Mitochondrial genome-derived microsatellites reveal genetic diversity and population structure in Callery pear populations.

Callery pear (Pyrus calleryana Decne.; PC) possesses many desirable characteristics valued in managed landscapes. This has driven the release of numerous cultivars, including both hybrids and selections derived from native populations. The extensive planting of PC cultivars in managed areas has contributed to the widespread occurrence of invasive individuals across a broad range of habitats in the eastern United States (US). Self-incompatibility, tolerance to various environmental conditions, pathogen and pest resistance, intraspecific hybridization among the cultivars, possible interspecific

Alternariol Detoxification by Rhodotorula taiwanensis P1 Isolated from a Pear and Its Mechanism.

Alternariol (AOH) is a mycotoxin produced mainly by Alternaria alternata on fruits including tomatoes and strawberries as well as cereal grains including wheat and soybeans. In this study, we isolated Rhodotorula taiwanensis (R. taiwanensis) P1 from a pear and investigated the effects of incubation time and temperature on AOH reduction rates and the mechanism involved in AOH detoxification by the yeast strain. The yeast strain showed a 76.10% AOH reduction rate from the initial level (1 μg/mL) at 30 °C after 48 h of incubation. The yeast cell-free filtrate from the yeast culture did not increa

Impact of phage treatment on fire blight disease outcome and floral microbiome composition.

With the increasing importance of alternative pesticides for the control of bacterial pathogens in agricultural and clinical settings, the use of bacteriophage viruses (phages) to reduce bacterial growth and prevent disease is gaining in popularity. Phages have been shown to be highly effective in killing bacterial cells both in vitro and across plant and animal host systems, although many questions remain about the predictability of their success across more realistic ecological conditions and in light of natural strain variation of pathogens. Furthermore, as phage application becomes more co

Whole Genome Sequence Resource of the Asian Pear Scab Pathogen Venturia nashicola.

Venturia nashicola, the cause of scab disease of Asian pears, is a host-specific, biotrophic fungus. It is restricted to Asia and is regarded as a quarantine threat outside this region. European pear displays nonhost resistance (NHR) to V. nashicola and Asian pears are nonhosts of V. pyrina (the cause of European pear scab disease). The host specificity of these two fungi is likely governed by differences in their effector arsenals, with a subset hypothesized to activate NHR. The Pyrus-Venturia pathosystem provides an opportunity to dissect the underlying genetics of nonhost interactions in th

On the early identification and characterization of pear blister canker viroid, apple dimple fruit viroid, peach latent mosaic viroid and chrysanthemum chlorotic mottle viroid.

In the 90's, pear blister canker viroid (PBCVd), apple dimple fruit viroid (ADFVd), peach latent mosaic viroid (PLMVd) and chrysanthemum chlorotic mottle viroid (CChMVd) were identified and characterized in the Ricardo Flores' laboratory. In these studies, the autonomous replication of these infectious RNAs and their involvement in the elicitation of diseases in their natural hosts were also shown. Their discovery was achieved by classical approaches based on the physical purification of the viroid RNAs from polyacrylamide gels followed by the sequencing of their genomic RNAs and by bioassays

Pear ACO genes encoding putative 1-aminocyclopropane-1-carboxylate oxidase homologs are functionally expressed during fruit ripening and involved in response to salicylic acid.

1-Aminocyclopropane-1-carboxylate (ACC) oxidase catalyzes the final reaction of the ethylene biosynthetic pathway, converting ACC into ethylene. Past studies have shown a possible link between ACC oxidase and salicylic acid during fruit ripening in pear, but the relationship has received no more than modest study at the gene expression level. In this study, two cDNA clones encoding putative ACC oxidase, PpACO1 and PpACO2, were isolated from a cDNA library constructed by our own laboratory and produced using mRNA from mesocarp of pear (Pyrus pyrifolia Nakai. cv.Whangkeumbae). One cDNA clone, de

Prevalence, Molecular Detection, and In Vitro Elimination of Latent Viruses and Viroids in Pome Fruit Germplasm of Kazakhstan.

Latent viral and viroid infections represent a major phytosanitary threat to pome fruit production because infected propagation material facilitates the long-term spread of pathogens in orchards. This study investigated the prevalence of viral and viroid pathogens in apple and pear orchards of southern and southeastern Kazakhstan and evaluated the efficiency of integrated in vitro sanitation approaches. A total of 34 samples collected from commercial orchards were analyzed using RT-qPCR. Apple stem pitting virus (ASPV) was the most prevalent pathogen detected across all surveyed regions, follo

A comprehensive dataset of Halyomorpha halys for automated detection and monitoring in orchards.

The Halyomorpha halys (HH) (Brown marmorated stink bug (BMSB)) is a globally invasive pest responsible for severe economic losses in fruit production, yet open, large-scale monitoring datasets remain scarce. Here we present a multi-year, multi-modal dataset acquired over three consecutive growing seasons in an experimental pear orchard in Carpi (Modena), Italy, designed to support research in entomology, agriculture, and Machine Learning (ML). The release comprises (i) three years of Unmanned Aerial Vehicle (UAV)-acquired RGB imagery annotated with bounding boxes for HH; (ii) one season of sta

Combined Effects of Radiative and Evaporative Cooling on Fruit Preservation under Solar Radiation: Sunburn Resistance and Temperature Stabilization.

Excessive solar radiation and high temperature often cause considerable loss and waste of fruits during transportation, retail, and storage. In the current study, a natural deep eutectic solvent-based polyacrylamide/poly(vinyl alcohol) hydrogel with nanoparticles (NPs/NADES@PAAm/PVA) is developed for fruit quality protection from solar radiation and high-temperature stress by achieving the combined effect of radiative and evaporative cooling. NPs/NADES@PAAm/PVA presents an average solar reflectance of ∼0.89 and an average emittance at the atmospheric window of ∼0.90. Besides, NPs/NADES@PAAm/PV

Cobinamide-based cyanide analysis by multiwavelength spectrometry in a liquid core waveguide.

A novel cyanide analyzer based on sensitive cobinamide chemistry relies on simultaneous reagent and sample injection and detection in a 50 cm liquid core waveguide (LCW) flow cell illuminated by a white light emitting diode. The transmitted light is read by a fiber-optic charge coupled device (CCD) spectrometer. Alkaline cobinamide (orange, lambda(max) = 510 nm) changes to violet (lambda(max) = 583 nm) upon reaction with cyanide. Multiwavelength detection permits built-in correction for artifact responses intrinsic to a single-line flow injection system and corrects for drift. With optimum cho

The PcbZIP44-PcWRKY70 Module Mediates Volatile Ester Biosynthesis Via PcAAT1 Transcriptional Regulation during Pear Postharvest Ripening.

Aroma profoundly influences the flavor profile and commercial appeal of fruits. While the biosynthetic pathways of volatiles are well understood, the regulatory mechanisms governing their transcription remain unclear. This study used 'Starkrimson' fruits (Pyrus communis L.) to explore molecular mechanisms of ester biosynthesis. Integration of gas chromatography-mass spectrometry analysis and RNA sequencing data revealed that the alcohol acyltransferase PcAAT1 is involved in ester synthesis. Experiments confirmed transcription factor PcbZIP44 as its upstream activator, and its physical interact

Phytochemical Composition and Quality Attributes of Pear Cultivars Grown Under Organic and Conventional Orchard Management: A Three-Year Study.

Organic fruit production is associated with a specific form of farm management: no artificial pesticides or mineral fertilizers are allowed. Only natural methods of fertilization and plant protection, including preventive practices, are used. The Organic Production Regulation describes all organic farming practices. Fruits from organic production are often perceived by consumers as safe and potentially beneficial to health. Pears contain many bioactive compounds from the polyphenol and carotenoid groups. In the present study, three pear cultivars, namely 'Alexander Lucas', 'Conference', and 'X

Alleviation of iron deficiency in pear by ammonium nitrate and nitric oxide.

BACKGROUND: Iron is essential for the growth and development of trace elements in plants, and iron deficiency can lead to leaf chlorosis. Ammonium and nitrate are the major forms of nitrogen present in soils. Ammonium nitrate alleviates the chlorosis of leaves caused by iron deficiency, but the mechanism is not clear in pear. RESULTS: Ammonium nitrate induced the increase of nitric oxide (NO) under iron deficiency. We further analyzed the effect of NO by exogenous NO treatment. The results showed that ammonium nitrate and NO increased the activity of ferric chelate reductase. NO induced the e

Effects of Exposure Time and Biological State on Acquisition and Accumulation of Erwinia amylovora by Drosophila melanogaster.

Fire blight, caused by the bacterium Erwinia amylovora, is a disease devastating the production of rosaceous crops, primarily apple and pear, with worldwide distribution. Fire blight begins in the spring when primary inoculum is produced as ooze, which consists of plant sap, E. amylovora, and exopolysaccharides. Ooze is believed to be transferred to healthy tissues by wind, rain, and insects. However, the mechanisms by which insects locate and transmit ooze are largely undocumented. The goals of this study were to investigate the biological factors affecting acquisition of E. amylovora from oo

Evaluation of the structure-bioactivity interaction in alginate-pectin films incorporated with Licania tomentosa seed extract for pear preservation.

Alginate-based films have limitations when used in isolation, including low mechanical strength, high water sensitivity, and reduced barrier properties, which can restrict their technological applicability. In this context, the present study investigated the effect of incorporating oiti seed extract on the structure and functionality of alginate-pectin biopolymer films. Small-angle X-ray scattering (SAXS) analysis revealed a concentration-dependent behavior of alginate in the extract dispersion. The profile of phenolic compounds detected in the extracts, such as vanillin (0.46 mg g-1), ferulic

Comprehensive analysis of volatile flavor components in pear fruit spanning the entire development stages.

The intricate pattern of flavor changes throughout the various stages of fruit development remains poorly understood. Here, we investigated the material dynamics underlying flavor formation, focusing on volatile and metabolite production across 8 fruit developmental and ripening stages of 'Yuluxiang' pear. A total of 154 volatile compounds were characterized by HS-GC-IMS combined with HS-SPME-GC-MS technology, mainly including aldehydes, ketones, and ester compounds in the young fruit and enlarging periods, and the ester compounds increase in the mature period. Notably, the expression of alcoh

Inhibitory activity and mechanism of aryl furan thioesters containing 1,3,4-thiadiazole against Erwinia amylovora.

BACKGROUND: The Gram-negative bacterium Erwinia amylovora induces fire blight disease in Rosaceae hosts, severely damaging pome fruit trees including pear, apple, and hawthorn cultivars. Traditional control methods, such as antibiotics and copper-based agents, are associated with plant damage and the emergence of resistant bacterial strains, and present significant challenges. This study aims to design and synthesize a novel eco-friendly bactericide. RESULTS: In this study, we designed and synthesized 30 small-molecule compounds by combining 1,3,4-thiadiazole, a key moiety of zinc thiazole, w
