Hepatic transcriptomics reveals immuno-metabolic interactions in juvenile channel catfish (Ictalurus punctatus) after Aeromonas hydrophila infection.

BACKGROUND: In aquaculture, disease outbreaks occur amidst complex husbandry factors like feeding schedule and incidental injury. The liver, a central immunometabolic organ, integrates these cues, but the systems-level transcriptional mechanisms governing its response to concurrent stress are poorly defined in channel catfish. We investigated how feeding status and physical injury modulate the hepatic transcriptome during early Aeromonas hydrophila infection in channel catfish. RESULTS: Juvenile channel catfish were assigned to four treatment groups reflecting combinations of feeding status (

Age-related reproductive performance and transcriptome profiling of testis in male blue catfish, Ictalurus furcatus.

Catfish farming is the largest aquaculture industry in the U.S., where hybrid catfish produced by channel catfish, Ictalurus punctatus ♀ × blue catfish, I. furcatus ♂, represent > 50% of the harvest. Previous studies indicated a high degree of variation in reproductive performance among individuals. Therefore, it is crucial to establish a connection between paternal age and reproductive success. This study investigated the reproduction of 103 blue catfish males aged 2 to 10 years. Morphometric data were collected, and blood was drawn to quantify testosterone (T), 11-ketotestosterone (11-KT), o

Morphological, Histological, and Molecular Description of Unicauda fimbrethilae n. sp. (Cnidaria: Myxosporea: Myxobolidae) from the Intestinal Tract of Channel Catfish Ictalurus punctatus.

The channel catfish Ictalurus punctatus is a known host for 10 species of Henneguya, but few other myxozoan genera are described from this species. Unicauda is a genus of myxozoan parasites within the family Myxobolidae that consists of 10 valid species from freshwater fish. Herein, we describe a novel species of Unicauda from the intestinal tract of farm-raised channel catfish in Mississippi. Myxospores were consistent with the genus Unicauda but exhibited a unique branching at the terminal end of the caudal process that has not previously been reported. Myxospores measured 90.39 ± 14.97 μm (

Acute thermal stress alters sperm swimming activity, viability, and gene expression in blue catfish, Ictalurus furcatus.

Fish sperm are vulnerable to changes in external environments, particularly temperature fluctuations. Such changes can directly impact sperm of externally fertilizing species, including blue catfish, Ictalurus furcatus, which in turn can affect physiological and molecular mechanisms. The objective of this study was to investigate how thermal stressors impact sperm kinematics, viability, and gene expression. Sperm from nine mature catfish were stored at 4-, 32- (representing pond temperature at end of spawning season), and 36°C (representing a pond heatwave) for 30-, 60-, and 90 min. Fresh sper

Spermatogenesis, hormonal fluctuations, and testicular gene expression changes in male blue catfish, Ictalurus furcatus, throughout pubescent and early adult stages of development.

Male blue catfish (Ictalurus furcatus) mature at 4-7 years and cannot be hand-stripped, making sperm collection a lethal procedure. Sperm are also highly variable in quality/quantity. We monitored blue catfish males for 34 months (2.3 to 4.9 years old; y/o) to quantify changes in body morphometrics, plasma androgens, osmolality, ions, testicular development, and gene expression. Body weight and length of fish ranged from 0.6 ± 0.2 to 3.2 ± 0.2 kg and 35.2 ± 2.1 to 58.9 ± 1.9 cm, respectively. Testes weight and gonadosomatic index (GSI) were highly variable within/between sampling points. Maxim

Using comparative transcriptome analysis to identify molecular response mechanisms to salinity stress in channel catfish (Ictalurus punctatus).

Channel catfish (Ictalurus punctatus) are an important global aquaculture species. To explore gene expression patterns and identify adaptive molecular mechanisms in catfish during salinity stress, we performed growth comparison and comparative transcriptome sequencing on liver tissue. Our study revealed that salinity stress has a significant impact on the growth, survival, and antioxidant system of channel catfish. 927 and 1356 significant DEGs were identified in L vs. C group and H vs. C group. Feed deprivation of channel catfish, Ictalurus punctatus (Rafinesque), influences organosomatic indices, chemical composition and susceptibility to Flavobacterium columnare.

Withholding feed has been suggested as a strategy to manage infectious disease of channel catfish, Ictalurus punctatus (Rafinesque). In a previous study, we demonstrated that deprivation of feed for as little as 7 days reduced innate resistance of catfish to Flavobacterium columnare. This study was conducted to evaluate the effect of feeding regimens [no feeding (NF), fed once every other day to satiation (FEOD) and fed once daily to satiation (FD)] on organosomatic indices, physiological changes and susceptibility of channel catfish to F. columnare. Fish that were not fed for 2 and 4 weeks ha

Elevated concentrations of organic and inorganic forms of iron in plant-based diets for channel catfish prevent anemia but damage liver and intestine, respectively, without impacting growth performance.

We compared the effects of using inorganic and organic forms of iron in plant-based diets on catfish performance in a feeding trial with 6-g catfish fingerlings. The objective was to determine whether dietary iron in excess of known requirements negatively affected the fish. Five diets supplemented with 0 (basal), 125, 250 mg Fe/kg of either FeSO4 or iron methionine were formulated. Weight gain, feed conversion ratio, hepatosomatic index, and survival were similar among diets. Plasma and intestine iron concentration was similar among diets. Effects of moderate exercise on hepatic amino acid and fatty acid composition, liver transcriptome, and intestinal microbiota in channel catfish (Ictalurus punctatus).

Previous research on swimming exercise in fish has focused on muscle building and quality of flesh. However, the effects of hepatic amino acid and fatty acid composition, liver gene expression profile, and intestinal microbiota are poorly understood. In this study, channel catfish (Ictalurus punctatus) were subjected to a 4-week swimming exercise, and liver transcriptome and intestinal microbiota analyses were performed to broaden our understanding of fish under exercise. When compared to non-exercised channel catfish (N-EXF), exercised channel catfish (EXF) had improved arachidonic acid (C20:

Identification of transcriptome markers in blue catfish cryopreserved sperm to predict male reproductive performance.

IN BRIEF: This study establishes a mechanistic link between sperm-specific transcriptomic profiles and hatch success, demonstrating that a six-gene panel can serve as a statistically robust predictor of hatch success in hybrid catfish aquaculture. ABSTRACT: In hybrid catfish aquaculture, hatch success varies significantly using cryopreserved sperm from different males, but phenotypic assays only explain a fraction of observed variations. We profiled the sperm transcriptome of 30 blue catfish (Ictalurus furcatus) males, whose cryopreserved sperm yielded a hatch success from 14 to 63%. Our tran

Tapeworms (Cestoda) of Ictalurid Catfishes (Siluriformes) in North America: Redescription of Type Species of Two Genera and Proposal of Essexiellinae n. Subfam.

Ictalurid catfishes (Siluriformes) in North America harbor proteocephalid tapeworms of the subfamily Corallobothriinae. Type species of 2 of 3 genera of these tapeworms from ictalurids are redescribed, based on museum and newly collected material. Essexiella fimbriata (Essex, 1928) is typified mainly by a wide, umbrella-shaped scolex with a metascolex formed by numerous folds of tissue, anteriorly directed suckers without sphincters, vitellarium bent inwards posteriorly, "flower-shaped" uterus (with anterior, lateral, and posterior diverticula), and a conspicuously pre-equatorial genital atriu

Cathelicidins enhance protection of channel catfish, Ictalurus punctatus, and channel catfish ♀ × blue catfish, Ictalurus furcatus ♂ hybrid catfish against Edwarsiella ictaluri infection.

Cathelicidins are a class of antimicrobial peptides (AMPs) known to possess rapid and direct antimicrobial activities against a variety of microorganisms. Recently identified cathelicidins derived from alligator and sea snake were found to be more effective in inhibiting microbial growth than other AMPs previously characterized. The ability of these two cathelicidins along with the peptides, cecropin and pleurocidin, to protect channel catfish (Ictalurus punctatus, Rafinesque) and hybrid catfish (I. punctatus ♀ × blue catfish, Ictalurus furcatus, Valenciennes ♂) against Edwardsiella ictaluri,

Epigenetic Landscape of H3K27ac, H3K27me3, H3K4me1, and H3K4me3 Marks in Channel Catfish Following Βeta Glucan Exposure.

Epigenetic regulation plays a central role in shaping innate immune responses following immunostimulant exposure. In this study, we characterized the epigenetic landscape of four histone modifications, H3K27ac, H3K27me3, H3K4me1, and H3K4me3, in the anterior kidney of Ictalurus punctatus (channel catfish) following β-glucan exposure. Using chromatin immunoprecipitation sequencing (ChIP-seq), we generated chromatin state maps and examined the regulatory potential of these histone marks in relation to transcriptional responses. Genes showing increased expression associated with H3K4me3 up-peaks

Passive immunization with anti-virulent Aeromonas hydrophila whole serum protects Channel Catfish against motile Aeromonas septicemia.

OBJECTIVE: Recently, we demonstrated the efficacy of oral bacterin virulent Aeromonas hydrophila (vAh) vaccines (S14-452 and ML09-119) in Channel Catfish Ictalurus punctatus, but gaps in knowledge exist with respect to protective immunity induced by vAh bacterins. Specific antibodies were proposed as a potential mechanism of protection; thus, the aim of the study was to assess whether passive immunization with anti-vAh whole serum provides protection upon vAh challenge. METHODS: The protective role of anti-vAh whole serum was determined by passive immunization of Channel Catfish, followed by
