BACKGROUND: The gold standard for the diagnosis of cholera is stool culture, but this requires laboratory facilities and takes at least 24 hours. A rapid diagnostic test (RDT) that can be used by minimally trained staff at treatment centers could potentially improve the reporting and management of cholera outbreaks.

The Pronto Gaucher kits and Pronto Gaucher RecTL Amplification Mix, marketed to identify mutations in the gene for glucocerebrosidase, are widely used for the diagnosis of Gaucher disease. Subjects genotyped using this kit have been reported with an allele including both the common N370S mutation and RecTL, a previously described Gaucher mutation arising from recombination between the glucocerebrosidase gene and pseudogene. Using direct sequencing and real-time PCR, we show that the RecTL, N370S allele is a false positive result, demonstrating possible pitfalls of diagnostic kits.

It was found possible to use method for preparation of antibody erythrocytic diagnosticums for detection of agents of influenza and arboviral infections. Test systems prepared by the developed method were superior in sensitivity to immunoreagents prepared with previously proposed amidol. Quantitative Buffy Coat analysis and blood smears were performed on a total of 47 blood samples. The technique showed 100% correlation with the blood smears in 9 samples containing babesia and 10 samples containing malaria, with some differential features distinguishing the two infections. Quantitative Buffy Coat analysis provides a simple and rapid method for the detection of parasitemia in cases of babesiosis.

A method for the determination of catecholamines (Cat-a-Kit; Upjohn Diagnostics) is discussed. It depends upon the enzymatic conversion of the catecholamines to their ring o-methylated analogues in the presence of s-adenosyl-L-methionine-methyl-14C and catechol-o-methyltransferase. Values obtained from the blood plasma of 16 tetraplegic and 11 healthy volunteers are reported. The advantages and disadvantages of the Cat-a-Kit are discussed.
