Ante- and post-mortem diagnosis of rabies using nucleic acid-amplification tests.

Sensitivity, specificity and short turn-around time nucleic acid-amplification tests (NATs) have been steadily improving. NATs have been employed in the diagnosis of rabies to distinct different strains, as well as to identify new lyssaviruses. NATs have advantages over traditional methods, such as the direct fluorescence antibody test. They can be applied to fluid samples and brain tissue that is substantially decomposed. NATs can be used as an alternative method for confirmation or exclusion of the diagnosis in a suspected rabies patient. Real-time PCR methods are more favored than conventio

An Integrated and Multi-Target Nucleic Acid Isothermal Analysis System for Rapid Diagnosis of Vulvovaginal Candidiasis.

Rapid identification of Candida species is significant for the diagnosis of vulvovaginal candidiasis (VVC). An integrated and multi-target system for the rapid, high-specificity, and high-sensitivity detection of four Candida species was developed. The system consists of a rapid sample processing cassette and a rapid nucleic acid analysis device. The cassette could process the Candida species to release nucleic acids in 15 min. The released nucleic acids were analyzed by the device as fast as within 30 min, using the loop-mediated isothermal amplification method. The four Candida species could

A Rationally Designed CRISPR/Cas12a Assay Using a Multimodal Reporter for Various Readouts.

The CRISPR/Cas systems offer a programmable platform for nucleic acid detection, and CRISPR/Cas-based diagnostics (CRISPR-Dx) have demonstrated the ability to target nucleic acids with greater accuracy and flexibility. However, due to the configuration of the reporter and the underlying labeling mechanism, almost all reported CRISPR-Dx rely on a single-option readout, resulting in limitations in end-point result readouts. This is also associated with high reagent consumption and delays in diagnostic reports due to protocol differences. Herein, we report for the first time a rationally designed

Rapid detection of human adenovirus subgroup B using recombinase polymerase amplification assay.

Human adenovirus subgroup B (HAdV B) is one of the major pathogens of human respiratory virus infections, which has considerable transmission and morbidity in a variety of populations. Therefore, rapid and specific detection of HAdV B in clinical samples is essential for diagnosis. This study aimed to develop a product for rapid nucleic acid detection of HAdV B using recombinase polymerase amplification assay (RPA) and validate the performance of this method by using clinical samples. Results showed that this method achieved a lower limit of detection (LOD) of 10 copies/μL and had no cross-rea

Isothermal nucleic acid amplification technology for rapid detection of virus.

While the research field and industrial market of in vitro diagnosis (IVD) thrived during and post the COVID-19 pandemic, the development of isothermal nucleic acid amplification test (INAAT) based rapid diagnosis was engendered in a global wised large measure as a problem-solving exercise. This review systematically analyzed the recent advances of INAAT strategies with practical case for the real-world scenario virus detection applications. With the qualities that make INAAT systems useful for making diagnosis relevant decisions, the key performance indicators and the cost-effectiveness of en

Detection of Vibrio cholerae by isothermal cross-priming amplification combined with nucleic acid detection strip analysis.

Vibrio cholerae is a water- and food-borne human pathogen, and V. cholerae serotypes O1 and O139 have attracted attention because of their severe pathogenesis. However, non-O1, non-O139 cholera vibrios (NCVs) were also recently recognized as having virulence properties. In this study, we developed a cross-priming amplification (CPA) method for the detection of all serotypes of V. cholerae. The specificity of the CPA method was tested using a panel of 60 different bacterial strains. All of the V. cholerae strains showed positive results, and 41 other types of bacteria gave negative results. The

[A CRISPR-Cas13a-based amplification-free electrochemical biosensor for rapid detection of bovine viral diarrhea virus].

Bovine viral diarrhea virus (BVDV), a major pathogen in the global bovine industry, causes diarrhea, fever, and reproductive disorders, leading to substantial economic losses. Developing the methods for rapid and accurate detection of BVDV is crucial for epidemic control. Current detection methods have notable limitations. PCR-based nucleic acid amplification techniques rely on sophisticated instruments and complex procedures. CRISPR-Cas13a systems, despite their high specificity, still require nucleic acid pre-amplification, which results in cumbersome workflows and contamination risks. To es

SiNWs-DNA nanoprobes functionalized microchannel for rapid and ultrasensitive virus sensing.

Accurate detection of low viral load infections is crucial for early diagnosis and effective prevention of virus transmission. However, most conventional virus detection methods are time-consuming and require complex nucleic acid amplification steps. Here, we present a glass microchannel sensor functionalized with silica nanowires-DNA (SiNWs-DNA) nanoprobes for simple, rapid, and ultrasensitive detection of Chikungunya virus (CHIKV). The detection is achieved via direct electrochemical signal acquisition after only a 9-min incubation. Without requiring nucleic acid amplification, the sensor ac

Specific and sensitive loop-mediated isothermal amplification (LAMP) method for Madurella strains, eumycetoma filamentous fungi causative agent.

BACKGROUND: Filamentous fungi of the genus Madurella are the primary causative agents of mycetoma, a disease observed in tropical and subtropical regions. Since early diagnostics based on a morphological approach are difficult and have many shortcomings, a molecular diagnostic method suitable for rural settings is required. In this study, we developed the loop-mediated isothermal amplification (LAMP) method to present a foundational technique of the diagnosis of Madurella spp. (M. mycetomatis, M. pseudomycetomatis, M. tropicana, and M. fahalii), the common causative organisms of eumycetoma. Bioinspired DNA sponge interface coupled with deoxyribozyme autography for nanoparticle-free nucleic acid detection on a strip.

Conventional nanoparticle-labeled lateral flow assays often face limitations such as aggregation in incompatible buffers, complex preparation procedures, and high costs. Here, we present a nanolabel-free lateral flow chromatographic platform that integrates lossless strand displacement amplification of deoxyribozyme with a barcoded DNA sponge-based capture interface. In this design, functional G-quadruplex structures serve as intrinsic reporting units instead of conventional nanoparticles, generating catalytic or fluorescent signals upon ligand binding. The DNA sponge network functions as a hi

Inner primer Blockers inhibit non-specific amplification in LAMP.

BACKGROUND: The frequent occurrence of non-specific amplification (NSA) in Loop-mediated Isothermal Amplification (LAMP) has significantly constrained its widespread adoption. When the amount of template is limited or absent, NSA products can be randomly generated, leading to unreliable and inaccurate detection results. The mechanisms underlying NSA remain poorly elucidated, resulting in mitigation strategies that inadequately address the fundamental causes and are therefore suboptimal. RESULTS: Non-template (primers-only) real-time LAMP assays were conducted with three published and three ro

UV-Assisted Hyperbranched Poly(β-amino ester) Modification of a Silica Membrane for Two-Step Microfluidic DNA Extraction from Blood.

Integrating nucleic acid extraction in amplification-based point-of-care diagnostics will be a significant feature for next-generation point-of-care virus detection devices. However, extracting DNA efficiently on a microfluidic chip poses many technological and commercialization challenges, including manual steps, multiple instruments, pretreatment processes, and the use of organic solvents (ethanol, IPA) that inhibit detection, which is not viable with routine testing such as viral load monitoring of transplant patients for post-operative care. This paper presents a microfluidic system capabl

Development of a rapid and sensitive one-pot recombinase polymerase amplification assay for the simultaneous detection of Bacillus anthracis and Yersinia pestis with naked eye result read-out.

Bacillus anthracis and Yersinia pestis are zoonotic bacteria serving as the etiological agents for anthrax and plague, respectively. Due to the rapidly progressing and lethal nature of anthrax and plague, fast and reliable detection is essential for effective disease control and management. Nucleic acid amplification-based approaches offer fast and sensitive pathogen detection. Isothermal amplification techniques (IAT) are being developed to address the limitations of the PCR-based nucleic acid amplification technology. Recombinase polymerase amplification (RPA) is an IAT with low operating te

Active Polymerase on Metal-Organic Frameworks: Space-Confined Amplification for the Extraction-Free Detection of Pathogens in Blood.

The presence of numerous inhibitors and genomic DNA in blood makes their direct use for analysis of clinical pathogens by nucleic acid amplification techniques difficult. Herein, an in situ biomimetic immobilized enzyme system featuring nucleic acid amplification within nanoconfined spaces and self-cleaning properties was constructed for rapid and direct absolute quantification of bacterial bloodstream infections in clinical blood samples without blood culture and extraction steps. The metal-organic framework (MOF) acts as an enzyme-protective carrier, providing 3D nanoconfined spaces, shieldi

Molecular studies of cerebrospinal fluid in human immunodeficiency virus type 1-associated opportunistic central nervous system diseases--an update.

Although the incidence of opportunistic central nervous system (CNS) diseases has markedly declined in developed countries following the advent of highly active antiretroviral therapies (HAARTs), they still represent a major diagnostic and therapeutic challenge over the world. The application of nucleic acid amplification techniques to the study of cerebrospinal fluid (CSF) has contributed substantially to their diagnosis. The detection of specific microbial genomes in the CSF is now the preferred test for some CNS opportunistic diseases, such as progressive multifocal leukoencephalopathy or c

Hybridization chain reaction-DNAzyme amplified switch microplate assay for ultrasensitive detection of magnesium ions.

It is well-recognized that metal ion contaminants present in food and the environment pose a serious threat to human health and contribute to huge economic losses. Therefore, the development of simple, rapid, sensitive, and on-site methods for the detection of metal ions has become an urgent need. Herein, we combined the isothermal hybridization chain reaction (HCR) and a DNAzyme to develop a dual-signal amplification sensing assay for ultrasensitive Mg2+ detection on microplates. In this assay, the linker DNA strand (LDNA) that triggered the formation of the HCR structure was immobilized on a

Accuracy and stability enhanced honey authenticity visual tracing method via false positive-eradicating PCR assisted nucleic acid-capturing lateral flow strip.

Honey authenticity guarantee is crucial for consumer health and fair-trading commerce. New visual false-positive-free molecular lateral flow strip (LFS), termed 5'-3' exonuclease activity -directed false positive-eradicating PCR assisted lateral flow strip (FPE-PCR-LFS) was developed. This FPE-PCR-LFS explored the availability of using a signal-probe as the mediator to integrate the efficient amplification module with visual LFS module. With the genomic DNA extracted from target honey, the designed signal probe would be hydrolyzed and exhausted by the 5'-3' exonuclease activity of Taq DNA poly

CRISPR/Cas12a-assisted visible fluorescence for pseudo dual nucleic acid detection based on an integrated chip.

BACKGROUND: A false negative result is one of the major problems in nucleic acid detection. Failure to screen positive samples for pathogens or viruses poses a risk to public health. This situation will lead to more serious consequences for infectious pathogens or viruses. At present, the common solution is to introduce exogenous or endogenous internal control. Because it amplifies and is detected separately from the target gene, it cannot avoid false negative results caused by DNA extraction failure or reagent inactivation. There is an urgent need for a simple and reliable method to solve the
