Placental mTOR complex 1 regulates fetal programming of obesity and insulin resistance in mice.

Fetal growth restriction, or low birth weight, is a strong determinant for eventual obesity and type 2 diabetes. Clinical studies suggest placental mechanistic target of rapamycin (mTOR) signaling regulates fetal birth weight and the metabolic health trajectory of the offspring. In the current study, we used a genetic model with loss of placental mTOR function (mTOR-KOPlacenta) to test the direct role of mTOR signaling on birth weight and metabolic health in the adult offspring. mTOR-KOPlacenta animals displayed reduced placental area and total weight, as well as fetal body weight at embryonic

Comparative analysis of high field MRI and histology for ex vivo whole organ imaging: assessment of placental functional morphology in a murine model.

OBJECTIVES: The purpose of our study was to evaluate MRI as a tool to examine placental morphology in a murine model in comparison to classical histology techniques. METHODS: Assessment of placental samples (n = 24) from C57Bl/6 J mice was performed using MR imaging and histomorphological analyses. To optimize image contrast for MRI, a protocol for gadolinium-mediated enhancement of placental tissue was established. To test method sensitivity, placental zone assessment with MRI and histology was applied to a model of prenatal stress exposure known to affect placental morphology (n = 10). Mean

Relevance of the regulation of the brain-placental axis to the nocturnal bottleneck of mammals.

INTRODUCTION: Evolutionary theory suggests that the ancestors of all placental animals were nocturnal. Visual perceptive function of mammalian brain has evolved extensively, but nearly 70 % of today's mammals are still nocturnal. While placental influence on brain development is known, if placenta plays a role in the visual perceptive function of mammalian brain remains untested. The present study aims to test this hypothesis. METHODS: In this study, single-nuclei RNA sequencing was performed to identify genes expressed in the pig placenta and fetal brain, and then compared with the orthologo

In Vivo Assessment of Placental Structure and Perfusion in Late-Gestation Pregnancies and Their Association With Fetal Growth.

Adequate placental structure and function are crucial for fetal growth, while placental dysfunction often leads to fetal growth restriction (FGR), associated with increased perinatal morbidity and mortality. Current FGR criteria, relying on sonographic biometry and Doppler assessments, typically fall short in sensitivity for diagnosing FGR, predicting perinatal outcomes, and assessing their association with placental pathology. Advanced MRI methods offer a unique opportunity to characterize placental structure and perfusion, and to assess their relationship with fetal growth. This study aims t

Marked Third-Trimester Placental Thickening Is Associated with Maternal Infectious, Hormonal, and Metabolic Burden: A Matched Case-Control Study.

Background: Increased placental thickness has been associated with adverse perinatal outcomes and fetal functional and structural abnormalities. However, whether marked third-trimester placental thickening is associated with a distinct maternal clinical profile compared with pregnancies with normal placental thickness remains insufficiently characterized. This study aimed to compare maternal characteristics, comorbidities, medication exposure, infection history, and fetal findings between pregnancies with marked placental thickening, defined as placental thickness ≥70 mm, and gestational-age-m

MV-Flow imaging for evaluating the placental function of gestational diabetes mellitus.

BACKGROUND: Gestational diabetes mellitus (GDM) is characterized by vascular dysfunction and is associated with adverse perinatal outcomes for both the mother and fetus. Microvascular index (MVI) is a non-invasive marker of microvascular function assessed by microvascular flow imaging (MV-Flow). This study aimed to evaluate the performance of conventional Doppler ultrasound and MV-Flow in delineating the placental microvessels and to explore the feasibility and potential clinical value of using MV-Flow for placental function in GDM women. METHODS: This study included women with singleton preg

Placental insufficiency in relation to postterm pregnancy and fetal postmaturity. Evaluation of fetoplacental function; management of the postterm gravida.

As pregnancy extends post term, incidence of placental insufficiency, fetal postmaturity (dysmaturity), and fetal perinatal death increases rapidly as a consequence of reduced respiratory and nutritive placental function. Despite a compensatory fetoplacental respiratory reserve capacity, fetal distress is observed in about one third of postterm pregnancies. On a biochemical level, placental pathophysiology in postterm-postmaturity pregnancies is not well understood. Postmaturity is correlated with increased incidence of placental lesions, fetal hypoxia-asphyxia, intrauterine growth retardation

High placental index and poor pregnancy outcomes: a retrospective study of 18,386 pregnancies.

INTRODUCTION: Our aim was to state the correlation between placental index and pregnancy outcomes or in vitro fertilization/intracytoplasmic sperm injection (IVF/ICSI) pregnancies. MATERIALS AND METHODS: We included in this retrospective study all singleton births in a third level clinic during the period 2001-2011 (n = 18,386). We divided placental index into quartiles and analyzed the differences between the groups in term of pregnancy outcomes. Then, we estimated crude and adjusted odds ratios (ORs) for placental index over the third centile of the distribution to correlate with pregnancy

Effect on low implantation of the placenta on maternal blood pressure and placental function.

Low implantation of the placenta may protect against the development of pregnancy-induced hypertension (PIH) and is associated with improved values in tests of placental function. PIH occurred in six of 201 (3%) consecutive patients with placenta praevia managed at the City Hospital, Nottingham, from 1 April 1973 to 30 June 1981. None of the six patients developed associated proteinuria. Of the total number of 24,549 patients delivered in the same hospital for the years 1974-1978, 3744 (15%) developed PIH. Of the patients with placenta praevia, 52% had serum levels of human placental lactogen

A prospective study of the oxytocin challenge test and newborn neurobehavioral outcome.

In a prospective study, infants of high-risk mothers delivered over a 1-year period were evaluated by clinical, biochemical, and behavioral methods. Of 67 newborns whose mothers had oxytocin challenge tests (OCTs), 54 were delivered after negative tests, and 13 after positive tests. Infants with positive OCTs had poor state organization and reflexive performance when compared with negative-OCT babies. These infants also showed evidence of intrauterine malnutrition, but did not have any greater asphyxiation than the negative OCT group. These results are consistent with the hypothesis that a pos

Association Between Third-Trimester Ultrasonography With Histopathological Changes in the Placenta Among Mothers With Gestational Diabetes Mellitus.

Background Gestational diabetes mellitus (GDM) is a prevalent metabolic disorder during pregnancy, associated with significant maternal and fetal complications. GDM affects placental structure and function, potentially compromising fetal development. Third-trimester ultrasonography (USG) serves as a non-invasive tool for fetal and placental assessment, while histopathological examination provides definitive evidence of placental changes. This study aimed to evaluate the correlation between third-trimester USG findings and placental histopathological changes in GDM pregnancies. Methodology A ho

Antithyroglobulin and Antiperoxidase Antibodies Can Negatively Influence Pregnancy Outcomes by Disturbing the Placentation Process and Triggering an Imbalance in Placental Angiogenic Factors.

Background/Objectives: Thyroid autoimmunity (TAI) affects about 15% of women of reproductive age and can negatively affect pregnancy outcomes. One possible mechanism for pregnancy complications can be attributed to a disturbed process of placentation caused by thyroid antibodies. To test this hypothesis, placental hormones and angiogenic factors in pregnant women with TAI were evaluated. Methods: Fifty-eight hypothyroid women positive for TPOAb/TgAb, thirty-three hypothyroid women negative for TPOAb/TgAb, and thirty-nine healthy controls were enrolled in this study. Animal Models to Study Placental Development and Function throughout Normal and Dysfunctional Human Pregnancy.

Abnormalities of placental development and function are known to underlie many pathologies of pregnancy, including spontaneous preterm birth, fetal growth restriction, and preeclampsia. A growing body of evidence also underscores the importance of placental dysfunction in the lifelong health of both mother and offspring. However, our knowledge regarding placental structure and function throughout pregnancy remains limited. Understanding the temporal growth and functionality of the human placenta throughout the entirety of gestation is important if we are to gain a better understanding of place

Placental Thickness Correlates with Severity-Weighted Fetal Dysfunction in the Third Trimester.

Background: Placental thickness has been associated with adverse perinatal outcomes, but the relationship to specific fetal abnormalities seems to not yet be well understood. This study investigates whether increased placental thickness correlates with the severity of fetal cardiac and extracardiac conditions using a structured classification and severity-weighted scoring system. Methods: We undertook a retrospective analysis of 1452 fetal echocardiograms conducted during the third trimester at a tertiary referral institution from the years 2022 to 2025. The diagnoses were categorized into fou

Intra-placental arterial Doppler: A marker of fetoplacental vascularity in late-onset placental disease?

INTRODUCTION: Late-gestation adverse pregnancy outcome is associated with reduced placental villous vascularity but rarely with a frankly abnormal umbilical artery Doppler waveform. The clinical utility of umbilical artery Doppler velocimetry in late gestation is limited by poor understanding of what aspect(s) of placental structure and function the impedance reflects. We hypothesized that placental arterial circulation impedance reflects placental vascularity and arterial function. MATERIAL AND METHODS: This was a secondary analysis of data from the FEMINA2 study, a study of pregnancy outcom

NDRG1 deficiency attenuates fetal growth and the intrauterine response to hypoxic injury.

Intrauterine mammalian development depends on the preservation of placental function. The expression of the protein N-myc downstream-regulated gene 1 (NDRG1) is increased in placentas of human pregnancies affected by fetal growth restriction and in hypoxic primary human trophoblasts, where NDRG1 attenuates cell injury. We sought to assess the function of placental NDRG1 in vivo and tested the hypothesis that NDRG1 deficiency in the mouse embryo impairs placental function and consequently intrauterine growth. We found that Ndrg1 knock-out embryos were growth restricted in comparison to wild-typ

Adiponectin and leptin levels in mothers, fetuses, and neonates with intrauterine growth restriction compared to those with appropriate gestational age.

OBJECTIVE: Intrauterine growth restriction (IUGR) is associated with impaired fetal growth and altered metabolic regulation. Adipokines such as adiponectin and leptin play key roles in energy homeostasis and placental function. This study aimed to evaluate adiponectin and leptin levels in maternal peripheral blood, umbilical cord blood, and neonatal peripheral blood, and to investigate associated changes in placental gene expression and DNA methylation of adipokine-related genes in IUGR compared to appropriate-for-gestational-age (AGA) pregnancies. MATERIALS AND METHODS: This case-control stu
