Successful 18-h acellular extracorporeal perfusion and replantation of porcine limbs - Histology versus nerve stimulation.

The current standard for composite tissue preservation is static cold storage (SCS) and is limited to 6 h until irreversible muscle damage occurs. Extracorporeal perfusion (ECP) is a promising technique for prolonged preservation, however, functional results have been scarcely researched. This article assessed neuromuscular function and compared results to histological alterations to predict muscle damage after ECP. Forelimbs of twelve Dutch landrace pigs were amputated and preserved by 4 h SCS at 4-6 °C (n = 6) or 18 h mid-thermic ECP with University of Wisconsin solution (n = 6). Limbs were

Modified perfusion fluid for renal transplantation in developing countries: our initial experience.

INTRODUCTION: Organ preservation is an important step in transplantation. Perfusion fluid plays a vital role in this matter. As in many other developing countries, perfusion fluid is not readily available and is costly. Alternatively, the commonly used Hartmann's solution does not have preservation quality. Hence, to meet the demand, a modified perfusion fluid was prepared following standard preservative flush principles. MATERIALS AND METHODS: Hartmann's solution was modified by adding potassium chloride, magnesium sulfate, mannitol, dexamethasone, and insulin. The whole process was performe

Vitality of human lip fibroblasts in milk, Hanks balanced salt solution and Viaspan storage media.

The purpose of the study was to compare the effect of storage in milk, Hanks balanced salt solution (HBSS) and Viaspan on the vitality of human lip fibroblasts. Confluent monolayers of the fibroblasts were grown in petri dishes. The growth medium was poured off and replaced in 27 dishes each with Viaspan, fresh whole milk and Hanks balanced salt solution respectively. Three of the original plates were analyzed at time zero to obtain the average number of cells each plate contained at confluence. At times ranging from 2 to 168 hours the average number of vital cells remaining was measured using

The influence of perfusion solution on renal graft viability assessment.

BACKGROUND: Kidneys from donors after cardiac or circulatory death are exposed to extended periods of both warm ischemia and intra-arterial cooling before organ recovery. Marshall's hypertonic citrate (HOC) and Bretschneider's histidine-tryptophan-ketoglutarate (HTK) preservation solutions are cheap, low viscosity preservation solutions used clinically for organ flushing. The aim of the present study was to evaluate the effects of these two solutions both on parameters used in clinical practice to assess organ viability prior to transplantation and histological evidence of ischemic injury afte

New preservation strategies for preventing liver grafts against cold ischemia reperfusion injury.

BACKGROUND: In spite of improvements in University of Wisconsin (UW) preservation solution, the injury from grafts during cold storage is an unresolved problem in liver transplantation. The aim of the present study was to evaluate the beneficial effect on ischemia-reperfusion injury associated with liver transplantation of the inversion of K(+) and Na(+) concentrations and the replacement of hydroxyethyl starch (HES) by polyethylene glycol (PEG) in UW preservation solution. METHODS: Using an orthotopic liver transplantation model, the effects on rat liver preservation of a modified preservati

Liver transplantation: current and potential applications of magnetic resonance spectroscopy.

Magnetic resonance spectroscopy (MRS) allows the noninvasive measurement of whole organ metabolism due to the presence of the MR-sensitive nucleus phosphorus 31 in adenosine triphosphate (ATP), its precursors, and break-down products. In small animal liver transplant studies it has been used to analyze the metabolic effects of cold and warm ischemia, hypothermic reperfusion, and the relative efficacy of different organ preservation solutions. In recent large animal studies MRS has been developed to provide continuous dynamic information on ATP metabolism during graft reperfusion and the bioene

Effect of preservation solutions for static cold storage on kidney transplantation outcomes: A National Registry Study.

This study aimed to evaluate how 5 preservation solutions for static cold storage affected kidney transplant outcomes. It included all first single kidney transplants during 2010-2014 from donations after brain death in the French national transplant registry, excluding preemptive transplants and transplants of kidneys preserved with a hypothermic perfusion machine. The effects of each preservation solution on delayed graft function (DGF) and 1-year transplant failure were evaluated with hierarchical multivariable logistic regression models. The study finally included 7640 transplanted kidneys

The isolated perfused rat liver: standardization of a time-honoured model.

For many years, the isolated perfused rat liver (IPRL) model has been used to investigate the physiology and pathophysiology of the rat liver. This in vitro model provides the opportunity to assess cellular injury and liver function in an isolated setting. This review offers an update of recent developments regarding the IPRL set-up as well as the viability parameters that are used, with regards to liver preservation and ischaemia and reperfusion mechanisms.A review of the literature was performed into studies regarding liver preservation or liver ischaemia and reperfusion. An overview of the

Hepatocellular uptake of cyclodextrin-complexed curcumin during liver preservation: A feasibility study.

The increasing demand for donor organs and the decreasing organ quality is prompting research toward new methods to reduce ischemia reperfusion injury (IRI). Several strategies have been proposed to protect preserved organs from this injury. Before curcumin/dextrin complex (CDC), a potent antioxidant and anti-inflammatory agent, can be used clinically we need to better understand the intracellular uptake under hypothermic conditions on a rat model of liver donation after circulatory death (DCD) and brain death (DBD). To be able to use the fluorescence of CDC for quantification the stability of

Successful extended lung preservation with UW solution.

Maximum preservation times of 4-6 hr continue to plague lung transplantation. The high-potassium colloid University of Wisconsin solution (UWS) has proved superior to the crystalloid modified Eurocollins' solution (ECS) for preservation of the liver, kidney, and pancreas. The purpose of this study was to compare UWS and ECS for extended lung preservation using a technique of combined pulmonary and bronchial artery perfusion. Simultaneous pulmonary artery and bronchial artery (via a closed aortic segment) perfusion was employed to harvest the lungs of ten mongrel dogs (wt 25-35 kg) using either

Stem Cell-Derived Mitochondria-Enriched Preservation Solutions as Organelle-Based Therapeutic Strategy for Cold Storage of Liver and Kidney.

Organ preservation remains a critical challenge in transplantation, primarily due to hypothermia-induced oxidative stress and metabolic dysfunction. Here, we report a mitochondria-enriched, cell-free preservation strategy by supplementing standard preservation solutions with freshly isolated mitochondria derived from human induced pluripotent stem cell-mesenchymal stem cells (MSC-mt). MSC-mt retained intact ultrastructure and functional biophysical properties. In vitro, MSC-mt were internalized by hepatocyte- and kidney-derived cells, reduced oxidative stress, preserved ATP levels, and attenua

Comparison of the University of Wisconsin preservation solution and other crystalloid perfusates in a 30-hour rabbit lung preservation model.

The University of Wisconsin solution, which contains a high potassium concentration (120 mmol/L), was evaluated for rabbit lung preservation by comparing it with a modified University of Wisconsin solution with low potassium (4 mmol/L), a low-potassium dextran solution (4 mmol/L), and simple surface cooling. In the first three groups rabbit lungs were flushed in situ with the solution (n = 5 in each group); then the lung-heart block was harvested and stored at 10 degrees C for 30 hours. In the surface cooling group the lungs were harvested without flushing and then simply immersed in saline an

Two-layer cold storage method for pancreas and islet cell transplantation.

The two-layer cold storage method (TLM) was first reported in 1988, consisting of a perfluorochemical (PFC) and initially Euro-Collins' solution, which was later replaced by University of Wisconsin solution (UW). PFC is a biologically inert liquid and acts as an oxygen-supplying agent. A pancreas preserved using the TLM is oxygenated through the PFC and substrates are supplied by the UW solution. This allows the pancreas preserved using the TLM to generate adenosine triphosphate during storage, prolonging the preservation time. In a canine model, the TLM was shown to repair and resuscitate war

Prolonged pancreas preservation using a simplified UW solution containing polyethylene glycol.

The University of Wisconsin solution is considered the most effective universal flush and cold storage solution to date, and is now being widely applied clinically in organ transplantation. The results of this study show that Cardiosol, a modified cardioplegic solution containing 5% polyethylene glycol (PEG20M, MW 17,000 daltons), is significantly superior to UW (P less than 0.001) in the flush perfusion and hypothermic storage of pancreases for more than 36 hr prior to transplantation into streptozotocin-induced diabetic rats. When the pancreases were stored in Cardiosol, the 1-week survival

New strategy for prolonging the preservation time of hearts for transplantation.

Our study concerned the findings that rat and rabbit heart transplants do not survive after six hours. They become dark, hard and fail to contract within 2 min after reperfusion and never regain their function. We tested the supplementation of solutions for heart transplant preservation with tetrahydrobiopterin (H4B) and L-arginine (L-ARG) to maintain the oxidative and reductive domains of the endocardial NO synthase. We decided to study the excised rabbit hearts preserved in Hank's balanced salt solution (HBSS) at 0 degrees C supplemented with different concentrations of H4B (0, 1, 5, 10 or 1

Limited Risk of Microbial Contamination During Hypothermic and Normothermic Machine Perfusion of Donor Kidneys.

BACKGROUND: Both hypothermic machine perfusion (HMP) and normothermic machine perfusion (NMP) are increasingly used for preservation of deceased donor kidneys. However, especially NMP can pose as a risk for microbial contamination of the kidney graft as the 37°C perfusate can act as a breeding ground for microbial contaminants. METHODS: In this study, we investigated the cultures of HMP and NMP perfusates of deceased donor kidneys. RESULTS: Between January 2021 and April 2024, a total of 99 perfusates were examined (73 HMP and 26 NMP perfusates)-. We found that contamination of HMP perfusate

The development of an extended normothermic ex vivo reperfusion model of the sheep uterus to evaluate organ quality after cold ischemia in relation to uterus transplantation.

INTRODUCTION: Uterus transplantation has recently proved that infertility in women with uterine factor infertility can be cured. It is still an experimental procedure with numerous critical details remaining to be established, including tolerance to warm and cold ischemic insults. In preparation for human uterus transplantation trials, most teams use the sheep as a model system for research and team training, since the vasculature and the uterus is of similar size as in the human. We, therefore, aimed to develop an ex vivo sheep uterus reperfusion platform that mimics the reperfusion situation

An innovative model of en bloc liver-pancreas-kidney preservation via ex vivo hypothermic acellular machine perfusion.

BACKGROUND: Liver machine perfusion (MP) has emerged as a promising organ preservation modality. Recent studies have shown that the addition of the kidneys to the circuit improves the biochemical environment and could benefit liver preservation. The aim of this study was to explore the technical and anatomical feasibility of en bloc liver-pancreas-kidney MP. We also examined the safety of ex vivo perfusion with a nonoxygen carrier solution and its effects on acid-base and metabolic parameters using this novel multivisceral perfusion platform. METHODS: Five multivisceral allografts, including

Transport media for avulsed teeth: a review.

Management protocols for avulsed teeth should include management of the pulp and periodontal ligament (PDL) cells in order to improve the long-term prognosis and survival of these teeth. The use of an inappropriate transport or storage medium potentially increases the risk of PDL cell necrosis, which can result in ankylosis and replacement resorption of the tooth root. Considering the critical role of these media, an informed choice of a suitable medium is essential for a favourable outcome. The literature regarding transport media for avulsed teeth was reviewed using PubMed/MEDLINE up to Febr
