Polyploidy in germ cell of brown bear Ursus arctos (Ursidae, Mammalia): A case report.

In this work, we present the first detailed study of the normal meiotic karyotype of the brown bear based on the analysis of synaptonemal complexes. Microphotographs of the key stages of meiotic prophase I were obtained. The level of meiotic recombination, features of XY sex bivalent assembly during meiotic prophase I were analyzed. A refined SC karyotype ideogram was constructed, consisting of 37 pachytene bivalents: 36 autosomal bivalents and and XY sex bivalent. We report a rare finding in mammalian germinal cells - a polyploid spermatocyte after genome duplication in the brown bear Ursus a

Evolution of the black muntjac (Muntiacus crinifrons) karyotype revealed by comparative chromosome painting.

The black muntjac (Muntiacus crinifrons) has an unusual karyotype of 2n = 8 in females and 2n = 9 in males. We have studied the evolution of this karyotype by hybridising chromosome-specific paints derived from flow-sorted chromosomes of the Chinese muntjac (M. reevesi, 2n = 46) to chromosomes of the black muntjac. The hybridisation pattern allowed us to infer chromosomal homologies between these two species. Tandem and centromeric fusions, reciprocal translocations, and insertions are involved in the reduction of the diploid number from 2n = 46 to 2n = 8, 9. The painting patterns further show

Complex Chromosomal Rearrangement Causes Male Azoospermia: A Case Report and Literature Review.

Background: Male carriers of complex chromosomal rearrangements (CCRs) may have decreased fertility and usually present with azoospermia, oligospermia or teratospermia. Methods: High-resolution karyotype analysis using G-banding on peripheral blood lymphocytes was performed in an azoospermic male. Copy number variations (CNVs) were detected by chromosomal microarray analysis, and genetic variations were determined by long-read nanopore sequencing with Sanger sequencing for breakpoint confirmation. Results: The karyotype of the patient was 46,XY,t(4;21)(p11;p11),t(5;6;14)(p13q22;p22q22;q22), wh

ATM-mediated double-strand break repair is required for meiotic genome stability at high temperature.

In eukaryotes, meiotic recombination maintains genome stability and creates genetic diversity. The conserved Ataxia-Telangiectasia Mutated (ATM) kinase regulates multiple processes in meiotic homologous recombination, including DNA double-strand break (DSB) formation and repair, synaptonemal complex organization, and crossover formation and distribution. However, its function in plant meiotic recombination under stressful environmental conditions remains poorly understood. In this study, we demonstrate that ATM is required for the maintenance of meiotic genome stability under heat stress in Ar

VerteBrain reveals novel neural and non-neural protein assemblies conserved across vertebrate evolution.

Protein-protein interactions underlie core brain functions, including neurotransmitter release, receptor activation, and intracellular signaling essential for learning, memory, and cognition. Here, we systematically map conserved brain protein interactions across five vertebrate species-rabbit, chicken, dolphin, pig, and mouse-using co-fractionation and immunoprecipitation mass spectrometry. From 2,197 biochemical fractions, we identify over 81,000 high-confidence interactions among 6,108 conserved proteins. This interaction map (VerteBrain) reveals both regulatory and structural complexes, in

Ultrastructure of spermatogenesis in the testis of Paragonimus heterotremus.

Lung fluke, Paragonimus heterotremus, is a flatworm causing pulmonary paragonimiasis in cats, dogs, and humans in Southeast Asia. We examined the ultrastructure of the testis of adult P. heterotremus with special attention to spermatogenesis and spermiogenesis using scanning and transmission electron microscopy. The full sequence of spermatogenesis and spermiogenesis, from the capsular basal lamina to the luminal surface, was demonstrated. The sequence comprises spermatogonia, spermatocytes with obvious nuclear synaptonemal complexes, spermatids, and eventual spermatozoa. Moreover, full steps

Acute nonlymphocytic leukemias following chemotherapy. Ultrastructural study of erythroblastic cell line and discussion of nuclear anomalies.

In 18 cases of drug-induced acute leukemia, electron microscopy brought out a number of anomalies found in all hematopoietic cell lines. They are described here for the erythroblastic line. Some of these anomalies occur in the cytoplasm (like siderosomes, iron-containing mitochondria, glycogen spots, various dense inclusions, and nuclear-cytoplasmic asynchronism; others occur in the nucleus (such as plurinucleated cells, fragility of nuclear membrane, and nuclear clefts and blebs). Nuclear clefts and blebs may also be found in granulocytic and megakaryocytic cell lines. Their structural aspect

SNI1/NSE5 is a cold-sensitive brake on class II crossovers in Arabidopsis.

Temperature reshapes meiotic recombination, but how environmental cues alter crossover pathway choice remains unclear. Here, using interval assays, genome-wide crossover maps, genetic mapping of recombination modifiers and targeted genetics, we show that cold shifts crossovers towards subtelomeric regions and increases class II pathway output. Genetic mapping identified a major temperature-responsive locus on chromosome 4, resolved to a 15.6-kb interval containing SNI1, the Arabidopsis NSE5 orthologue and a component of the SMC5/6 complex. Reduced SNI1 function alters the temperature response

The Drosophila CPEB Protein Orb Specifies Oocyte Fate by a 3'UTR-Dependent Autoregulatory Loop.

orb encodes one of the two fly CPEB proteins. These widely conserved proteins bind to the 3'UTRs of target messenger RNAs (mRNAs) and activate or repress their translation. We show here that a positive autoregulatory loop driven by the orb gene propels the specification of oocyte identity in Drosophila egg chambers. Oocyte fate specification is mediated by a 3'UTR-dependent mechanism that concentrates orb mRNAs and proteins in one of the two pro-oocytes in the 16-cell germline cyst. When the orb 3'UTR is deleted, orb mRNA and protein fail to localize and all 16 cells become nurse cells. In wil

Close association between vasa-positive germ plasm granules and mitochondria correlates with cytoplasmic localization of 12S and 16S mtrRNAs during zebrafish spermatogenesis.

The phenomenon of the cytoplasmic localisation of mitochondrial ribosomal subunits (12 S mitochondrial rRNA and 16 S mitochondrial rRNA) has been discovered by scientific teams working with spermatogenic cells of mice. Previous reports showed that the release of mitochondrial substance occurs during interaction of mitochondria with the germ plasm granules (GG). To determine if the interplay between the vasa-positive GG and the mitochondria is associated with cytoplasmic localisation of mtrRNAs, we studied the spermatogenic cells of zebrafish, Danio rerio. It was revealed that in type A undiffe

Multi-omics analysis reveals fluorinated liquid crystal monomers BDPrB/EDPrB exposure disrupts the steroid biosynthesis function via inhibition of CYP17A1.

Fluorinated liquid crystal monomers (F-LCMs) are emerging environmental contaminants that have been detected in human urine, breast milk, and serum. However, their potential impacts on reproductive health remain unclear. In this study, we investigated the reproductive toxicity of two major F-LCMs-BDPrB and EDPrB-using a 70-day exposure model in male ICR mice. Multi-omics analyses of testicular tissue and serum showed that both compounds reduced testosterone and increased progesterone levels. Further mechanistic studies revealed that BDPrB and EDPrB inhibit CYP17A1 enzyme activity by directly b

A homozygous frameshift variant in SYCP2 caused meiotic arrest and non-obstructive azoospermia.

Genetic causation for the majority of non-obstructive azoospermia (NOA) remains unclear. Mutations in synaptonemal complex (SC)-associated genes could cause meiotic arrest and NOA. Previous studies showed that heterozygous truncating variants in SYCP2 encoding a protein essential for SC formation, are associated with non-obstructive azoospermia and severe oligozoospermia. Herein, we showed a homozygous loss-of-function variant in SYCP2 (c.2689_2690insT) in an NOA-affected patient. And this variant was inherited from heterozygous parental carriers by natural reproduction. HE, IF, and meiotic ch

The mouse "tool box" for meiotic studies.

Besides availability, there are numerous advantages to using mice for meiotic studies: (1) techniques exist for enriching the population of spermatocytes at particular meiotic stages; (2) spermatogenesis in mice is highly synchronized and testis sections afford a highly accurate method of staging meiotic events; (3) knock-out mice provide a rich source of meiotic mutants. Coupled with antibody localization techniques these tools, singularly and in combination, provide multiple means of temporally and physically dissecting meiosis.

CRL2(LRR-1) E3-ligase regulates proliferation and progression through meiosis in the Caenorhabditis elegans germline.

The ubiquitin-proteolytic system controls the stability of proteins in space and time. In this study, using a temperature-sensitive mutant allele of the cul-2 gene, we show that CRL2(LRR-1) (CUL-2 RING E3 ubiquitin-ligase and the Leucine Rich Repeat 1 substrate recognition subunit) acts at multiple levels to control germline development. CRL2(LRR-1) promotes germ cell proliferation by counteracting the DNA replication ATL-1 checkpoint pathway. CRL2(LRR-1) also participates in the mitotic proliferation/meiotic entry decision, presumably controlling the stability of meiotic promoting factors in

Uniparental Disomy Reveals Hidden Genetic Causes of Congenital Heart Disease.

Congenital heart disease (CHD) affects ~1% of live births, yet the genetic basis of many cases remains unresolved. Uniparental disomy (UPD), the inheritance of both homologous chromosomes from one parent, is often overlooked. We developed TrioMix-UPD, an integrated short- and long-read sequencing framework for UPD detection and classification. Applying it to 3,740 CHD trios, we identified 12 UPD events, representing a 6.57-fold enrichment relative to the general population. Both advanced maternal age and enrichment of rare inherited variants in synaptonemal complex genes implicated meiotic chr

Initiation of meiosis from human iPSCs under defined conditions through identification of regulatory factors.

Meiotic failure is a major cause of infertility, but the lack of an in vitro model of human meiosis is a barrier to understanding its mechanism. Here, we establish a method to initiate meiosis directly from male or female human-induced pluripotent stem cells (iPSCs). DNMT1 inhibition, retinoid signaling activation, and overexpression of regulatory factors (antiapoptotic BCL2 and promeiotic HOXB5, BOLL, or MEIOC) rapidly activates meiosis over a 15-day protocol. Our protocol bypasses the primordial germ cell stage and directly generates cells expressing genes similar to meiotic oogonia, includi

Pericentric inversion in human chromosome 1 and the risk for male sterility.

A pericentric inversion in chromosome 1 of a severely oligospermic human male is reported. Pachytene analysis in microspread preparations shows an absence of full loop formation in the inversion bivalent and only the rare occurrence of a partial loop. The majority of cells exhibit extensive asynapsis across the inverted segment, or a normal looking synaptonemal complex indicative of heterologous pairing along the length of the inversion. Crossing over is reduced in the No 1 bivalent with only a rare chiasma being seen in the inverted region at metaphase I. Males heterozygous for a pericentric

Expression analysis of genes encoding TEX11, TEX12, TEX14 and TEX15 in testis tissues of men with non-obstructive azoospermia.

OBJECTIVE: Spermatogenesis is a complex process controlled by a plethora of genes. Changes in expression and function of these genes may thus lead to spermatogenic deficiency and male infertility. TEX11, TEX12, TEX14 and TEX15 are germ cell-specific genes expressed in the testis. TEX11, involved in the initiation and maintenance of chromosome synapses in meiotic chromosomes, has been shown to be essential for meiosis and fertility in males. TEX14, a component of intercellular bridges in germ cells, is required for spermatogenesis and fertility. TEX12 and TEX15 are essential for correct assembl
