Comparison of effects of propranolol versus pindolol on sinus rate and pacing frequency in sick sinus syndrome.

Beta blockers in patients with sick sinus syndrome (SSS) may prevent supraventricular arrhythmias, systemic hypertension and myocardial ischemia, but may cause excessive depression of sinus node function. In 8 patients with SSS and a permanent pacemaker, the effect of chronic oral pindolol on sinus rate and pacing frequency was compared with that of propranolol in a double-blind crossover trial. In all patients the pacemaker was programmed to a rate of < or = 50 beats/min. Holter monitors, obtained at baseline and on each drug, were used to calculate peak ambulatory sinus rate, number of paced

Comparative effects of propranolol and pindolol on small and large arteries and veins of the forearm circulation in hypertensive man.

Brachial artery diameter (pulsed Doppler method), forearm vascular resistance, and venous tone (plethysmographic method) were studied in 18 patients with sustained essential hypertension. Hemodynamic parameters were reevaluated after 3 months of treatment by propranolol (9 patients) or pindolol (9 patients). For the same decrease in pressure, propranolol decreased heart rate significantly while pindolol did not, indicating the role of intrinsic sympathomimetic activity. After pindolol, forearm vascular resistance and venous tone significantly decreased while brachial artery cross-sectional are

Chronic effects of pindolol on the arterioles, large arteries, and veins of the forearm in mild to moderate essential hypertension.

The effects of pindolol were studied on the large arteries, arterioles, and veins of the forearm in 13 patients with essential hypertension after 12 weeks of dosing. The methods used were pulsed Doppler velocimetry of the brachial artery and strain gauge mercury-in-silicone rubber plethysmography of the forearm. Compared with placebo baseline values, chronic pindolol significantly decreased systolic and diastolic blood pressure (P less than 0.001), pulse pressure (P less than 0.001), and pulse rate (P less than 0.05). A significant increase was observed in brachial artery diameter (P less than

Effect of the beta-receptor blocker pindolol on survival in HgCl2 induced acute renal failure in dogs.

The effect of the beta receptor blocker pindolol on survival was investigated in HgCl2 intoxicated dogs. A single injection of 100 microgram/kg b.w. pindolol intravenously (i.v.) caused a significant rise in urinary sodium excretion and a significant decrease of plasma renin activity (PRA) and urinary norepinephrine (NE) and epinephrine (E) excretion in control dogs. A single injection of 3 mg/kg HgCl2 i.v. resulted in death of the animals within 3-5 days. Pretreatment with the above dose of pindolol increased length of survival 4-8 days, two dogs recovering from acute renal failure (ARF). The

Effect of single doses of pindolol and d-fenfluramine on flumazenil-induced anxiety in panic disorder patients.

The effects of the 5-HT1A receptor blocker pindolol and the 5-HT releasing and uptake blocking agent d-fenfluramine, both used as indirect serotonin agonists, on flumazenil-induced acute anxiety reactions were studied in panic disorder patients to test the hypothesis that serotonin (5-HT) inhibits neural systems mediating panic attacks. Thirty never treated or drug free PD patients (16 females) aged 22-49 y (mean ± SD, 32.9 ± 8) received single doses of d-fenfluramine (n = 10; 30 mg, p.o.), pindolol (n = 10; 5 mg, p.o.), or placebo (n = 10) 90 and 45 min before a challenge test with flumazenil

Effective termination of reentrant supraventricular tachycardia by single dose oral combination therapy with pindolol and verapamil.

We evaluated the efficacy of single oral dose combining 20 mg pindolol and 120 mg verapamil in termination of paroxysmal supraventricular tachycardia (SVT) in 12 patients with recurrent symptomatic tachycardia. All had electrically inducible SVT lasting longer than 30 minutes. Patients were administered placebo or crushed pindolol and verapamil on 2 consecutive days after tachycardia was electrically induced and allowed to sustain for 30 minutes. With placebo, SVT lasted 186 +/- 18 minutes (mean +/- SEM); five patients converted spontaneously within 121 to 180 minutes. With pindolol and verapa

Pindolol--a new beta-adrenergic blocking agent with intrinsic sympathomimetic activity in the management of mild and moderate hypertension.

Pindolol, a well-tolerated noncardioselective beta-adrenergic blocking drug, effectively reduced blood pressure in patients with mild to moderate hypertension without causing orthostatic hypotension. In common with other beta-adrenergic blocking agents, pindolol blocked the normal increase in the standing pulse rate. The drug, however, did not decrease the supine pulse rate, a feature that can be interpreted as evidence of the postulated intrinsic sympathomimetic activity of pindolol.

Supercritical fluid chromatography-tandem mass spectrometry for the enantioselective determination of propranolol and pindolol in mouse blood by serial sampling.

Packed-column supercritical fluid chromatography (pSFC) coupled to an atmospheric pressure chemical ionization (APCI) source and a tandem mass spectrometer (MS/MS) with minimum sample pretreatment was explored for the rapid and enantioselective determination of (R,S)-propranolol in mouse blood. Serial bleeding of mice is advantageous for the reduction of animal usage, dosing errors, and animal-to-animal variation. Intrinsic sympathomimetic activity of (-)-pindolol mediated through a (-)-propranolol-resistant site of the beta1-adrenoceptor in human atrium and recombinant receptors.

The beta-blocker (-)-pindolol produces intrinsic sympathomimetic activity manifested clinically by cardiostimulation, but the beta-adrenoceptor subtype, which mediates these effects, is unknown. Recent work indicates the existence of a (-)-propranolol-resistant site of the cardiac beta(1)-adrenoceptor and we propose that it mediates the cardiostimulation evoked by (-)-pindolol. We compared the interaction of (-)-pindolol both with human atrial myocardium and with recombinant beta(1)-adrenoceptors. The effects of (-)-pindolol on paced human atrial trabeculae were studied in the presence of 3-is

Acute hemodynamic and neurohumoral effects of pindolol: a beta-adrenoceptor antagonist with high intrinsic sympathomimetic activity in patients with dilated cardiomyopathy.

It has been claimed that beta-adrenoceptor antagonists produce clinical improvement and increase longevity in patients with idiopathic dilated cardiomyopathy. Dilated heart is critically dependent upon adrenergic support to maintain forward output. Acute withdrawal of such support, even with small doses of beta-blockers, may worsen myocardial function sufficiently to limit their widespread use. Pindolol (P), a potent beta-adrenoceptor antagonist, possesses high intrinsic sympathomimetic activity. Administration of P to patients with dilated cardiomyopathy may protect against the effects of hig

Pindolol antagonizes the effects on hippocampal rhythmical slow activity of clonidine, baclofen and 8-OH-DPAT, but not chlordiazepoxide and sodium amylobarbitone.

Buspirone, benzodiazepines, barbiturates and ethanol all reliably reduce the frequency of reticular-elicited hippocampal rhythmical slow activity. In the present experiments we tested a number of drugs which are not usually used for treating generalized anxiety disorders but which have been reported to have some anxiolytic properties. Clonidine (0.3 mg/kg, i.p.), baclofen (6 mg/kg, i.p.) and 8-hydroxy-di-n-propylamino tetralin (8-OH-DPAT) (2.5 mg/kg, i.p.) all reduced the frequency of rhythmical slow activity. The effect of all three drugs was reduced by the 5-hydroxytryptamine 1a antagonist p

A short-term double-blind randomized controlled pilot trial with active or placebo pindolol in patients treated with venlafaxine for major depression.

BACKGROUND: Pindolol has been widely investigated as an augmenter of antidepressant drug response. Results have been inconsistent. In this study, we used pindolol together with venlafaxine because of its ability to achieve a rapid onset of serotonin transporter blockade. AIMS: The object of this study was thus to investigate if pindolol augments the antidepressant response to venlafaxine. METHODS: Patients with major depression were randomized to either active or placebo pindolol 20 mg retard daily dosage and concomitantly treated with venlafaxine for 19 days. Depression severity was evaluat

Effects of pindolol, befunolol and melanin treated with adrenergic beta-blocking agents on lysosomal enzymes in bovine ciliary body and iris in vitro.

We studied biochemically the effects of pindolol, befunolol and melanin treated with beta-blockers on lysosomal enzymes in the ammonium sulfate fraction of the bovine ciliary body and iris in vitro. Acid phosphatase, beta-D-glucuronidase and alpha-D-mannosidase were not inhibited by the beta-blockers. N-acetyl-beta-D-glucosaminidase and alpha-L-fucosidase activities were inhibited by pindolol and befunolol at high concentrations. After centrifugation of the enzyme fraction incubated with melanin, the enzyme activity in the supernatant decreased, possibly as a result of the affinity of lysosoma

Failure of intravenous pindolol to reduce the hemodynamic determinants of myocardial oxygen demand or enzymatically determined infarct size in acute myocardial infarction.

Pindolol, a beta blocker with intrinsic sympathomimetic activity, was investigated in a randomised controlled trial of 100 patients presenting within 12 hours of uncomplicated acute myocardial infarction. Pindolol was given intravenously for 24 hours and orally for 48 hours to achieve serum levels above 10 ng/ml. Heart rate and arterial pressure, both systolic and diastolic, fell to a similar degree in actively treated and control patients. There was no significant difference between systolic blood pressure-heart rate product in actively treated and control patients during the first 72 hours o

Exploration of low-dose ozone-activated periodate (O₃/IO₄⁻) for rapid degradation of the β-Blocker pindolol: efficiency and radical mechanism.

The emergence of recalcitrant organic pollutants poses a significant threat to aquatic environments. PI-based AOPs have been widely used in water treatment and show great potential in degrading these pollutants. This study pioneers the application of low-dose ozone-activated periodate (O₃/IO₄⁻), a novel advanced oxidation process (AOP), for rapid degradation of β-blocker contaminants exemplified by pindolol (PIN). The O₃/PI system exhibits broad pH applicability. Under optimal conditions of 0.175 mg/L O₃, 0.1 mM IO₄⁻, and pH 7.0, the system achieved 99.2 % PIN degradation within 20 s (kobs = 0

Serotonin-1A receptors in the dorsal periaqueductal gray matter mediate the panicolytic-like effect of pindolol and paroxetine combination in the elevated T-maze.

The β-adrenergic blocker and 5-HT(1A) receptor antagonist pindolol has been combined with selective serotonin reuptake inhibitors (SSRIs) in patients with depressive and anxiety disorders to shorten the onset of the clinical action and/or increase the proportion of responders. The results of a previous study have shown that pindolol potentiates the panicolytic effect of paroxetine in rats submitted to the elevated T-maze (ETM). Since reported evidence has implicated the 5-HT(1A) receptors of the dorsal periaqueductal gray matter (DPAG) in the panicolytic effect of antidepressants, rats treated

No effect of Pindolol on postural hypotension in type 1 (insulin-dependent) diabetic patients with autonomic neuropathy. A randomised double-blind controlled study.

Orthostatic hypotension is one of the most troublesome symptoms in diabetic autonomic neuropathy. Some reports have suggested Pindolol - a beta-adrenoceptor antagonist with intrinsic sympathomimetic activity - to be effective in the treatment of this condition. In order to elucidate the value of this therapy we performed a double-blind placebo controlled cross-over study with Pindolol (15 mg/day). Eight Type 1 (insulin-dependent) diabetic patients with autonomic neuropathy and signs and symptoms of orthostatic hypotension (systolic blood pressure decrease greater than 30 mm Hg when standing) p
