The effect of propranolol, prindolol, practolol and verapamil versus placebo on exercise induced tachycardia in patients with ventricular preexcitation.

Propranolol, prindolol, practolol and verapamil have been given orally in a fixed dose to subjects with preexcitation syndrome in a cross over study. The effect on exercise induced tachycardia, prevention of tachyarrhythmias and normalization of heart rate after exercise was investigated. Propranolol, practolol and prindolol significantly (p less than 0.01-0.05) reduced the heart rate at rest and during exercise and favourably influenced the normalization of the heart rate after exercise. The effect on effort tolerance (work done till the submaximal heart rate was reached) was significantly be

Improvement in prognosis of myocardial infarction by long-term beta-adrenoreceptor blockade using practolol. A multicentre international study.

In a large-scale double-blind controlled trial of practolol (200 mg twice daily) in the long-term prophylactic treatment of 3038 patients recovering from acute myocardial infarction treatment was started one to four weeks after the acute attack. The trial was originally planned to include 4000 patients treated for at least a year but had to be terminated prematurely because of the serious oculocutaneous and peritoneal reactions reported elsewhere. Nevertheless, important findings, probably applicable to other beta-adrenoreceptor antagonists, have emerged in relation to mortality and morbidity.

An experimental study on canine A-V junctional pacemaker--evaluation of the automaticity by overdrive suppression and autonomic blockade by practolol and atropine.

1. The automaticity of the A-V junction was evaluated in 15 awake dogs with experimentally induced A-V junctional rhythm. 2. The spontaneous heart rate of these dogs ranged from 54 to 112 beats a minute, showing about 1.5-2.5 times slower than that of control dogs with sinus rhythm. The duration of asystole after overdrive in these dogs prolonged significantly in accordance with an increase in the frequency of stimulation for overdrive, and its mean +/- SD attained 4.7 +/- 1.1 seconds after overdrive at a rate of 2.5 times the spontaneous heart rate. 3. By administration of atropine (0.04 mg/k

Beta-adrenoreceptor antagonists enhance the benzodiazepine-induced suppression of footshock induced fighting in mice.

Aggressive behavior in the footshock induced fighting test (FIFT) was studied after treatment with benzodiazepines (diazepam, nitrazepam, temazepam, medazepam) given alone or in combination with beta-adrenoreceptor antagonists (propranolol, oxprenolol, practolol). With the combinations of those drugs it was found that diazepam (0.5 mg/kg) with propranolol, oxprenolol, and practolol (20 mg/kg); nitrazepam (0.25 mg/kg) with propranolol (10 mg/kg) and nitrazepam (0.1 mg/kg) with practolol (20 mg/kg); temazepam (0.5 mg/kg) with propranolol (10 mg/kg) and temazepam (1 mg/kg) with oxprenolol and pra

Effect of cardioselective beta-blockade on heart function and chest pain in acute myocardial infarction.

Systolic time intervals and the a/H ratio were recorded in 20 patients with uncomplicated acute myocardial infarction over a period of five days. The initial high heart rate and systolic blood pressure and the short PEP and ICT indicating a sympathetic overactivity were spontaneously normalized during the first week of infarction. LVET was reduced indicating a fall in stroke volume and the a/H ratio was unchanged at the high levels suggestive of elevated preload or LVEDP. In 10 patients with acute myocardial infarction and recurrent chest pain recordings on noninvasive parameters were made bef

Peyronie's disease and systemic lupus erythematosus syndrome associated with metoprolol administration: a case report.

Peyronie's disease (induratio penis plastica) and a syndrome resembling systemic lupus erythematosus (S.L.E.) contemporaneously developed in a 50-year-old man six months after initiation of treatment with metoprolol. In view of the improvement in symptoms and signs on withdrawal of metoprolol, it seems likely that the drug was responsible for both these conditions. Peyronie's disease has been observed in relation to treatment with other adrenergic blocking agents, including practolol, and the S.L.E. syndrome is thought to be a possible untoward effect of practolol therapy. The relationship bet

Hypothalamic action of adrenoreceptor blocking agents.

In cats anaesthetized with pentobarbital sodium, the posterior hypothalamus was superfused and electrically stimulated with a push-pull cannula. The pressor response to stimulation of this hypothalamic area was inhibited when the hypothalamus was superfused with drugs blocking either alpha-adrenoreceptors (piperoxan, tolazoline), or beta-adrenoreceptors--(+/-)-propranolol, (-)-propranolol, practolol, sotalol, metoprolol. (+)-Propranolol and a concentration of procaine equianaesthetic to propranolol were ineffective. During superfusion with tolazoline in the presence of practolol the inhibition

Sclerosing peritonitis associated with keratoconjunctivitis sicca, pleurisy, and secretory otitis media.

A male case of sclerosing peritonitis of unknown cause, associated with keratoconjunctivitis sicca, pleurisy, and secretory otitis media, is presented. These unusual complications are very similar to the adverse reactions to the beta-adrenergic blocking agent, practolol, which is known to cause sclerosing peritonitis. This similarity suggests an etiological association between the sclerosing peritonitis of this case and that caused by practolol. The sclerosing peritonitis of this case may be considered to occur as part of some systemic disorder, and thus casts doubt on the hitherto accepted hy

Hypotensive action of beta-blocking drugs injected into the cerebral ventricle of the rat.

Pindolol, propranolol, practolol, N-isopropyl-p-nitrophenyl-ethanolamine (INPEA), d and l-alprenolol were injected into the lateral cerebral ventricle of the anaesthetized rat. Except for INPEA, which has no action at the doses used, these various beta-blocking agents produce a dose-related fall in blood pressure. D-alprenolol and practolol seem to be the least active of the hypotensive agents. Our results show some relationship between the beta-blocking potency of the drugs studied and the hypotension induced when they were injected in the lateral ventricle.

Effect of two types of beta-adrenergic blockade on gastric acid secretion during basal conditions and during histamine stimulation on non-vagotomized and in vagotomized gastric fistula dogs.

The effect of beta-adrenoreceptor blockade by propranolol and practolol on basal secretion and on submaximally histamine-stimulated gastric acid secretion was studied in conscious non-vagotomized and in vagotomized gastric fistula dogs. Neither propranolol (0.5 mg/kg) nor practolol (1.0 mg/kg), both intravenously, had a significant influence on basal or histamine-stimulated gastric acid secretion in non-vagotomized and vagotomized dogs. It is concluded that beta-adrenergic blockade has no influence on basal or histamine-stimulated gastric acid secretion in dogs. Furthermore, no differences wer

Beta-adrenergic blockade as adjunctive oral therapy in patients with chronic atrial fibrillation.

In many patients with chronic atrial fibrillation, it is difficult to prevent an excessive ventricular rate under stress, even with high levels of digoxin in the blood. The effect of adding beta-adrenergic blockade with practolol to digoxin on the heart rate at rest and during low-grade controlled exercise was investigated in 28 patients with chronic atrial fibrillation and in ten normal control subjects who were receiving maintenance dosages (0.25 to 0.75 mg) of digoxin. In atrial fibrillation, therapy with practolol decreased the mean heart rate at rest from 99.8 beats per minute to 77.5 bea

Transmission blockade and stimulation of ganglionic adenylate cyclase by catecholamines.

Isolated rat superior cervical ganglia treated with isoproterenol and related drugs show an increase in ganglionic cyclic adenosine 3':5'-monophosphate (cAMP) and a block of transmission. For isoproterenol, the maximum increase in cAMP occurred at 1 X 10(-6) M, a concentration without effect on transmission. Approximately 5 X 10(-4) M isoproterenol was required to reduce the ganglionic compound action potential by 50%. Dopamine, in contrast to isoproterenol, had no effect on the content of cAMP but depressed transmission. The maximum increase in cAMP produced by norepinephrine occurred with 5

Effect of some beta-adrenoceptor blockers and of (+) propranolol on adrenaline-induced pulmonary edema in mice.

(+) Propranolol is considered to prevent adrenaline-induced pulmonary edema (A.P.E.) due to the beta-adrenoceptor blockade. However, local anaesthetics also are known to prevent pulmonary edema. To assess the role of beta-adrenergic blockade in A.P.E., the effect of a beta 1-blocker possessing local anaesthetic action (+/- Metaprolol) and a beta 1-blocker possessing no local anaesthetic action (+/- practolol) was studied along with propranolol derivatives. The study revealed that (+), (-), (+/-) propranolol and (+/-) metaprolol completely prevented A.P.E. whereas (+/-) practolol did not. This

Beta-adrenoceptors in human lung, bronchus and lymphocytes.

1 The ability of propranolol, metoprolol, acebutolol, atenolol and practolol to reverse the stimulant effect of 10-5 M isoprenaline on lymphocyte cyclic AMP levels was determined. The ratios of doses required to produce 50% inhibition as compared with propranolol were 1:316 for metoprolol 1:1780 for acebutolol and 1:2820 for atenolol. No ratio could be calculated for practolol as it never produced more than 35% inhibition possibly because of its intrinsic sympathomimetic activity. This rank order of antagonist potencies suggest the lymphocyte β-receptor has β2 selectivity. 2 For peripheral air

Effects of diet-induced hypokalaemia on the efficacy of antiarrhythmic drugs against ventricular arrhythmias evoked by coronary artery ligation in the anaesthetised rat.

The antiarrhythmic efficacies of intravenous quinidine (Q), disopyramide (D), prenylamine (P), bepridil (B), and practolol were compared in normokalaemic and dietary-induced hypokalaemic anaesthetised rats. Hypokalaemia markedly increased the severity of arrhythmias induced by coronary artery ligation and reduced the antiarrhythmic efficacy of all five drugs tested. After anaesthesia, hypokalaemia was associated with hypotension and bradycardia but not QTc prolongation. However, bradycardia was not seen in conscious hypokalaemic animals. Papillary muscles taken from hypokalaemic rats displayed

Intravenous beta-blockade in coronary heart disease: is cardioselectivity or intrinsic sympathomimetic activity hemodynamically useful?

Beta-blocking drugs are increasingly prescribed in coronary heart disease, but controversy surrounds the hemodynamic relevance of their ancillary pharmacologic properties--cardioselectivity and intrinsic sympathomimetic activity. We therefore compared the effects of four intravenous beta-adrenoreceptor antagonists with different ancillary properties on left ventricular function in 24 patients with coronary heart disease. All four drugs depressed the relation between left ventricular filling pressure and cardiac output at rest and during exercise. However, practolol and oxprenolol, which have i

Effect of isoprenaline on bethanechol-stimulated gastric antral motility in dogs with gastric fistula.

The purpose of the present study was to evaluate the effect of isoprenaline on gastric antral motility in conscious dogs with gastric fistula, using intraluminal strain-gauge transducers. Infusion of bethanechol increased the motility for both frequency and strength. Isoprenaline, a beta 1- and beta 2-agonist, was used alone and in conjunction with selective blockade of beta 1 and beta 2 receptors. The stimulated antral motility was dose-dependently inhibited by isoprenaline. The effect was significantly blocked by the beta 1 + beta 2-adrenoceptor blocker propranolol and by using in conjunctio
