Conditioned immune responses and the neural-immune connection in animals and humans.

Continuous drug treatment is essential for managing immune-related clinical conditions. However, the significant adverse side-effects associated with immunosuppressive drugs call for alternative or complementary treatment approaches. In experimental animals, as well as in healthy individuals and patient populations, controlled drug-dose reduction has been shown to be achievable via taste-immune associative learning paradigms without compromising efficacy of treatment. A systematic use of such conditioned immunopharmacological responses may therefore be considered as a supportive treatment tool

Neuroimmune interactions in the development and chronification of migraine headache.

Migraine is highly prevalent and debilitating. The persistent headaches in this condition are thought to arise from the activation and sensitization of the trigeminovascular pathway. Both clinical and animal model studies have suggested that neuroimmune interactions contribute to the pathophysiology of migraine headache. In this review, we first summarize the findings from human studies implicating the dysregulation of the immune system in migraine, including genetic analyses, measurement of circulatory factors, and neuroimaging data. We next discuss recent advances from rodent studies aimed a

The Neuroimmune System and the Cerebellum.

The recognition that there is an innate immune system of the brain, referred to as the neuroimmune system, that preforms many functions comparable to that of the peripheral immune system is a relatively new concept and much is yet to be learned. The main cellular components of the neuroimmune system are the glial cells of the brain, primarily microglia and astrocytes. These cell types preform many functions through secretion of signaling factors initially known as immune factors but referred to as neuroimmune factors when produced by cells of the brain. The immune functions of glial cells play

The role of the immune system in the generation of neuropathic pain.

Persistent pain is a sequela of several neurological conditions with a primary immune basis, such as Guillain-Barré syndrome and multiple sclerosis. Additionally, diverse forms of injury to the peripheral or the central nervous systems--whether traumatic, metabolic, or toxic--result in substantial recruitment and activation of immune cells. This response involves the innate immune system, but evidence also exists of T-lymphocyte recruitment, and in some patient cohorts antibodies to neuronal antigens have been reported. Mediators released by immune cells, such as cytokines, sensitise nocicepti

Mast Cells in Neuroimmune Interactions: Mechanisms, Pathophysiological Roles, and Therapeutic Implications.

Mast cells (MCs) are versatile immune cells that play a key role in neuroimmune interactions due to their strategic localization in barrier tissues and perineuronal sites, including the meninges, choroid plexus, thalamus, hypothalamus, and hippocampus within the central nervous system (CNS), as well as peripheral sites such as the skin, gastrointestinal tract, and dorsal root ganglia. They rapidly release stored and newly synthesized mediators, including histamine, tryptase, tumor necrosis factor-α (TNF-α), interleukin-1β (IL-1β), substance P, and prostaglandins, which regulate nerve signaling

The neuroimmune axis: Integrating two systems to decipher itch.

An age-old question in the field of itch research is "why do we experience itch?" Though the physiological reason for acute itch may be clear in the setting of irritants or infections, the mechanisms of pathologic itch remain largely elusive. Ongoing research into the molecular and cellular drivers of chronic pruritus has revealed a palpable synergy between the nervous and immune systems. Classic immune signaling molecules directly activate sensory neurons to evoke itch, while neuropeptides concurrently modulate local immune responses. This bidirectional crosstalk between two widespread system

Neural regulatory networks: challenges and opportunities in the treatment of psoriasis.

Psoriasis is a chronic systemic inflammatory skin disease characterized by complex pathogenic mechanisms involving various cellular interactions. The nervous system significantly influences the development and exacerbation of psoriatic lesions, particularly in innervated regions. Despite the recognized importance of these interactions, a comprehensive understanding of the underlying mechanisms between different cell types and the nervous system in psoriasis is still lacking. This review systematically explores the intricate regulatory interactions between multiple cellular components and the n

Targeted spleen modulation: a novel strategy for next-generation disease immunotherapy.

The spleen, the largest lymphatic organ, comprises a diverse array of immunocytes in approximately one quarter of the body, including T cells, B cells, natural killer cells, and myeloid cells (such as dendritic cells, neutrophils, myeloid-derived suppressor cells, and macrophages). These immune cells undergo dynamic transitions and mobilization, enabling the spleen to execute a wide range of immunological functions. The spleen's structural organization and multicellular composition, along with its reservoir of lymphocytes, facilitate the capture and clearance of blood-borne antigens while also

Reimagining the meninges from a neuroimmune perspective: a boundary, but not peripheral.

Recent advances in neuroscience have transformed our understanding of the meninges, the layers surrounding the central nervous system (CNS). Two key findings have advanced our understanding: researchers identified cranial bone marrow as a reservoir for meningeal immune cells, and rediscovered a brain lymphatic system. Once viewed merely as a protective barrier, the meninges are now recognized as a dynamic interface crucial for neuroimmune interactions. This shift in perspective highlights their unique role in maintaining CNS balance, shaping brain development, and regulating responses to injur

Prevention of immunosuppression in stressed mice by neurotropin(NSP).

It is well known that the immune function can be compromised by stress. To investigate immune function in mice stressed by experimental restraint or unavoidable and opioid dependent stress, we evaluated the changes in total body weight and in organ weights (liver, spleen and thymus) of these animals, as well as the phagocytic activity of macrophages, the cytotoxicity of T cells and inhibitory effects on tumor growth and changes in T cell subset populations. At the same time we evaluated the effects of Neurotropin (NSP), a substance extracted from the inflamed skin of rabbits inoculated with th

Dissecting PANoptosis in the Nervous System: A Unified Cell-Death Mechanism Driving Neuroimmune Activation and Chronic Neuroinflammation.

PANoptosis, a collective form of programmed cell death that includes apoptosis, necroptosis, and pyroptosis, is turning out to be a key player in the neuroimmune activation and sustaining chronic neuroinflammation in the nervous system. PANoptosis, in contrast to single cell death mechanisms, is a web of events coordinating neuronal death, glial cell changes, and inflammatory signals, being implicated in the initiation and progression of neurodegenerative and neuroinflammatory diseases. This review compiles current knowledge of the molecular pathways of PANoptotic signaling, its interaction wi

The pineal gland in multiple sclerosis.

Multiple sclerosis (MS) is a chronic demyelinating disease of unknown etiology. Clinical, neurochemical, and neuroradiological data implicate the pineal gland in the pathophysiology of MS. To investigate the relationship of MS to the pineal gland further, we surveyed the prevalence of pineal calcification (PC) on CT scan in a cohort of 29 MS patients (7 men, 22 women, mean age: 40.1 years, SD = 8.9) who were admitted consecutively to a neurological service for acute exacerbation of symptoms. For the purpose of comparison, we also surveyed the prevalence of choroid plexus calcification (CPC) in

Reduced Sympathetic Innervation in Endometriosis is Associated to Semaphorin 3C and 3F Expression.

Endometriosis is a chronic inflammatory disease and one of the most common causes of pelvic pain. The mechanisms underlying pain emergence or chronic inflammation during endometriosis remain unknown. Several chronic inflammatory diseases including endometriosis show reduced amounts of noradrenergic nerve fibers. The source of the affected innervation is still unclear. Semaphorins represent potential elicitors, due to their known role as axonal guidance cues, and are suggested as nerve repellent factors in different chronic inflammatory diseases. Therefore, semaphorins might influence the progr

Effects on peripheral and central nervous system of key inflammatory intercellular signalling peptides and proteins in psoriasis.

Psoriasis is a chronic systemic inflammatory cutaneous disease. Where the immune system plays an important role in its pathogenesis, with key inflammatory intercellular signalling peptides and proteins including IL-17 and IL-23. The psychoneurological system also figures prominently in development of psoriasis. There is a high prevalence of comorbidity between psoriasis and mental health disorders such as depression, anxiety and mania. Patients with psoriasis often suffer from pathological pain in the lesions, and their neurological accidents could improve the lesions in innervated areas. The

Antidepressant imipramine diminishes stress-induced inflammation in the periphery and central nervous system and related anxiety- and depressive- like behaviors.

In order to relieve anxiety and depression accompanying stress, physicians resort to tricyclic antidepressants, such as imipramine. We had previously shown that imipramine reversed stress-induced social avoidance behavior, and down-regulated microglial activation 24days after stress cessation. To further characterize the effects of imipramine on stress induced neuroimmune dysregulation and associated changes in behavior, the aims of this study were to determine if imipramine 1) ameliorated stress-induced inflammation in the periphery and central nervous system, and 2) prevented stress related

Treatment of cytokine-induced depression.

A high proportion of cancer and hepatitis C patients who receive cytokine immunotherapy develop symptoms of depression that are indistinguishable from those found in major depressive disorders. These symptoms are alleviated by anti-depressant treatment. Moreover, preventive treatment with anti-depressants, in particular selective serotonin reuptake inhibitors (SSRIs) attenuates IFN-alpha-associated symptoms of depression, anxiety, and neurotoxicity. The intermediate mechanisms of these effects are still unclear. Studies suggest that the state of depression is associated with an increase in pla

Emerging mechanisms of ILC2-neuronal crosstalk across the body.

Emerging roles of ILC2 in neuronal crosstalk across organs. In a series of studies published in 2025, ILC2 were shown to regulate nervous system function across a range of organs across the body. Within the intestinal tract, ILC2-derived cytokines acted on enteric neurons to orchestrate immunity to helminths (Wang et al). Intestinal ILC2 are triggered to migrate to the pancreas via enteric neuronal-derived norepinephrine (NE), with pancreatic ILC2 mediating key roles in glucose homeostasis (Sestan et al). ILC2-derived cytokines in the dorsal root ganglia of the sciatic nerve controlled neurose

Neuro-immune Crosstalk in Atopic Dermatitis: A Multiaxial Regulatory Network and Novel Therapeutic Perspectives.

Atopic dermatitis (AD) is a chronic inflammatory disorder driven by complex neuro‑immune interactions beyond classical immune dysregulation. Previous reviews have largely focused on local peripheral neuro‑immune circuits mediating itch and inflammation, but they have not provided an integrative framework connecting peripheral, systemic, and psychological axes. Here, we propose a “multi‑axial neuro‑immune regulatory network” unifying cutaneous, central, and gut–brain–skin pathways to explain the systemic and psychosomatic nature of AD. We highlight neuroplastic remodeling, psycho-neuro-immunolo
