Lysosome DMT1-Mediated Renal Cell Ferroptosis Induced by Ochratoxin A.

The mycotoxin ochratoxin A (OTA) was reported to induce ferroptosis in renal cells, but the role of lysosomal DMT1 remained unclear. In the current study, the induction of ferroptosis was first determined by assessments of cell viability, mitochondrial phenotypes, Fe2+ content, and lipid peroxidation levels in NRK-52E and 293T cells. At both mRNA and protein expression levels, OTA exposure resulted in an increase in DMT1 levels. Interestingly, lysosomes were found to colocalize with Fe2+ upon exposure to OTA. Lysosome inhibitor BafA1 reduced OTA-induced ferroptosis and activation of DMT1 and F

[Development of an Analytical Method for Simultaneous Determination of Deoxynivalenol and Ochratoxin A in Wheat].

A simultaneous analytical method was developed for the determination of deoxynivalenol (DON) and ochratoxin A (OTA) in wheat. Analytes were purified from wheat extracts by a multifunctional column and quantified by LC-MS/MS. An interlaboratory study was conducted to validate the analytical method using three spiked and two contaminated wheat samples. The ranges of mean recoveries of DON and OTA were 88-89% and 91-96%, respectively. The RSDs for repeatability and reproducibility were all ≤15%. Furthermore, the levels of DON and OTA in artificially contaminated wheat samples were analyzed by the

Central role of Sigma-1 receptor in ochratoxin A-induced ferroptosis.

Ochratoxin A (OTA), a secondary fungal metabolite known for its nephrotoxic effects, is prevalent in various feeds and food items. Our recent study suggests that OTA-induced nephrotoxicity is linked to the Sigma-1 receptor (Sig-1R)-mediated mitochondrial pathway apoptosis in human proximal tubule epithelial-originated kidney-2 (HK-2) cells. However, the contribution of Sig-1R to OTA-induced nephrotoxicity involving other forms of regulated cell death, such as ferroptosis, remains unexplored. In this investigation, cell viability, malondialdehyde (MDA) levels, glutathione (GSH) levels, and prot

Detection of ochratoxin A by quantum dots-based fluorescent immunochromatographic assay.

Ochratoxin A (OTA) is a toxic metabolite produced mainly by Aspergillus and Penicillium species. A quantitative method was developed for the rapid, simple, and sensitive detection of OTA in corn by quantum dots-based fluorescent immunochromatographic assay (QDs-ICA). The CdSe/ZnS QDs-labelled anti-OTA monoclonal antibody (mAb) conjugates were designed as the fluorescent signal probe. The QDs-ICA included the designation of test line (T line) and control line (C line), which were sprayed with optimal concentrations of the OTA-OVA and staphylococcal protein-A (SPA), respectively. Under the optim

Epithelial-fibroblast cross talk aggravates the impact of the nephrotoxin ochratoxin A.

BACKGROUND: Chronic nephropathies result from different pathogenic agents, including nutritional factors triggering vicious pathophysiological cycles. Ochratoxin A (OTA) is a globally occurring nephrotoxic mycotoxin detectable in a variety of foodstuff and suspected to cause tubulointerstitial damage. The underlying mechanisms are not sufficiently understood, compromising risk assessment. Because crosstalk of proximal tubule cells with fibroblasts is crucial for tubulointerstitial damage, we investigated the effects of OTA in co-culture of these two cell types. METHODS: Rat renal proximal tub

MicroRNA profiling in kidney in pigs fed ochratoxin A contaminated diet.

OTA is a toxic metabolite produced by fungus belonging to Aspergillus and Penicillium genera. Kidney is the main target of this toxin; OTA is considered as one of the etiological factors at the origin of the human Balkan endemic nephropathy. microRNA are short non-coding transcrips (18-22 nucleotides in length) regulating key cellular processes. Various miRNAs have been established to play important roles in development of renal carcinoma and urothelial cancer. The objective of this study is to analyse the miRNA profiling in the kidney of piglets experimentally intoxicated with feed contaminat

Ameliorative Influence of Ajwa Dates on Ochratoxin A-Induced Testis Toxicity.

Ajwa dates (Phoenix dactylifera), cultivated particularly in Al-Madina Al-Monawarh in Saudi Arabia, and considered as a rich source of natural antioxidants such as anthocyanins, carotenoids and phenolics. It is believed that Ajwa dates have a wide range of protective effects. For this reason, this work aimed to investigate the ameliorative effect of Ajwa dates aqueous extract against ochratoxin A (OTA)-induced testicular lesions in rats. Rats were divided into four groups and treated for 28 days. Group I served as normal control, group II (OTA exposed) treated orally with 289 μg/kg/day. Group

Microfabricated biosensor for the simultaneous amperometric and luminescence detection and monitoring of Ochratoxin A.

The low molecular weight hapten, Ochratoxin A (OTA), is a natural carcinogenic mycotoxin produced by Aspergillus and Penicillium fungi and so it commonly appears in wines, other foods, and in the environment. An amperometric biosensor has been developed that uses the immobilized synthetic peptide, NFO4; which possesses a high binding affinity and thus provides for molecular recognition of OTA; simulating the mycotoxin-specific antibody. Biotransducers were produced from a microlithographically fabricated electrochemical cell-on-a-chip that uses the microdisc electrode array working electrode f

Novel fluoroimmunoassays for detecting ochratoxin A using CdTe quantum dots.

Novel direct and indirect competitive fluorescence-linked immunosorbent assays (cFLISA and icFLISA) for detection of ochratoxin A (OTA) were described using CdTe quantum dots (QDs) as fluorescent label. CdTe QDs were successfully synthesized, which had an emission wavelength of 615 nm. The high purity monoclonal antibody against OTA was prepared through cell thawing and the octylic acid-ammonium sulfate method. The OTA MAbs were successfully coupled with CdTe QDs, and which also retained the original biological activity. The 50% inhibition values (IC50 ) of the cFLISA and icFLISA were 0.630 ng

Effects of ochratoxin A on the mouse immune system after subchronic exposure.

The effects on the immune system of oral, subchronic exposure to ochratoxin A (OA) at 6, 250 or 2600 micrograms/kg diet were studied in female Balb/c mice. After 28 days of exposure, antibody production plague-forming cells/spleen, was suppressed in a dose-dependent manner which was significant in the two highest exposure groups. In addition, a decrease in thymocyte cell counts was seen in the 250-micrograms/kg group. After 90 days exposure, flow cytometry analysis of thymic lymphocyte subpopulations revealed a decreased proportion of mature (CD4+ or CD8+) cells. Furthermore, the mitogenic res

Triple-readout immunoassay based on copper ion trigger for the detection of ochratoxin A.

BACKGROUND: Ochratoxin A (OTA) is a widespread mycotoxin with nephrotoxic, neurotoxic and immunotoxic properties. It exhibits elevated thermal stability and a prolonged half-life and is resistant to removal. Long-term exposure to OTA increases the risk of carcinogenesis and influences human health. The detection methods of single signal mode had poor anti-interference ability and were prone to false positive results, while the detection method of multi-signal mode has higher adaptability to conditions and can offset environmental interference through self-correction. Therefore, proposing multi

Unveiling the hub genes associated with ochratoxin A-induced hepatotoxicity in broiler chickens.

Ochratoxin A (OTA) widely exists in raw food and feed materials and can induce liver damage and toxicity. However, the mechanisms of OTA-induced hepatotoxicity were largely unknown. Thus, our study aimed to uncover the vital genes relevant to OTA-induced hepatotoxicity in broiler chickens. Gene expression data of chicken embryo primary hepatocytes (CEPHs) in OTA-treated and control groups were obtained from the GEO database. Totally 1407 differentially expressed genes (DEGs) were selected, of which 850 and 557 genes were up- and downregulated in OTA-treated CEPHs. Gene ontology (GO) enrichment

Effects of Aspergillus niger Infection on the Quality of Jujube and Ochratoxin A Cumulative Effect.

The jujube is one of the most popular fruits in China because of its delicious taste and high nutritional value. It has a long history of usage as an important food or traditional medicine. However, the jujube is easily infected by fungi, which causes economic losses and threatens human health. When the jujube was infected by Aspergillus niger (H1), the changes in nutritional qualities were determined, such as the content of total acid, vitamin C, reducing sugar, etc. In addition, the ability of A. niger (H1) to produce ochratoxin A (OTA) in different inoculation times and culture media was ev

A reproductive and developmental screening study of the fungal toxin ochratoxin A in Fischer rats.

The presence of the mycotoxin ochratoxin A (OTA) in cereal grains is due to the growth of toxigenic Penicillium mold on stored crops. Human exposure to OTA is higher in infants, toddlers, and children than in adolescents and adults, based on exposure assessments of ng OTA consumed/kg body weight/day. Ochratoxin A is nephrotoxic and teratogenic in animals, but its effects on juveniles exposed during the reproduction and development period have not been studied. To address this, Fischer rats were exposed to 0, 0.16, 0.4, 1.0, or 2.5 mg OTA/kg diet throughout breeding, gestation, and lactation an

Ochratoxin A promotes porcine circovirus type 2 replication in vitro and in vivo.

Ochratoxin A (OTA), a worldwide mycotoxin found in food and feeds, is a potent nephrotoxin in animals and humans. Porcine circovirus-associated disease (PCVAD), including porcine dermatitis and nephropathy syndrome, is a worldwide swine disease. To date, little is known concerning the relationship between OTA and porcine circovirus type 2 (PCV2), the primary causative agent of PCVAD. The effects of OTA on PCV2 replication and their mechanisms were investigated in vitro and in vivo. The results in vitro showed that low doses of OTA significantly increased PCV2 DNA copies and the number of infec
