Single-atom electrosynthesis of glycine from glyphosate wastewater.

The extensive application of glyphosate (PMG) has led to the accumulation of contaminated effluent, posing environmental risks and losing valuable chemical resources. Although electrochemical conversion represents a promising pathway for waste valorization, selectively transforming PMG into a specific high-value product remains challenging. Here, we introduce an integrated electrochemical system to efficiently convert waste PMG to value-added glycine through collaborative redesign of anode and cathode reactions. Isolated iron atoms anchored on copper nanowires (Fe1Cu NWs) to form the efficient

Architecture and assembly mechanism of native glycine receptors.

Glycine receptors (GlyRs) are pentameric, 'Cys-loop' receptors that form chloride-permeable channels and mediate fast inhibitory signalling throughout the central nervous system1,2. In the spinal cord and brainstem, GlyRs regulate locomotion and cause movement disorders when mutated2,3. However, the stoichiometry of native GlyRs and the mechanism by which they are assembled remain unclear, despite extensive investigation4-8. Here we report cryo-electron microscopy structures of native GlyRs from pig spinal cord and brainstem, revealing structural insights into heteromeric receptors and their p

Structure and Pharmacologic Modulation of Inhibitory Glycine Receptors.

Glycine receptors (GlyR) are inhibitory Cys-loop ion channels that contribute to the control of excitability along the central nervous system (CNS). GlyR are found in the spinal cord and brain stem, and more recently they were reported in higher regions of the CNS such as the hippocampus and nucleus accumbens. GlyR are involved in motor coordination, respiratory rhythms, pain transmission, and sensory processing, and they are targets for relevant physiologic and pharmacologic modulators. Several studies with protein crystallography and cryoelectron microscopy have shed light on the residues an

Chimeric GABAA/glycine receptors: expression and barbiturate pharmacology.

GABAA and glycine receptors are close relatives in the "gene superfamily" of ligand-gated ion channels, but have distinctly different pharmacology. For example, barbiturates have two effects on GABAA receptors (GABAA-R): at low micromolar concentrations (2-5 microM), the anesthetic barbiturate methohexital potentiates submaximal chloride current responses to GABA; at higher concentrations (20-50 microM), the barbiturate causes direct gating of the channel in the absence of agonist. Neither of these barbiturate effects is seen on the glycine receptor (Gly-R). In order to study the structural pa

Glycine transport in rat brain and liver mitochondria.

Transport of glycine by rat brain and liver mitochondria has been investigated by both [14C]glycine uptake and swelling experiments. Glycine enters mitochondria passively down its concentration gradient by a respiratory-independent carrier-mediated process. This view is supported by the following observations: (a) glycine inside the mitochondria reaches the incubation medium concentration; (b) mitochondria swell in the presence of isoomotic solutions of glycine in a concentration-dependent fashion; (c) the uptake of glycine is not influenced by respiratory inhibitors such as KCN or by uncouple

[The human utilization of orally administered 15N-glycine].

In 7 convalescent patients we studied utilisation of a single dose of 15N-labelled glycine. In intervals we estimated total-N, urea-N, NH3-N and their 15N-abundances (urine). The following results were obtained. The peak of ammonium-15N appeared 25 minutes earlier than the peak of urea-15 N. The cumulative elimination of total-15N amounted to 25% of uptake after 24 hours and to 34.4% after 6 days. 12 hours after application 53% of the total amount, excreted within 6 days, were already eliminated. 24 hours after administration 80% of total-15N was urea-15N and 2.8% NH3-15N. Further we found cor

Glycine removal from both glycinergic and glutamatergic synapses into astrocytic processes by glycine transporter 1 in the adult mouse spinal cord.

Glycine (Gly) and γ-aminobutyric acid (GABA) are inhibitory neurotransmitters in the spinal cord. Released Gly is removed by two Gly transporters (GlyTs): GlyT1 and GlyT2. We have previously demonstrated the localization of GlyT2, which reuptakes Gly into presynaptic terminals. In the present study, we focused on GlyT1, which terminates glycinergic transmission by uptaking Gly into astrocytes. GlyT1 may also regulate glutamatergic transmission by controlling the concentration of Gly, which is a co-agonist of N-methyl D-aspartate receptors in the forebrain, where glycinergic synapses are sparse

Immunomodulatory Effects of Copper Bis-Glycinate In Vitro.

Copper functions as a cofactor and antioxidants in a large number of enzymes that are important for cellular respiration and the nervous system. In the last century scholars have explored copper's relationship with the immune system, with copper deficiency drastically upsetting the overall function of the immune system, as seen in symptoms such as increased susceptibility to pathogens, decreased proliferation of lymphocytes, and impaired function of both cytotoxic T lymphocytes and helper T cells. Among copper's various forms, copper bis-glycinate (Cbg) has been used as an official EU-approved

Determination of glycine and threonine in topical dermatological preparations.

In the present study, a single HPLC method was developed for the determination of glycine and threonine in cicatrizants. Two different preparations of a cream and an ointment, and the corresponding bandages, onto which the formulations were applied, were studied. The method involved matrix solubilisation with dichloromethane, liquid-liquid isolation of gly and thr with aqueous 1N NaOH, and derivatization with phenylisothiocyanate. Reversed-phase HPLC separation was carried out by gradient elution with 20mM aqueous NaClO4 and acetonitrile (from 90% to 30% aqueous NaClO4 in 10 min) on a LiChrosp

Disorders of the inhibitory glycine receptor: the spastic mouse.

The mutant mouse spastic suffers from a motor disorder of autosomal recessive inheritance which is characterized by tremor, myoclonic episodes, and a disturbed righting response. The most prominent alteration in the mutant is a substantial deficit of postsynaptic glycine receptor channels resulting in a dramatic reduction of glycinergic synaptic inhibition. Function and structure of the glycine receptor protein appear unaffected, which argues for a regulatory rather than a structural effect of the spastic mutation. It appears that other alterations in the spastic mouse are secondary to this fu

Glycine is a transmitter in the human and chimpanzee cochlear nuclei.

INTRODUCTION: Auditory information is relayed from the cochlea via the eighth cranial nerve to the dorsal and ventral cochlear nuclei (DCN, VCN). The organization, neurochemistry and circuitry of the cochlear nuclei (CN) have been studied in many species. It is well-established that glycine is an inhibitory transmitter in the CN of rodents and cats, with glycinergic cells in the DCN and VCN. There are, however, major differences in the laminar and cellular organization of the DCN between humans (and other primates) and rodents and cats. We therefore asked whether there might also be difference

A Piezoelectric Ionic Cocrystal of Glycine and Sulfamic Acid.

Cocrystallization of two or more molecular compounds can dramatically change the physicochemical properties of a functional molecule without the need for chemical modification. For example, coformers can enhance the mechanical stability, processability, and solubility of pharmaceutical compounds to enable better medicines. Here, we demonstrate that amino acid cocrystals can enhance functional electromechanical properties in simple, sustainable materials as exemplified by glycine and sulfamic acid. These coformers crystallize independently in centrosymmetric space groups when they are grown as

The Influence of Mineral Matrices on the Thermal Behavior of Glycine.

On the Hadean-Early Archean Earth, the first islands must have provided hot and dry environments for abiotically formed organic molecules. The heat sources, mainly volcanism and meteorite impacts, were also available on Mars during the Noachian period. In recent work simulating this scenario, we have shown that neat glycine forms a black, sparingly water-soluble polymer ("thermomelanoid") when dry-heated at 200 °C under pure nitrogen. The present study explores whether relevant minerals and mineral mixtures can change this thermal behavior. Most experiments were conducted at 200 or 250 °C for

[Preservation of hepatocytes of the rat by active transport of glycine].

The measurement of the active accumulation of specific substrates against a concentration gradient in tissue fragments in vitro has proved to be a simple but excellent procedure to ascertain the viability of several cell types and, in parallel with routine microscopy, has become a reliable tool in assaying the biochemical and functional modifications of a tissue under physiopathological conditions. It also has the added advantage of using tissues that are not pretreated in any way such as the preparative procedures used in cell isolation methods. The majority of investigations employing hepato

Action and localization of glycine and taurine in the cat retina.

The effects on retinal ganglion cells of iontophoretically applied glycine, taurine and strychnine were studied in the optically intact eye of the cat. Glycine and taurine suppressed the light-evoked discharge of all on-centre and off-centre brisk ganglion cells, regardless of the visual stimulus used. Strychnine blocked the action of externally applied glycine and taurine. The light-evoked response of all ganglion cells was raised by strychnine. The tonic discharge of the light response was suppressed or raised by the drugs more than the phasic response. A population of amacrine cells, which

The effect of phenoxybenzamine on the gastrin response to glycine stimulation.

The nature of adrenergic mechanism involved in gastrin release in response to local stimuli and feeding is still unclear. In the present study, the effect of phenoxybenzamine, a potent alpha-blocking agent, on the gastrin release in response to glycine perfusion (pH 7.0) of an isolated canine antral pouch was examined. The experiment was repeated with truncal vagotomy performed before glycine perfusion. It was found that gastrin concentrations measured via right gastroepiploic vein were significantly increased during phenoxybenzamine infusion with and without truncal vagotomy. This finding ind
