is an antigenic target for pharmacologically active autoantibodies in patients with idiopathic dilated cardiomyopathy. To extend these observations to cover patients with the same disease from different geographical origins or to patients with other cardiac diseases, peptides corresponding to the sequences of the second extracellular loops of the human M2 muscarinic receptors and beta adrenoceptors were used as antigens in an enzyme immunoassay. Sera from patients from Sweden and Japan with

Recombinant tissue plasminogen activator (rt-PA) for veno-occlusive liver disease in pediatric autologous bone marrow transplant patients.

The pathogenesis of veno-occlusive disease (VOD) of the liver appears to be secondary to endothelial damage of terminal hepatic venules, which leads to activation of the coagulation cascade, fibrin deposition, and eventual fibrous obliteration of the hepatic venules. Patients with VOD usually present with jaundice, hepatomegaly, weight gain, and ascites. This complication is usually associated with a high mortality rate. We report here the frequency and treatment of VOD in our autologous bone marrow transplant (BMT) patient population. Three of 15 (20%) children (median age 9 years) developed

Local thiamet-G delivery by a thermosensitive hydrogel confers ischemic cardiac repair via myeloid M2-like activation in a STAT6 O-GlcNAcylation-dependent manner.

Infarct healing requires a dynamic and orchestrated inflammatory reaction following myocardial infarction (MI). While an uncontrolled excessive inflammatory response exaggerates ischemic injury post-MI, M2-like reparative macrophages may facilitate inflammation regression and promote myocardial healing. However, how protein post-translational modification regulates post-MI cardiac repair and dynamic myeloid activation remains unknown. Here we show that M2-like reparative, but not M1-like inflammatory activation, is enhanced by pharmacologically-induced hyper-O-GlcNAcylation. Mechanistically, m

Associations of Biomarkers of Kidney Tubule Health with Retinal Microvascular Signs: The Multi-Ethnic Study of Atherosclerosis.

KEY POINTS: Kidney injury molecule-1 and soluble urokinase plasminogen activator receptor are associated with retinal microvascular changes in individuals without CKD, diabetes, or cardiovascular disease. Tubule biomarkers may reveal microvascular pathways linking kidney dysfunction to cardiovascular risk beyond eGFR or albumin-to-creatinine ratio. BACKGROUND: CKD is strongly associated with cardiovascular disease (CVD), yet the etiology responsible for this link remains elusive. Novel blood and urine biomarkers reflecting kidney tubule dysfunction and injury may provide novel insights to mec

Process-dependent structural and functional modification of yeast protein: Alkaline extraction-acid precipitation versus high-pressure homogenization-assisted pH shifting.

Limited redispersibility constrains commercial yeast-protein ingredients. This study compared preparation efficiency, structure, and function of yeast protein prepared by alkaline extraction-acid precipitation (ASAP) and high-pressure homogenization-assisted pH shift (HPHS). ASAP achieved higher product yield (69.23% ± 1.42%) and protein recovery (57.81% ± 2.42%) than HPHS. ASAP-YP exhibited dense sheet-like morphology, higher β-turn content (55.71%), and preserved reactive groups, whereas HPHS-YP formed porous aggregates with higher α-helix content and lower sulfhydryl availability.
