Hygienic safety of alcohol-based hand disinfectants and skin antiseptics.

PURPOSE: The aim of this study was to evaluate the overall risk of hand disinfectants and skin antiseptics to become contaminated with bacterial spores throughout the production process and the subsequent in-use period, hence posing a public health risk. METHODS: Microbiological assessment of primary packaging material was carried out and long-term survival of bacterial spores in alcohol was assessed using sporulated B. subtilis ATCC 6633 as a standard. In-use contamination of alcohol-based formulations was tested by repeated use over 12 months under practical conditions and microbiological a

Metabolic Gadolinium Labeling of Clostridium novyi-Nontoxic for Magnetic Resonance Imaging Visualization of Spores and Germinated Bacteria.

Bacterial cancer therapy using Clostridium novyi-nontoxic (C. novyi-NT) has emerged as a promising strategy to selectively eradicate hypoxic and necrotic tumor regions. However, noninvasive visualization of injected spores and germinated bacteria remains a major barrier to clinical translation. Here, we developed metabolically responsive gadolinium-labeled C. novyi-NT (Gd-C. novyi-NT) to enable magnetic resonance imaging (MRI)-based tracking of spore localization and bacterial germination. Gd ions were electrostatically condensed onto the negatively charged spore surface, forming Gd clusters t

Development of a filter to prevent infections with spore-forming bacteria in injecting drug users.

BACKGROUND: In heroin injectors, there have been a number of outbreaks caused by spore-forming bacteria, causing serious infections such as anthrax or botulism. These are, most likely, caused by injecting contaminated heroin, and our aim was to develop a filter that efficiently removes these bacteria and is also likely to be acceptable for use by people who inject drugs (i.e. quick, simple and not spoil the hit). METHODS: A prototype filter was designed and different filter membranes were tested to assess the volume of liquid retained, filtration time and efficiency of the filter at removing

Vulnerability of Bacillus spores and of related genera to physical impaction injury with particular reference to spread-plating.

AIMS: To examine whether bacterial spores are vulnerable to impaction injury during standard spread-plating or to other modes of physical impaction. METHODS AND RESULTS: Employing heat-challenged spores of Bacillus pumilus, Bacillus subtilis, Bacillus thuringiensis, Lysinibacillus, Paenibacillus and Brevibacillus spp. from day-4 to day-10 nutrient agar (NA) plates in 50% ethanol, plating the spore suspension to the extent of just drying the agar surface on fresh NA (50-60 s; SP-B) was tested in comparison with the spreader-independent approach of spotting-and-tilt-spreading (SATS), or a brief

Inactivation of spores by nonthermal plasmas.

Bacterial and fungal spore contamination in different industries has a greater economic impact. Because of the remarkable resistance of spores to most physical and chemical microbicidal agents, their inactivation need special attention during sterilization processes. Heat and chemical sporicides are not always well suited for different sterilization/decontamination applications and carries inherent risks. In recent years, novel nonthermal agents including nonthermal plasmas are emerging as effective sporicides against a broad spectrum of bacterial and fungal spores. The present review discusse

Detection of Bacillus subtilis spores in water by means of broadband coherent anti-Stokes Raman spectroscopy.

Broadband coherent anti-Stokes Raman scattering (CARS) spectroscopy is used for detection of bacterial spores in aqueous solution. Polarization CARS spectroscopy is employed to suppress the non-resonant background. CARS spectrum recorded in the spectral region from 700 to 1900 cm(-1) exhibits all the characteristic features of spontaneous Raman spectrum taken for a solid powder and resembles that one of the dipicolinic acid, which is considered to be the major component of bacterial spores, including anthrax.

A Stochastic Intracellular Model of Anthrax Infection With Spore Germination Heterogeneity.

We present a stochastic mathematical model of the intracellular infection dynamics of Bacillus anthracis in macrophages. Following inhalation of B. anthracis spores, these are ingested by alveolar phagocytes. Ingested spores then begin to germinate and divide intracellularly. This can lead to the eventual death of the host cell and the extracellular release of bacterial progeny. Some macrophages successfully eliminate the intracellular bacteria and will recover. Here, a stochastic birth-and-death process with catastrophe is proposed, which includes the mechanism of spore germination and matura

Oral Treatment With Ileal Spores Triggers Immunometabolic Shifts in Chicken Gut.

The animal gut is a major site affecting productivity via its role in mediating functions like food conversion and pathogen colonization. Live microorganisms like probiotics are widely used to improve poultry productivity. However, given that chicks receive their microbiota from the environment at-hatch, a bacterial treatment that can stimulate gut immune maturation in early life can benefit animal health. Thus, our lab has begun investigating alternative means to improve poultry health via single inoculation with microbial spores. In this study, we orally-inoculated day-old chicks with ileal

Effects of Low Temperature, Freeze-Thaw Cycles, and Healing Conditions on Viability of Non-Ureolytic Bacteria in Biological Self-Healing Concrete.

The capacity of biological self-healing concrete (BSHC) to repair cracks relies on the sustained viability and metabolic function of bacteria embedded within the concrete. BSHC structures face significant risk in cold climates due to low temperatures and freeze-thaw (FT) cycles, during which freezing water can generate internal pressure that damages bacterial cells and diminishes their activity. A special feature of this study is the incorporation of bacterial spores within expanded clay aggregates, tested under varying environmental conditions. The viability of bacterial spores was measured u

YpeB dimerization may be required to stabilize SleB for effective germination of Bacillus anthracis spores.

BACKGROUND: Bacillus cells faced with unfavorable environmental conditions undergo an asymmetric division process ultimately leading to the formation of the bacterial spore. In some instances the spore serves as an infectious agent; such is the case with the spore of Bacillus anthracis and the disease anthrax. Spores are resistant to a variety of environment conditions including traditional decontamination techniques due to the formation of specialized cellular structures. One such structure, the spore cortex, is a thick layer of modified peptidoglycan that contributes to spore dormancy throug

Comparison of four commercial DNA extraction kits for the recovery of Bacillus spp. spore DNA from spiked powder samples.

Bacillus spp. include human pathogens such as Bacillus anthracis, the causative agent of anthrax and a biothreat agent. Bacillus spp. form spores that are physically highly resistant and may remain active over sample handling. We tested four commercial DNA extraction kits (QIAamp DNA Mini Kit, RTP Pathogen Kit, ZR Fungal/Bacterial DNA MiniPrep, and genesig Easy DNA/RNA Extraction kit) for sample inactivation and DNA recovery from two powders (icing sugar and potato flour) spiked with Bacillus thuringiensis spores. The DNA was analysed using a B. thuringiensis-specific real-time PCR assay. The

Toxicity study of SARS-CoV-2 Spike protein expressed on Bacillus subtilis spores.

BACKGROUND: The SARS-CoV-2 spike protein is the primary antigen used in most COVID-19 vaccines. To improve protection, especially at mucosal surfaces, a robust and safe delivery system capable of inducing both systemic and mucosal immunity is needed. Bacillus subtilis spores offer a promising platform due to their stability, safety, and capacity for protein display. A study to evaluate the potential acute oral toxicity of recombinant B. subtilis spores expressing SARS-CoV-2 spike proteins was undertaken. RESULTS: B. subtilis spore was used to express the SARS-CoV-2 S1 domain spike proteins du

Activation and inactivation of Bacillus pumilus spores by kiloelectron volt X-ray irradiation.

In this study, we investigated the inactivation efficacy of endospore-forming bacteria, Bacillus pumilus, irradiated by low-energy X-rays of different beam qualities. The different low-energy X-rays studied had cut-off energies of 50, 100 and 150 keV. Bacillus pumilus spores (in biological indicator strips) were irradiated at step doses between 6.5 to 390 Gy. The resulting bacteria populations were then quantified by a pour plate method. Results showed that X-rays of lower energies were more effective in inactivating bacterial spores. In addition, an increment in bacterial population was obser

Composite Sampling Approaches for Bacillus anthracis Surrogate Extracted from Soil.

Any release of anthrax spores in the U.S. would require action to decontaminate the site and restore its use and operations as rapidly as possible. The remediation activity would require environmental sampling, both initially to determine the extent of contamination (hazard mapping) and post-decon to determine that the site is free of contamination (clearance sampling). Whether the spore contamination is within a building or outdoors, collecting and analyzing what could be thousands of samples can become the factor that limits the pace of restoring operations. To address this sampling and anal

The Use of Germinants to Potentiate the Sensitivity of Bacillus anthracis Spores to Peracetic Acid.

Elimination of Bacillus anthracis spores from the environment is a difficult and costly process due in part to the toxicity of current sporicidal agents. For this reason we investigated the ability of the spore germinants L-alanine (100 mM) and inosine (5 mM) to reduce the concentration of peracetic acid (PAA) required to inactivate B. anthracis spores. While L-alanine significantly enhanced (p = 0.0085) the bactericidal activity of 500 ppm PAA the same was not true for inosine suggesting some form of negative interaction. In contrast the germinant combination proved most effective at 100 ppm

Urea-mercaptoethanol-soluble protein from spores of Bacillus thuringiensis and other species.

Treatment with urea-mercaptoethanol of purified spores of Bacillus thuringiensis, other Bacillus species, and Clostridium roseum solubilizes a protein fraction between 5 and 12% of the dry weight of the spores. This fraction behaves identically to the crystal protein of B. thuringiensis on acrylamide-gel electrophoresis. The protein from all of the Bacillus species shows partial homology with crystal protein, using the Ouchterlony immunodiffusion technique. A further fraction, similar in amount, can be removed from spores of B. thuringiensis by the addition of sodium lauryl sulfate to the urea

Chaperone protein DnaK-mediated regulation of spore coat assembly in Bacillus subtilis.

Bacterial spores pose a significant challenge to the food industry due to their ability to germinate from dormancy and their extreme resistance. Spores are encased by a multilayered proteinaceous structure termed the spore coat. Previous studies on spore coat assembly have predominantly focused on the interactions among coat proteins, while the involvement of broader protein quality control systems remains poorly understood. Here, we demonstrate that the molecular chaperone DnaK is essential for the proper assembly of the inner spore coat in Bacillus subtilis. Deletion of dnaK leads to misloca

Application of the real-time PCR for the detection of airborne microbial pathogens in reference to the anthrax spores.

To establish the rapid detection method of airborne bacterial spores, we examined Bacillus anthracis spores by real-time PCR. One hundred liters of air were trapped on a filter of an air monitor device. After it was suspended in PBS, spores of B. anthracis were artificially added. The suspension was also heated at 95 degrees C for 15 min and used for real-time PCR using anthrax-specific primers. A single cell of B. anthracis was detected by real-time PCR within 1 h. Our results provide evidence that anthrax spores from the atmosphere can be detected rapidly, suggesting that real-time PCR provi

Application of gaseous ozone for inactivation of Bacillus subtilis spores.

The effectiveness of gaseous ozone (O3) as a disinfectant was tested on Bacillus subtilis spores, which share the same physiological characteristics as Bacillus anthracis spores that cause the anthrax disease. Spores dried on surfaces of different carrier material were exposed to O3 gas in the range of 500-5000 ppm and at relative humidity (RH) of 70-95%. Gaseous O3 was found to be very effective against the B. subtilis spores, and at O3 concentrations as low as 3 mg/L (1500 ppm), approximately 3-log inactivation was obtained within 4 hr of exposure. The inactivation curves consisted of a shor
