Optimized Ex Vivo Human Liver Slice Culture Maintains Extended Viability and Function for Hepatotoxicity Testing.

Human precision-cut liver slices (hPCLS) are widely used to study liver physiology and disease, but their short lifespan limits long-term functional studies and drug testing. Here, we describe an optimized ex vivo culture system, hPCLS-EV (Extended Viability), that maintains tissue viability, cellular composition, and liver-specific functions for up to five days. Optimization combined reduced slice thickness, high oxygen partial pressure, and air-liquid interface culture, preserving hepatocyte integrity, stromal stability, and immune cell populations. Functional assays confirmed sustained albu

Organoid and Spheroid Tumor Models: Techniques and Applications.

Techniques to develop three-dimensional cell culture models are rapidly expanding to bridge the gap between conventional cell culture and animal models. Organoid and spheroid cultures have distinct and overlapping purposes and differ in cellular sources and protocol for establishment. Spheroids are of lower complexity structurally but are simple and popular models for drug screening. Organoids histologically and genetically resemble the original tumor from which they were derived. Ease of generation, ability for long-term culture and cryopreservation make organoids suitable for a wide range of

Mimicking the Articular Joint with In Vitro Models.

Treating joint diseases remains a significant clinical challenge. Conventional in vitro cultures and animal models have been helpful, but suffer from limited predictive power for the human response. Advanced models are therefore required to mimic the complex biological interactions within the human joint. However, the intricate structure of the joint microenvironment and the complex nature of joint diseases have challenged the development of in vitro models that can faithfully mimic the in vivo physiological and pathological environments. In this review, we discuss the current in vitro models

An ex vivo human placental explant model for controlled exposure studies.

The human placental explant model preserves the placenta's natural structure, enabling studies on whole-tissue response to exposures or growth conditions. It outlines procedures for placental collection, isolation, and culturing, followed by exposure to conditions of interest. Additionally, it details sample collection of explant tissue and culture supernatant after exposure for downstream assessments of placental function. This protocol provides a framework for evaluating ex vivo responses of human placental tissue. For complete details on the use and execution of this protocol, please refer

Protocol for ex vivo culture of adult mouse testis tissue using a gel-based approach.

Testicular function relies on tightly regulated cell-cell interactions, including Sertoli-Leydig cell communication and Sertoli cells nurturing germ cells. Here, we present a 48-h ex vivo culture protocol for adult mouse testis tissue using a gel-based approach. We describe experimental design, dissection, and examination of treatment-induced outcomes, in particular, stimulatory effects on germ cells. We outline procedures for quantifying apoptosis and germ cell proliferation in the tissue and measurements of reproductive hormones and paracrine factors secreted to the culture medium. For compl

A comparison of callus induction and plant regeneration from different embryo explants of triticale (x Triticosecale wittmack).

Immature, mature and endosperm-supported mature embryos of six triticale cultivars (BDMT-98-8S, Melez-2001, Mikham-2002, Presto, Tacettin Bey and Tatlicak-97) were cultured in vitro to compare the levels of callus induction and plant regeneration. Immature embryos, 15-18 days after anthesis, were aseptically excised and placed with the scutellum upwards on a callus culture medium consisting of Murashige and Skoog (MS) mineral salts supplemented with 2 mg l(-1) 2,4-dichlorophenoxyacetic acid (2,4-D). Mature embryos were aseptically excised from the imbibed seeds and placed scutellum up on MS me

Comparison of Mycobacterial Culture Yield from Pleural Tissue Using Pleuroscopic Cryobiopsy versus Forceps Biopsy in Patients with Tuberculous Pleurisy.

INTRODUCTION: The diagnostic yield of mycobacterial culture from pleural tissue in tuberculous pleurisy is high and practical. However, the yield rates of mycobacterial culture from pleural tissue obtained through different pleural biopsy techniques via medical pleuroscopy remain unclear. This study aimed to compare the efficiency of mycobacterial culture from pleural tissue obtained by cryobiopsy and forceps biopsy during medical pleuroscopy for diagnosing tuberculous pleurisy. METHODS: This retrospective study included 84 patients with tuberculous pleurisy who underwent mycobacterial cultur

Stem cell culture and differentiation in 3-D scaffolds.

Conventional two-dimensional (2-D) cultivation are easy to utilize for human pluripotent stem (hPS) cell cultivation in standard techniques and are important for analysis or development of the signal pathways to keep pluripotent state of hPS cells cultivated on 2-D cell culture materials. However, the most efficient protocol to prepare hPS cells is the cell culture in a three dimensional (3-D) cultivation unit because huge numbers of hPS cells should be utilized in clinical treatment. Some 3-D cultivation strategies for hPS cells are considered: (a) microencapsulated cell cultivation in suspen

Graft union formation in artichoke grafting onto wild and cultivated cardoon: an anatomical study.

In order to develop a non-chemical method such as grafting effective against well-known artichoke soil borne diseases, an anatomical study of union formation in artichoke grafted onto selected wild and cultivated cardoon rootstocks, both resistant to Verticillium wilt, was performed. The cardoon accessions Belgio (cultivated cardoon) and Sardo (wild cardoon) were selected as rootstocks for grafting combinations with the artichoke cv. Romolo. Grafting experiments were carried out in the autumn and spring. The anatomical investigation of grafting union formation was conducted by scanning electro

Tissue culture methods for testing the toxicity of ocular plastic materials.

A screening method using tissue culture techniques determined inertness of ocular alloplastics. Secondary rabbit kidney cells revealed the presence of toxic constituents by absence of growth or cytopathic changes, or both. The optical portions of the lenses studied were nontoxic, whereas the polyamide loops of one lens were toxic. A keratoprosthesis that had been in situ in a patient for two years was highly toxic in culture. A neutralized agar overlay technique was not sufficiently sensitive to screen anterior chamber implants. The tissue culture technique, in combination with chemical, optic

An Introduction to Plant Cell, Tissue, and Organ Culture: Current Status and Perspectives.

Plant cell, tissue, and organ cultures (PCTOC) have been used as experimental systems in basic research, allowing gene function demonstration through gene overexpression or repression and investigating the processes involved in embryogenesis and organogenesis or those related to the potential production of secondary metabolites, among others. On the other hand, PCTOC has also been applied at the commercial level for the vegetative multiplication (micropropagation) of diverse plant species, mainly ornamentals but also horticultural crops such as potato or fruit and tree species, and to produce

Hollow Fiber Bioreactors for In Vivo-like Mammalian Tissue Culture.

Tissue culture has been used for over 100 years to study cells and responses ex vivo. The convention of this technique is the growth of anchorage dependent cells on the 2-dimensional surface of tissue culture plastic. More recently, there is a growing body of data demonstrating more in vivo-like behaviors of cells grown in 3-dimensional culture systems. This manuscript describes in detail the set-up and operation of a hollow fiber bioreactor system for the in vivo-like culture of mammalian cells. The hollow fiber bioreactor system delivers media to the cells in a manner akin to the delivery of

Advances in tissue engineering; a novel technology making use of an in vivo vascularized chamber.

INTRODUCTION: Regenerative therapies aim to repair or replace damaged tissues. Clinical applications may vary from organ replacement, to breast reconstruction or limb regeneration. In growing 3-dimensional tissue constructs, access to a nourishing vascular network is essential. METHODS: In rats, an arteriovenous loop was surgically established from the femoral vessels and positioned inside a perforated chamber in the groin. Chambers were filled with 2 ml Adipogel, a novel acellular tissue engineering gel -containing growth factors and polymers. Constructs were harvested at 6 and 12 weeks (n=6

Effect of medium composition on in vitro ovary culture of cucumber.

Ovary culture is among the most effective techniques for haploid induction in horticultural crops, including cucumber. The application of doubled haploid technology can significantly accelerate cultivar development by enabling the rapid production of homozygous inbred lines. This study evaluated the effects of culture medium combinations of plant growth hormones (2,4-D, NAA, IAA, IBA, PUT, BAP, KIN, TDZ) on haploid induction in cucumber via ovary culture. Four induction and five regeneration media were tested across four cucumber varieties: Beith Alpha, PTK40, Botanik, and Sardes. The highest

3D Adipose Tissue Culture Links the Organotypic Microenvironment to Improved Adipogenesis.

Obesity and type 2 diabetes are strongly associated with adipose tissue dysfunction and impaired adipogenesis. Understanding the molecular underpinnings that control adipogenesis is thus of fundamental importance for the development of novel therapeutics against metabolic disorders. However, translational approaches are hampered as current models do not accurately recapitulate adipogenesis. Here, a scaffold-free versatile 3D adipocyte culture platform with chemically defined conditions is presented in which primary human preadipocytes accurately recapitulate adipogenesis. Following differentia

Effect of Rotation on Scaffold Motion and Cell Growth in Rotating Bioreactors.

Efficient use of different bioreactor designs to improve cell growth in three-dimensional scaffolds requires an understanding of their mechanism of action. To address this for rotating wall vessel bioreactors, fluid and scaffold motion were investigated experimentally at different rotation speeds and vessel fill volumes. Low cost bioreactors with single and dual axis rotation were developed to investigate the effect of these systems on human osteoblast proliferation in free floating and constrained collagen-glycosaminoglycan porous scaffolds. A range of scaffold motions (free fall, periodic os
