Asymmetric distribution of type IV pili triggered by directional light in unicellular cyanobacteria.

The type IV pili (T4P) system is a supermolecular machine observed in prokaryotes. Cells repeat the cycle of T4P extension, surface attachment, and retraction to drive twitching motility. Although the properties of T4P as a motor have been scrutinized with biophysics techniques, the mechanism of regulation remains unclear. Here we provided the framework of the T4P dynamics at the single-cell level in Synechocystis sp. PCC6803, which can recognize light direction. We demonstrated that the dynamics was detected by fluorescent beads under an optical microscope and controlled by blue light that in

Circular RNA expression profile and functional analysis of circUvrag in light-induced photoreceptor degeneration.

BACKGROUND: Circular RNAs (circRNAs) are implicated in retinal pathophysiology; however, their expression profiles and functions in photoreceptor apoptosis are largely unknown. We explored circRNA-expression profiles and circUvrag (host gene: Uvrag, ultraviolet radiation resistance associated gene) function in light-induced photoreceptor apoptosis. METHODS: Sprague-Dawley rats and 661 W photoreceptor cells were exposed to blue light to establish light-induced photoreceptor degeneration. Differentially expressed circRNAs were identified using microarrays. Potential functions of dysregulated ci

An AIE-based fluorescence probe for assessing treatment response of bacteria-infected wound.

The integration of precise bacterial detection with effective sterilization, while mitigating the risk of bacterial resistance, is of paramount importance in clinical medicine and public health. Herein, we present a novel aggregation-induced emission (AIE) fluorescent probe, PTZ-PD, engineered for the simultaneous detection and eradication of bacteria in animal models. PTZ-PD features a donor-acceptor (D-A) architecture composed of a phenothiazine donor and a quinoline propionitrile acceptor, which endows the probe with long-wavelength emission (λem ≈ 680 nm) and remarkable AIE characteristics

Ultraviolet-induced fluorescence mapping of facial porphyrin and sebum using deep-learning segmentation.

BACKGROUND: Porphyrins and sebum, which fluoresce under ultraviolet (UV) light, are important features closely linked to acne development. However, traditional detection methods are often unreliable as they are sensitive to variations in lighting conditions and skin tone. OBJECTIVES: While deep learning-based segmentation has shown promise, comparative studies for this specific application remain limited. This study aims to address this gap by evaluating and comparing two distinct deep learning approaches for the automated segmentation of UV-induced fluorescence. METHODS: We evaluated two de

The use of 7-amino actinomycin D in identifying apoptosis: simplicity of use and broad spectrum of application compared with other techniques.

The detection and quantitation of apoptotic cells is becoming increasingly important in the investigation of the role of apoptosis in cellular proliferation and differentiation. The pathogenesis of hematologic disorders such as aplastic anemia and the development of neoplasia are believed to involve dysregulation of apoptosis. To quantitate accurately the proportion of apoptosis cells within different cell types of a heterogeneous cell population such as blood or bone marrow, a method is required that combines the analysis of large numbers of cells with concurrent immunophenotyping of cell sur

Paper-based fluorescent sensor based on molecularly imprinted polymers and upconversion nanoparticles for selective and sensitive D-proline detection in human serum and saliva.

D-Proline has been reported as a potential biomarker for gastric (stomach) cancer, creating a demand for practical and rapid analytical approaches for early diagnosis using simple and non-invasive clinical samples. In this work, a microfluidic paper-based analytical device (µPAD) integrating molecularly imprinted polymer-functionalized upconversion nanoparticles (UCNPs@MIPs) was developed for the selective fluorescence-based detection of D-proline. Owing to near-infrared excitation (980 nm), UCNPs provide background-free fluorescence with high photostability, while MIPs serve as artificial rec

Detection and quantification of tetracyclines by whole cell biosensors.

Three different mini-Tn5 plasmids, containing a tetracycline-inducible promoter, Ptet and a regulatory gene, tetR, in operon fusions with a reporter gene system (lacZYA, luxCDABE or gfp), were constructed. These biosensor constructs responded to low levels of tetracyclines by producing beta-galactosidase, light or green fluorescent protein. They did so in a quantitative manner, thus enabling the quantification of tetracyclines in the immediate surroundings of the biosensor organism. All three constructs were transferred successfully to different gram-negative bacteria by conjugation. An Escher

Fluorescence Transduction of Liquid Crystal Ordering Transitions for Biosensing.

Liquid crystal (LC) ordering transitions are exquisitely sensitive to molecular interactions at aqueous interfaces and have long served as the basis for optical biosensors. However, the readout of these transitions has almost exclusively relied on polarized-light optical microscopy, which limits quantification and hinders practical deployment. Here, we report a fluorescence-based transduction scheme that converts LC ordering transitions to quantitative optical outputs. Our strategy employs amphiphilic block copolymers bearing aggregation-induced emission (AIE) motifs that undergo dynamic coval

Comparative effects of fluoride-containing and remineralization-promoting agents on artificial gastric juice-induced bovine enamel erosion susceptibility: an in vitro study.

BACKGROUND: Gastroesophageal reflux disease (GERD) is a major risk factor for erosive tooth wear because repeated exposure of enamel to gastric acid promotes demineralization. This in vitro study evaluated the protective effects of fluoride-containing and remineralization-promoting agents on enamel erosion induced by simulated gastric acid challenge. METHODS: Eighty-four bovine enamel specimens were randomly assigned to seven groups (n = 12/group): a sound enamel control group, an erosion control group treated with distilled water, and five treatment groups receiving Natural-F gel (2.09% NaF,

A comparison of the sensitivity of photodamage assays in rat basophilic leukemia cells.

Many aspects of cellular function or physiology can be used to indicate the level of damage resulting from the application of potentially deleterious agents such as drugs, solvents or even light. The dose required to reach a specific biological endpoint will necessarily depend on the characteristics of the damage induced by the agent. By using multiple biological probes, it is possible to get a more complete description of the type of damage induced. Photodamage was induced in rat basophilic leukemia cells by either 254-nm UVC light exposure or rose bengal photosensitization. Damage was measur

Mouse retinal cell behaviour in space and time using light sheet fluorescence microscopy.

As the general population ages, more people are affected by eye diseases, such as retinopathies. It is therefore critical to improve imaging of eye disease mouse models. Here, we demonstrate that 1) rapid, quantitative 3D and 4D (time lapse) imaging of cellular and subcellular processes in the mouse eye is feasible, with and without tissue clearing, using light-sheet fluorescent microscopy (LSFM); 2) flat-mounting retinas for confocal microscopy significantly distorts tissue morphology, confirmed by quantitative correlative LSFM-Confocal imaging of vessels; 3) LSFM readily reveals new features

Photoactivatable NPOM-caged split-activator CRISPR/Cas12a biosensor for amplification-free imaging of deoxynivalenol-induced miR-155 dynamics and lateral-flow sensing.

MicroRNA-155 (miR-155) is an immune-associated small RNA biomarker involved in macrophage responses to environmental stressors such as deoxynivalenol (DON). However, achieving amplification-free miRNA sensing with low background and precise temporal regulation remains challenging. Here, we develop a photoactivatable split-activator CRISPR/Cas12a biosensor for miR-155 detection without target nucleic acid amplification and for dynamic analysis of toxin-induced cellular responses. The sensing system integrates an NPOM-caged crRNA with a split-complementary activator, enabling optical control of

Nanobody-targeted photodynamic therapy induces significant tumor regression of trastuzumab-resistant HER2-positive breast cancer, after a single treatment session.

RATIONALE: A substantial number of breast cancer patients with an overexpression of the human epidermal growth factor receptor 2 (HER2) have residual disease after neoadjuvant therapy or become resistant to trastuzumab. Photodynamic therapy (PDT) using nanobodies targeted to HER2 is a promising treatment option for these patients. Here we investigate the in vitro and in vivo antitumor efficacy of HER2-targeted nanobody-photosensitizer (PS) conjugate PDT. METHODS: Nanobodies targeting HER2 were obtained from phage display selections. Monovalent nanobodies were engineered into a biparatopic con

Ultrastructural evaluation of adverse effects on dentine formation from systemic fluoride application in an experimental mouse model.

AIM: Fluoride is widely used in dentistry for its caries prevention. To reduce dental caries, the optimal fluoride concentration of public water supplies in the United States is 0.7 ppm. However, excessive systemic fluoride consumption can lead to dental/enamel fluorosis. Numerous studies have explored the effects of fluoride on enamel and enamel-forming cells. However, research on systemic fluoride's impact on dentine is limited, particularly the effect of fluoride on the structure of the dentine-pulp complex. Therefore, this study aimed to identify how excessive fluoride affects dentine micr

Microchip laser-induced fluorescence detection of proteins at submicrogram per milliliter levels mediated by dynamic labeling under pseudonative conditions.

We have previously demonstrated on-column dynamic labeling of protein-SDS complexes on capillaries and microchips for laser-induced fluorescence (LIF) detection using both a commercially available fluor and a protein separation buffer. Upon binding to hydrophobic moieties (of the analyte or separation buffer), the fluor undergoes a conformational change allowing fluorescence detection at 590 nm following excitation with 488-nm light. Our original work showed on-chip limits of detection (LOD) comparable with those using UV detection (1 x 10(-5) M) on capillaries-falling significantly short of t

FLIM Reveals Red Light-Induced Changes in Murine Hair Follicles.

Hair loss, particularly androgenetic alopecia (AGA) and alopecia areata (AA), is a prevalent condition with widespread psychosocial impact. Recently, low-level laser therapy (LLLT) has emerged as a promising non-invasive therapeutic alternative due to its bioregulatory effects and favorable safety profile compared to conventional pharmacological treatments. In this study, we employed fluorescence lifetime imaging microscopy (FLIM) to investigate the effects of red-light irradiation on hair follicle dynamics and the cutaneous microenvironment in a C57BL/6 mouse model. A hair regeneration model

Mechanochemical Acceptor Engineering for NIR-I and NIR-II Fluorophores Enabling Orthogonal Ureter-Vascular Image-Guided Surgery.

Accurate intraoperative identification of the ureters and surrounding vasculature is essential for preventing iatrogenic injuries during laparoscopic surgery. While fluorescence imaging (FLI) offers a promising solution, conventional single-channel systems lack the capacity for multistructure anatomical differentiation. Here, we present a dual-channel near-infrared (NIR) FLI strategy facilitated by mechanochemistry-assisted acceptor engineering, which allows the subsequent screening of spectrally orthogonal fluorophores. Two of the resulting NIR fluorophores, TQx-FTA (NIR-I, λem = 755 nm) and
