Gross and histopathological examination was performed on seven captive magnificent tree frogs (Litoria splendida) and one green tree frog (Litoria caerulea) that had died or been humanely destroyed while naturally infected with Mahaffey Road virus, a Bohle iridovirus-like ranavirus. Necropsy examination revealed skin lesions consisting of multiple small pale or haemorrhagic papules and ulcers in most frogs. Other common gross findings were perineural haemorrhage affecting the spinal nerves, hydrocoelom, hepatomegaly and splenomegaly with pinpoint pale foci throughout the parenchyma. On histolo

Infectious diseases, particularly those caused by members of the genus Ranavirus (Iridoviridae), have caused mass mortality events in amphibian populations worldwide. Frog virus 3 (FV3) infects a wide range of amphibian hosts and results in high mortality. Despite the global prevalence of FV3, its origin and routes of spread remain unclear, largely because of limited genetic data from limited geographic regions. This study aimed to classify the ranaviruses isolated in the Republic of Korea through whole-genome sequencing. We collected amphibian carcasses and both symptomatic and asymptomatic l

Amphibians are susceptible to a multitude of skin disorders, many of which can appear grossly similar. The most common clinical presentations include hyperemia, discoloration, dermal mass, ulceration, and necrosis. Many amphibian skin diseases are related to captive husbandry. The diagnostic process starts with environmental evaluations, a full history, physical examination and sampling for direct observation, histology, polymerase chain reaction testing, and bacterial and fungal culture. This review emphasizes the main conditions encountered in amphibian dermatology.

The flattened musk turtle (Sternotherus depressus) is a stream-dwelling species that inhabits a small range in the Upper Black Warrior River water basin system in AL and is classified as critically endangered by the International Union for Conservation of Nature. The stripe-necked musk turtle (Sternotherus peltifer) has a broader range, surrounding and slightly overlapping that of the flattened musk turtle in the southeast United States, and is classified as least concern. Ongoing conservation efforts for flattened musk turtles have, thus far, been limited to population surveys; however, the c

The highly endangered European pond turtle (Emys orbicularis) was reintroduced in Switzerland in 2010. Up until 2019, no routine medical examinations have been carried out prior to its release or during recapture events. The aim of this study was to assess the health status of captive and free-living Emys orbicularis populations in Switzerland, taking into account the most important and frequently occurring health threats to freshwater turtles. A total of 141 European pond turtles, including captive (n = 89) and free-living (n = 52) individuals, underwent clinical examination (n = 136), choana

Dredging is commonly used to maintain navigational channels in freshwater lakes such as Lake Erie, and the dredged material may be discarded in confined disposal facilities (CDFs). The effects of these CDFs on wildlife health are largely unknown. We compared health assessments of painted turtles (Chrysemys picta) at two sites in southwestern Lake Erie, Ohio, USA in May 2022: CDF for dredged material (n=27), and a protected coastal marsh (PCM; n=24). Each turtle underwent a physical examination, blood collection for hematology and plasma biochemistry profiles, and testing for Chlamydia spp., he

A mass mortality event of captive Hong Kong warty newts Paramesotriton hongkongensis with non-granulomatous necrotic lesions occurred in Taipei Zoo, Taiwan, in 2014. Clinically, the sick newts were lethargic and often covered with water mold Saprolegnia sp. on the skin of the body trunk or extremities. Predominant pathological findings were multifocal non-granulomatous necrotic lesions in the liver, spleen, and kidneys and severe skin infection with Saprolegnia sp., with deep invasion and involvement of underlying muscles. The possibility of ranavirus infection was ruled out by negative PCR re

The collection and storage of swab samples for molecular diagnostics is a routine component of wildlife health surveillance. The suitability of different sample storage conditions for maximizing the recovery of pathogen DNA in most species has not been assessed; therefore, the aim of this study was to identify a preferred storage method for swabs collected for the detection of frog virus 3 (FV3), a significant chelonian pathogen. Sterile swabs were inoculated in triplicate with a plasmid containing known quantities of FV3 DNA from 100 to 107 copies. Swabs were then stored under one of the foll

In 2025, a severe mass mortality outbreak struck captive Geochelone sulcata at a zoo in Guangdong Province, China. To trace the causative agent, identify the viral strain and characterize associated pathological lesions, respiratory tract samples collected from diseased tortoises were subjected to a combined technical workflow. Metagenomic high-throughput sequencing was first applied to screen for potential pathogens, followed by virus isolation via cell culture. Transmission electron microscopy (TEM) was used to observe viral morphology. The major capsid protein (MCP) gene was amplified by PC

A captive 'survival assurance' population of 56 endangered boreal toads Anaxyrus boreas boreas, housed within a cosmopolitan collection of amphibians originating from Southeast Asia and other locations, experienced high mortality (91%) in April to July 2010. Histological examination demonstrated lesions consistent with ranaviral disease, including multicentric necrosis of skin, kidney, liver, spleen, and hematopoietic tissue, vasculitis, and myriad basophilic intracytoplasmic inclusion bodies. Initial confirmation of ranavirus infection was made by Taqman real-time PCR analysis of a portion of

Emerging infectious diseases are major threats to amphibian biodiversity. Ranaviruses (Family Iridoviridae) are a group of globally emerging infections that infect poikilothermic vertebrates. In the United Kingdom, the emergence of this group of viruses has been followed by the decline in populations of common frogs (Rana temporaria) across the country, especially in the southeast of England. Simple susceptible-infected (SI) and combinatorial models have been developed to explain the spread of ranavirus in populations of common frogs; here, we use those models as a basis for developing a simpl

Largemouth bass virus (LMBV), belonging to the genus Ranavirus, causes high mortality and heavy economic losses in largemouth bass aquaculture. In the present study, a novel cell line, designated as MsF, was established from the fin of largemouth bass (Micropterus salmoides), and applied to investigate the characteristics of cell death induced by LMBV. MsF cells showed susceptibility to LMBV, evidenced by the occurrence of a cytopathic effect (CPE), increased viral gene transcription, protein synthesis, and viral titers. In LMBV-infected MsF cells, two or more virus assembly sites were observe

The global decline in amphibian populations is a major environmental issue. Chytridiomycosis, Ranaviruses and the red-leg syndrome have been identified in unusual mortality events. However, these infections do not account for all causes of declining amphibian populations. Moreover, several cases of amphibian mortality are difficult to solve without resorting to an interdisciplinary approach. Two cases of unusual mortality in Rana temporaria occurred at two high-mountain ponds (northwest Italy) in April and May 2021. Water and frog samples were analysed to understand the possible causes respons

Eastern box turtles (Terrapene carolina carolina) are declining range-wide due to anthropogenic factors, including the illegal wildlife trade. Infectious diseases that cause high morbidity and mortality, such as frog virus 3 (family Iridoviridae, genus Ranavirus, species Rana1), may also threaten this species in managed-care and free-ranging settings. High rates of infectious disease transmission are documented within the illegal wildlife trade; however, the impact of diseases on health, survival, and ultimate disposition of confiscated turtles is not thoroughly described. In July 2021, 96 eas

The global amphibian declines are compounded by infections with members of the Ranavirus genus such as Frog Virus 3 (FV3). Premetamorphic anuran amphibians are believed to be significantly more susceptible to FV3 while this pathogen targets the kidneys of both pre- and postmetamorphic animals. Paradoxically, FV3-challenged Xenopus laevis tadpoles exhibit lower kidney viral loads than adult frogs. Presently, we demonstrate that X. laevis tadpoles are intrinsically more resistant to FV3 kidney infections than cohort-matched metamorphic and postmetamorphic froglets and that this resistance appear

Ranaviruses infect and have been associated with mass mortality events in fish, amphibians and reptiles and are capable of interclass transmission. Eastern water dragons (EWDs), a semi-aquatic squamate, have an overlapping distribution with several species shown to be susceptible to Bohle iridovirus (BIV). However, this species has not been previously investigated, and no known mass mortalities have occurred in wild populations. Here we report the experimental infection of juvenile EWDs with BIV to investigate a water-dwelling lizards' susceptibility to a ranaviral strain present in northern Q

During an on-going amphibian ecology study, a free-ranging American bullfrog (Rana catesbeiana) metamorph was captured in a pitfall trap adjacent to a constructed farm pond at the Plateau Research and Education Center (PREC) on the Cumberland Plateau near Crossville, Tennessee, USA. Grossly, the right eye was approximately 50% the size of the left. Stereo and light microscopic examination revealed two granulomas within the orbit. Electron microscopic examination revealed virus particles scattered throughout one structure but mostly aggregated toward the center. Subsequent PCR and sequencing (G

Ranaviruses have been observed with increasing frequency amongst poikilothermic vertebrate hosts. The impact of ranaviruses upon amphibian populations has remained largely unknown. A gene probe for Bohle iridovirus (BIV) based upon primers designed to detect epizootic haematopoietic necrosis virus (EHNV) was constructed. A PCR and dot-blot system was used successfully in screening for the presence of BIV nucleic acid in digested formalin-fixed, paraffin-embedded amphibian tissues. Juvenile frogs were more susceptible to BIV than adults. In experimental challenges and epizootics in captive frog

Infections by Frog Virus 3 (FV3) and other ranavirus genus members are significantly contributing to global amphibian decline. The Xenopus laevis frog is an ideal research platform upon which to study the roles of distinct frog leukocyte populations during FV3 infections. Frog macrophages (MΦs) are integrally involved during FV3 infection, as they facilitate viral dissemination and persistence but also participate in immune defense against this pathogen. In turn, MΦ differentiation and functionality depend on the colony-stimulating factor-1 receptor (CSF-1R), which is ligated by CSF-1 and iter
