Anticoagulant rodenticide exposure in endangered American badgers (Taxidea taxus) and fishers (Pekania pennanti) from British Columbia, Canada, 1998 to 2018.

Anticoagulant rodenticide (AR) concentrations in livers of 69 samples of two mustelid species, collected 1998 to 2018 from locations in British Columbia, Canada are presented. The American badger Taxidea taxus is endangered in Canada, and fisher Pekania pennanti populations are at-risk. Using a high performance liquid chromatography method, we measured liver concentrations of the following second generation ARs (SGARs): brodifacoum, bromadiolone, difethialone and the first generation (FGAR) compounds: warfarin, diphacinone and chlorophacinone in 59 badgers and 10 fishers. Of the 59 badgers ana

Extracorporeal life support and digoxin-specific Fab fragments for successful management of Taxus baccata intoxication with low output and ventricular arrhythmia.

INTRODUCTION: Yew plants are evergreen shrubs which are widely spread throughout the northern hemisphere. Taxane alkaloid derivatives, mainly taxine B, represent the main toxins of Taxus baccata and are highly cardiotoxic. Due to the lack of randomized clinical trials, case reports on accidental or suicidal yew intoxications build the only source of knowledge of clinical treatment options. CASE REPORT: We report the case of a suicidal yew ingestion admitted to our hospital under prolonged cardiopulmonary resuscitation due to pulseless electrical activity. Impact of obesity on revascularization and restenosis rates after bare-metal and drug-eluting stent implantation (from the TAXUS-IV trial).

The effect of obesity on repeat coronary revascularization and restenosis in patients who undergo stent implantation has not been reported. We therefore examined the database from the multicenter randomized TAXUS-IV trial to determine the effect of body mass index (BMI) on outcomes after bare-metal and drug-eluting stent implantation. In TAXUS-IV, patients were randomized to receive a slow-release, polymer-based, paclitaxel-eluting stent or a bare-metal stent. Outcomes were stratified by baseline BMI. Of the 1,307 randomized patients who had documented BMI, 233 (17.8%) had normal weight (BMI <

Relative abundance of American badger (Taxidea taxus) and red fox (Vulpes vulpes) in landscapes with high and low rodenticide poisoning levels.

Over the past decade, extensive poisoning campaigns have been conducted in southern Saskatchewan to control Richardson's ground squirrel Spermophilus richardsonii (Sabine, 1822) populations. Such campaigns might impact on predator abundance by decreasing prey levels, and also through secondary poisoning. Using spotlighting, we investigated the relative abundance of American badgers Taxidea taxus (Schreber, 1777) and red fox Vulpes vulpes (Linnaeus, 1758) in 2 study areas with road access and crops, but with different levels of poisoning. In the study area with relatively low poisoning (19.6% o

Large-scale purification of 13-dehydroxybaccatin III and 10-deacetylpaclitaxel, semi-synthetic precursors of paclitaxel, from cell cultures of Taxus chinensis.

The taxane derivatives 13-dehydroxybaccatin III (13-DHB III) and 10-deacetylpaclitaxel (10-DAP) can be used as semi-synthetic precursors of paclitaxel. These precursors were isolated in a simple and economical procedure during purification of paclitaxel from plant cell culture extracts. No additional costs for cell culture or extraction by silica-gel low-pressure chromatography were incurred. Precipitation from dichloromethane/n-hexane followed by HPLC on ODS and silica-gel resins resulted in paclitaxel of 99.5% purity with 80% overall yield. ODS low-pressure chromatography and THF/n-hexane pr

Long-term follow-up of the platinum chromium TAXUS Element (ION) stent: The PERSEUS Workhorse and Small Vessel trial five-year results.

BACKGROUND: The TAXUS Element (ION) platinum chromium paclitaxel-eluting stent (PtCr-PES) incorporates a thin (81 μm) strut design with a similar polymer and drug dose density as prior PES. The pivotal PERSEUS trial program consisted of two studies: PERSEUS Workhorse (WH) and PERSEUS Small Vessel (SV). The PERSEUS WH trial demonstrated the PtCr-PES to be non-inferior to the predicate TAXUS Express PES (TE-PES) for target lesion failure (TLF) at 1 year and in-segment angiographic percent diameter stenosis at 9 months. The PERSEUS SV trial demonstrated the PtCr-PES to be superior to a historical

Comparison of in-hospital and long-term outcomes between a Cypher stent and a Taxus stent in Chinese diabetic patients with coronary artery disease.

BACKGROUND: The sirolimus and paclitaxel distribution patterns and tissue residence time may be modified in atherosclerotic lesions for patients with diabetes, and the biological mechanisms of action for these agents differ significantly. Previous clinical trials have yielded discrepant results of major adverse cardiac events and restenosis between a sirolimus-eluting stent and a paclitaxel-eluting stent in coronary artery disease. Therefore, this study was conducted to compare in-hospital and long-term clinical outcomes between patients receiving sirolimus-eluting stent (Cypher or Cypher Sele

Engineering of 10-Deacetylbaccatin III-10-β-O-Acetyltransferase From Taxus Species for Efficient Acetylating Non-Natural Substrates Into Taxol in Nicotiana benthamiana.

7-β-xylosyl-10-deacetyltaxol (XDT) is much more abundant than the anticancer drug Taxol in Taxus species and is usually regarded as the by-product of Taxol. It could be enzymatically transformed into 10-deacetyltaxol (DT), and the latter could be further converted into Taxol. The enzyme 10-deacetylbaccatin III-10-β-O-acetyltransferase (DBAT) can acetylate the non-natural substrate DT into Taxol, but the conversion efficiency was extremely low. Herein, we globally redesigned DBATcus from Taxus cuspidata to improve its efficiency in DT acetylation through combinatorial protein engineering strate

Comparison of a polymer-free rapamycin-eluting stent (YUKON) with a polymer-based paclitaxel-eluting stent (TAXUS) in real-world coronary artery lesions.

BACKGROUND: In selected patient cohorts the polymer-free rapamycin-eluting YUKON stent (A) has demonstrated noninferiority compared with the polymer-based paclitaxel-eluting TAXUS stent (B). To test for equivalency in unselected real-world patients with coronary lesions of various complexities, we retrospectively compared both stent designs. METHODS: A total of 410 patients with symptomatic CAD were successfully treated with A (n = 205) or with B (n = 205). Taxoids-rich extract from Taxus wallichiana alleviates high-fat diet-induced insulin resistance in C57BL/6 mice through inhibition of low-grade inflammation.

The needle powder of Taxus wallichiana is in use for the management of diabetes and inflammation-related complications in the Indian and Chinese Systems of Traditional Medicine but the lack of proper pharmacological intervention has prompted us to investigate the pharmacological mechanism against inflammation-induced insulin resistance in high-fat diet-fed C57BL/6 mice. Hexane (Tw-H), chloroform (Tw-C), and ethyl acetate (Tw-EA) extracts were prepared from a needle of T. wallichiana and its effect on glucose uptake against TNF-α-induced insulin resistance in skeletal muscle cells was studied.

Impact of platelet glycoprotein IIb/IIIa Inhibition on the paclitaxel-eluting stent in patients with stable or unstable angina pectoris or provocable myocardial ischemia (a TAXUS IV substudy).

Whether the benefits that glycoprotein IIb/IIIa inhibitors confer in patients who undergo bare metal stent implantation extend to drug-eluting stents is unknown. We performed a prespecified subgroup analysis of the TAXUS IV study population to examine the effect of procedural glycoprotein IIb/IIIa inhibition during paclitaxel-eluting stent implantation on periprocedural creatine kinase-MB (CK-MB) levels. Neutrophil-to-Lymphocyte Ratio Correlation With the Synergy Between Percutaneous Coronary Intervention With Taxus and Cardiac Surgery (SYNTAX) Score in Patients With Non-ST-Segment Elevation Acute Coronary Syndrome.

BACKGROUND: The neutrophil-to-lymphocyte ratio (NLR) is a simple inflammatory marker that may reflect the extent of coronary atherosclerotic complexity. This study examined the relationship between NLR and angiographic disease severity in individuals presenting with non-ST-segment elevation acute coronary syndrome (NSTE-ACS), using the synergy between percutaneous coronary intervention with taxus and cardiac surgery (SYNTAX) score as an anatomical reference standard. Long-term outcomes of percutaneous coronary intervention for unprotected left main coronary artery according to the synergy between percutaneous coronary intervention with taxus and cardiac surgery (SYNTAX) score.

BACKGROUND: Percutaneous coronary intervention (PCI) of unprotected left-main coronary artery (uLMCA) is increasingly performed. Although it is recommended to calculate the SYNTAX score, if uLMCA PCI is considered, data of outcomes by the SYNTAX score are scarce, particularly among patients with high score (SYNTAX ≥ 33). OBJECTIVE: To evaluate long-term outcomes of uLMCA PCI according to SYNTAX score in real-world practice. METHODS: Patients following PCI of uLMCA throughout 2006-2018, in a tertiary hospital, were included and classified into three groups according to the SYNTAX I score. The
