This Review Article summarizes outcomes from the ILSI Europe expert workshop on plant-based proteins in infant formula, held in November 2024. Experts from academia, clinical nutrition, and food science evaluated the current use and future potential of plant-based protein sources in infant formula, considering nutritional adequacy, allergenicity, sustainability, processing technologies, and regulatory constraints. While soy and hydrolyzed rice proteins are already approved and in use, emerging sources such as pea, lentil, and faba beans show promise but require further validation of their amin

Legumes establish symbiosis with rhizobia, forming nitrogen-fixing nodules. The central role of reactive oxygen species (ROS) and reactive nitrogen species (RNS) in nodule biology has been clearly established. Recently, hydrogen sulfide (H2S) and other reactive sulfur species (RSS) have emerged as novel signaling molecules in animals and plants. A major mechanism by which ROS, RNS, and RSS fulfil their signaling role is the post-translational modification of proteins. To identify possible functions of H2S in nodule development and senescence, we used the tag-switch method to quantify changes i

Next to cow's milk and eggs, plant foods, i.e., legumes, tree nuts and cereal grains, most often sensitise atopic children. Storage proteins constitutes the most relevant protein fraction of plant foods, causing primary sensitisation. They exhibit strong allergenic properties and immunogenicity. Our goal was to analyse sensitisation to 26 plant storage proteins in a group of 76 children aged 0-5 years with chronic symptoms of atopic dermatitis using Allergy Explorer ALEX2 and to discover changes in serum protein-peptide patterns in allergic patients with the use of MALDI-TOF-MS. We reported th

BACKGROUND: The human (Homo sapiens) chemokine-like protein macrophage migration inhibitory factor (HsMIF) is a pivotal mediator of inflammatory, infectious and immune diseases including septic shock, colitis, malaria, rheumatoid arthritis, and atherosclerosis, as well as tumorigenesis. HsMIF has been found to exhibit several sequential and three-dimensional sequence motifs that in addition to its receptor binding sites include catalytic sites for oxidoreductase and tautomerase activity, which provide this 12.5 kDa protein with a remarkable functional complexity. Herbivore elicitors, most of which identified thus far are from the oral secretions (OS) of herbivores, are important in herbivore-induced plant defense. However, whether and how larval OS of the striped stem borer (SSB), Chilo suppressalis, a devastating pest on rice, induce plant defenses remains largely unknown. We found that treating plants with SSB OS enhanced levels of phosphorylated OsMPK3/4/6, JA, JA-Ile, ethylene, and ABA in rice, which subsequently induced the production of defense compounds, such as trypsin protease inhibitors, and decreased the growth of SSB larvae. Digestion by pr

Curcumin (Cur) is a hydrophobic nutraceutical with poor water solubility and low oral bioavailability. This study elucidates how plant protein molecular traits shape the assembly, structure, and digestive fate of Cur-loaded nanoparticles. Using a pH-shifting strategy, zein was employed as a hydrophobic core, while soluble legume protein fractions (Sup) or whey protein isolate (WPI, W) served as shell components. The resulting core-shell nanoparticles exhibited tunable particle sizes (40-130 nm), high uniformity (PDI < 0.18), and excellent loading capacity (up to 316.3 μg/mgprotein). Core-shell

Valsa mali negative strand RNA virus 1 (VmNSRV1), a fungal virus infecting Valsa mali and belonging to the Phenuiviridae family, was recently shown to be capable of naturally infecting plants. Its RNA2 encodes a homologue of a plant virus movement protein (MP) that can complement MP-deficient virus vectors and localizes to plasmodesmata, allowing natural cross-kingdom infection. To determine whether this property is a common feature among mycoviruses possessing distant MP homologues or an isolate-specific trait, we investigated Trichoderma gamsii cogu-like virus 1 (TgClV1), a Phenuiviridae mem

Ribosomes are essential cellular machines that translate genetic information into functional proteins. Ribosomes require massive nutrient investments, accounting for as much as 50 % of organic phosphorus and 25 % of organic nitrogen in leaves. Optimizing ribosome levels could therefore reduce crop plant fertilizer requirements, an urgent goal for agricultural sustainability. Disruptions to ribosome biogenesis often cause surprising developmental defects, however, and there is substantial confusion and debate among plant geneticists about how to interpret mutant phenotypes caused by defective r

Alternative routes for producing animal proteins are crucial for reducing the reliance on traditional livestock farming, which contributes significantly to greenhouse gas emissions, deforestation, and water consumption. Myoglobin (Mb) is an important oxygen-binding hemoprotein found in vertebrate muscle which enhances the nutritional and sensorial properties of meat. Due to its unique functionality, Mb has been heterologously expressed in a variety of organisms, although only transient expression in Nicotiana benthamiana has been reported for higher plants. In this study, we used chloroplast t

Overexpression of pathogen-induced cysteine-rich transmembrane proteins (PCMs) in Arabidopsis thaliana enhances resistance against biotrophic pathogens and stimulates hypocotyl growth, suggesting a potential role for PCMs in connecting both biological processes. Plants possess a sophisticated immune system to protect themselves against pathogen attack. The defense hormone salicylic acid (SA) is an important player in the plant immune gene regulatory network. Using RNA-seq time series data of Arabidopsis thaliana leaves treated with SA, we identified a largely uncharacterized SA-responsive gene

Plants are constantly exposed to environmental stresses during their growth and development. Owing to their immobility, plants possess stress-sensing abilities and adaptive responses to cope with the abiotic and biotic stresses caused by extreme temperatures, drought, flooding, salinity, heavy metals and pathogens. Acyl-CoA-binding proteins (ACBPs), a family of conserved proteins among prokaryotes and eukaryotes, bind to a variety of acyl-CoA esters with different affinities and play a role in the transport and maintenance of subcellular acyl-CoA pools. In plants, studies have revealed ACBP fu

Cold shock domain proteins (CSDPs) participate in plant development and resistance, but the underlying molecular mechanisms are poorly understood. In this study, we demonstrated that the CSDPs, including EsCSDP1, EsCSDP2, and EsCSDP3, from the extremophyte Eutrema salsugineum possess all basic properties of RNA chaperones. EsCSDP1-3 melt secondary structures in RNAs with various nucleotide sequences and exhibit RNA chaperone activity in vitro. EsCSDP1 and EsCSDP3 were shown to have higher RNA melting activity, whereasile EsCSDP2 had higher RNA chaperone activity. We demonstrated that higher RN

BACKGROUND: The enterocytes of the intestinal epithelium are regularly exposed to potentially harmful substances of dietary origin, such as lectins. Expression of heat shock proteins (HSPs) by this epithelium may be part of a protective mechanism developed by intestinal epithelial cells to deal with noxious components in the intestinal lumen.
AIM: To investigate if the lectins PHA, a lectin from kidney beans (Phaseolus vulgaris) and WGA, a lectin from wheat germ (Triticum aestivum) could modify the heat shock response in gut epithelial cells and to establish the extent of this effect.

Brome mosaic virus (BMV), a positive-strand RNA virus, encodes two replication proteins: the 2a protein, which contains polymerase-like sequences, and the 1a protein, with N-terminal putative capping and C-terminal helicase-like sequences. These two proteins are part of a multisubunit complex which is necessary for viral RNA replication. We have previously shown that the yeast two-hybrid assay consistently duplicated results obtained from in vivo RNA replication assays and biochemical assays of protein-protein interaction, thus permitting the identification of additional interacting domains. The increasing global demand for sustainable and health-promoting foods has positioned plant-based proteins as key components in the development of dairy alternatives. This review critically examines plant protein sources: legumes, cereals, pseudocereals, oilseeds, and nuts not merely by composition, but by their comparative suitability for specific dairy applications, evaluating protein quality using the Digestible Indispensable Amino Acid Score (DIAAS), digestibility, and source-specific antinutritional factors (phytates in cereals; tannins, trypsin inhibitors, and lectins in legumes and oil

Mental disorders are a severe health problem, and the number of patients is growing worldwide. Increased anxiety and decreased motivation due to excessive mental stress further accelerated the severity of the problem. Enzymatic digestion of food proteins produces bioactive peptides with various physiological functions, some of which exhibit neuromodulatory effects with oral administration. Recently, studies reported that some peptides produced from plant proteins such as soybeans, leaves, and grains exhibit emotional regulatory functions such as strong anxiolytic-like and antidepressant-like e

BACKGROUND: Leucine-rich repeat extensins (LRXs) are extracellular proteins consisting of an N-terminal leucine-rich repeat (LRR) domain and a C-terminal extensin domain containing the typical features of this class of structural hydroxyproline-rich glycoproteins (HRGPs). The LRR domain is likely to bind an interaction partner, whereas the extensin domain has an anchoring function to insolubilize the protein in the cell wall. Based on the analysis of the root hair-expressed LRX1 and LRX2 of Arabidopsis thaliana, LRX proteins are important for cell wall development. The importance of LRX protei

Horticultural plants play a vital role in agricultural production and daily life. Post-translational modifications are essential for regulating various cellular processes, but the functions of SUMOylation, an increasingly significant post-translational modification, remain to be thoroughly explored in horticultural plants. In this review, we summarize the current research advancements regarding protein SUMOylation in horticultural plants and offer perspectives for future investigations. We begin by introducing the SUMOylation system in horticultural plants. Subsequently, we focus on the roles

As sessile organisms, plants rely on environmental cues to make key developmental decisions. These decisions are rarely based on a single time point but instead emerge through the integration of past and present experiences, a process broadly referred to as molecular memory. Canonical epigenetic mechanisms operating at the chromatin and RNA levels are often viewed as major drivers of plant molecular memory, yet whether plants also exploit changes in protein state remains an open and compelling question. In this opinion piece, we speculate that prion-like proteins (PrLPs) may function as memory
