Iron Metabolism Disorders Associated With the Severity of Pyogenic Liver Abscess.

Pyogenic liver abscess (PLA) is a serious intra-abdominal infection with high morbidity and mortality. Current diagnostic methods are time-consuming, inefficient and high-cost. Rapid identification/assessment of PLA and early recognition of bacterial liver abscess (BLA) improve prognosis. Iron metabolism disorders play a critical role in infections, but their role in PLA (especially ferritin) remains unclear. Notably, dietary nutrition, as a modulator of iron metabolism/immune function, may influence PLA progression, with limited research. In this study, we collected clinical data of 125 PLA p

Disorders of Iron Metabolism: A "Sharp Edge" of Deoxynivalenol-Induced Hepatotoxicity.

BACKGROUND/OBJECTIVES: Deoxynivalenol (DON), known as vomitoxin, is one of the most common mycotoxins produced by Fusarium graminearum, with high detection rates in feed worldwide. Ferroptosis is a novel mode of cell death characterized by lipid peroxidation and the accumulation of reactive oxygen species. Although it has been demonstrated that DON can induce ferroptosis in the liver, the specific mechanisms and pathways are still unknown. The aim of this experiment was to investigate that DON can induce iron metabolism disorders in the livers of mice, thereby triggering ferroptosis and causin

Genetic disorders affecting proteins of iron and copper metabolism: clinical implications.

Iron and copper are essential transition metals that permit the facile transfer of electrons in a series of critical biochemical pathways. Recent work has identified the specific proteins involved in the absorption, transport, utilization, and storage of iron and copper. Remarkable progress is being made in understanding the molecular basis of disorders of human iron and copper metabolism. This review describes these proteins and examines the clinical consequences of new insights into the pathophysiology of genetic abnormalities affecting iron and copper metabolisms. Hereditary hemochromatosis

[Microelementosis with iron metabolism disorder: the nomenclature and classification].

In the discussion article the basic rules of human microelementosis theory (A. P. Avtsyn et al., 1991) and the review of modern data devoted to molecular-genetic mechanisms of iron homeostasis are represented. The conception of iron microelementosis (siderosis) with appropriate nomenclature and classification has been formulated. The main types of siderosis have been singled out as well hyposiderosis (a deficiency of iron), hypersiderosis (redundant accumulation of iron) and dissiderosis (an imbalance, redistribution of iron between functional and storage pools). In addition each type of sider

Aceruloplasminemia: molecular characterization of this disorder of iron metabolism.

Ceruloplasmin is an abundant alpha 2-serum glycoprotein that contains 95% of the copper found in the plasma of vertebrate species. We report here on the identification of a genetic defect in the ceruloplasmin gene in a patient previously noted to have a total absence of circulating serum ceruloplasmin in association with late-onset retinal and basal ganglia degeneration. In this patient T2 (transverse relaxation time)-weighted magnetic resonance imaging of the brain revealed basal ganglia densities consistent with iron deposition, and liver biopsy confirmed the presence of excess iron. Immunochemical and mass-spectrometry-based serum hepcidin assays for iron metabolism disorders.

BACKGROUND: Hepcidin is an iron-regulatory peptide hormone that consists of 3 isoforms: bioactive hepcidin-25, and inactive hepcidin-22 and hepcidin-20. Hepcidin is instrumental in the diagnosis and monitoring of iron metabolism disorders, but reliable methods for its quantification in serum are sparse, as is knowledge of their relative analytical strengths and clinical utility. METHODS: We developed a competitive (c)-ELISA and an immunocapture TOF mass-spectrometry (IC-TOF-MS) assay. Exploiting these 2 methods and our previously described weak cation exchange (WCX)-TOF-MS assay, we measured

Stress-Induced Dysregulation of Brain Iron Metabolism and Its Links to Neurological Disorders.

Stress is a non-specific systemic response to internal or external challenges. Recent studies show that stress can disrupt iron metabolism and that iron dyshomeostasis is implicated in many diseases-particularly within the nervous system, where iron distribution and regulation intersect tightly with oxidative stress and inflammation. Activation of the hypothalamic-pituitary-adrenal (HPA) axis by stress can upregulate hepatic hepcidin and reprogram systemic iron fluxes, leading to functional iron deficiency and, in the brain, reduced iron availability, which affects myelination and neurotransmi

Assessing causal relationships between iron level, disorders of iron metabolism, and hepatocellular carcinoma: a Mendelian randomisation study : The first author.

BACKGROUND: Emerging evidence indicates that elevated systemic iron levels and dysregulated iron metabolism are closely associated with the pathogenesis of hepatocellular carcinoma (HCC). Observational studies have linked iron overload (e.g., hereditary hemochromatosis) to an increased risk of HCC, potentially mediated through iron-induced oxidative stress, DNA damage, and chronic inflammatory responses. However, the causal relationship between specific iron-related factors-such as organ-specific iron accumulation, systemic iron metabolism disorders, or exogenous iron exposure (dietary/drug ir

Inherited microcytic anemias due to disorders of iron and heme metabolism: An updated clinical review.

Microcytic anemia is among the most common hematological abnormalities in clinical practice and is usually attributable to iron deficiency, thalassemia traits, or anemia of inflammation. A small but clinically important subset of patients, however, has inherited disorders of iron metabolism or heme synthesis presenting with persistent, unexplained, familial, or iron-refractory microcytosis. Over the past decade, advances in the understanding of the hepcidin-ferroportin axis, erythroid iron regulation, standardized hepcidin assays, and next-generation sequencing have substantially refined diagn

Lif-deficiency promote systemic Iron metabolism disorders and increases the susceptibility of osteoblasts to ferroptosis.

Leukemia inhibitory factor (LIF) is a multifunctional cytokine that plays a crucial role in various biological processes. However, LIF involvement in iron metabolism remains almost unexplored. This study aimed to explore the impact of LIF on systemic iron transportation and its potential role in ferroptosis in osteoblasts. We observed that the Lif-deficient (Lif-/-) mice is characterized by a reduction in bone mass and a decrease in bone mineral density compared with wild-type (WT) mice. Energy-dispersive X-ray spectroscopy revealed a marked increase in iron content on the surface of femurs fr

[Comparison between serum ferritin and induced sideruria in the diagnosis of disorders of iron metabolism].

The A. compare the values of serum ferritin and Desferrioxamine (DFO)-induced sideruria on 73 patients affected by different diseases. The results may be divided into three groups: a first one concerning patients with low serum ferritin and low DFO-induced sideruria, a second one of patients with normal serum ferritin and normal DFO-induced sideruria; in the third group of patients the A. found high values of serum ferritin and high or normal values of DFO-induced sideruria. The different behaviour observed in this III group of patients mostly affected by malignancy may be ascribed, in all tho

Disordered Maternal and Fetal Iron Metabolism Occurs in Preterm Births in Human.

BACKGROUND: Increasing evidence reveals that iron deficiency during pregnancy causes adverse pregnancy outcomes. Thus far, the mechanisms underlying iron deficiency-associated preterm birth are mostly limited to animal studies. Whether the suggested mechanisms exist in human requires further investigation. The goal of this study was to characterize the iron metabolism in both the maternal side and fetal side in pregnant women with preterm birth. METHODS: Serum and placenta samples were collected from 42 pregnant women divided into four groups according to the gestational week. Indicators of i

Iron metabolism disorder regulated by BMP signaling in hypoxic pulmonary hypertension.

BACKGROUNDS AND AIMS: Unexplained iron deficiency is associated with poorer survival in patients with pulmonary hypertension (PH). Bone morphogenetic protein (BMP) signaling and BMP protein type II receptor (BMPR2) expression are important in the pathogenesis of PH. BMP6 in hepatocytes is a central transcriptional regulator of the iron hormone hepcidin that controls systemic iron balance. This study aimed to investigate the effects of BMP signaling on iron metabolism and its implication in hypoxia-induced PH. METHODS AND RESULTS: PH was induced in Sprague-Dawley Rats under hypoxia for 4 weeks
