Acidification-based mineral weathering mechanism involves a glucose/methanol/choline oxidoreductase in Caballeronia mineralivorans PML1(12).

UNLABELLED: While mineral weathering (MWe) plays a key role in plant growth promotion and soil fertility, the molecular mechanisms and the genes used by bacteria to weather minerals remain poorly characterized. Acidification-based dissolution is considered the primary mechanism used by bacteria. This mechanism is historically associated with the conversion of glucose to protons and gluconic acid through the action of particular glucose dehydrogenases (GDH) dependent on the pyrroquinoline quinone (PQQ) cofactor. Recently, bacteria lacking the GDH-PQQ system have been shown to perform the same e

Dietary pyrroloquinoline quinone alleviated intestinal damage of juvenile yellow catfish (Pelteobagrus fulvidraco) induced by oxidized fish oil: insight from intestinal digestive enzyme, antioxidant status, morphology, inflammation, and microbiota.

Feeding oxidized fish oil to cultured fish harms their intestine, but whether pyrroloquinoline quinone (PQQ) can counteract this damage had not been explored. This 56-day study investigated the protective effects of dietary PQQ against oxidized fish oil-induced intestinal injury in juvenile yellow catfish (Pelteobagrus fulvidraco). Four hundred and eighty yellow catfish (initial weight approximately 12 g) were randomly assigned to four groups in triplicate: control (fresh fish oil), PQQ (fresh oil + 5 mg/kg PQQ), OFO (oxidized oil), and PFO (oxidized oil + 5 mg/kg PQQ). Fish fed OFO diet showe

Synthetic Biology Strategies for Harnessing Bacterial Glucose Oxidation Pathways.

As the most important carbon source for bioproduction, glucose is transported and enters the central carbon metabolic pathway directly in most microorganisms. However, certain bacteria utilize membrane-bound dehydrogenases to oxidize glucose to gluconic acid (GA) in the periplasmic space. GA is subsequently converted either to 5-keto-d-gluconate by PQQ-dependent gluconate dehydrogenase (GADH) or to 2-keto-d-gluconate by FAD-dependent GADH, with the latter further oxidized to 2,5-diketo-d-gluconate. This review systematically examines the composition, distribution, physiological functions, and

Serum anti-Müllerian hormone response to pyrroloquinoline quinone supplementation in healthy women: no overall change and exploratory subgroup findings.

INTRODUCTION: Oocyte maturation requires substantial ATP, and the resulting oxidative stress may impair follicular development. Pyrroloquinoline quinone (PQQ), an antioxidant compound, protects mitochondria and promotes follicular development in animal models; however, its effects on human ovarian function remain unclear. METHODS: This single-arm, open-label study prospectively evaluated the effects of oral PQQ supplementation on ovarian reserve and related clinical and biochemical outcomes. The primary outcome was serum anti-Müllerian hormone (AMH). Secondary outcomes included luteinizing ho

Effects of pyrroloquinoline quinone supplementation on growth performance and small intestine characteristics in weaned pigs.

This study was conducted to explore the effect of graded levels of pyrroloquinoline quinone disodium (PQQ·Na2) on the performance and intestinal development of weaned pigs. A total of 216 pigs weaned at 28 d were assigned in a randomized complete block design to 6 diets containing 0, 1.5, 3.0, 4.5, 6.0, or 7.5 mg/kg PQQ·Na2 for 28 d. Performance, diarrhea incidence, intestinal morphology, redox status, cytokines, and the expression of tight junction proteins were determined. Pigs had increased ADG (linear, P < 0.01), G:F (quadratic, P < 0.01), and lower diarrhea incidence (P < 0.01) with the i

Model studies on calcium-containing quinoprotein alcohol dehydrogenases. Catalytic role of Ca2+ for the oxidation of alcohols by coenzyme PQQ (4,5-dihydro-4,5-dioxo-1H-pyrrolo[2,3-f]quinoline-2, 7,9-tricarboxylic acid).

Mechanistic studies on the action of calcium-containing quinoprotein alcohol dehydrogenases have been performed by using a series of PQQ model compounds in anhydrous organic media. The PQQ model compounds are shown to form 1:1 complexes with a series of alkaline earth metal ions by spectroscopic methods and theoretical calculations. The site of coordination of the PQQ molecule to the metal ions in solution is indicated to be the same as in the case of enzymatic systems. It has also been found that Ca2+ binds to the quinone most strongly among the alkaline earth metal ions. Formation of the C-5

Exogenous Pyrroloquinoline Quinone Application Enhances Low-Phosphorus Adaptation in Rice by Modulating Redox Homeostasis Under Variable Light.

Phosphorus (P) bioavailability in soil is often limited, restricting plant growth. Pyrroloquinoline quinone (PQQ) is a natural quinone compound with potent redox-cycling capabilities. In this study, we investigated the physiological effects of exogenously applied PQQ on rice adaptation to P deficiency. A P-sensitive rice cultivar (Meixiangzhan) was treated with 0.1 μM PQQ under different light intensities (High Light, HL: 800 μmol m-2 s-1; Low Light, LL: 300 μmol m-2 s-1) and P supplies (Sufficient P, SP; Deficient P, DP). The effects of PQQ on rice growth, P uptake, photosynthesis, and the an

Pyrroloquinoline Quinone Protects Against Light-Induced Retinal Damage in Association with the Suppression of c-Fos Signalling.

Age-related macular degeneration (AMD) is a progressive retinal disorder characterised by oxidative stress and inflammation. Although pyrroloquinoline quinone (PQQ) has been reported to exert neuroprotective effects, its specific efficacy in in vivo models of AMD pathophysiology has not yet been elucidated. In this study, we evaluated the protective effects of PQQ against all-trans-retinal (ATR)-induced cytotoxicity in ARPE-19 cells and light-induced photoreceptor degeneration in rats. Pretreatment of ARPE-19 cells with PQQ dose-dependently mitigated ATR-induced cytotoxicity. In the in vivo mo

Expanding Aldopentose Catalysis of a Novel PQQ-Dependent Glucose Dehydrogenase without Compromising Native d-Glucose Activity.

Lignocellulosic biomass is an abundant renewable resource that is rich in aldohexoses and aldopentoses. Efficient biocatalysts for aldopentose oxidation remain limited, and few enzymes exhibit high catalytic efficiency toward both aldohexoses and aldopentoses. Here, a naturally promiscuous pyrroloquinoline quinone-dependent glucose dehydrogenase (PQQ-GDH) from Pseudomonas fragi NL20W was selected as an engineering template and tailored by semirational design to expand aldopentose oxidation while preserving native d-glucose activity. The V706P/L723 M mutant exhibited markedly enhanced catalytic

Pyrroloquinoline Quinone Prevents Estrogen Deficiency-Induced Osteoporosis by Inhibiting Oxidative Stress and Osteocyte Senescence.

Accumulating studies have shown that oxidative stress increases with aging, which is related to the pathophysiology of postmenopausal osteoporosis. Pyrroloquinoline quinone (PQQ) is a natural anti-oxidant with anti-oxidative and anti-aging effects. However, it is unclear whether PQQ has a protective role against estrogen deficiency-induced osteoporosis. Here, we evaluated the efficacy of PQQ on bone mineral density, bone microarchitecture, bone turnover and biomechanical strength in ovariectomy (OVX)-induced osteoporosis mouse model. Although dietary PQQ supplement did not affect serum E2 leve

Construction of nerve guide conduits from cellulose/soy protein composite membranes combined with Schwann cells and pyrroloquinoline quinone for the repair of peripheral nerve defect.

Regeneration and functional reconstruction of peripheral nerve defects remained a significant clinical challenge. Nerve guide conduits, with seed cells or neurotrophic factors (NTFs), had been widely used to improve the repair and regeneration of injured peripheral nerve. Pyrroloquinoline quinone (PQQ) was an antioxidant that can stimulate nerve growth factors (NGFs) synthesis and accelerate the Schwann cells (SCs) proliferation and growth. In present study, three kinds of nerve guide conduits were constructed: one from cellulose/SPI hollow tube (CSC), another from CSC combined with SCs (CSSC)

Multi-spectroscopic, thermodynamic and molecular simulation studies on binding of pyrroloquinoline quinone with DNA: coexistence of intercalation and groove binding modes.

The interaction between enzyme-like pyrroloquinoline quinone (PQQ) and calf-thymus DNA (CT-DNA) has been investigated by means of multi-spectroscopic (UV-Vis, fluorescence and circular dichroism), isothermal titration calorimetric (ITC), viscometry and molecular docking and metadynamics simulation techniques. Absorption spectral data suggested the formation of a PQQ/CT-DNA complex, which quenched the fluorescence of PQQ via the dynamic quenching process. The results of CD spectral studies coupled with viscosity measurements, competitive binding assays with Hoechst 33258 and ethidium bromide (E

Quinone-Based Mediator Immobilized Mesoporous Electrodes for Bioelectrocatalysis of Glucose Dehydrogenase.

Bioelectrocatalysis, which couples enzymatic catalytic and electrode reactions, is the basic principle behind enzyme electrochemical biosensors, biofuel cells, and bioelectrosynthesis systems. Electron mediators facilitate the transfer of electrons between electrodes and oxidoreductases during catalytic reactions, so ideally the mediator remains near the electrode for higher catalytic efficiency. Additionally, electrode materials with three-dimensional space at the enzyme volume mesoscale are expected to improve electrode performance (e.g., increased current density and stability). Engineered neuronal exosomes mediate α-synuclein clearance to ameliorate Parkinson's disease.

Parkinson's disease (PD) is the second most common neurodegenerative disorder after Alzheimer's disease. A hallmark pathological feature of PD is the abnormal aggregation of α-synuclein (αSyn) into insoluble Lewy bodies. Consequently, developing strategies to inhibit αSyn aggregation in the brain has been a major research focus for PD treatment. This study developed a therapeutic approach using engineered neuronal exosomes. These exosomes were modified to extend their blood circulation half-life to 3.8 h and enhance targeting, with a 2.15 ± 0.09% brain signal proportion (vs. 0.78 ± 0.07% for f

Enhanced rat sciatic nerve regeneration through silicon tubes filled with pyrroloquinoline quinone.

Pyrroloquinoline quinone (PQQ) is an antioxidant that also stimulates nerve growth factor (NGF) synthesis and secretion. In an earlier pilot study in our laboratory, Schwann cell growth was accelerated, and NGF mRNA expression and NGF secretion were promoted. The present study was designed to explore the possible nerve-inducing effect of PQQ on a nerve tube model over a 1-cm segmental deficit. An 8-mm sciatic nerve deficit was created in a rat model and bridged by a 1-cm silicone tube. Then,10 mul of 0.03 mmol/l PQQ were perfused into the silicone chamber in the PQQ group. Pyrroloquinoline Quinone Preconditioning Alleviates Ischemic Cerebral Injury Through Antioxidant and Anti-Inflammatory Mechanisms.

The underlying pathological mechanism of ischemic stroke is complex, with oxidative stress and inflammation being two key factors that are intertwined and mutually influential. They also serve as important potential targets for the intervention of cerebral ischemia. Pyrroloquinoline quinone (PQQ) is known for its neuroprotective properties and the ability to modulate immune system function. Previous studies have demonstrated that PQQ mitigates brain infarction in rodent models of cerebral ischemia; however, the neuroprotective mechanisms underlying PQQ's effects against ischemic brain injury a
