Novel duck reovirus (NDRV) causes high morbidity in ducklings, and recovered ducklings are often remarkably stunted in growth. In this study, four NDRV strains were isolated from the NDRV outbreaks that occurred in different regions of Shandong province, China. The biological characteristics and pathogenicity of the four NDRV strains were elucidated, and the N20 was identified as a naturally attenuated strain. Three-day-old ducklings were immunized with live N20 strain (100 ELD50/duck), and challenged with 104.52 ELD50 of virulent N19 strain at 7 days post immunization. The vaccinated duckling

Avian reoviruses are the causative agent of viral arthritis/tenosynovitis in chickens and turkeys. Clinical signs of disease include swelling of the hock joints accompanied by lesions in the gastrocnemius and digital flexor tendons causing lameness in addition to hydropericardium. The economic impact is significant as it results in poor weight gain, increased feed conversion ratios and condemnations at the processing plant. Vaccination with both live attenuated and inactivated oil emulsion vaccines have been used successfully for decades to control the disease. Current commercial vaccine strai

Mitochondrial metabolic homeostasis serves as a critical checkpoint that integrates cellular bioenergetics with innate immunity. While viruses often subvert mitochondrial functions to manipulate cell fate, the specific viral determinants that directly reprogram host metabolic enzymes to drive inflammatory injury remain incompletely defined. Here, using infection with a highly virulent novel duck reovirus (NDRV) strain as a model of systemic immunopathology, we identify the viral capsid protein σC as a metabolic virulence factor. We show that σC binds to the mitochondrial targeting sequence (MT

In recent years, outbreak of the novel duck reovirus (NDRV) disease has occurred frequently in duck populations. Due to its rapid spreading, absence of effective control methods, and high treatment costs, the NDRV disease has caused huge losses to waterfowl breeding in China. As reported, four non-structural (NS) proteins are encoded by the NDRV genome, among which the σNS protein is an RNA-binding protein that can improve the stability of bound RNA by forming oligomers (Adams and Cory, 1998). Nevertheless, the mechanism by which it facilitates reovirus replication remains ambiguous. According

BACKGROUND: In China, Newly emerging duck reovirus (NDRV) variants have been causing major disease problems in cherry valley ducks. NDRV has the potential to cause high morbidity and 5-50% mortality rates. Severe hemorrhagic-necrosis in the liver and spleen were commonly seen in NDRV affected ducks. The availability of upgraded methods for rapid diagnosis of newly emerging DRV variants is crucial for successful DRV infection control and prevention.
RESULTS: In this study, we present a TaqMan-based real-time PCR assay (RT-qPCR) for the detection of NDRV infection. Using the conserved regions w

The Muscovy duck reovirus (MDRV) is a highly pathogenic virus that causes substantial economic losses in the Muscovy duck industry. While MDRV poses a significant threat to Muscovy ducklings, no vaccine candidates are available to date to alleviate MDRV infection throughout the world. The present study presents efforts toward establishing an attenuated vaccine for MDRV. For this purpose, a live attenuated vaccine strain named CA was obtained via alternate propagation of the MDRV isolate MW9710 in both Muscovy duck embryo fibroblasts (MDEFs) and chicken embryo fibroblasts (CEFs) for 90 passages

Waterfowl reovirus (WRVs) includes two genotypes, Genotype I (Muscovy duck reovirus, MDRV) and Genotype II (novel duck reovirus, NDRV). They have the same clinical manifestations, but different pathogenicity, and can co-infect individual hosts, resulting in diagnostic ambiguity that cannot be solved by single-target detection. Here, we report a single-tube dual-channel platform coupling recombinase polymerase amplification (RPA) with orthogonal CRISPR effectors: LbCas12a targeting the conserved S4 gene of MDRV via a ROX-labeled ssDNA reporter, and LwaCas13a targeting the S1 gene of NDRV via a

BACKGROUND: Turkey arthritis reovirus (TARV) causes arthritic lameness in market-age turkeys. Since 2011, highly pathogenic TARV strains have caused significant economic losses in the turkey industry due to increased culling, reduced market weights, and decreased carcass quality, necessitating more effective control measures. Autogenous vaccine prevention strategies have been inefficacious partly due to a limited understanding of age-related susceptibility of turkeys to TARV. This study investigated age-related host and gut microbiota responses to TARV infection in commercial turkeys derived f

Infection with novel duck reovirus (NDRV) results in severe splenic necrosis, leading to immunosuppression, secondary infections with other pathogens, and impairment of the immune effect of the vaccine. However, little is known about NDRV-induced spleen injury and its antagonistic mechanism on the host immune response. In this study, we conducted pathological and comparative transcriptomic analyses of NDRV-infected duck spleens. Our findings elucidated the histopathological progression of splenic necrotic foci formation following NDRV infection and identified splenic macrophages as the primary

A novel strain of duck reovirus (DRV) associated with a high mortality in Pekin ducklings in China, 2013, was isolated and characterized. This strain (designated as HN5d) grew well in Vero cells and produced marked cytopathic effects. HN5d contains 10 dsRNA genome segments, a typical feature of avian orthoreovirus. Following cloning, sequencing, and sequence analysis of the genome segments, a unique deletion of 18 amino acids was found in the sigma C protein of HN5d when compared with that of the recent Chinese waterfowl reoviruses (e.g., DRV 091). Phylogenetic analysis of cDNA amplicons of se

The complete sequence of a reovirus, strain 815-12 associated with necrotic focus formation in the liver and spleen of Muscovy ducklings in China, was determined and compared with sequences of other duck-, goose-, and chicken-origin reoviruses. The 815-12 genome comprised 22,969 bp with 10 dsRNA segments ranging from 1125 bp (S4) to 3958 bp (L1), all of which (except S4) were almost identical in length to the cognate segments of other waterfowl and chicken isolates. Detailed analyses revealed that 815-12 and other waterfowl isolates contained the conserved 3'-terminal pentanucleotide sequence

To investigate the immunogenic cross-reactivity among different novel duck reovirus isolates, animals immunized with monovalent inactivated duck reovirus were challenged with homologous and heterologous viruses, and serum samples were collected after immunization for cross-neutralization test. The results showed that the virus neutralizing titer against homologous virus was significantly higher than that against heterologous virus(p < 0.000001), which indicated that there was an important difference in antigenicity between DE13 and WL01 strains, which affected the specificity of protective vir

INTRODUCTION: Ducks rank among the most important sources of animal protein globally, yet hepatic and splenic hemorrhage and necrosis in Muscovy ducks present a critical challenge to the poultry industry. The causes behind such diseases are often multifaceted, involving both established and newly emerging pathogens.
METHODS: In this study, we leveraged metatranscriptomic sequencing to profile the intestinal viral communities of healthy and diseased Muscovy ducks from a Guangdong Province farm that experienced a hepatic and splenic hemorrhage in June 2024.
RESULTS: Our findings revealed marke

Condemnations in slaughterhouses are a recurring cause of losses for the poultry industry. This study aimed to evaluate the involvement of avian reovirus (ARV) in arthritis-related condemnations in a broiler slaughterhouse under Brazilian Federal Inspection. A total of 150 pelvic limbs condemned for arthritis were collected from the inspection line at the time of condemnation, originating from 30 flocks not vaccinated against ARV. For the detection of ARV, real-time polymerase chain reaction following reverse transcription (RT-qPCR) was performed on a pool of five gastrocnemius tendons from ea

A major cause of lameness in turkeys is reoviral arthritis, driven by turkey reovirus (TRV) infection. In the U.S., TRV was first isolated in the 1980s but re-emerged as a significant pathogen causing arthritis in 2011. Since then, TRV outbreaks have spread nationwide across turkey-producing regions and have occasionally resulted in hepatitis-associated pathotypes. The absence of a consistently effective commercial vaccine continues to hinder disease control efforts. To better understand TRV's evolutionary trajectory and transmission dynamics, we analyzed 211 complete TRV genome sequences coll

This study conducted a comprehensive survey of Fowl Adenovirus (FAdV), Astrovirus (CAstV), and Avian Reovirus (ARV) in Brazilian poultry, aiming to understand their epidemiological aspects. A total of 1,988 commercial flocks were analyzed across various Brazilian states, resulting in 4,568 analyses. The findings indicated a 25.5 % positivity rate among flocks showing suggestive symptoms, with at least one virus detected in all industrial poultry-producing states. Single-agent infections were more prevalent than co-infections. Among states with over 100 flocks analyzed, the highest FAdV prevale

The newly emerged disease, duck beak atrophy and dwarfism syndrome (BADS), is caused by novel goose parovirus (N-GPV). Although N-GPV infection has severe consequences, few methods for detecting this virus have been developed. Therefore, the availability of rapid and reliable molecular diagnostic methods would aid future studies of this novel virus. Clinical specimens from 138 suspected cases of N-GPV infection and 120 cloacal swabs from breeding ducks were used in this study. The targeted sequence of N-GPV cloned into the pMD18-T vector was used to generate the N-GPV DNA standard curve. The s

Runting-stunting syndrome (RSS) is an enteric disease of young chickens with complex aetiopathogenesis. It is often linked to the isolation of multiple virus species and histologically characterized by cystic enteropathy. We investigated the correlation between the detection frequency of avian reovirus, astrovirus, parvovirus, and rotavirus A and the presence of microscopic lesions consistent with cystic enteropathy in 123 broiler chicken submissions to the Alabama State Laboratory System (Auburn, AL, USA). Multi-viral detections were found to be predictive of cystic enteropathy with an increa

INTRODUCTION: Avian reovirus (ARV) infection causes viral arthritis in broiler chickens, leading to lameness and substantial economic loss in poultry production. Current conventional diagnostic methods detect infection only after clinical signs appear, limiting the ability to prevent disease progression. Hence, this study utilized serum metabolomics to identify early biomarkers of ARV infection prior to tissue damage.
METHODS: A total of 150 serum samples were collected from ARV-challenged broilers at 24, 48, and 72 h post-infection, along with uninfected controls. The samples underwent liqui
